    Expectation Accuracy, Cost Behavior, and Slippery Prices

    This study investigates the relation between the accuracy of managerial demand expectations and cost behavior. Based on a unique dataset of 4,107 firm-year observations from 737 companies in Denmark, this paper first shows that cost stickiness is substantially stronger for unforeseen negative demand shocks than for expected demand shocks, which is in line with the argument that adjustment costs are typically higher when the time horizon for making the adjustments is shorter. Second, we find empirical evidence that omitting selling price changes can mislead researchers’ inferences about resource adjustments and cost stickiness when sales are used as a proxy for activity level, which is a common practice in the existing literature. Finally, we show a moderating effect of managerial expectation accuracy on the relationship between demand volatility and cost elasticity. This helps reconciling the ongoing debate on whether companies increase or decrease cost elasticity in response to demand volatility

    Europium nanoparticle-based high performing immunoassay for the screening of treponemal antibodies

    Treponema pallidum subspecies pallidum (Tp) is the causative agent of syphilis which mainly spreads through sexual contact, blood transfusion and perinatal route. In order to curtail the spread of the infection and to clinically manage the disease, timely, accurate and reliable diagnosis is very important. We have developed an immunoassay for the detection of treponemal antibodies in human serum or plasma samples. In vivo biotinylated and non-biotinylated versions of the recombinant antigen were designed by the fusion of three Tp-specific antigens namely Tp15, Tp17 and Tp47. These fusion antigens were expressed in E. coli and purified using single-step metal affinity chromatography. Biotinylated fusion antigen immobilized on streptavidin coated plate was used to capture the treponemal antibodies and the non-biotinylated antigen coated on europium nanoparticles was used as tracer. Assays with two different incubation times of 10 min and 1 h were developed, and following the incubation the europium fluorescence was measured using time-resolved fluorometry. The developed Time-resolved Fluorometric (TRF) immunoassays were evaluated with in-house and commercial serum/plasma sample panels. For well-established treponemal antibodies positive or negative samples, the sensitivity of TRF immunoassay with 10 min incubation time was 97.4% and of TRF immunoassay with 1 h incubation time was 98.7% and the specificities of both the TRF immunoassays were 99.2%. For the samples with discordant results with the reference assays, both the TRF immunoassays showed better specificity than the Enzygnost syphilis enzyme immunoassay as a screening test. The two different incubation times did not have any significant effect on the signal to cutoff (S/Co) ratios obtained with the two immunoassays (p  =  0.06). Our results indicate that the developed immunoassay with a short incubation time of 10 min has the potential to be used in clinical laboratories and in blood-bank settings as a screening test for treponemal antibodies

    CA125 : A superior prognostic biomarker for colorectal cancer compared to CEA, CA19-9 or CA242

    OBJECTIVES: The tumor stage represents the single most important prognostic factor for colorectal cancer (CRC), although more accurate prognostics remain much needed. Previously, we identified CA125 as an independent significant prognostic factor, which we have further validated along with CEA, CA19-9, and CA242 in a large cohort of CRC patients. METHODS: Using enzyme-linked immunosorbent assays, we analyzed preoperative serum samples in 322 CRC patients operated on between 1998 and 2003. RESULTS: Using the Spearman's rho model, we calculated the correlation between our previous findings on MUC16 and CA125, for which the correlation coefficient was 0.808 (p 67, with stage I-II or III-IV, and both colon and rectal cancer exhibited poor prognoses. In the multivariate analysis, we used clinical pathological variables in the model, where age, gender, and stage served as the background characteristics. We dichotomized CA125 using the Youden maximal cutoff point, and the median values for CEA, CA19-9, and CA242. CA125 emerged as the only marker remaining significant and independent together with stage, location, and age (HR 1.91; 95% CI 1.24-2.95; p 0.003). CONCLUSIONS: CA125 represents a significant and independent prognostic factor in CRC patients, superior to CEA. Furthermore, CA242 served as a better prognostic marker than both CEA and CA19-9. We recommend including both CA125 and CA242 in prognostic clinical trials among CRC patients.Peer reviewe

    Simultaneous detection of Human Immunodeficiency Virus 1 and Hepatitis B virus infections using a dual-label time-resolved fluorometric assay

    A highly specific and novel dual-label time-resolved immunofluorometric assay was developed exploiting the unique emission wavelengths of the intrinsically fluorescent terbium (Tb3+) and europium (Eu3+) tracers for the simultaneous detection of human immunodeficiency virus 1 (HIV-1) and hepatitis B virus (HBV) infections, respectively. HIV-1 infection was detected using a double antigen sandwich format wherein anti-HIV-1 antibodies were captured using an in vivo biotinylated version of a chimeric HIV-1 antigen and revealed using the same antigen labeled with Tb3+ chelate. Hepatitis B surface antigen (HBsAg), which served as the marker of HBV infection, was detected in a double antibody sandwich using two monoclonal antibodies (mAbs), one chemically biotinylated to capture, and the other labeled with Eu3+ nanoparticles, to reveal. The performance of the assay was evaluated using a collection (n = 60) of in-house and commercially available human sera panels. This evaluation showed the dual-label assay to possess high degrees of specificity and sensitivity, comparable to those of commercially available, single analyte-specific kits for the detection of HBsAg antigen and anti-HIV antibodies. This work demonstrates the feasibility of developing a potentially time- and resource-saving multiplex assay for screening serum samples for multiple infections in a blood bank setting

    Detection of bladder cancer with aberrantly fucosylated ITGA3

    We describe a simple, non-invasive assay to identify fucosylated-glycoisoform of integrin alpha-3 (ITGA3) directly from unprocessed urine. ITGA3 was detected directly from the urine of bladder cancer (BlCa) (n = 13) and benign prostatic hyperplasia (BPH) (n = 9) patients with the use of lectins coated on europium-doped-nanoparticles (Eu3+-NPs). Lectin Ulex europaeus agglutinin-I (UEA) showed enhanced binding with BlCa-derived ITGA3. The evaluation with individual samples showed that a glycovariant ITGA3-UEA assay could significantly discriminate BlCa from BPH patients (p = 0.007). The detection of aberrantly fucosylated-isoform of ITGA3 from urine can be used to distinguish BlCa from age-matched benign controls in a simple sandwich assay

    Small molecule tandem organic photovoltaic cells incorporating an α-NPD optical spacer layer

    We report an improvement in power conversion efficiency in a small molecule tandem organic photovoltaic (OPV) device by the optimisation of current balancing of the sub-cells using an optical spacer layer. A co-deposited layer of N,N’-bis(1-naphthyl)-N,N′-diphenyl-1,1’-biphenyl-4,4’-diamine (α-NPD) and molybdenum oxide was used as the optical spacer layer and provided a highly transparent and conductive layer. Optical simulations showed the addition of the optical spacer in a boron subphthalocyanine (SubPc)/C60 based tandem OPV device increased the SubPc absorption in the front sub-cell and resulted in current balancing through the device. Fabricated tandem OPV devices showed similar trends, with the power conversion efficiency increasing from 2.3% to 4.2% with the addition of an optimised optical spacer thickness. External quantum efficiency and total absorption efficiency measurements back up the optical model data which attribute the increased performance to improved SubPc absorption in the front sub-cell, balancing the photocurrents of the two sub-cells
