    Assessment of the anti-Listerial properties of Garcinia kola (Heckel) seeds

    A follow-up of traditional medicinal plants uses is an important tool in highlighting their therapeutic potentials, as they have been found to be a source of a wide range of bioactive compounds that can be used as base compounds for new pharmaceutical drugs. This study therefore focuses on assessing the anti-Listerial properties of the seeds of Garcinia kola (Heckel) plant, which is a traditional medicinal plant of west and central African origin, and was and is still used to traditionally treat several ailments. Four different solvents crude extracts of the seeds were assessed for their anti-Listerial activities in-vitro, against a panel of 42 Listeria bacteria, which included Listeria monocytogenes, Listeria ivanovii and Listeria grayi species. At 10 mg/ml concentration the aqueous extract had activity against 29% of the test isolates while the other three crude extracts namely dichloromethane, n-hexane and the methanol extracts had activity against 45% of the test bacteria. The minimum inhibitory concentration (MIC) ranges of the extracts were 0.079-0.313 mg/ml for the dichloromethane extract; 0.079-0.625 mg/ml for the n-hexane extract; 0.157-0.625 mg/ml for the methanol extract; and 10->10 mg/ml for the aqueous extract. The minimum bactericidal concentration (MBC) ranges of the extracts were 0.625–10 mg/ml for both the n-hexane and the dichloromethane extract; 5-10 mg/ml for the methanol extract; and those for the aqueous extract were above 10 mg/ml against all the susceptible Listeria isolates. The rate of kill analysis was then determined for the three most active crude extracts that is excluding the aqueous extract and it was assessed against four representative Listeria species namely L. monocytogenes (LAL 8), L. grayi (LAL 15), L. ivanovii (LEL 30) and L. ivanovii (LEL 18). All the three extracts showed a general trend of being concentration and time dependent in their rate of kill profiles such that most bacteria cells were killed at the highest test concentration of 4× MIC value after the maximum exposure time of 2 h. The n-hexane, dichloromethane and methanol extracts were bactericidal against 4, 3 and 1 isolates out of the four test Listeria isolates respectively

    The n-Hexane, dichloromethane, methanol and aqueous crude extracts of Garcinia kola (Heckel) seeds were screened for their anti-Vibrio activities against 50 Vibrio bacteria isolated from wastewater final effluents. The 50 isolates consisted of different Vibrio species namely V. fluvialis (14), V. vulnificus (12), V. parahaemolyticus (12), V. metschnikovii (3) and 9 others unidentified to the specie level. The n-Hexane, dichloromethane and methanol extracts had activities against 16 (32 percent) of the Vibrio isolates, while the aqueous extracts had activities against 12 (24 percent) all at a screening concentration of 10 mg/ml. The minimum inhibitory concentrations (MICs) were 0.313-0.625 mg/ml, 0.313-0.625 mg/ml, 0.313-2.5 mg/ml and 10 mg/ml for n-Hexane, dichloromethane, methanol and aqueous extracts respectively. Rate of kill studies were carried out against three different Vibrio species namely V. vulnificus (AL042), V. parahaemolyticus (AL049) and V. fluvialis ( AL040) using the n-Hexane, dichloromethane and methanol extracts at 1× to 4 × MICs and 2 hour exposure. About 96.3 percent, 82.2 percent, and 78.1 percent (V. fluvialis AL040); 92.6 percent, 87.8 percent and 68.9 percent (V. parahaemolyticus AL049); and 91.6 percent, 64.4 percent, 60 percent (V. vulnificus AL042) of the bacteria were killed by the crude n-Hexane, dichloromethane and methanol extracts respectively after 2 hour exposure time at 4× MIC. The patterns of activity were bacteriostatic, with the n-Hexane extracts being most effective in activity. We conclude that the Garcinia kola seeds have promise in the treatment and management of infections caused by Vibrio species

    Differentiation in higher education : a case study of Lesotho

    Differentiation has become a crucial policy driver in higher education systems that attempts to respond to global pressures for a highly skilled labour force, employ new technologies and adapt to unpredictable or volatile global product markets and rapid technological change. In order to be globally competitive, nations have to place knowledge production, accumulation, transfer and application at the centre of their national development strategies. This study investigates the case of Lesotho, a relatively small African country with a colonial past that has left a lasting imprint, both enabling and constraining, on many aspects of its education system. The aim of this explanatory case study was to identify differentiating trends within Lesotho's higher education system, focusing on two institutional types: the polytechnic and the university. These institutions were investigated at both institutional and programme level. The research question was "How differentiated is the polytechnic-university binary division in the public sector higher education system in Lesotho?" The sample for the study comprised two schools and two faculties at the polytechnic and the university respectively. One programme under each school or faculty was investigated

    Identification of the Anti Listerial

    Partially purified fractions of the n-hexane extract of Garcinia kola seeds were obtained through column chromatography and their constituents were identified through the use of gas chromatography coupled to mass spectrometry (GC-MS). Three fractions were obtained by elution with benzene as the mobile phase and silica gel 60 as the stationery phase and these were named Benz1, Benz2, and Benz3 in the order of their elution. The antiListerial activities of these fractions were assessed through MIC determination and only Benz2 and Benz3 were found to be active with MIC’s ranging from 0.625 to 2.5 mg/mL. The results of the GC-MS analysis showed Benz2 to have 9 compounds whilst Benz3 had 7 compounds, with the major compounds in both fractions being 9,19-Cyclolanost-24-en-3-ol, (3.β.) and 9,19-Cyclolanostan-3-ol,24-methylene-, (3.β.). The Benz2 fraction was found to have mainly indifferent interactions with ampicillin and penicillin G whilst mainly additive interactions were observed with ciprofloxacin. The Benz3 fraction’s interactions were found to be 50% synergistic with penicillin G and 25% synergistic with ciprofloxacin and ampicillin. A commercially available 9,19-Cyclolanost-24-en-3-ol, (3.β.) was found not to exhibit any antiListerial activities at maximum test concentrations of 5 mg/mL, suggesting that the compound could be acting in synergy with the other compounds in the eluted fractions of Garcinia kola seeds

    The bacteria profiles of wounds in diabetic patients hospitalized in northern Kwazulu-Natal, South Africa

    Diabetic wound infections still remain a health concern such that correct identification of bacteria is essential in monitoring the spread of the infections as well as in the administration of the correct treatment. This study therefore focuses on isolating and identifying bacteria present in diabetic wounds of hospitalized patients in northern KwaZulu-Natal and assessing their distribution.The wound specimen were collected and swabbed onto selective and differential media. The bacteria identities were presumptively ascertained through biochemical characterization (Gram-stain, catalase test, oxidase test and API) and then confirmed through 16S rDNA sequencing.A total of 42 isolates were recovered from 83% of the patients sampled from the three participating hospitals (X, Y, and Z). Gram-negative bacilli from Enterobacteriaceaewere predominant followed by Staphylococci spp and Enterococcus faecaliswith 43% polymicrobial cases from hospital Z and 29% from hospital X. Distribution of some opportunistic pathogens and nosocomially-acquired pathogens were also observed across the patients with five bacterial identities distributed among hospital X and Z. The adverse effects associated with the recovered bacteria in diabetic wounds pose a serious health concern and preventive measure should be taken.Keywords: Diabetes mellitus, wounds, bacteria, infectio

    In Vitro Anti-Listerial Activities of Crude n-Hexane and Aqueous Extracts of Garcinia kola (heckel) Seeds

    We assessed the anti-Listerial activities of crude n-hexane and aqueous extracts of Garcinia kola seeds against a panel of 42 Listeria isolates previously isolated from wastewater effluents in the Eastern Cape Province of South Africa and belonging to Listeria monocytogenes, Listeria grayi and Listeria ivanovii species. The n-hexane fraction was active against 45% of the test bacteria with zones of inhibition ranging between 8–17 mm, while the aqueous fraction was active against 29% with zones of inhibition ranging between 8–11 mm. The minimum inhibitory concentrations (MIC) were within the ranges of 0.079–0.625 mg/mL for the n-hexane extract and 10 to >10 mg/mL for the aqueous extract. The rate of kill experiment carried out for the n-hexane extract only, revealed complete elimination of the initial bacterial population for L. grayi (LAL 15) at 3× and 4× MIC after 90 and 60 min; L. monocytogenes (LAL 8) at 3× and 4× MIC after 60 and 15 min; L. ivanovii (LEL 18) at 3× and 4× MIC after 120 and 15 min; L. ivanovii (LEL 30) at 2, 3 and 4× MIC values after 105, 90 and 15 min exposure time respectively. The rate of kill activities were time- and concentration-dependant and the extract proved to be bactericidal as it achieved a more than 3log10 decrease in viable cell counts after 2 h exposure time for all of the four test organisms at 3× and 4× MIC values. The results therefore show the potential presence of anti-Listerial compounds in Garcinia kola seeds that can be exploited in effective anti-Listerial chemotherapy

    Effect of Moringa oleifera Lam. leaf powder on the pharmacokinetics of nevirapine in HIV-infected adults: a one sequence cross-over study

    Moringa oleifera Lam., an herb commonly consumed by HIV-infected people on antiretroviral therapy, inhibits cytochrome P450 3A4, 1A2 and 2D6 activity in vitro; and may alter the pharmacokinetics (PK) of antiretroviral drugs metabolized via the same pathways. However, in vitro drug interaction activity may not translate to a clinically significant effect. Therefore, the effect of moringa leaf powder on the PK of nevirapine in HIV-infected people was investigated. Adult patients at steady-state dosing with nevirapine were admitted for 12-h intensive PK sampling following a 21-day herbal medicine washout. Blood sampling was repeated after 14 days of nevirapine and moringa (1.85 g leaf powder/day) co-administration. Nevirapine plasma concentrations were determined by liquid chromatography-tandem mass spectrometry. To assess the effect of moringa on nevirapine PK, the change in nevirapine area under the plasma concentration–time curve (AUC) was determined. The mean difference in pre- and post-moringa nevirapine, maximum concentration (Cmax) and concentration at 12 h (C12h) were also calculated. The PK parameters were compared by assessing the post/pre geometric mean ratios (GMRs) and associated 90% confidence intervals (CIs). Pharmacokinetics analyses were performed on the results from 11 participants for whom complete data were obtained. The post/pre GMRs and associated 90% CIs for nevirapine were 1.07 (1.00–1.14) for the AUC; 1.06 (0.98–1.16) for Cmax and 1.03 (0.92–1.16) for C12h. Co-administration of Moringa oleifera Lam. leaf powder at the traditional dose did not significantly alter the steady-state PK of nevirapine.     THE ANTIBACTERIAL ACTIVITIES OF SOME PLANT DERIVED TRITERPENES

    Background: The increase in the prevalence of multi-drug resistant bacteria has necessitated the search for new antimicrobials from alternative sources such as traditional medicinal plants. Materials and Methods: The agar well diffusion method was employed to determine the susceptibilities of four plant derived triterpenes namely, 3β-hydroxylanosta-9, 24-dien-21-oic acid (RA5), and methyl-3β-hydroxylanosta-9, 24-dien-21oate (RA3), a mixture of oleanolic acid and betulinic acid (SF1) and a mixture of 3β-acetonyloleanolic acid and 3β-acetonylbetulinic acid (SF2), at a concentration of 10 mg/ml against seven Escherichia coli, one Bacillus cereus, five Enterococcus and nine Vibrio bacteria. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were determined through the micro-broth dilution assay. The checkerboard method was used to determine the antibiotictriterpene interactions while the cytosolic lactate dehydrogenase test was used to determine the membrane damaging potentials of the triterpenes in comparison to 3% Triton X-100. Results: The triterpenes RA3, RA5, SF1 and SF2 had activities against 86.4%, 54.6%, 22.7% and 9.09% of the test bacteria respectively. SF1 had the lowest MIC values ranging 0.625-10 mg/ml with lower MIC values being noted against Gram negative bacteria in comparison to Gram positive bacteria; this trend was also noted among the activities of RA3 and RA5 although they had higher MIC value ranges of 1.25-10 mg/ml and 5- 10 mg/ml respectively. MBC studies proved the triterpenes to be mostly bacteriostatic. The interaction studies with ciprofloxacin were mainly ranging between indifference and antagonism. RA3 alone showed minimal membrane damaging potential with the levels of cytosolic lactate dehydrogenase released ranging from 1-36% in comparison to 3%Triton X-100 against E.coli (DSM-8695) and V. vulnificus (AL 042). Conclusion: The results hereby show the potential that the test triterpenes have as antibacterial agents, especially against the Gram negative bacteria namely E. coli and Vibrio bacteri

    The experiences of expatriate mothers regarding pregnancy, childbirth and motherhood in the host city Cape Town, South Africa

    The study explored the experiences of expatriate mothers' pregnancy, childbirth and motherhood in a host city. This included identifying how expatriate mothers access public healthcare, and adapt to motherhood in a host country in a subsequent pregnancy. These expatriate mothers had previously experienced childbirth in their country of origin. A descriptive qualitative approach was utilised as it describes individuals' lived experiences. Purposive sampling was chosen and I had an in-depth conversation with nine expatriate mothers. Thematic analysis was used to identify themes and sub-themes. Findings: The participants established that antenatal care was easily accessible. Mothers were confronted with a lack of respectful care in labour wards. They also had personal challenges. This all led to the development of the four themes. These are: expatriate mothers' need for support; organised antenatal healthcare; high cost of living; as well as labour and childbirth challenges in Cape Town (public healthcare). These revealed mothers need support to manage motherhood in a different setting. Further, the expatriate mothers rely on their husbands as their main support in the host city. Recommendations: An emphasis on teaching of respectful maternity care in midwifery is needed, the availability of more and highly skilled and caring midwives and the need for support groups for expatriate mothers
