    Effects of in vitro short- and long-term treatment with telomerase inhibitor in U-251 glioma cells

    BACKGROUND: The inhibition of the enzyme telomerase (TERT) has been widely investigated as a new pharmacological approach for cancer treatment, but its real potential and the biochemical consequences are not totally understood. OBJECTIVE: Here, we investigated the effects of the telomerase inhibitor MST-312 on a human glioma cell line after both short- and long-term (290 days) treatments. METHODS: Effects on cell growth, viability, cell cycle, morphology, cell death and genes expression were assessed. RESULTS: We found that short-term treatment promoted cell cycle arrest followed by apoptosis. Importantly, cells with telomerase knock-down revealed that the toxic effects of MST-312 are partially TERT dependent. In contrast, although the long-term treatment decreased cell proliferation at first, it also caused adaptations potentially related to treatment resistance and tumor aggressiveness after long time of exposition. CONCLUSIONS: Despite the short-term effects of telomerase inhibition not being due to telomere erosion, they are at least partially related to the enzyme inhibition, which may represent an important strategy to pave the way for tumor growth control, especially through modulation of the non-canonical functions of telomerase. On the other hand, long-term exposure to the inhibitor had the potential to induce cell adaptations with possible negative clinical implications

Participants in the ChAdOx1 nCoV-19 group received two doses containing 5 × 1010 viral particles (standard dose; SD/SD cohort); a subset in the UK trial received a half dose as their first dose (low dose) and a standard dose as their second dose (LD/SD cohort). The primary efficacy analysis included symptomatic COVID-19 in seronegative participants with a nucleic acid amplification test-positive swab more than 14 days after a second dose of vaccine. Participants were analysed according to treatment received, with data cutoff on Nov 4, 2020. Vaccine efficacy was calculated as 1 - relative risk derived from a robust Poisson regression model adjusted for age. From 21 days after the first dose, there were ten cases hospitalised for COVID-19, all in the control arm; two were classified as severe COVID-19, including one death. There were 74 341 person-months of safety follow-up (median 3·4 months, IQR 1·3–4·8): 175 severe adverse events occurred in 168 participants, 84 events in the ChAdOx1 nCoV-19 group and 91 in the control group.     Desenvolvimento de nanocarreadores com base em carbonato de cálcio visando a entrega de short interfering RNA em câncer de mama

    O câncer de mama, segundo tipo de câncer mais comum entre mulheres, é ainda uma das principais causas de mortalidade no mundo. Em consequência, houve um aumento no número de pesquisas para melhorar os procedimentos terapêuticos convencionais, que incluem a pesquisa com a terapia por interferência de RNA (RNAi). Para tal, o desenvolvimento de novos nanocarreadores que sejam eficientes na proteção e na liberação do ativo é necessário. O objetivo deste estudo foi a preparação de nanopartículas inorgânico-orgânicas baseadas em carbonato de cálcio e copolímero contendo polietileno (glicol), através dos métodos de difusão a gás (método 1) e adição direta (método 2), para posterior aplicação como carreadores de short interfering RNA (siRNA) a serem utilizados na terapia anticâncer. As nanopartículas foram formadas por auto-associação, em um sistema fechado, para que houvesse a reação de carbonatação. Após a preparação, a suspensão de nanopartículas foi caracterizada físico-quimicamente. As medidas de espalhamento dinâmico de luz (DLS – dynamic light scattering) indicaram uma distribuição de tamanho com diâmetro médio em 474,3 ± 299,8 nm pelo método 1; e 618,4 ± 444,2 nm pelo método 2; e índice de polidispersão (PdI – polydispersity index) de 0,5292 ± 0,27 e 0,596 ± 0,20, para os respectivos métodos. Os histogramas de tamanho por intensidade mostraram distribuições bimodais, sugerindo a formação de duas apresentações com pico de tamanho em 9,4 e 1738,0 nm pelo método 1; e 7,4 e 766,7 nm pelo método 2, indicando que as partículas se apresentavam polidispersas, o que pode influenciar diretamente na sua estabilidade. Os resultados obtidos abrem nova perspectiva para a preparação de nanopartículas híbridas baseadas em carbonato de cálcio, no entanto é necessário melhorar o método para obtê-las com tamanho e PdI ideais para aplicação biomédica

    Avaliação de metodologias para a reutilização de membranas de nanocelulose bacteriana como scaffold para cultura celular

    Biofilmes bacterianos constituídos por matrizes biocompatíveis têm sido estudados como scaffolds celulares a serem aplicados na engenharia de tecidos. Em especial, membranas de nanocelulose bacteriana têm sido testadas in vitro e in vivo e descartadas. Mesmo com o reconhecimento da importância do reaproveitamento de materiais, ainda existem poucas metodologias para reutilizar este recurso. Este trabalho teve como objetivo avaliar metodologias de reutilização de membranas de nanocelulose bacteriana (BNC – bacterial nanocellulose membranes), previamente usadas em testes biológicos, com a finalidade de serem novamente viáveis como scaffold na cultura de células, evitando gastos dispendiosos e tempo de preparação de novas membranas. Para tal, membranas utilizadas em experimentos de projetos de doutorado que fazem o desenvolvimento de pele artificial foram reservadas para a tentativa de recuperação. Foram estabelecidos ensaios de higienização com base em métodos de purificação inicial, ou seja, métodos de retirada da bactéria do biofilme, deixando a membrana limpa e pronta para uso. As metodologias testadas baseiam-se em aplicação de pH extremo para degradação de materiais celulares, seguido de lavagem e esterilização. Para avaliar os resultados, as membranas foram fotografadas através de um microscópio óptico invertido. A espessura das membranas foi aferida por meio de um paquímetro; também foi calculada a perda de água das membranas, sendo esses parâmetros definidos antes e depois dos ensaios de limpeza. Em comparação microscópica feita da membrana sem e com o tratamento, verificou-se que após o tratamento não haviam restos ou resíduos celulares. Não foi observada alteração relevante na espessura da membrana, resultado semelhante ao do potencial de perda de água. Além disso, foi verificada a viabilidade de fibroblastos cultivados nas membranas recuperadas. Verificou-se que os métodos testados podem ser úteis na recuperação de membranas de nanocelulose utilizadas para nova aplicação, sendo o aproveitamento de grande relevância considerando custo, tempo, recurso e infraestrutura necessários para produção dos biofilmes como material biotecnológico de aplicação em engenharia de tecidos
