In the first chapter, co-authored with Dr. Christian Julliard, we study the impact of option expiration on underlying stock volatility. We find a negative direct effect on stock realized volatility and a positive and significant effect on stock implied volatility. Moreover, a positive spillover effect on stocks with no options expiring on a given expiration date is observed. Two possible explanations are discussed, namely investors’ delta hedging and stock pinning around option expiration dates. Both seem to affect stock volatility. Finally, we implement a trading strategy that takes advantage of these findings. The second chapter studies the investment behaviour of mutual funds during financial bubbles. I find that mutual funds over-invest in bubble sectors during the run-up and withdraw money right before the collapse. This result is robust across different benchmark specifications and across fund styles. I also document that this strategy generates a positive and significant alpha (4% on an annual basis), with respect to both a risk-neutral expectation and the Fama-French factors. The paper provides evidence supporting the theory that mutual funds ride the bubble rather than causing it. It also demonstrates that mutual fund holdings can predict the future returns of a sector over a short to medium horizon. Building on the previous findings, the third chapter studies the fee setting behaviour of mutual funds during financial bubbles. It shows that, besides the well-documented persistence of fees, mutual funds charge higher fees during price run-ups. Two theoretical models support the finding: the first shows that investors’ sensitivity to fees decreases during bubble episodes, while the second demonstrates that the increase in fees translates into a higher mark-up over marginal cost

Rad9/53BP1 promotes DNA repair via crossover recombination by limiting the Sgs1 and Mph1 helicases

The DNA damage checkpoint (DDC) is often robustly activated during the homologous recombination (HR) repair of DNA double strand breaks (DSBs). DDC activation controls several HR repair factors by phosphorylation, preventing premature segregation of entangled chromosomes formed during HR repair. The DDC mediator 53BP1/Rad9 limits the nucleolytic processing (resection) of a DSB, controlling the formation of the 3\u2032 single-stranded DNA (ssDNA) filament needed for recombination, from yeast to human. Here we show that Rad9 promotes stable annealing between the recombinogenic filament and the donor template in yeast, limiting strand rejection by the Sgs1 and Mph1 helicases. This regulation allows repair by long tract gene conversion, crossover recombination and break-induced replication (BIR), only after DDC activation. These findings shed light on how cells couple DDC with the choice and effectiveness of HR sub-pathways, with implications for genome instability and cancer

As novas tecnologias e os cegos em situações interativas e a compensação social

Dissertação (mestrado) - Universidade Federal de Santa Catarina, Centro de Ciências da Educação.O desenvolvimento e a acelerada implementação das novas tecnologias, associados ao processo de compensação social ou dialética, podem constituir-se em fatores desencadeadores e suporte ao processo de inclusão social de pessoas com história de cegueira. Nesta dissertação discutem-se estes aspectos, buscando-se a sustentação nos aportes da teoria histórico-cultural de Vigotski e seus colaboradores, para daí enfocar questões específicas, sobretudo a díade compensação/deficiência no cotidiano dos sujeitos cegos. A metodologia de pesquisa utilizada inclui a história de dois sujeitos cegos, evidenciando de que forma as alternativas sociais, em especial aquelas relacionadas às novas tecnologias, contribuem para o processo de compensação como força motriz que leva à superação das faltas/carências/deficiências. Trata-se de apreender a inclusão e redesenhar as questões do preconceito, da discriminação, da estigmatização, refletindo sobre o significado e as possibilidades de superação da deficiência dentro do contexto da diversidade social que envolve as pessoas com história de cegueira

PIBID – Articulando práticas pedagógicas promissoras

Neste trabalho buscamos fornecer informações sobre o cumprimento dos objetivos propostos pelo Programa Institucional de Bolsa de Iniciação à Docência (PIBID) da Universidade do Oeste de Santa Catarina, Campus de Joaçaba, SC, subprojeto Pedagogia. A pesquisa tem caráter qualitativo, incluindo a análise de opiniões dos envolvidos no Programa das Escolas de Educação Básica Frei Bruno de Joaçaba, São José de Herval d’ Oeste, Mater Dolorum de Capinzal e Silvio Santos, da cidade de Ouro, SC. Os resultados indicam que os objetivos do programa estão sendo alcançados com sucesso, e indicam que, com a implantação do Projeto PIBID nas Unidades Escolares atendidas, os alunos do 1º ao 5º ano passaram a ter atendimento com aulas diferenciadas e de acordo com as dificuldades individuais na alfabetização, produção textual, raciocínio lógico matemático, utilizando do lúdico; houve superação de dificuldades de ortografia, produção de textos, leituras, cálculos, interpretação e elevação da autoestima. Todos os jogos, atividades e demais materiais elaborados ou selecionados são utilizados sempre após o planejamento semanal das bolsistas, planejados conforme as dificuldades apresentadas pelas crianças em sala de aula e/ou no atendimento individualizado, ou de reforço escolar. Os resultados alcançados atendem o domínio da leitura e da escrita bem como a autoestima,  a vontade de produção instiga a querer mais e melhorar a qualidade de ensino e aprendizagem dentro e fora da sala de aula e a valorizar as bolsistas, como alunas acadêmicas, propiciando maior incentivo à profissão e atuação na prática escolar. Por fim, apontamos a importância dessa experiência, pois projetos dessa natureza possibilitam rever práticas pedagógicas desenvolvidas nas escolas com vistas à elevação da qualidade do ensino e enriquecem a formação inicial de professores para atuarem nos anos iniciais do ensino fundamental. Formation and nucleolytic processing of Cas9-induced DNA breaks in human cells quantified by droplet digital PCR

Cas9 endonuclease from S. pyogenes is widely used to induce controlled double strand breaks (DSB) at desired genomic loci for gene editing. Here, we describe a droplet digital PCR (ddPCR) method to precisely quantify the kinetic of formation and 5\u2032-end nucleolytic processing of Cas9-induced DSB in different human cells lines. Notably, DSB processing is a finely regulated process, which dictates the choice between non-homologous end joining (NHEJ) and homology directed repair (HDR). This step of DSB repair is also a relevant point to be taken into consideration to improve Cas9-mediated technology. Indeed, by this protocol, we show that processing of Cas9-induced DSB is impaired by CTIP or BRCA1 depletion, while it is accelerated after down-regulation of DNAPKcs and 53BP1, two DSB repair key factors. In conclusion, the method we describe here can be used to study DSB repair mechanisms, with direct utility for molecularly optimising the knock-out/in outcomes in genome manipulation

Expression of dendritic and T cells in potentially malignant lesions and oral squamous cell carcinoma

O sistema imune, por sua vez, desempenha importante papel no reconhecimento de células malignas precursoras. No entanto, as células tumorais através do mecanismo de imunoedição são capazes de escapar da imuno vigilância. Objetivo: Avaliar a expressão de células dendríticas de Langerhans (CDL) imaturas e maduras, células dendríticas plasmocitoides (CD plasmocitoides) e linfócitos T em displasia epitelial oral (DE), leucoplasia verrucosa proliferativa (LVP) e carcinoma espinocelular bucal (CEC) e correlacionar essa expressão com os dados clínicos disponíveis. Material e métodos: Cinquenta casos de CEC, 48 casos de DE e 15 casos de LVP foram submetidos a técnica imunoistoquímica para os anticorpos CD1a, CD83, CD303 e CD8. Os resultados dessas imunomarcações foram avaliados através da análise quantitativa de cinco campos consecutivos de cada lâmina utilizando um aumento de 400x. Resultados: A análise mostrou diminuição significativa da expressão de CD83 no grupo CEC em comparação ao grupo DE. CD303 mostrou aumento significativo no grupo CEC em comparação ao grupo DE. A expressão de CD8 não apresentou diferença significativa entre os grupos avaliados. Para o grupo LVP a expressão de CD1a e CD8 apresentou aumento gradual nas lesões de hiperqueratose, displasia leve/moderada e displasia epitelial intensa. A expressão de CD83 apresentou diminuição gradual nas lesões de hiperqueratose, displasia leve/moderada e displasia epitelial intensa. Não houve correlação estatística com os dados clínicos. Conclusão: O aumento no número de CDL maduras no grupo DE em relação ao grupo CEC sugere que essas células possam ter migrado até os linfonodos para dar início a resposta imune. O aumento no número de CD plasmocitoides no grupo CEC, sugere que essas células desempenham papel importante na defesa do organismo contra lesões tumorais, no entanto não está claro se promovendo ou inibindo a progressão tumoral. No grupo LVP a expressão de CDL imaturas e linfócitos T CD8 sugere que o sistema imune encontra-se vigilante e ativo, respectivamente. No entanto, a expressão de CDL maduras sugere que pode haver algum tipo de falha na resposta imuneAbstract: Oral squamous cell carcinomas (OSCC) may arise from potential malignant lesions, such as leukoplakia. The immune system plays an important role recognizing tumor precursor cells. However, due to the immunoediting mechanisms cancer cells are able to escape the immune system surveillance. Objective: Evaluate the profile of imature and mature dendritic Langerhans cells (DLC), plasmacytoid dendritic cells (plasmacytoid DC) and T cells in epithelial displasya (OED), proliferative verrucous leukoplakia (PVL) and OSCC and correlate these fndings with clinical data available. Study design: Fifty cases of OSCC, 48 of OED and 15 of PVL were immunostained for CD1a, CD83, CD303 and CD8 followed by a quantitative analysis. Results: Analysis revealed a significant decrease in the number of CD83 positive cells in OSCC compared with OED group. CD303 positivity was significantly increased in the OSCC group when compared to OED whilst there was no statistical difference in CD8 positivity between OSCC and OED. In the PVL group analysis revealed increased expression of CD1a from hyperkeratosis to severe epithelial dysplasia and decreased expression of CD83 from hyperkeratosis to severe epithelial dysplasia. The expression of CD8 increased from hyperkeratosis to severe epithelial dysplasia. No statisticant correlation was found for clinical data. Conclusion: The numbers of mature DLC were decreased in OSCC compared with OED lesions suggesting that maybe these cells might have migrated to lymph nodes to present the tumour antigen and activate the immune system. The numbers of plasmacytoid DC was significantly increased in the OSCC group compared with the OED group. This suggests they may play an important role in the defence against tumours although it is not clear whether this is promoting or inhibiting malignant progression. Experimental benchmarking of Monte Carlo simulations for radiotherapy dosimetry using monochromatic X-ray beams in the presence of metal-based compounds

The local dose deposition obtained in X-ray radiotherapy can be increased by the presence of metal-based compounds in the irradiated tissues. This finding is strongly enhanced if the radiation energy is chosen in the kiloelectronvolt energy range, due to the proximity to the absorption edge. In this study, we present a MC application developed with the toolkit Geant4 to investigate the dosimetric distribution of a uniform monochromatic X-ray beam, and benchmark it against experimental measurements. Two validation studies were performed, using a commercial PTW RW3 water-equivalent slab phantom for radiotherapy, and a custom-made PMMA phantom conceived to assess the influence of high atomic number compounds on the dose profile, such as iodine and gadolinium at different concentrations. An agreement within 9% among simulations and experimental data was found for the monochromatic energies considered, which were in the range of 30–140 keV; the agreement was better than 5% for depths <60 mm. A dose enhancement was observed in the calculations, corresponding to the regions containing the contrast agents. Dose enhancement factors (DEFs) were calculated, and the highest values were found for energies higher than the corresponding K-edges of iodine and gadolinium. The in-silico results are in line with the empirical findings, which suggest that Geant4 can be satisfactorily used as a tool for the calculation of the percentage depth dose (PDD) at the energies considered in this study in the presence of contrast agents
