    ERP effects of subject-verb agreement violations in patients with Broca's aphasia

    Contains fulltext : 13805.pdf (publisher's version ) (Open Access)This article presents electrophysiological data on on-line syntactic processing during auditory sentence comprehension in patients with Broca's aphasia. Event-related brain potentials (ERPs) were recorded from the scalp while subjects listened to sentences that were either syntactically correct or contained violations of subject-verb agreement. Three groups of subjects were tested: Broca patients (n = 10), nonaphasic patients with a right-hemisphere (RH) lesion (n = 5), and healthy agedmatched controls (n = 12). The healthy, control subjects showed a P600/SPS effect as response to the agreement violations. The nonaphasic patients with an RH lesion showed essentially the same pattern. The overall group of Broca patients did not show this sensitivity. However, the sensitivity was modulated by the severity of the syntactic comprehension impairment. The largest deviation from the standard P600/SPS effect was found in the patients with the relatively more severe syntactic comprehension impairment. In addition, ERPs to tones in a classical tone oddball paradigm were also recorded. Similar to the normal control subjects and RH patients, the group of Broca patients showed a P300 effect in the tone oddball condition. This indicates that aphasia in itself does not lead to a general reduction in all cognitive ERP effects. It was concluded that deviations from the standard P600/SPS effect in the Broca patients reflected difficulties with on-line maintaining of number information across clausal boundaries for establishing subject-verb agreement

    Novel application of multi-stimuli network inference to synovial fibroblasts of rheumatoid arthritis patients

    BACKGROUND: Network inference of gene expression data is an important challenge in systems biology. Novel algorithms may provide more detailed gene regulatory networks (GRN) for complex, chronic inflammatory diseases such as rheumatoid arthritis (RA), in which activated synovial fibroblasts (SFBs) play a major role. Since the detailed mechanisms underlying this activation are still unclear, simultaneous investigation of multi-stimuli activation of SFBs offers the possibility to elucidate the regulatory effects of multiple mediators and to gain new insights into disease pathogenesis. METHODS: A GRN was therefore inferred from RA-SFBs treated with 4 different stimuli (IL-1 β, TNF- α, TGF- β, and PDGF-D). Data from time series microarray experiments (0, 1, 2, 4, 12 h; Affymetrix HG-U133 Plus 2.0) were batch-corrected applying ‘ComBat’, analyzed for differentially expressed genes over time with ‘Limma’, and used for the inference of a robust GRN with NetGenerator V2.0, a heuristic ordinary differential equation-based method with soft integration of prior knowledge. RESULTS: Using all genes differentially expressed over time in RA-SFBs for any stimulus, and selecting the genes belonging to the most significant gene ontology (GO) term, i.e., ‘cartilage development’, a dynamic, robust, moderately complex multi-stimuli GRN was generated with 24 genes and 57 edges in total, 31 of which were gene-to-gene edges. Prior literature-based knowledge derived from Pathway Studio or manual searches was reflected in the final network by 25/57 confirmed edges (44%). The model contained known network motifs crucial for dynamic cellular behavior, e.g., cross-talk among pathways, positive feed-back loops, and positive feed-forward motifs (including suppression of the transcriptional repressor OSR2 by all 4 stimuli. CONCLUSION: A multi-stimuli GRN highly concordant with literature data was successfully generated by network inference from the gene expression of stimulated RA-SFBs. The GRN showed high reliability, since 10 predicted edges were independently validated by literature findings post network inference. The selected GO term ‘cartilage development’ contained a number of differentiation markers, growth factors, and transcription factors with potential relevance for RA. Finally, the model provided new insight into the response of RA-SFBs to multiple stimuli implicated in the pathogenesis of RA, in particular to the ‘novel’ potent growth factor PDGF-D

    Observational Constraints on the Catastrophic Disruption Rate of Small Main Belt Asteroids

    We have calculated 90% confidence limits on the steady-state rate of catastrophic disruptions of main belt asteroids in terms of the absolute magnitude at which one catastrophic disruption occurs per year (HCL) as a function of the post-disruption increase in brightness (delta m) and subsequent brightness decay rate (tau). The confidence limits were calculated using the brightest unknown main belt asteroid (V = 18.5) detected with the Pan-STARRS1 (Pan-STARRS1) telescope. We measured the Pan-STARRS1's catastrophic disruption detection efficiency over a 453-day interval using the Pan-STARRS moving object processing system (MOPS) and a simple model for the catastrophic disruption event's photometric behavior in a small aperture centered on the catastrophic disruption event. Our simplistic catastrophic disruption model suggests that delta m = 20 mag and 0.01 mag d-1 < tau < 0.1 mag d-1 which would imply that H0 = 28 -- strongly inconsistent with H0,B2005 = 23.26 +/- 0.02 predicted by Bottke et al. (2005) using purely collisional models. We postulate that the solution to the discrepancy is that > 99% of main belt catastrophic disruptions in the size range to which this study was sensitive (100 m) are not impact-generated, but are instead due to fainter rotational breakups, of which the recent discoveries of disrupted asteroids P/2013 P5 and P/2013 R3 are probable examples. We estimate that current and upcoming asteroid surveys may discover up to 10 catastrophic disruptions/year brighter than V = 18.5.Comment: 61 Pages, 10 Figures, 3 Table

    Lightcurves of Type Ia Supernovae from Near the Time of Explosion

    We present a set of 11 type Ia supernova (SN Ia) lightcurves with dense, pre-maximum sampling. These supernovae (SNe), in galaxies behind the Large Magellanic Cloud (LMC), were discovered by the SuperMACHO survey. The SNe span a redshift range of z = 0.11 - 0.35. Our lightcurves contain some of the earliest pre-maximum observations of SNe Ia to date. We also give a functional model that describes the SN Ia lightcurve shape (in our VR-band). Our function uses the "expanding fireball" model of Goldhaber et al. (1998) to describe the rising lightcurve immediately after explosion but constrains it to smoothly join the remainder of the lightcurve. We fit this model to a composite observed VR-band lightcurve of three SNe between redshifts of 0.135 to 0.165. These SNe have not been K-corrected or adjusted to account for reddening. In this redshift range, the observed VR-band most closely matches the rest frame V-band. Using the best fit to our functional description of the lightcurve, we find the time between explosion and observed VR-band maximum to be 17.6+-1.3(stat)+-0.07(sys) rest-frame days for a SN Ia with a VR-band Delta m_{-10} of 0.52mag.
