    Activation of the Listeria monocytogenes Virulence Program by a Reducing Environment.

    Upon entry into the host cell cytosol, the facultative intracellular pathogen Listeria monocytogenes coordinates the expression of numerous essential virulence factors by allosteric binding of glutathione (GSH) to the Crp-Fnr family transcriptional regulator PrfA. Here, we report that robust virulence gene expression can be recapitulated by growing bacteria in a synthetic medium containing GSH or other chemical reducing agents. Bacteria grown under these conditions were 45-fold more virulent in an acute murine infection model and conferred greater immunity to a subsequent lethal challenge than bacteria grown in conventional media. During cultivation in vitro, PrfA activation was completely dependent on the intracellular levels of GSH, as a glutathione synthase mutant (ΔgshF) was activated by exogenous GSH but not reducing agents. PrfA activation was repressed in a synthetic medium supplemented with oligopeptides, but the repression was relieved by stimulation of the stringent response. These data suggest that cytosolic L. monocytogenes interprets a combination of metabolic and redox cues as a signal to initiate robust virulence gene expression in vivoIMPORTANCE Intracellular pathogens are responsible for much of the worldwide morbidity and mortality from infectious diseases. These pathogens have evolved various strategies to proliferate within individual cells of the host and avoid the host immune response. Through cellular invasion or the use of specialized secretion machinery, all intracellular pathogens must access the host cell cytosol to establish their replicative niches. Determining how these pathogens sense and respond to the intracellular compartment to establish a successful infection is critical to our basic understanding of the pathogenesis of each organism and for the rational design of therapeutic interventions. Listeria monocytogenes is a model intracellular pathogen with robust in vitro and in vivo infection models. Studies of the host-sensing and downstream signaling mechanisms evolved by L. monocytogenes often describe themes of pathogenesis that are broadly applicable to less tractable pathogens.     Exploring a string-like landscape

    We explore inflationary trajectories within randomly-generated two-dimensional potentials, considered as a toy model of the string landscape. Both the background and perturbation equations are solved numerically, the latter using the two-field formalism of Peterson and Tegmark which fully incorporates the effect of isocurvature perturbations. Sufficient inflation is a rare event, occurring for only roughly one in 10510^5 potentials. For models generating sufficient inflation, we find that the majority of runs satisfy current constraints from WMAP. The scalar spectral index is less than 1 in all runs. The tensor-to-scalar ratio is below the current limit, while typically large enough to be detected by next-generation CMB experiments and perhaps also by Planck. In many cases the inflationary consistency equation is broken by the effect of isocurvature modes.Comment: 24 pages with 8 figures incorporated, matches version accepted by JCA

    Dietary supplementation with pollen enhances survival and Collembola boosts fitness of a web-building spider

    Uncertainties exist about the value of non-prey food for predators that are commonly food-limited, and the dietary conditions where non-prey foods are beneficial for carnivorous species. Prior studies show that large quantities of pollen grains are intercepted in the webs of web-building spiders. We examined the nutritional benefits of pollen as a non-prey food for a common ground-dwelling, sheet web-building spider, Mermessus fradeorum (Berland) (Araneae: Linyphiidae). These predators were provided diets of prey or no prey in the presence and absence of pollen. Treatment effects were quantified by measuring predator body nutrient composition, survival, body size, and offspring production. Per unit dry weight, pollen had less nitrogen and lipids than prey, although relative quantities of these nutrients per meal were not measured. Dietary treatments altered the body tissue composition of the spiders, leading to the highest N content and lipid reserves in spiders provided with Collembola. Supplementing diets with pollen increased both juvenile and adult survival, and the greatest survivorship and offspring production was observed when spiders were provided diets of Collembola supplemented with pollen. Our results show that Collembola are high-quality prey for spiders and pollen has positive effects on nutritional status and survival of a carnivorous species. Foraging on plant material potentially promotes population growth at early and late developmental stages by supplementing diets of poor-quality prey, and preventing starvation when prey are scarce

    Completion of the Long Duration Wear Test of the NASA HERMeS Hall Thruster

    The NASA Hall Effect Rocket with Magnetic Shielding (HERMeS) 12.5-kW Hall thruster has been the subject of extensive technology maturation by NASA GRC and JPL in preparation for development into a flight propulsion system. As part of this effort, a series of wear tests have been conducted to identify erosion phenomena and the accompanying failure modes as well as to validate service-life models for magnetically-shielded thrusters. This work presents a summary of the results obtained during the Long Duration Wear Test (LDWT), which was the third in this wear test series. The LDWT accumulated approximately 3,570 hours of operation and had the overall goal to identify and correct design or facility issues prior to the flight qualification campaign. Thruster performance, stability, and plume properties were invariant throughout the duration of the LDWT and consistent with measurements acquired during previous HERMeS performance and wear characterizations. Average erosion rates of a carbon-carbon composite pole cover were found to match those measured with graphite to within the empirical uncertainty while the previously observed time-dependence of pole cover erosion rates was linked to changes in pole cover roughness. Azimuthal variations in keeper wear rate were observed including deposition on one of the azimuthal-facing sides of the keeper mask. This strongly suggests the presence of an azimuthal component in the process driving keeper erosion

    The Black Hole Mass of NGC 4151. II. Stellar Dynamical Measurement from Near-Infrared Integral Field Spectroscopy

    We present a revised measurement of the mass of the central black hole (Mbh) in the Seyfert 1 galaxy NGC 4151. The new stellar dynamical mass measurement is derived by applying an axisymmetric orbit-superposition code to near-infrared integral field data obtained using adaptive optics with the Gemini NIFS spectrograph. When our models attempt to fit both the NIFS kinematics and additional low spatial resolution kinematics, our results depend sensitively on how chi-squared is computed--probably a consequence of complex bar kinematics that manifest immediately outside the nuclear region. The most robust results are obtained when only the high spatial resolution kinematic constraints in the nuclear region are included in the fit. Our best estimates for the BH mass and H-band mass-to-light ratio are Mbh~(3.76+/-1.15)E7 Msun (1-sigma error) and M/L(H-band)~0.34+/-0.03 Msun/Lsun (3-sigma error), respectively (the quoted errors reflect the model uncertainties). Our BH mass measurement is consistent with estimates from both reverberation mapping (3.57[+0.45/-0.37]E7 Msun) and gas kinematics (3.0[+0.75/-2.2]E7 Msun; 1-sigma errors), and our best-fit mass-to-light ratio is consistent with the photometric estimate of M/L(H-band)=0.4+/-0.2 Msun/Lsun. The NIFS kinematics give a central bulge velocity dispersion sigma_c=116+/-3 km/s, bringing this object slightly closer to the M-sigma relation for quiescent galaxies. Although NGC 4151 is one of only a few Seyfert 1 galaxies in which it is possible to obtain a direct dynamical BH mass measurement--and thus, an independent calibration of the reverberation mapping mass scale--the complex bar kinematics makes it less than ideally suited for this purpose.Comment: 21 pages, 15 figures.     Identification of a transporter complex responsible for the cytosolic entry of nitrogen-containing bisphosphonates

    Nitrogen-containing-bisphosphonates (N-BPs) are widely prescribed to treat osteoporosis and other bone-related diseases. Although previous studies established that N-BPs function by inhibiting the mevalonate pathway in osteoclasts, the mechanism by which N-BPs enter the cytosol from the extracellular space to reach their molecular target is not understood. Here we implemented a CRISPRi-mediated genome-wide screen and identified SLC37A3 (solute carrier family 37 member A3) as a gene required for the action of N-BPs in mammalian cells. We observed that SLC37A3 forms a complex with ATRAID (all-trans retinoic acid-induced differentiation factor), a previously identified genetic target of N-BPs. SLC37A3 and ATRAID localize to lysosomes and are required for releasing N-BP molecules that have trafficked to lysosomes through fluid-phase endocytosis into the cytosol. Our results elucidate the route by which N-BPs are delivered to their molecular target, addressing a key aspect of the mechanism of action of N-BPs that may have significant clinical relevance

Maternal obesity and physical inactivity have been identified as correlates of overweight and obesity and physical inactivity in older preadolescents; however, no study has explored this relationship in Hispanic preadolescents. Furthermore, the relation between maternal physical activity (PA) and blood pressure (BP) in Hispanic preadolescents has not been examined. Purpose. This study aimed to assess the associations between Hispanic mothers’ PA and body mass index (BMI) and their preadolescents’ PA, screen time, BP, and BMI. Methods. Data of 118 mother-child (aged 2–10 years) dyads enrolled in a crosssectional study of metabolic syndrome in Hispanic preadolescents at a community health center in Johnson City, TN were used.     Trophic Relationships of Demersal Fishes in the Shrimping Zone Off Alvarado Lagoon, Veracruz, Mexico

    The diet of demersal piscivorous fishes captured as bycatch of the commercial shrimping fleet off the Alvarado lagoonal system, Veracruz, Mexico, was studied. Nine collections distributed throughout the nortes (windy), wet, and dry seasons were made from November 1993 to January 1995. Sampling yielded a total of 646 fishes representing 10 families and 14 species, of which 44.9% had empty digestive tracts and were excluded from analysis. Trichiurus lepturus and Synodus foetens were the most abundant demersal predators in the collections. Differences in food consumption of the 7 most abundant predators were observed among the 3 seasons, with the greatest variety of prey (20 species) taken during the nortes season and the lowest variety (9 species) during the dry season. Five distinct trophic guilds were determined based on an index of relative importance of prey. Prey type and location of prey within the water column helped determine guild classification. The occurrence of different trophic guilds may allow for decreased competition for food resources on the continental shelf off Alvarado, Mexico
