    Antioxidants, radical-scavenging and protein carbonylation inhibition capacity of six monocultivar virgin olive oils in Istria (Croatia)

    Six monocultivar virgin olive oils (VOOs) produced from five autochthonous (Buža, Buža Puntoža, Istarska Bjelica, Porečka Rosulja, and Rosinjola) and one referent cultivar (Leccino), were investigated. The mass fractions of orthodiphenols (ORT) and tocopherols (TOC) were analysed by VIS spectroscopy and HPLC. The radical-scavenging capacity was evaluated by electron spin resonance spectroscopy (ESR) using galvinoxyl free radical and the DPPH test. Results revealed a high level of total TOC in VOOs of Buža Puntoža (243 mg kg−1) and Porečka Rosulja (325 mg kg−1). VOOs contained in 100 g from 79% (Istarska Bjelica) to 261% (Porečka Rosulja) of recommended daily allowance of α-tocopherol. The mass fraction of ORT in Istarska Bjelica and Rosinjola was >250 mg kg−1 (on average 30–40% higher in comparison to other studied cultivars). Istarska Bjelica and Porečka Rosulja showed the highest antioxidant capacity in both methods of measurement. The highest capacity to inhibit protein carbonylation (PC) in response to oxidative stress (54–56%) was displayed by Buža Puntoža, Leccino, and Buža. High level of positive linear correlation between ORT mass fraction and radical-scavenging capacity measured by DPPH test (r=0.768), as well as strong negative correlation between PC inhibition and mass fraction of ORT (r= −0.697), were observed

    Ampelographic characterisation of grapevine accessions denominated 'Refošk', 'Refosco', 'Teran' and 'Terrano' (Vitis vinifera L.) from Slovenia, Croatia and Italy

    Grapevines denominated 'Refošk', 'Refosco', 'Teran' and 'Terrano have been cultivated in the area of western Slovenia, north-western Croatia and north-eastern Italy for centuries. Despite historical documents reporting the longstanding tradition of grapevine cultivation and winemaking, the denomination and origin of these varieties is still questionable. The aims of this work were to study the genetic identity and relationship of the grapevine accessions denominated 'Refošk', 'Refosco', 'Teran' and 'Teranno' that have been traditionally cultivated in Slovenia, Croatia and Italy. For this purpose, 9 SSR loci were analysed to fingerprint 53 accessions with denominations or similar true-to-type morphologies of 'Refošk' and 'Teran'. The grapevine variety 'Refošk' cultivated in Slovenia and most accessions denominated 'Teran' in Croatia showed identical genotypes in all analysed SSR markers, and can therefore be used as synonyms. Five accessions showed identical genotypes to 'Refosco dal peduncolo rosso' variety, however five other genotyped accessions suggested individual profiles, and can be characterized as clonal mutants of true-to-type 'Refošk'/'Teran'. Accessions 'Sladki Teran' and 'Ref5/31' shared 56 % and 61 % of the alleles with true-to-type profiles of 'Refošk'/'Teran' and their parentage analysis strongly suggested a full-siblings relationship. Obtained results contribute to the understanding of the genetic diversity of grapevine varieties cultivated in this part of Europe

    Effect of in vitro gastrointestinal digestion on the total phenolic contents and antioxidant activity of wild Mediterranean edible plant extracts

    The recent interest in wild edible plants is associated with their health benefits, which are mainly due to their richness in antioxidant compounds, particularly phenolics. Nevertheless, some of these compounds are metabolized after ingestion, being transformed into metabolites frequently with lower antioxidant activity. The aim of the present study was to evaluate the influence of the digestive process on the total phenolic contents and antioxidant activity of extracts from four wild edible plants used in the Mediterranean diet (Beta maritima L., Plantago major L., Oxalis pes-caprae L. and Scolymus hispanicus L.). HPLC-DAD analysis revealed that S. hispanicus is characterized by the presence of caffeoylquinic acids, dicaffeoylquinic acids and flavonol derivatives, P. major by high amounts of verbascoside, B. maritima possesses 2,4-dihydroxybenzoic acid, 5-O-caffeoylquinic acid, quercetin derivatives and kaempferol-3-O-rutinoside, and O. pes-caprae extract contains hydroxycinnamic acids and flavone derivatives. Total phenolic contents were determined by Folin-Ciocalteu assay, and antioxidant activity by the ABTS, DPPH, ORAC and FRAP assays. Phenolic contents of P. major and S. hispanicus extracts were not affected by digestion, but they significantly decreased in B. maritima after both phases of digestion process and in O. pes-caprae after the gastric phase. The antioxidant activity results varied with the extract and the method used to evaluate the activity.     Viral Diversity in Autochthonous Croatian Grapevine Cultivars

    A survey was conducted on nine autochthonous grapevine cultivars grown along the Croatian coastal region. In total, 48 vines (44 from germplasm collection, 4 from vineyards) originating from 23 sites were tested for 26 viruses using molecular methods. Results revealed high infection rates with Grapevine leafroll-associated virus 3 (GLRaV-3); Grapevine virus A (GVA, both 91.7%); Grapevine fleck virus (GFkV, 87.5%); and Grapevine rupestris stem pitting-associated virus (GRSPaV, 83.3%). Other detected viruses were: Grapevine fanleaf virus (GFLV); Grapevine leafroll-associated viruses 1, 2, and strains of 4 (GLRaV-1, GLRaV-2, GLRaV-4); Grapevine viruses B, D, F (GVB, GVD, GVF); Grapevine red globe virus (GRGV); Grapevine vein feathering virus (GVFV); Grapevine Syrah virus 1 (GSyV-1); and Grapevine Pinot gris virus (GPGV). No virus-free vine was found. Mixed infections were determined in all vines, the number of viruses in a single vine ranged from three to nine. GLRaV-3 variant typing confirmed presence of group I, II, and III. Four vines with leaf deformation and mottling were positive for GPGV. Seven viruses (GLRaV-4-like group, GVD, GVE, GVF, GRGV, GSyV-1, and GVFV) were detected for the first time in Croatia.
