High-resolution stable isotope stratigraphy of the upper Cambrian and Ordovician in the Argentine Precordillera: Carbon isotope excursions and correlations
We report the occurrence of important carbon isotope excursions in early Paleozoic formations of the Eastern and Central Argentine Precordillera. The Steptoean positive isotope carbon excursion (SPICE) is known from North America, Kazakhstan, South China, Australia and South America, and the negative isotope carbon excursion (SNICE) has been described for the first time in South America. We report here the record of the SPICE and SNICE in a single section in the Eastern Precordillera. In the Central Precordillera, a minor middle Darriwilian positive carbon isotope excursion (MDICE) and a late Sandbian positive isotope carbon excursion, the GICE (~ + 3‰; C. bicornis biozone) are reported from two sections. One pre-GICE positive carbon-isotope excursion (Sandbian Sa1, N. gracilis biozone) in the Central Precordillera with a δ13C peak of ~ + 2‰ is, perhaps, equivalent to the positive Spechts Ferry δ13C excursion of North America. A positive δ13C excursion (~ 6‰; N. persculptus biozone) recorded at the base of the late Hirnantian La Chilca Formation probably corresponds to HICE. These carbon-isotope excursions resulted from more than one factor where paleoceanographic events probably played a major role: (a) sea-level fall and vigorous fluctuations in the Steptoean (SPICE), (b) sea-level rise in the Sunwaptan (SNICE), (c) important transgression in the Sandbian (pre-GICE and GICE), and (d) sea-level fall in the late Hirnantian (HICE). In the Darriwilian and Sandbian stages, organic burial has led to a large 12C sequestration in the deep anoxic ocean with saline bottom water, recorded by the graptoliferous black shales of the Gualcamayo and Los Azules formations in Central Precordillera, helped the building of the MDICE and GICE anomalies. δ18O values for the Upper Cambrian carbonates are likely near-primary isotope signals that point to progressive cooling from the SPICE to the SNICE, whereas for Sandbian carbonates they suggest strong temperature fluctuations. The δ13C peak of the GICE coincides with a cooler period with temperatures warming up towards the late Hirnantian to Rhuddanian. The Upper Cambrian to the Middle Ordovician of the Precordillera recorded a decrease of seawater 87Sr/86Sr ratios in accordance to the global picture. This decrease probably reflects the influence of widespread volcanic activity from arc terranes in low-latitude settings along eastern margins of Laurentia and in the Argentine Precordillera in the Early to Middle Ordovician. Five carbonate samples (from San Juan, Gualcamayo and Los Azules formations) yielded εNd values that along seven already published values seem to plot along the Nd isotopic evolution trend of the Iapetus Ocean. Together, the record of global Upper Cambrian and Ordovician carbon-isotope excursions in the Argentine Precordillera is a valuable proxy in refining Early Paleozoic stratigraphy, establishing of regional/global high-resolution correlations, and sea-level change history in South America.Fil: Sial, A. A straightforward multiallelic significance test for the Hardy-Weinberg equilibrium law
Much forensic inference based upon DNA evidence is made assuming Hardy-Weinberg Equilibrium (HWE) for the genetic loci being used. Several statistical tests to detect and measure deviation from HWE have been devised, and their limitations become more obvious when testing for deviation within multiallelic DNA loci. The most popular methods-Chi-square and Likelihood-ratio tests-are based on asymptotic results and cannot guarantee a good performance in the presence of low frequency genotypes. Since the parameter space dimension increases at a quadratic rate on the number of alleles, some authors suggest applying sequential methods, where the multiallelic case is reformulated as a sequence of “biallelic” tests. However, in this approach it is not obvious how to assess the general evidence of the original hypothesis; nor is it clear how to establish the significance level for its acceptance/rejection. In this work, we introduce a straightforward method for the multiallelic HWE test, which overcomes the aforementioned issues of sequential methods. The core theory for the proposed method is given by the Full Bayesian Significance Test (FBST), an intuitive Bayesian approach which does not assign positive probabilities to zero measure sets when testing sharp hypotheses. We compare FBST performance to Chi-square, Likelihood-ratio and Markov chain tests, in three numerical experiments. The results suggest that FBST is a robust and high performance method for the HWE test, even in the presence of several alleles and small sample sizes
Characterization and pattern recognition of color images of dermatological ulcers: a pilot study
We present color image processing methods for the char\-ac\-te\-ri\-za\-tion of images of dermatological lesions for the purpose of content-based image retrieval (CBIR) and computer-aided di\-ag\-no\-sis. The intended application is to segment the images and perform classification and analysis of the tissue composition of skin lesions or ulcers, in terms of granulation (red), fibrin (yel\-low), necrotic (black), callous (white), and mixed tissue composition. The images were analyzed and classified by an expert dermatologist following the red-yellow-black-white model. Automatic segmentation was performed by means of clustering using Gauss\-ian mixture modeling, and its performance was evaluated by deriving the Jaccard coefficient between the automatically and manually segmented images. Statistical texture features were derived from cooccurrence matrices of RGB, HSI, Lab, and Luv color components. A retrieval engine was implemented using the k-nearest-neighbor classifier and the Euclidean, Man\-hat\-tan, and Chebyshev distance metrics. Classification was performed by means of a metaclassifier using logistic regression. The average Jaccard coefficient after the segmentation step between the automatically and manually segmented images was 0.560, with a standard deviation of 0.220. The performance in CBIR was mea\-sured in terms of precision of retrieval, with average values of up to 0.617 obtained with the Chebyshev distance. The metaclassifier yielded an average area under the receiver operating char\-ac\-ter\-is\-tic curve of 0.772
Comparative highly efficient production of β-glucan by Lasiodiplodia theobromae CCT 3966 and its multiscale characterization
Lasiodiplodan, a (1→6)-β-d-glucan, is an exopolysaccharide with high commercial value and many applications in food, pharmaceuticals, and cosmetics. Current industrial production of β-glucans from crops is mostly by chemical routes generating hazardous and toxic waste. Therefore, alternative sustainable and eco-friendly pathways are highly desirable. Here, we have studied the lasiodiplodan production from sugarcane bagasse (SCB), a major lignocellulosic agricultural residue, by Lasiodiplodia theobromae CCT 3966. Lasiodiplodan accumulated on SCB hydrolysate (carbon source) supplemented with soybean bran or rice bran (nitrogen source) was 16.2 [6.8 × 103 Da] and 22.0 [7.6 × 103 Da] g/L, respectively. Lasiodiplodan showed high purity, low solubility, pseudoplastic behavior and was composed of glucose units. Moreover, the exopolysaccharides were substantially amorphous with moderate thermal stability and similar degradation temperatures. To our knowledge, this is the first report on the highest production of SCB-based lasiodiplodan to date. L. theobromae, as a microbial cell factory, demonstrated the commercial potential for the sustainable production of lasiodiplodan from renewable biomass feedstock
Systematic comparison of the effects of Alpha-synuclein mutations on its oligomerization and aggregation

Furthermore, mutations in the gene encoding for ASYN are associated with familial and sporadic forms of PD, suggesting this protein plays a central role in the disease. However, the precise contribution of ASYN to neuronal dysfunction and death is unclear. There is intense debate about the nature of the toxic species of ASYN and little is known about the molecular determinants of oligomerization and aggregation of ASYN in the cell. In order to clarify the effects of different mutations on the propensity of ASYN to oligomerize and aggregate, we assembled a panel of 19 ASYN variants and compared their behaviour. We found that familial mutants linked to PD (A30P, E46K, H50Q, G51D and A53T) exhibited identical propensities to oligomerize in living cells, but had distinct abilities to form inclusions. While the A30P mutant reduced the percentage of cells with inclusions, the E46K mutant had the opposite effect. Interestingly, artificial proline mutants designed to interfere with the helical structure of the N-terminal domain, showed increased propensity to form oligomeric species rather than inclusions. Moreover, lysine substitution mutants increased oligomerization and altered the pattern of aggregation. Altogether, our data shed light into the molecular effects of ASYN mutations in a cellular context, and established a common ground for the study of genetic and pharmacological modulators of the aggregation process, opening new perspectives for therapeutic intervention in PD and other synucleinopathies.This work was supported by the DFG Center for Nanoscale Microscopy and Molecular Physiology of the Brain (CNMPB).info:eu-repo/semantics/publishedVersio
Predictors of rescue percutaneous coronary intervention after pharmacoinvasive strategy in women
AbstractBackgroundPharmacoinvasive therapy (PIT) is feasible in patients with acute myocardial infarction with ST-segment elevation (STEMI) when timely primary percutaneous coronary intervention (PCI) is unavailable. In this study, we compared women who underwent successful reperfusion PIT with those who required rescue PCI, to identify potential predictors of thrombolytic failure.MethodsFrom January 2010 to November 2014, 327 consecutive women with STEMI were referred to a tertiary hospital, 206 after successful thrombolysis (63%) and 121 who required rescue PCI. The groups were compared regarding demographic, clinical and angiographic outcomes, and clinical (TIMI, GRACE, and ZWOLLE CADILLAC) and bleeding (CRUSADE) risk scores. A multivariate logistic regression model was used to identify predictors of thrombolytic failure.ResultsThere was no significant difference between the demographic characteristics or the medical history of the groups. Rescue PCI group had significantly higher values of the evaluated scores. Clinical hospital complications and mortality (2.5% vs. 22.0%; p < 0.0001) were more frequent in rescue PCI group. The independent variables associated with rescue PCI were pain-to-needle time > 3h (OR: 3.07, 95%CI: 1.64 to 5.75; p < 0.0001), ZWOLLE score (OR: 1.25; 95%CI: 1.14 to 1.37; p = 0.0001) and creatinine clearance (OR: 1.009, 95%CI: 1.0 to 1.02; p = 0.04).ConclusionsWomen with STEMI who underwent PIT and who required rescue PCI had significantly higher mortality compared to those who achieved initial success of PIT with elective PCI. Pain-to-needle time > 3h, ZWOLLE score and creatinine clearance were independent predictors of the need for rescue PCI
Characterization of street food consumption in palermo: possible effects on health
<p>Abstract</p> <p>Background</p> <p>Street Food (SF) consists of out-of-home food consumption and has old, historical roots with complex social-economic and cultural implications. Despite the emergence of modern fast food, traditional SF persists worldwide, but the relationship of SF consumption with overall health, well-being, and obesity is unknown.</p> <p>Methods</p> <p>This is an observational, cross-sectional study. The study was performed in Palermo, the largest town of Sicily, Italy. Two groups were identified: consumers of SF (n = 687) and conventional restaurant food (RES) consumers (n = 315). Study subjects answered a questionnaire concerning their health conditions, nutritional preferences, frequency of consumption of SF and a score relative to SF consumption ranging from 0 to 20 was calculated.</p> <p>Results</p> <p>Body mass index (BMI, kg/m<sup>2</sup>) was significantly and independently correlated with the score of street food consumption (r = 0,103; p < 0.002). The prevalence of different diseases, including hypertension and type 2 diabetes, and the use of medications did not differ between the two groups. Milza (a sandwich stuffed with thin slice of bovine spleen and lung) consumers had a significantly higher prevalence of hypertension (12.2% vs 6.2% in non consumers; p < 0.005) and in this subgroup the use of anti-hypertensive drugs was inversely correlated with the frequency of milza consumption (r = 0.11; P = 0.010).</p> <p>Conclusions</p> <p>This study suggests that SF consumption in Palermo is associated with a higher BMI and higher prevalence of hypertension in milza consumers. Further studies should evaluate whether frequent SF consumers have unfavourable metabolic and cardiovascular profile.</p
