AUTO3D: Novel view synthesis through unsupervisely learned variational viewpoint and global 3D representation
This paper targets on learning-based novel view synthesis from a single or
limited 2D images without the pose supervision. In the viewer-centered
coordinates, we construct an end-to-end trainable conditional variational
framework to disentangle the unsupervisely learned relative-pose/rotation and
implicit global 3D representation (shape, texture and the origin of
viewer-centered coordinates, etc.). The global appearance of the 3D object is
given by several appearance-describing images taken from any number of
viewpoints. Our spatial correlation module extracts a global 3D representation
from the appearance-describing images in a permutation invariant manner. Our
system can achieve implicitly 3D understanding without explicitly 3D
reconstruction. With an unsupervisely learned viewer-centered
relative-pose/rotation code, the decoder can hallucinate the novel view
continuously by sampling the relative-pose in a prior distribution. In various
applications, we demonstrate that our model can achieve comparable or even
better results than pose/3D model-supervised learning-based novel view
synthesis (NVS) methods with any number of input views.Comment: ECCV 202
Correlation of interfacial bonding mechanism and equilibrium conductance of molecular junctions
We report theoretical investigations on the role of interfacial bonding
mechanism and its resulting structures to quantum transport in molecular wires.
Two bonding mechanisms for the Au-S bond in an
Au(111)/1,4-benzenedithiol(BDT)/Au(111) junction were identified by ab initio
calculation, confirmed by a recent experiment, which, we showed, critically
control charge conduction. It was found, for Au/ BDT/Au junctions, the hydrogen
atom, bound by a dative bond to the Sulfur, is energetically non-dissociative
after the interface formation. The calculated conductance and junction
breakdown forces of H-non-dissociative Au/BDT/Au devices are consistent with
the experimental values, while the H-dissociated devices, with the interface
governed by typical covalent bonding, give conductance more than an order of
magnitude larger. By examining the scattering states that traverse the
junctions, we have revealed that mechanical and electric properties of a
junction have strong correlation with the bonding configuration. This work
clearly demonstrates that the interfacial details, rather than previously
believed many-body effects, is of vital importance for correctly predicting
equilibrium conductance of molecular junctions; and manifests that the
interfacial contact must be carefully understood for investigating quantum

Chromosome and DNA methylation dynamics during meiosis in autotetraploid Arabidopsis arenosa
Variation in chromosome number due to
polyploidy can seriously compromise meiotic stability. In
autopolyploids, the presence of more than two homologous
chromosomes may result in complex pairing patterns
and subsequent anomalous chromosome
segregation. In this context, chromocenter, centromeric,
telomeric and ribosomal DNA locus topology and DNA
methylation patterns were investigated in the natural
autotetraploid, Arabidopsis arenosa. The data show that
homologous chromosome recognition and association
initiates at telomeric domains in premeiotic interphase,
followed by quadrivalent pairing of ribosomal 45S RNA
gene loci (known as NORs) at leptotene. On the other hand, centromeric regions at early leptotene show pairwise
associations rather than associations in fours. These
pairwise associations are maintained throughout prophase
I, and therefore likely to be related to the diploid-like
behavior of A. arenosa chromosomes at metaphase I,
where only bivalents are observed. In anthers, both cells
at somatic interphase as well as at premeiotic interphase
show 5-methylcytosine (5-mC) dispersed throughout the
nucleus, contrasting with a preferential co-localization
with chromocenters observed in vegetative nuclei. These
results show for the first time that nuclear distribution
patterns of 5-mC are simultaneously reshuffled in meiocytes
and anther somatic cells. During prophase I, 5-mC
is detected in extended chromatin fibers and chromocenters
but interestingly is excluded from the NORs what
correlates with the pairing patter
Large-Scale Atomistic Simulations of Environmental Effects on the Formation and Properties of Molecular Junctions
Using an updated simulation tool, we examine molecular junctions comprised of
benzene-1,4-dithiolate bonded between gold nanotips, focusing on the importance
of environmental factors and inter-electrode distance on the formation and
structure of bridged molecules. We investigate the complex relationship between
monolayer density and tip separation, finding that the formation of
multi-molecule junctions is favored at low monolayer density, while
single-molecule junctions are favored at high density. We demonstrate that tip
geometry and monolayer interactions, two factors that are often neglected in
simulation, affect the bonding geometry and tilt angle of bridged molecules. We
further show that the structures of bridged molecules at 298 and 77 K are

Advanced Technologies for Oral Controlled Release: Cyclodextrins for oral controlled release
Cyclodextrins (CDs) are used in oral pharmaceutical formulations, by means of inclusion complexes formation, with the following advantages for the drugs: (1) solubility, dissolution rate, stability and bioavailability enhancement; (2) to modify the drug release site and/or time profile; and (3) to reduce or prevent gastrointestinal side effects and unpleasant smell or taste, to prevent drug-drug or drug-additive interactions, or even to convert oil and liquid drugs into microcrystalline or amorphous powders. A more recent trend focuses on the use of CDs as nanocarriers, a strategy that aims to design versatile delivery systems that can encapsulate drugs with better physicochemical properties for oral delivery. Thus, the aim of this work was to review the applications of the CDs and their hydrophilic derivatives on the solubility enhancement of poorly water soluble drugs in order to increase their dissolution rate and get immediate release, as well as their ability to control (to prolong or to delay) the release of drugs from solid dosage forms, either as complexes with the hydrophilic (e.g. as osmotic pumps) and/ or hydrophobic CDs. New controlled delivery systems based on nanotechonology carriers (nanoparticles and conjugates) have also been reviewed
Confirmation of the Chinese version of the Problematic Internet Use Questionnaire Short Form (PIUQ-SF)
Although an increasing number of studies have focused on problematic Internet use, numerous measures exist with only modest investigation into their reliability and validity, both within and across different cultures (Király et al. 2015). The most frequently used questionnaire is the Internet Addiction Test (Young 1998). However, its factor structure appears to be unstable (e.g., Fernandez-Villa et al. 2015; Pontesetal. 2014). Koronczai et al. (2011) suggested that a suitable measure should fit six basic requirements. More specifically, it should be (i) comprehensive, that is examining more, possibly all, aspects of problematic Internet use; (ii) as concise as possible, in order to be able to assess the more impulsive population, and to use in time-limited surveys (or having different forms with different length). Such a measure should be (iii) reliable and valid for different methods of data collection (e.g. online, paper-and-pencil self-rating, face-to-face); as well as (iv) for different age groups (e.g. adolescents and adults), and (v) in different cultural settings. Finally, (vi) cut-off scores should be defined on the basis of clinical examination. The Problematic Internet Use Questionnaire (PIUQ) (Demetrovics et al. 2008) fulfills several of the aforementioned criteria. The 18-item PIUQ is a comprehensive measure that assesses three basic aspects of problematic Internet use (i.e., obsession, neglect, and control disorder). The 18-item and the shorter 9-item versions of the PIUQ have a reliable structure, and are suitable for time-limited surveys and have proved to be valid across various methods of data collection (i.e., online as well as paper-pencil) and age groups (Demetrovics et al. 2008; Koronczai et al. 2011). However, its implementation has so far mostly taken place in Western individualistic cultures (Kelley and Gruber 2010, 2013; Koronczai et al. 2011; Zahodne et al. 2011. To date, the PIUQ has also been applied in a survey in Iran with good Cronbach’s alpha coefficients and subscales (Mazhari 2012a, 2012b). Consequently, validation of the questionnaire in a culture that is significantly different from the Western culture is needed. Metal A and Metal B Sites of Nuclear RNA Polymerases Pol IV and Pol V Are Required for siRNA-Dependent DNA Methylation and Gene Silencing
Plants are unique among eukaryotes in having five multi-subunit nuclear RNA polymerases: the ubiquitous RNA polymerases I, II and III plus two plant-specific activities, nuclear RNA polymerases IV and V (previously known as Polymerases IVa and IVb). Pol IV and Pol V are not required for viability but play non-redundant roles in small interfering RNA (siRNA)-mediated pathways, including a pathway that silences retrotransposons and endogenous repeats via siRNA-directed DNA methylation. RNA polymerase activity has not been demonstrated for Polymerases IV or V in vitro, making it unclear whether they are catalytically active enzymes. Their largest and second-largest subunit sequences have diverged considerably from Pol I, II and III in the vicinity of the catalytic center, yet retain the invariant Metal A and Metal B amino acid motifs that bind magnesium ions essential for RNA polymerization. By using site-directed mutagenesis in conjunction with in vivo functional assays, we show that the Metal A and Metal B motifs of Polymerases IV and V are essential for siRNA production, siRNA-directed DNA methylation, retrotransposon silencing, and the punctate nuclear localization patterns typical of both polymerases. Collectively, these data show that the minimal core sequences of polymerase active sites, the Metal A and B sites, are essential for Pol IV and Pol V biological functions, implying that both are catalytically active
Withdrawal of infliximab therapy in ankylosing spondylitis in persistent clinical remission, results from the REMINEA study
Altres ajuts: This work is conducted under the umbrella of the Rheumatology Society of Catalonia and supported by Merck Research Laboratories.Background: Recent data suggest that anti-TNF doses can be reduced in ankylosing spondylitis (AS) patients. Some authors even propose withdrawing treatment in patients in clinical remission; however, at present there is no evidence to support this. Objective: To assess how long AS patients with persistent clinical remission remained free of flares after anti-TNF withdrawal and to evaluate the effects of treatment reintroduction. We also analyze the characteristics of patients who did not present clinical relapse. Methods: Multicenter, prospective, observational study of a cohort of patients with active AS who had received infliximab as a first anti-TNF treatment and who presented persistent remission (more than 6 months). We recorded at baseline and every 6-8 weeks over the 12-month period the age, gender, disease duration, peripheral arthritis or enthesitis, HLA-B27 status, BASDAI, CRP, ESR, BASFI, and three visual analogue scales, spine global pain, spinal night time pain, and patient's global assessment. Results: Thirty-six out of 107 patients (34%) presented persistent remission and were included in our study. After treatment withdrawal, 21 of these 36 patients (58%) presented clinical relapse during follow-up. Infliximab therapy was reintroduced and only 52% achieved clinical remission, as they had before the discontinuation of infliximab; in an additional 10%, reintroduction of infliximab was ineffective, obliging us to change the anti-TNF therapy. No clinical or biological factors were associated with the occurrence of relapse during the follow-up. Conclusions: Two thirds of patients in clinical remission presented clinical relapse shortly after infliximab withdrawal. Although the reintroduction of infliximab treatment was safe, half of the patients did not present the same clinical response that they had achieved prior to treatment withdrawal
IDN2 and Its Paralogs Form a Complex Required for RNA–Directed DNA Methylation
IDN2/RDM12 has been previously identified as a component of the RNA–directed DNA methylation (RdDM) machinery in Arabidopsis thaliana, but how it functions in RdDM remains unknown. By affinity purification of IDN2, we co-purified two IDN2 paralogs IDP1 and IDP2 (IDN2 PARALOG 1 and 2). The coiled-coil domain between the XS and XH domains of IDN2 is essential for IDN2 homodimerization, whereas the IDN2 C-terminal XH domain but not the coiled-coil domain is required for IDN2 interaction with IDP1 and IDP2. By introducing the wild-type IDN2 sequence and its mutated derivatives into the idn2 mutant for complementation testing, we demonstrated that the previously uncharacterized IDN2 XH domain is required for the IDN2-IDP1/IDP2 complex formation as well as for IDN2 function. IDP1 is required for de novo DNA methylation, siRNA accumulation, and transcriptional gene silencing, whereas IDP2 has partially overlapping roles with IDP1. Unlike IDN2, IDP1 and IDP2 are incapable of binding double-stranded RNA, suggesting that the roles of IDP1 and IDP2 are different from those of IDN2 in the IDN2-IDP1/IDP2 complex and that IDP1 and IDP2 are essential for the functioning of the complex in RdDM
Incomplete homogenization of 18 S ribosomal DNA coding regions in Arabidopsis thaliana
<p>Abstract</p> <p>Background</p> <p>As a result of concerted evolution, coding regions of ribosomal DNA sequences are highly conserved within species and variation is generally thought to be limited to a few nucleotides. However, rDNA sequence variation has not been systematically examined in plant genomes, including that of the model plant <it>Arabidopsis thaliana </it>whose genome was the first to be sequenced.</p> <p>Findings</p> <p>Both genomic and transcribed 18 S sequences were sampled and revealed that most deviation from the consensus sequence was limited to single nucleotide substitutions except for a variant with a 270 bp deletion from position 456 to 725 in <it>Arabidopsis </it>numbering. The deletion maps to the functionally important and highly conserved 530 loop or helix18 in the structure of <it>E. coli </it>16 S. The expression of the deletion variant is tightly controlled during developmental growth stages. Transcripts were not detectable in young seedlings but could be amplified from RNA extracts of mature leaves, stems, flowers and roots of <it>Arabidopsis thaliana </it>ecotype Columbia. We also show polymorphism for the deletion variant among four <it>Arabidopsis </it>ecotypes examined.</p> <p>Conclusion</p> <p>Despite a strong purifying selection that might be expected against functionally impaired rDNAs, the newly identified variant is maintained in the <it>Arabidopsis </it>genome. The expression of the variant and the polymorphism displayed by <it>Arabidopsis </it>ecotypes suggest a transition state in concerted evolution.</p
