    Synergistic effects of glyphosate- and 2,4-D-based pesticides mixtures on Rhinella arenarum larvae

    Glyphosate and 2,4-D are two herbicides commonly used together. Since there is little information about the interactions between these pesticides, the aim of this study was to evaluate the single and joint lethal toxicity of the glyphosate-based herbicide (GBH) ATANOR® (43.8% of glyphosate, isopropylamine salt) and the 2,4-D-based herbicide (2,4-DBH) Así Max 50® (602000 mg/L of 2,4-D) on Rhinella arenarum larvae. Equitoxic and non-equitoxic mixtures were prepared according to the recommendation for their combination and analyzed with a fixed ratio design at different exposure times and levels of lethality (LC10, LC50, and LC90). GBH (504h-LC50=38.67 mg ae/L) was significantly more toxic than 2,4-DBH (504h-LC50=250.31 mg ae/L) and their toxicity was time-dependent. At 48h, the equitoxic mixture toxicity was additive and from the 96h was antagonistic at LC10 and LC50 effect level. The non-equitoxic mixture toxicity was additive at LC10 effect level from the 48h to the 168h, and synergistic from the 240h. At LC50 and LC90 effect level, the mixture interaction resulted synergistic for all exposure times. This is the first study to report the synergistic interactions between GBH and 2,4-DBH on amphibians, alerting about its negative impact on aquatic ecosystems.Fil: Peluso, Julieta.     Very early rheumatoid arthritis is the major predictor of major outcomes: clinical ACR remission and radiographic non-progression

    OBJECTIVES: To identify predictors of clinical remission as well as of no x-ray progression in a cohort of early rheumatoid arthritis (ERA) treated with a tight-control protocol. METHODS: A total of 121 consecutive patients with ERA were treated to reach European League Against Rheumatism (EULAR) and/or American College of Rheumatology (ACR) clinical remission with methotrexate (MTX) for 3 months, then with a combination with anti-tumour necrosis factor if the patient did not achieve a 44-joint Disease Activity Score (DAS44) ≤2.4. At baseline and after 12 months all the patients had hand and foot joint radiographs. Very early rheumatoid arthritis (VERA) was defined as a disease with symptoms of less than 12 weeks. RESULTS: In all, 46.3% of the patients reached DAS remission and 24.8% achieved ACR remission. More than 60% of patients reached remission with MTX. Male sex and an erythrocyte sedimentation rate <35 mm/h at onset arose as significant predictors of EULAR remission, while VERA disease was the only predictor of ACR remission. At baseline, 28.1% of the patients were erosive. Multivariate analysis demonstrated that the only independent predictor of erosiveness was 'not having VERA disease'. After 12 months, VERA was the only factor predicting a lack of new erosions. CONCLUSIONS: VERA represents the best therapeutic opportunity in clinical practice to achieve a complete remission and to stop the erosive course of rheumatoid arthritis

    Body composition and metabolic status of Italian and Spanish university students: relationship with fruit and vegetable consumption

    Most university students do not follow recommendations for fruit and vegetable intake, with a consequent increase in the prevalence of cardiovascular disease (CVD) risk factors. The aim of this study was to compare obesity prevalence and biomarkers of metabolic status between Italian and Spanish university students, in relation with the consumption of fruits and vegetables. Food consumption, adherence to a Mediterranean diet (MD), level of physical activity (PA), blood glucose, total cholesterol, triglycerides and ketones, blood pressure, and body composition were evaluated. Among CVD risk factors, only glucose was significantly higher in Spaniards (SP), and only 3.1% of SP presented ketosis. SP had a higher percentage of energy from fat. Although adherence to MD and fruit and vegetable consumption did not differ between Italians and SP, students who consumed at least four servings of fruit and vegetables (FV group) showed better values for pressure and metabolic parameters than the no FV group. We observed an association between consumption of fruit and PA. Students who consumed more vegetables than fruit reported a better body composition profile and lower glucose concentrations. As previously suggested, in addition to PA, two servings of fruit and three servings of vegetables per day should be recommended

    LPS-induced IL-8 activation in human intestinal epithelial cells is accompanied by specific histone H3 acetylation and methylation changes

    <p>Abstract</p> <p>Background</p> <p>The release of LPS by bacteria stimulates both immune and specific epithelial cell types to release inflammatory mediators. It is known that LPS induces the release of IL-8 by intestinal mucosal cells. Because it is now emerging that bacteria may induce alteration of epigenetic patterns in host cells, we have investigated whether LPS-induced IL-8 activation in human intestinal epithelial cells involves changes of histone modifications and/or DNA methylation at IL-8 gene regulatory region.</p> <p>Results</p> <p>RT-PCR analysis showed that IL-8 mRNA levels rapidly increase after exposure of HT-29 cells to LPS. DNA demethylating agents had no effects on IL-8 expression, suggesting that DNA methylation was not involved in IL-8 gene regulation. Consistently we found that 5 CpG sites located around IL-8 transcription start site (-83, -7, +73, +119, +191) were unmethylated on both lower and upper strand either in LPS treated or in untreated HT-29 cells, as well as in normal intestinal mucosa.</p> <p>Conversely, pretreatment of HT-29 cells with deacetylase inhibitors strengthened the LPS-mediated IL-8 activation. Inhibitors of histone deacetylases could induce IL-8 mRNA expression also in the absence of LPS, suggesting that chromatin modifications could be involved in IL-8 gene regulation. Chromatin immunoprecipitation analyses showed that, concurrently with IL-8 activation, transient specific changes in H3 acetylation and H3K4, H3K9 and H3K27 methylation occurred at IL-8 gene promoter during LPS stimulation. Changes of H3-acetyl, H3K4me2 and H3K9me2 levels occurred early, transiently and corresponded to transcriptional activity, while changes of H3K27me3 levels at IL-8 gene occurred later and were long lasting.</p> <p>Conclusion</p> <p>The results showed that specific chromatin modifications occurring at IL-8 gene, including histone H3 acetylation and methylation, mark LPS-mediated IL-8 activation in intestinal epithelial cells while it is unlikely that DNA methylation of IL-8 promoter is directly involved in IL-8 gene regulation in these cells.</p

    An Unsupervised Spectrogram Cross-Correlation Method to Assess ELM Triggering Efficiency by Pellets

    The high confinement mode (H-mode) is considered the optimal regime for the production of energy through nuclear fusion for industrial purposes since it allows to increase the energy confinement time of the plasma roughly by a factor of two. Consequently, it has been selected at the moment as the standard scenario for the next generation of devices, such as ITER. However, pressure-driven edge instabilities, known as edge localized modes (ELMs), are a distinct feature of this plasma regime. Their extrapolated thermal and particle peak loads on the plasma-facing components (PFC) of the next generation of devices are expected to be so high as to damage such structures, compromising the normal operations of the reactors themselves. Consequently, the induced loads have to be controlled; this can be achieved by mitigating ELMs. A possibility then lays in increasing the ELMs frequency to lower the loads on the PFCs. As already demonstrated at JET, the pellet pacing of ELMs is considered one of the most promising techniques for such scope, and its optimization is therefore of great interest for present and future operations of nuclear fusion facilities. In this work, we suggest a method to access primary pieces of information to perform statistics, assess and characterize the pacing efficiency. The method, tested on JET data, is based on the clustering (k-means) of convoluted signals, using so-called spectrogram cross-correlation, between the measured pellets and ELMs time traces. Results have also been obtained by taking advantage of a new type of diagnostic for measuring the ELMs dynamic, based on synthetic diamond sensors, faster than the standard spectroscopic cameras used at JET

    Estudio de indicadores fisicoquímicos y toxicológicos del agua superficial del bajo Río Paraná

    La cuenca baja del Río Paraná comprende un sistema de humedales que albergan un reservorio vital de biodiversidad y uno de los ecosistemas más productivos del planeta, brindando además importantes beneficios económicos y sociales. En el tramo final del bajo Paraná se desarrollan actividades agropecuarias e industriales que generan importantes impactos ambientales. En particular, el Arroyo de la Cruz (S1) atraviesa el partido de Campana que se caracteriza por una alta actividad industrial. El Río Arrecifes (S2) se caracteriza por atravesar grandes extensiones de cultivos. Por otro lado, los anfibios se destacan como valiosos indicadores de la calidad ambiental, desempeñando múltiples roles en los ecosistemas. El objetivo del presente trabajo fue evaluar en forma preliminar la calidad del agua superficial en S1 y S2 midiendo parámetros fisicoquímicos (Temperatura, Oxígeno Disuelto, pH, profundidad, conductividad, sólidos suspendidos totales, DBO5, DQO, amonio, fosfatos, nitratos, nitritos, cloruros), metales (As, Cr, Cu, Ni, Pb, Zn), y mediante bioensayos estandarizados de toxicidad (ANFITOX) con un anfibio autóctono Rhinella arenarum(Ra), por exposición de 10 embriones (E.4) o larvas (E.25), por triplicado, a diluciones de las muestras compuestas de agua superficial de cada sitio durante 504h y a un control negativo de Solución ANFITOX. Se destacaron los bajos valores de OD de los dos sitios: S1=3,4 y S2=4,1(mg/L). En cuanto a los metales, las concentraciones de Cu fueron mayores a lo reglamentado (decreto 831/93). Cabe destacar la letalidad observada por exposición de organismos en ambos estadios a las muestras ambientales. En el periodo embrionario, S1 causó mayor mortalidad, mientras que en la etapa larval, S2 fue más tóxica. Este estudio alerta sobre la degradación del Río Paraná en su cuenca baja y las herramientas empleadas podrían usarse para la toma de decisiones a fin de proteger los humedales.Fil: Peluso, Julieta.     Quality of life in elderly with dizziness

    Dizziness is frequent in elderly people. AIMS: To evaluate the Quality of Life (QoL) in elderly subjects with dizziness, relate it with gender and age. MATERIAL AND METHOD: A prospective study comprising 120 elderly patients with dizziness evaluated with Brazilian versions of the Whoqol-bref and the dizziness handicap inventory (DHI). The factor analysis (FA), the Mann Whitney and Kruskal Wallis tests, and the Spearman correlation were applied to study the results. RESULTS: The most compromised domains were the DHI physical domain and the Whoqol-bref physical and environment domains. FA resulted in 3 factors in the DHI and 5 factors in the Whoqol-bref. There was a moderate correlation (-0.596) in the total scores of both instruments. Males had a better QoL in the environment perception and introspectivity and health perception factors of the Whoqol-bref test. Females had a better QoL in the functionality perception factor of the Whoqol-bref test. There were no significant age differences. CONCLUSIONS: Elderly patients with dizziness have a worse QoL. Elderly females with dizziness have worse QoL scores in environment perception and introspectivity and health perception and better QoL in the functionality perception factor compared to elderly males.Tontura é muito frequente em idosos. OBJETIVOS: Avaliar a qualidade de vida (QV) em idosos vestibulopatas e relacioná-la com o gênero e a faixa etária. MATERIAL E MÉTODO: Estudo prospectivo que incluiu série de 120 casos de idosos com tontura que foram avaliados por meio das versões brasileiras dos questionários Whoqol-bref e Dizziness Handicap Inventory (DHI). Os resultados obtidos foram submetidos à Análise Fatorial (AF) e aos testes de Mann Whitney, Kruskal Wallis e correlação de Spearman. RESULTADOS: O domínio físico avaliado pelo DHI, e o físico e o ambiental do Whoqol-bref foram os mais comprometidos. A AF resultou em 3 fatores no DHI e 5 no Whoqol-bref: Houve moderada correlação (-0,596) entre o escore total de ambos os instrumentos. Os homens apresentaram melhor QV nos fatores percepção ambiental e introspectividade e percepção da saúde do Whoqol-bref. As idosas obtiveram melhor QV no fator percepção da funcionalidade do Whoqol-bref. Não houve diferenças significantes entre os fatores dos dois instrumentos por faixa etária. CONCLUSÕES: Idosos vestibulopatas apresentam QV prejudicada.     Identification of bi-allelic LFNG variants in three patients and further clinical and molecular refinement of spondylocostal dysostosis 3

    : Spondylocostal dysostosis (SCD), a condition characterized by multiple segmentation defects of the vertebrae and rib malformations, is caused by bi-allelic variants in one of the genes involved in the Notch signaling pathway that tunes the "segmentation clock" of somitogenesis: DLL3, HES7, LFNG, MESP2, RIPPLY2, and TBX6. To date, seven individuals with LFNG variants have been reported in the literature. In this study we describe two newborns and one fetus with SCD, who were found by trio-based exome sequencing (trio-ES) to carry homozygous (c.822-5C>T) or compound heterozygous (c.[863dup];[1063G>A]) and (c.[521G>T];[890T>G]) variants in LFNG. Notably, the c.822-5C>T change, affecting the polypyrimidine tract of intron 5, is the first non-coding variant reported in LFNG. This study further refines the clinical and molecular features of spondylocostal dysostosis 3 and adds to the numerous investigations supporting the usefulness of trio-ES approach in prenatal and neonatal settings

    Protection against Pseudomonas aeruginosa lung infection in mice by recombinant OprF-pulsed dendritic cell immunization

    BACKGROUND: The Pseudomonas aeruginosa major constitutive outer membrane porin protein F (OprF) has been shown to be a protective antigen and was previously used to activate an immunological response in a mouse model of lung pneumonia. The purpose of our study was to demonstrate the ability of mouse dendritic cells pulsed with purified or recombinant OprF to protect mice against P. aeruginosa infection and inflammation.Both native (n-OprF), isolated and purified from PAO1 bacterial strain, and recombinant (histidin-conjugated) OprF (His-OprF), obtained by cloning of the oprF gene into the pET28a expression vector, were used to stimulate dendritic cells in vitro before adoptive transfer into prospective recipient mice with P. aeruginosa pulmonary infection. RESULTS: Similar to n-OprF, His-OprF activated dendritic cells in vitro, inducing the costimulatory molecule expression as well as cytokine production. Upon adoptive transfer in vivo, porin-pulsed dendritic cells (DCs) induced Th1-mediated resistance to infection and associated inflammatory pathology caused by either the PAO1 strain or a clinically-isolated mucoid strain. CONCLUSIONS: This study highlights the pivotal contribution of DCs to vaccine-induced protection against P. aeruginosa infection and associated inflammation
