Bielefeld: Bielefeld University; 2015.What factors drive adult personality development? Several theoretical perspectives have been offered that propose different sources of influence on personality development, such as genetic and environmental factors, developmental tasks, life events, and peers. From a systems perspective, developmental tasks, life events, and peers constitute more or less external factors of influence on personality development. However, at some point, many of these external influences will be translated by the individual into the form of a deliberate goal to change the self. Exploring this idea, researchers have by now become increasingly interested in the roles of motivation and self-regulation for personality development. Research findings support the assumption that goals to change oneâs personality traits play an important role in shaping current thoughts, feelings, and behaviors, and thus potentially influence even the developmental pattern of personality traits over time. The overall aim of this dissertation is to investigate processes of personality trait development from a micro- and a macro-perspective while focusing on goals as driving mechanisms in intentional personality development. This overall aim was accomplished by implementing an experience-sampling design within the broader timeframe of longitudinal data collection as well as capturing personality and goal characteristics at multiple time-points.
After a general theoretical introduction (chapter 1), the second chapter presents macro-perspective findings on motivational determinants of personality trait development with respect to mean-level changes as well as inter-individual differences in change. The analyses carried out in this dissertation yielded support for the effect on personality trait change both of goals to change oneself and of broader major life goals. These results establish investment in goals at different levels of the definitional hierarchy as underlying processes of personality development. While short-term goals to change oneself exerted influence on short-term changes, major life goals were predictive of more long-term trait changes.
The experience-sampling study presented in the third chapter addresses developmental processes at the micro-level and examines both unique and interaction effects of change goal characteristics on trait-related behavior. It builds on previous research that identified different social role contexts as relevant predictors of trait-related behavior. My results indicate that the importance and perceived feasibility of personal goals functioned as underlying micro-processes that may explain the effects of social roles on trait-related behavior. Findings that close the gap between analyses from the micro- and macro-perspective seem to suggest that there are behavioral investment effects on mean-level changes in corresponding personality traits.
In sum, the research findings from this multiple-time-scale design confirm the meaningful influence of deliberate goals on both variability in personality-related behavior and development of personality traits. Thus, the results point to the relevance of the recently suggested perspective on personality development that highlights self-regulation and motivational factors as important driving forces of personality development.
The fourth chapter will give a critical reflection and theoretical integration of my overall results and indicate profitable areas for future research. The dissertation is rounded off by a general conclusion in the fifth chapter
Zygosity Determination in Twin Studies: a Validation of Zygosity Questionnaires Using DNA in the German TwinLife Study

A correct determination of zygosity is essential for the estimation of heritability and environmental influences on phenotypes. Therefore, the present study examined the validity of two similarity questionnaires used in the German TwinLife study, in which data from 4,097 twin pairs and their families were assessed: twin childrenâs zygosities were determined with the Zygosity Questionnaire for Young Twins, which was administered in parent-report form. For adolescent twins, the Self Report Zygosity Questionnaire was used. For the present validation analyses, DNA samples of N = 328 twin pairs were collected via buccal swabs. In this DNA subsample, questionnaires were filled out by parents for n = 212 (aged 4 to 12 years) twin pairs while self-reports were collected from n = 116 adolescent twins (16 to 23 years of age). Using DNA-based zygosity as criteria, correct classification rates of 97% for parent- and 92% for self-reports were established and cross-validated. Additionally, classification rates based on a single item and variants of questionnaire based zygosity determination used in other twin studies were calculated and compared. Implications of incorrectly classified zygosity on genetic and environmental estimates in twin studies are discussed
Gene set of nuclear-encoded mitochondrial regulators is enriched for common inherited variation in obesity
There are hints of an altered mitochondrial function in obesity. Nuclear-encoded genes are relevant for mitochondrial function (3 gene sets of known relevant pathways: (1) 16 nuclear regulators of mitochondrial genes, (2) 91 genes for oxidative phosphorylation and (3) 966 nuclear-encoded mitochondrial genes). Gene set enrichment analysis (GSEA) showed no association with type 2 diabetes mellitus in these gene sets. Here we performed a GSEA for the same gene sets for obesity. Genome wide association study (GWAS) data from a case-control approach on 453 extremely obese children and adolescents and 435 lean adult controls were used for GSEA. For independent confirmation, we analyzed 705 obesity GWAS trios (extremely obese child and both biological parents) and a population-based GWAS sample (KORA F4, nâ=â1,743). A meta-analysis was performed on all three samples. In each sample, the distribution of significance levels between the respective gene set and those of all genes was compared using the leading-edge-fraction-comparison test (cut-offs between the 50(th) and 95(th) percentile of the set of all gene-wise corrected p-values) as implemented in the MAGENTA software. In the case-control sample, significant enrichment of associations with obesity was observed above the 50(th) percentile for the set of the 16 nuclear regulators of mitochondrial genes (p(GSEA,50)â=â0.0103). This finding was not confirmed in the trios (p(GSEA,50)â=â0.5991), but in KORA (p(GSEA,50)â=â0.0398). The meta-analysis again indicated a trend for enrichment (p(MAGENTA,50)â=â0.1052, p(MAGENTA,75)â=â0.0251). The GSEA revealed that weak association signals for obesity might be enriched in the gene set of 16 nuclear regulators of mitochondrial genes

Development of Immune-Specific Interaction Potentials and Their Application in the Multi-Agent-System VaccImm
Peptide vaccination in cancer therapy is a promising alternative to conventional methods. However, the parameters for this personalized treatment are difficult to access experimentally. In this respect, in silico models can help to narrow down the parameter space or to explain certain phenomena at a systems level. Herein, we develop two empirical interaction potentials specific to B-cell and T-cell receptor complexes and validate their applicability in comparison to a more general potential. The interaction potentials are applied to the model VaccImm which simulates the immune response against solid tumors under peptide vaccination therapy. This multi-agent system is derived from another immune system simulator (C-ImmSim) and now includes a module that enables the amino acid sequence of immune receptors and their ligands to be taken into account. The multi-agent approach is combined with approved methods for prediction of major histocompatibility complex (MHC)-binding peptides and the newly developed interaction potentials. In the analysis, we critically assess the impact of the different modules on the simulation with VaccImm and how they influence each other. In addition, we explore the reasons for failures in inducing an immune response by examining the activation states of the immune cell populations in detail
Exposure to Apoptotic Activated CD4+ T Cells Induces Maturation and APOBEC3G- Mediated Inhibition of HIV-1 Infection in Dendritic Cells
Dendritic cells (DCs) are activated by signaling via pathogen-specific receptors or exposure to inflammatory mediators. Here we show that co-culturing DCs with apoptotic HIV-infected activated CD4+ T cells (ApoInf) or apoptotic uninfected activated CD4+ T cells (ApoAct) induced expression of co-stimulatory molecules and cytokine release. In addition, we measured a reduced HIV infection rate in DCs after co-culture with ApoAct. A prerequisite for reduced HIV infection in DCs was activation of CD4+ T cells before apoptosis induction. DCs exposed to ApoAct or ApoInf secreted MIP-1α, MIP-1ÎČ, MCP-1, and TNF-α; this effect was retained in the presence of exogenous HIV. The ApoAct-mediated induction of co-stimulatory CD86 molecules and reduction of HIV infection in DCs were partially abrogated after blocking TNF-α using monoclonal antibodies. APOBEC3G expression in DCs was increased in co-cultures of DCs and ApoAct but not by apoptotic resting CD4+ T cells (ApoRest). Silencing of APOBEC3G in DC abrogated the HIV inhibitory effect mediated by ApoAct. Sequence analyses of an env region revealed significant induction of G-to-A hypermutations in the context of GG or GA dinucleotides in DNA isolated from DCs exposed to HIV and ApoAct. Thus, ApoAct-mediated DC maturation resulted in induction of APOBEC3G that was important for inhibition of HIV-infection in DCs. These findings underscore the complexity of differential DC responses evoked upon interaction with resting as compared with activated dying cells during HIV infection
Variability and quasi-decadal changes in the methane budget overthe period 2000â2012
Following the recent Global Carbon Project (GCP)
synthesis of the decadal methane (CH4/ budget over 2000â
2012 (Saunois et al., 2016), we analyse here the same dataset
with a focus on quasi-decadal and inter-annual variability in
CH4 emissions. The GCP dataset integrates results from topdown
studies (exploiting atmospheric observations within an
atmospheric inverse-modelling framework) and bottom-up
models (including process-based models for estimating land
surface emissions and atmospheric chemistry), inventories of
anthropogenic emissions, and data-driven approaches.The annual global methane emissions from top-down studies,
which by construction match the observed methane
growth rate within their uncertainties, all show an increase in
total methane emissions over the period 2000â2012, but this
increase is not linear over the 13 years. Despite differences
between individual studies, the mean emission anomaly of the top-down ensemble shows no significant trend in total
methane emissions over the period 2000â2006, during
the plateau of atmospheric methane mole fractions, and also
over the period 2008â2012, during the renewed atmospheric
methane increase. However, the top-down ensemble mean
produces an emission shift between 2006 and 2008, leading
to 22 [16â32] Tg CH4 yr1 higher methane emissions
over the period 2008â2012 compared to 2002â2006. This
emission increase mostly originated from the tropics, with
a smaller contribution from mid-latitudes and no significant
change from boreal regions.
The regional contributions remain uncertain in top-down
studies. Tropical South America and South and East Asia
seem to contribute the most to the emission increase in the
tropics. However, these two regions have only limited atmospheric
measurements and remain therefore poorly constrained.
The sectorial partitioning of this emission increase between
the periods 2002â2006 and 2008â2012 differs from
one atmospheric inversion study to another. However, all topdown
studies suggest smaller changes in fossil fuel emissions
(from oil, gas, and coal industries) compared to the
mean of the bottom-up inventories included in this study.
This difference is partly driven by a smaller emission change
in China from the top-down studies compared to the estimate
in the Emission Database for Global Atmospheric Research
(EDGARv4.2) inventory, which should be revised to smaller
values in a near future. We apply isotopic signatures to the
emission changes estimated for individual studies based on
five emission sectors and find that for six individual top-down
studies (out of eight) the average isotopic signature of the
emission changes is not consistent with the observed change
in atmospheric 13CH4. However, the partitioning in emission
change derived from the ensemble mean is consistent with
this isotopic constraint. At the global scale, the top-down ensemble
mean suggests that the dominant contribution to the
resumed atmospheric CH4 growth after 2006 comes from microbial
sources (more from agriculture and waste sectors than
from natural wetlands), with an uncertain but smaller contribution
from fossil CH4 emissions. In addition, a decrease in
biomass burning emissions (in agreement with the biomass
burning emission databases) makes the balance of sources
consistent with atmospheric 13CH4 observations.
In most of the top-down studies included here, OH concentrations
are considered constant over the years (seasonal variations
but without any inter-annual variability). As a result,
the methane loss (in particular through OH oxidation) varies
mainly through the change in methane concentrations and not
its oxidants. For these reasons, changes in the methane loss
could not be properly investigated in this study, although it
may play a significant role in the recent atmospheric methane

We first perform a large genetic association study by merging all available case-control datasets and by-proxy study results (discovery nâ=â409,435 and validation size nâ=â58,190). Here, we add six variants associated with Alzheimer's disease risk (near APP, CHRNE, PRKD3/NDUFAF7, PLCG2 and two exonic variants in the SHARPIN gene). Assessment of the polygenic risk score and stratifying by APOE reveal a 4 to 5.5 years difference in median age at onset of Alzheimer's disease patients in APOE É4 carriers. Because of this study, the underlying mechanisms of APP can be studied to refine the amyloid cascade and the polygenic risk score provides a tool to select individuals at high risk of Alzheimer's disease
