    Highly efficient xylem transport of arsenite in the arsenic hyperaccumulator Pteris vittata

    The hyperaccumulator Pteris vittata translocates arsenic (As) from roots to fronds efficiently, but the form of As translocated in xylem and the main location of arsenate reduction have not been resolved. Here, P. vittata was exposed to 5 mu M arsenate or arsenite for 1-24 h, with or without 100 mu M phosphate. Arsenic speciation was determined in xylem sap, roots, fronds and nutrient solutions by high-performance liquid chromatography (HPLC) linked to inductively coupled plasma mass spectrometry (ICP-MS). The xylem sap As concentration was 18-73 times that in the nutrient solution. In both arsenate- and arsenite-treated plants, arsenite was the predominant species in the xylem sap, accounting for 93-98% of the total As. A portion of arsenate taken up by roots (30-40% of root As) was reduced to arsenite rapidly. The majority (c. 80%) of As in fronds was arsenite. Phosphate inhibited arsenate uptake, but not As translocation. More As was translocated to fronds in the arsenite-treated than in the arsenate-treated plants. There was little arsenite efflux from roots to the external solution. Roots are the main location of arsenate reduction in P. vittata. Arsenite is highly mobile in xylem transport, possibly because of efficient xylem loading, little complexation with thiols in roots, and little efflux to the external medium

    Shift Work Is Not Associated with High Blood Pressure or Prevalence of Hypertension

    Background: Working mostly at night has been suggested to be associated with upset of chronobiological rhythms and high blood pressure, but the evidence from epidemiological studies is weak. Methods: In a cross-sectional survey, we evaluated the association between shift work and blood pressure, pre-hypertension and hypertension. In total, 493 nurses, nurse technicians and assistants, were selected at random in a large general hospital setting. Hypertension was diagnosed by the mean of four automatic blood pressure readings 140/90mmHgoruseofbloodpressureloweringagents,andpre−hypertensionbysystolicbloodpressure140/ 90 mmHg or use of blood pressure lowering agents, and pre-hypertension by systolic blood pressure 120–139 or diastolic blood pressure $80–89 mmHg. Risk factors for hypertension were evaluated by a standardized questionnaire and anthropometric measurements. The association between the shift of work and blood pressure, pre-hypertension and hypertension was explored using univariate and multivariate analyses that controlled for risk factors for hypertension by covariance analysis and modified Poisson regression. Results: The mean age of the participants was 34.369.4 years and 88.2 % were women. Night shift workers were older, more frequently married or divorced, and less educated. The prevalence of hypertension in the whole sample was 16%, and 28% had pre-hypertension. Blood pressure (after adjustment for confounding) was not different in day and night shift workers.     The role of the rice aquaporin Lsi1 in arsenite efflux from roots

    When supplied with arsenate (As(V)), plant roots extrude a substantial amount of arsenite (As(III)) to the external medium through as yet unidentified pathways. The rice (Oryza sativa) silicon transporter Lsi1 (OsNIP2;1, an aquaporin channel) is the major entry route of arsenite into rice roots. Whether Lsi1 also mediates arsenite efflux was investigated. Expression of Lsi1 in Xenopus laevis oocytes enhanced arsenite efflux, indicating that Lsi1 facilitates arsenite transport bidirectionally. Arsenite was the predominant arsenic species in arsenate-exposed rice plants. During 24-h exposure to 5 mu M arsenate, rice roots extruded arsenite to the external medium rapidly, accounting for 60-90% of the arsenate uptake. A rice mutant defective in Lsi1 (lsi1) extruded significantly less arsenite than the wild-type rice and, as a result, accumulated more arsenite in the roots. By contrast, Lsi2 mutation had little effect on arsenite efflux to the external medium. We conclude that Lsi1 plays a role in arsenite efflux in rice roots exposed to arsenate. However, this pathway accounts for only 15-20% of the total efflux, suggesting the existence of other efflux transporters

    The interface of syntax with pragmatics and prosody in children with Autism Spectrum Disorders

    In order to study problems of individuals with Autism Spectrum Disorders (ASD) with morphosyntax, we investigated twenty high-functioning Greek-speaking children (mean age:6;11) and twenty age- and language-matched typically developing children on environments that allow or forbid object clitics or their corresponding noun phrase. Children with ASD fell behind typically developing in comprehending and producing simple clitics and producing noun phrases in focus structures. The two groups performed similarly in comprehending and producing clitics in clitic left dislocation and in producing noun phrases in non-focus structures. We argue that children with ASD have difficulties at the interface of(morpho)syntax with pragmatics and prosody, namely, distinguishing a discourse prominent element, and considering intonation relevant for a particular interpretation that excludes clitics

    Arsenite efflux is not enhanced in the arsenate-tolerant phenotype of Holcus lanatus

    P>Arsenate tolerance in Holcus lanatus is achieved mainly through suppressed arsenate uptake. We recently showed that plant roots can rapidly efflux arsenite to the external medium. Here, we tested whether arsenite efflux is a component of the adaptive arsenate tolerance in H. lanatus. Tolerant and nontolerant phenotypes were exposed to different arsenate concentrations with or without phosphate for 24 h, and arsenic (As) speciation was determined in nutrient solutions, roots and xylem sap. At the same arsenate exposure concentration, the nontolerant phenotype took up more arsenate and effluxed more arsenite than the tolerant phenotype. However, arsenite efflux was proportional to arsenate uptake and was not enhanced in the tolerant phenotype. Within 2-24 h, most (80-100%) of the arsenate taken up was effluxed to the medium as arsenite. About 86-95% of the As in the roots and majority of the As in xylem sap (c. 66%) was present as arsenite, and there were no significant differences between phenotypes. Arsenite efflux is not adaptively enhanced in the tolerant phenotype H. lanatus, but it could be a basal tolerance mechanism to greatly decrease cellular As burden in both phenotypes.     Direct Visualization of Dislocation Dynamics in Grain Boundary Scars

    Mesoscale objects with unusual structural features may serve as the analogues of atoms in the design of larger-scale materials with novel optical, electronic or mechanical behaviour. In this paper we investigate the structural features and the equilibrium dynamics of micron-scale spherical crystals formed by polystyrene particles adsorbed on the surface of a spherical water droplet. The ground state of sufficiently large crystals possesses finite-length grain boundaries (scars). We determine the elastic response of the crystal by measuring single-particle diffusion and quantify the fluctuations of individual dislocations about their equilibrium positions within a scar determining the dislocation spring constants. We observe rapid dislocation glide with fluctuations over the barriers separating one local Peierls minimum from the next and rather weak binding of dislocations to their associated scars. The long-distance (renormalised) dislocation diffusion glide constant is extracted directly from the experimental data and is found to be moderately faster than single particle diffusion. We are also able to determine the parameters of the Peierls potential induced by the underlying crystalline lattice.Comment: 11 pages, 4 figures, pdf forma

    The Integrated Genomic Landscape of Thymic Epithelial Tumors

    Thymic epithelial tumors (TETs) are one of the rarest adult malignancies. Among TETs, thymoma is the most predominant, characterized by a unique association with autoimmune diseases, followed by thymic carcinoma, which is less common but more clinically aggressive. Using multi-platform omics analyses on 117 TETs, we define four subtypes of these tumors defined by genomic hallmarks and an association with survival and World Health Organization histological subtype. We further demonstrate a marked prevalence of a thymoma-specific mutated oncogene, GTF2I, and explore its biological effects on multi-platform analysis. We further observe enrichment of mutations in HRAS, NRAS, and TP53. Last, we identify a molecular link between thymoma and the autoimmune disease myasthenia gravis, characterized by tumoral overexpression of muscle autoantigens, and increased aneuploidy

    Austenite in Transformation-Induced Plasticity Steel Subjected to Multiple Isothermal Heat Treatments

    The thermodynamic limit to the progress of the bainite reaction in steels containing a cementite inhibitor often leaves large quantities of thermally or mechanically unstable austenite. Such austenite is not effective in delaying the onset of plastic instabilities during the course of deformation. In such circumstances, it is useful to conduct isothermal transformation at a high temperature where the rate of reaction is relatively rapid, followed by a lower temperature step that permits more bainite to be generated. This in turn increases the stability of the refined austenite, which then transforms gently over a large range of strain during a tensile test. A significant corollary is that the two-step heat treatments are unnecessary in low-carbon steels, where the bainite reaction is able to proceed to a greater extent before reaching the thermodynamic limit. Furthermore, the two-step process can be counterproductive in low carbon steel, because the austenite content is reduced to a level below which it does not enhance the mechanical properties.
