    Coplas, diablos y carnaval

    La Copla has remained in America for centuries expressing the life of the original peoples in the Andes. It is, in the words of Leda Valladares, of collective songs accompanied by Cajas, whistles and rattles that appear in Carnival time with a strength of the accent and the voice expelled in spurts, and where the figure of the ¨diablo¨ is the one who presides all this Party space and joy. In South America, during the process of conquest and colonization, the Catholic Church began a period of catechism proposing to make known the images of both the Virgin Mary, Jesus and the saints, as well as that of angels and devils. Thus, these figures with different conceptions and assimilations were repeated from the colonial era to the present day in different parts of the American continent. The objective of this work is to make a comparison of the couplets of the Argentine Northwest, with the Couplets of the Cajamarquino Carnival of Peru, where the figure of the carnival devil represents a time when the most significant features of Andean social life become “bottoms-up”, which is then reflected in the lyrics and music of these songs.La Copla se ha mantenido en América durante siglos expresando la vida de los pueblos originarios en los Andes. Se trata, al decir de Leda Valladares, de cantos colectivos acompañados con cajas, silbatos y matracas que aparecen en tiempo de Carnaval con una pujanza del acento y la voz expulsada a borbotones y en donde la figura del “diablo” es quien preside todo este espacio de fiesta y alegría. En América, durante el proceso de conquista y colonización, la iglesia católica comenzó un momento de catequización proponiendo dar a conocer las imágenes, tanto de la Virgen María, Jesús y los santos, como también, la de los ángeles y de los diablos. Es así, que estas figuras con distintas concepciones y asimilaciones se repitieron desde la época colonial hasta nuestros días en distintos puntos del continente americano. El objetivo de este trabajo es realizar una comparación de las coplas del Noroeste Argentino, con las Coplas del Carnaval Cajamarquino, de Perú en donde la figura del diablo carnavalero representa un tiempo en que los rasgos más significativos de la vida social andina “se ponen de cabeza” reflejándose en la letra y música de estos cantos

    Sustainable Tourism and Facilities Preferences: The Sustainable Tourist Stay Scale ({STSS}) Validation

    This study aims to start the development of the Sustainable Tourist Stay Scale (STSS), a self-report instrument designed to measure tourists' preferences regarding the degree to which they accept accommodation and programs in tourism facilities with sustainable characteristics. The research involved a total sample of 621 participants aged 18 to 74 (m = 41.75%; f = 58.25%). According to the literature and the available data, we considered the possibility that young people (millennials) and adults within the same sample may show peculiarities concerning the sustainability issues. We carried out three subsequent analyses: (1) an explorative factor analysis; (2) a confirmatory factor analysis via structural equation modelling; (3) the test of the structural invariance between young people and adults. The results supported a three-factor scale solution and they are discussed with reference to their potential practical applications to better understanding the preference for a sustainable stay

    Psychometric properties of a new instrument for the measurement of the perceived quality of distance learning during the coronavirus disease 2019 (COVID-19) pandemic

    Introduction: The lockdown restrictions due to the COVID-19 pandemic forced many students to use distance learning. Few studies have examined the psychological effects of distance learning during the pandemic on university or on non-university students using a specific psychometric tool. The principal aim of this study was the construction and validation of a new psychometric tool, the Perceived Quality of Distance Learning (PQDL), to measure students’ appreciation and reaction to distance learning. The connection between anxiety, depression, perceived self-efficacy, and students’ perception of distance learning was analyzed to assess the nomological validity of the new scale. Method: The sample consists of 429 students who attended university or training courses. The factor structure of the new instrument was analyzed through Exploratory and Confirmatory Factor Analyses and its nomological validity was analyzed through regression analysis. Conclusion: The results showed that PQDL consists of two subscales: Distance Learning Organization and Cognitive-Emotive Reaction to Distance Learning. Higher student’s ability to organize and plan distance learning and higher student’s positive cognitive-emotive reaction to distance learning, higher student’s perceived quality of distance learning. Anxiety and depression scores were negatively correlated with students’ perceived quality of distance learning. Furthermore, students’ perceived emotional self-efficacy of negative emotions and perceived scholastic self-efficacy were positively correlated with students’ perceived quality of distance learning. These data indicate that PQDL is a reliable questionnaire to assess student’s perceived quality of distance learning

    The mediating role of positive and negative affect in the relationship between death anxiety and Italian students’ perceptions of distance learning quality during the COVID-19 pandemic

    Recent data show that death anxiety and negative affect (NA) have become increasingly relevant because of the COVID-19 pandemic. The constant reminders of mortality through mass media and social media have contributed to this trend. Simultaneously, students have experienced a sudden and radical shift from face-to-face to online teaching, reducing direct human interactions and increasing anxiety. Death anxiety is often associated with mental illnesses and maladaptive mood states such as depression, anxiety, and NA. Despite this, few studies have investigated the effect of death anxiety, positive affect (PA), and NA on students’ perceived quality of distance learning. The present study aims to investigate the association among death anxiety, PA, NA, and the perceived quality of distance learning in a sample of 429 students attending university or training courses. Positive and negative affect were assessed through the positive and negative affect scales; death anxiety was measured through the death anxiety scale, and the perceived quality of distance learning was evaluated through the perceived quality of distance learning questionnaire. Zero-order correlation coefficients were calculated among the examined variables. To study the mediating role of positive and negative affect in the relationship between students’ perceptions of distance learning quality and death anxiety, we employed multiple regression analyses. Our findings indicate a significant association between death anxiety and cognitive–emotive reactions to distance learning. Lower levels of death anxiety are associated with PA, while higher levels are related to NA. Moreover, PA and NA act as mediators in the relationship between death anxiety and a positive reaction to distance learning. In conclusion, our findings highlight that PA and NA partially mediate the relationship between death anxiety and the perceived quality of distance learning

    Intermittent theta-burst stimulation rescues dopamine-dependent corticostriatal synaptic plasticity and motor behavior in experimental parkinsonism. Possible role of glial activity.

    Background: Recent studies support the therapeutic utility of repetitive transcranial magnetic stimulation in Parkinson's disease (PD), whose progression is correlated with loss of corticostriatal long-term potentiation and long-term depression. Glial cell activation is also a feature of PD that is gaining increasing attention in the field because astrocytes play a role in chronic neuroinflammatory responses but are also able to manage dopamine (DA) levels. Methods: Intermittent theta-burst stimulation protocol was applied to study the effect of therapeutic neuromodulation on striatal DA levels measured by means of in vivo microdialysis in 6-hydroxydopamine-hemilesioned rats. Effects on corticostriatal synaptic plasticity were studied through in vitro intracellular and whole-cell patch clamp recordings while stepping test and CatWalk were used to test motor behavior. Immunohistochemical analyses were performed to analyze morphological changes in neurons and glial cells. Results: Acute theta-burst stimulation induced an increase in striatal DA levels in hemiparkinsonian rats, 80 minutes post-treatment, correlated with full recovery of plasticity and amelioration of motor performances. With the same timing, immediate early gene activation was restricted to striatal spiny neurons. Intense astrocytic and microglial responses were also significantly reduced 80 minutes following theta-burst stimulation. Conclusion: Taken together, these results provide a first glimpse on physiological adaptations that occur in the parkinsonian striatum following intermittent theta-burst stimulation and may help to disclose the real potential of this technique in treating PD and preventing DA replacement therapy-associated disturbances

    Human papillomavirus (HPV) DNA in penile carcinomas in Argentina: Analysis of primary tumors and lymph nodes

    Among sexually transmitted diseases, infection by human papillomavirus (HPV) has become one of the most important. On the other hand, though epidemiological data show that some HPV types are closely associated with cervical cancer, few reports have been found with reference to penile carcinoma because of its rare occurrence. The aim of this study was to investigate the relationship between HPV infection and penile cancer in Argentina. A retrospective study was carried out on 38 white men with penile squamous-cell carcinoma. Sixty-five archival fixed biopsies taken from 34 primary penile tumors, 25 nodal metastases, 1 skin “satellite” metastasis and 5 histologically normal lymph nodes were used as specimens. HPV detection and typing were carried out by the polymerase chain reaction (PCR) using generic primers, combined with single-stranded conformational polymorphism (SSCP) analysis. HPV DNA was found in 71% patients, corresponding 81% of them to “high risk” types, with predominance of HPV 18. Both primary tumors and metastases showed concordance of HPV occurrence and type in both lesions. In 3 patients, HPV 16 was detected not only in primary tumors and metastases, but also in histologically normal lymph nodes. Our data indicate that most penile carcinomas in Argentine patients are etiologically related to HPV, especially to “high risk” genital types. The agreement in HPV detection between primary tumors and metastases suggests a potential viral role in tumor progression.     Association of COMT, BDNF and 5-HTT functional polymorphisms with personality characteristics.

    Background: The real impact of genetic factors on personality is still unknown, even if in literature about 50% of variance in personality traits are considered genetically determined. The determination of the genetic variance in personality traits could promote psychological well-being and the prevention of psychopathologies, because there are many experimental evidences showing that mental illness is associated to personality. Numerous studies have showed that Catechol-O-methyltransferase (COMT), brain derived neurotrophic factor (BDNF) and serotonin transporter (5-HTT) are genes whose variants are associated with personality traits. This aim of this study is the investigation of the association between personality traits and 5-HTTLPR/rs255315-HTT promoter variant, COMT Val158Met and BDNF Val66Met gene polymorphisms. Methods: The sample was composed by 132 healthy female students. Genomic DNA was extracted from buccal swab, while personality was assessed with Cloninger's Temperament and Character Inventory-Revised (TCI-R). Linear discriminant analysis was used to analyze how personality characteristics can differentiate individuals in relation to their genetic polymorphisms. Results: Data showed that the temperament trait Reward Dependence discriminated individuals with different BDNF variants; Novelty Seeking and Harm Avoidance discriminated individuals with different 5HTTLPR variants; Persistence discriminated individuals with different COMT variants. Conclusions: Since these traits are connected to psychological diseases as depression, social anxiety, anorexia and obsessive-compulsive disorders of personality, the study of their genetic component can be used as intermediary issue to better define the connection between genes and predisposition toward maladaptive behavior and mental illness

    Mitochondrial DNA ancestry, HPV infection and the risk of cervical cancer in a multiethnic population of northeastern Argentina

    Background: Misiones Province in northeastern Argentina is considered to be a region with a high prevalence of HPV infection and a high mortality rate due to cervical cancer. The reasons for this epidemiological trend are not completely understood. To gain insight into this problem, we explored the relationship between mitochondrial DNA (mtDNA) ancestry, HPV infection, and development of cervical lesions/cancer in women from the city of Posadas in Misiones Province. Methods: Two hundred and sixty-one women, including 92 cases of patients diagnosed with cervical lesions and 169 controls, were analyzed. mtDNA ancestry was assessed through HVS1 sequencing, while the detection and typing of HPV infection was conducted through nested multiplex PCR analysis. Multivariate logistic regression was conducted with the resulting data to estimate the odds ratios (ORs) adjusted by socio-demographic variables. Results: The study participants showed 68.6% Amerindian, 26.1% European and 5.3% African mtDNA ancestry, respectively. Multiple regression analysis showed that women with African mtDNAs were three times more likely to develop a cervical lesion than those with Native American or European mtDNAs [OR of 3.8 (1.2–11.5) for ancestry and OR of 3.5 (1.0–12.0) for L haplogroups], although the associated p values were not significant when tested under more complex multivariate models. HPV infection and the development of cervical lesions/cancer were significant for all tested models, with the highest OR values for HPV16 [OR of 24.2 (9.3–62.7)] and HPV-58 [OR of 19.0 (2.4–147.7)]. Conclusion: HPV infection remains a central risk factor for cervical cancer in the Posadas population. The potential role of African mtDNA ancestry opens a new avenue for future medical association studies in multiethnic populations, and will require further confirmation in large-scale studies.Fil: Badano, Ines.     Phosphoproteomic Landscaping Identifies Non-canonical cKIT Signaling in Polycythemia Vera Erythroid Progenitors

    Although stem cell factor (SCF)/cKIT interaction plays key functions in erythropoiesis, cKIT signaling in human erythroid cells is still poorly defined. To provide new insights into cKIT-mediated erythroid expansion in development and disease, we performed phosphoproteomic profiling of primary erythroid progenitors from adult blood (AB), cord blood (CB), and Polycythemia Vera (PV) at steady-state and upon SCF stimulation. While AB and CB, respectively, activated transient or sustained canonical cKIT-signaling, PV showed a non-canonical signaling including increased mTOR and ERK1 and decreased DEPTOR. Accordingly, screening of FDA-approved compounds showed increased PV sensitivity to JAK, cKIT, and MEK inhibitors. Moreover, differently from AB and CB, in PV the mature 145kDa-cKIT constitutively associated with the tetraspanin CD63 and was not endocytosed upon SCF stimulation, contributing to unrestrained cKIT signaling. These results identify a clinically exploitable variegation of cKIT signaling/metabolism that may contribute to the great erythroid output occurring during development and in PV

    HIV-1 tat protein enters dysfunctional endothelial cells via integrins and renders them permissive to virus replication

    Previous work has shown that the Tat protein of Human Immunodeficiency Virus (HIV)-1 is released by acutely infected cells in a biologically active form and enters dendritic cells upon the binding of its arginine-glycine-aspartic acid (RGD) domain to the α5β1, αvβ3, and αvβ5 integrins. The up-regulation/activation of these integrins occurs in endothelial cells exposed to inflammatory cytokines that are increased in HIV-infected individuals, leading to endothelial cell dysfunction. Here, we show that inflammatory cytokine-activated endothelial cells selectively bind and rapidly take up nano-micromolar concentrations of Tat, as determined by flow cytometry. Protein oxidation and low temperatures reduce Tat entry, suggesting a conformation- and energy-dependent process. Consistently, Tat entry is competed out by RGD-Tat peptides or integrin natural ligands, and it is blocked by anti-α5β1, -αvβ3, and -αvβ5 antibodies. Moreover, modelling-docking calculations identify a low-energy Tat-αvβ3 integrin complex in which Tat makes contacts with both the αv and β3 chains. It is noteworthy that internalized Tat induces HIV replication in inflammatory cytokine-treated, but not untreated, endothelial cells. Thus, endothelial cell dysfunction driven by inflammatory cytokines renders the vascular system a target of Tat, which makes endothelial cells permissive to HIV replication, adding a further layer of complexity to functionally cure and/or eradicate HIV infection
