    Tracing the role of endogenous carbon in denitrification using wine industry by-product as an external electron donor: coupling isotopic tools with mathematical modeling

    Nitrate removal through enhanced biological denitrification (EBD), consisting of the inoculation of an external electron donor, is a feasible solution for the recovery of groundwater quality. In this context, liquid waste from wine industries (wine industry by-products, WIB) may be feasible for use as a reactant to enhance heterotrophic denitrification. To address the feasibility of WIB as electron donor to promote denitrification, as well as to evaluate the role of biomass as a secondary organic C source, a flow-through experiment was carried out. Chemical and isotopic characterization was performed and coupled with mathematical modeling. Complete nitrate attenuation with no nitrite accumulation was successfully achieved after 10 days. Four different C/N molar ratios (7.0, 2.0, 1.0 and 0) were tested. Progressive decrease of the C/N ratio reduced the remaining C in the outflow and favored biomass migration, producing significant changes in dispersivity in the reactor, which favored efficient nitrate degradation. The applied mathematical model described the general trends for nitrate, ethanol, dissolved organic carbon (DOC) and dissolved inorganic carbon (DIC) concentrations. This model shows how the biomass present in the system is degraded to dissolved organic C (DOCen) and becomes the main source of DOC for a C/N ratio between 1.0 and 0. The isotopic model developed for organic and inorganic carbon also describes the general trends of d13C of ethanol, DOC and DIC in the outflow water. The study of the evolution of the isotopic fractionation of organic C using a Rayleigh distillation model shows the shift in the organic carbon source from the WIB to the biomass and is in agreement with the isotopic fractionation values used to calibrate the model. Isotopic fractionations (e) of C-ethanol and C-DOCen were -1‰ and -5‰ (model) and -3.3‰ and -4.8‰ (Rayleigh), respectively. In addition, an inverse isotopic fractionation of +10‰ was observed for biomass degradation to DOCen. Overall, WIB can efficiently promote nitrate reduction in EBD treatments.     Synergies between Geomatics and Health Sciences for the creation of new virtual materials for teaching podiatry

    [EN] Thermography as a methodology for quantitative analysis is not usually addressed in the degrees of the university programs of the Health Sciences branch, with the consequent deficiencies in technological tools in the training of future graduates. Therefore, this manuscript proposes a novel approach for the acquisition of advanced skills in the Health Sciences degree of podiatry, through the application of techniques and tools from Geomatics engineering and based on free/open-source software solutions. This strategy uses 3D visualization techniques on thermographic images to improve the interpretation and understanding of thermographic images related to the physiological and pathological analysis of the lower extremity. The specific workflow for the generation of didactic material related to this objective is proposed for asynchronous and e-learning programs, so that these virtual materials can be easily deployed on the institutional based-on Moodle platform, allowing students to enrich the learning.     Characterization of Human Herpesvirus 8 genomic integration and amplification events in a primary effusion lymphoma cell line

    In this study, we investigated the integration of Human Herpesvirus 8 (HHV-8) into the human genome using the primary effusion lymphoma (PEL) cell line BC-3. Through next-generation sequencing (NGS) data from multiple independent sequencing runs, we identified two highly supported HHV-8 integrants. These integrants encompassed a region of human chromosome 12 that was amplified approximately 16-fold between the junctions. Significantly, these events could represent the first known instance of HHV-8 integration into a hybrid human-viral extrachromosomal chimeric circular DNA (eccDNA). The amplified fragment contained partial or complete copies of various human genes, including SELPLG and CORO1C. Analysis of long-read Nanopore data indicated that the CpGs at the SELPLG promoter were mostly unmethylated, suggesting that the additional copies of SELPLG within this eccDNA are likely transcriptionally active. Our findings suggest that viral insertion and eccDNA amplification could be crucial mechanisms in the development of HHV-8-related cancers. In conclusion, our study provides valuable insights into the molecular mechanisms involved in HHV-8-induced oncogenesis and emphasizes the importance of investigating viral integration and eccDNAs in cancer development. Furthermore, we highlight the necessity of employing multiple independent sequencing approaches to validate integration events and avoid false positives derived from library construction artifacts

    Predicting long-term disease control in transplant-ineligible patients with multiple myeloma: impact of an MGUS-like signature

Therefore, to gain further insights in this endpoint, a population of 498 newly diagnosed transplant-ineligible patients enrolled in two Spanish trials (GEM2005MAS65 and GEM2010MAS65), has been analyzed. Among the 435 patients included in this post-hoc study, 18.6% remained alive and progression free after 5 years of treatment initiation. In these patients, overall survival (OS) rate at 10 years was 60.8% as compared with 11.8% for those progressing within the first 5 years. Hemoglobin (Hb) ≥ 12 g/dl (OR 2.74, p = 0.001) and MGUS-like profile (OR 4.18, p = 0.005) were the two baseline variables associated with long-term disease-free survival. Upon including depth of response (and MRD), Hb ≥ 12 g/dl (OR 2.27) and MGUS-like signature (OR 7.48) retained their predictive value along with MRD negativity (OR 5.18). This study shows that despite the use of novel agents, the probability of disease control at 5 years is still restricted to a small fraction (18.6%) of elderly MM patients. Since this endpoint is associated with higher rates of OS, this study provides important information about diagnostic and post-treatment biomarkers helpful in predicting the likelihood of disease control at 5 years

    Search on speech from spoken queries: the Multi-domain International ALBAYZIN 2018 Query-by-Example Spoken Term Detection Evaluation

    [Abstract] The huge amount of information stored in audio and video repositories makes search on speech (SoS) a priority area nowadays. Within SoS, Query-by-Example Spoken Term Detection (QbE STD) aims to retrieve data from a speech repository given a spoken query. Research on this area is continuously fostered with the organization of QbE STD evaluations. This paper presents a multi-domain internationally open evaluation for QbE STD in Spanish. The evaluation aims at retrieving the speech files that contain the queries, providing their start and end times, and a score that reflects the confidence given to the detection. Three different Spanish speech databases that encompass different domains have been employed in the evaluation: MAVIR database, which comprises a set of talks from workshops; RTVE database, which includes broadcast television (TV) shows; and COREMAH database, which contains 2-people spontaneous speech conversations about different topics. The evaluation has been designed carefully so that several analyses of the main results can be carried out. We present the evaluation itself, the three databases, the evaluation metrics, the systems submitted to the evaluation, the results, and the detailed post-evaluation analyses based on some query properties (within-vocabulary/out-of-vocabulary queries, single-word/multi-word queries, and native/foreign queries). Fusion results of the primary systems submitted to the evaluation are also presented. Three different teams took part in the evaluation, and ten different systems were submitted. The results suggest that the QbE STD task is still in progress, and the performance of these systems is highly sensitive to changes in the data domain.     Secondary aroma: influence of wine microorganisms in their aroma profile

    Aroma profile is one of the main features for the acceptance of wine. Yeasts and bacteria are the responsible organisms to carry out both, alcoholic and malolactic fermentation. Alcoholic fermentation is in turn, responsible for transforming grape juice into wine and providing secondary aromas. Secondary aroma can be influenced by different factors; however, the influence of the microorganisms is one of the main agents affecting final wine aroma profile. Saccharomyces cerevisiae has historically been the most used yeast for winemaking process for its specific characteristics: high fermentative metabolism and kinetics, low acetic acid production, resistance to high levels of sugar, ethanol, sulfur dioxide and also, the production of pleasant aromatic compounds. Nevertheless, in the last years, the use of non-saccharomyces yeasts has been progressively growing according to their capacity to enhance aroma complexity and interact with S. cerevisiae, especially in mixed cultures.     State-of-the-art of analytical techniques to determine food fraud in olive oils

    The benefits of the food industry compared to other sectors are much lower, which is why producers are tempted to commit fraud. Although it is a bad practice committed with a wide variety of foods, it is worth noting the case of olive oil because it is a product of great value and with a high percentage of fraud. It is for all these reasons that the authenticity of olive oil has become a major problem for producers, consumers, and legislators. To avoid such fraud, it is necessary to develop analytical techniques to detect them. In this review, we performed a complete analysis about the available instrumentation used in olive fraud which comprised spectroscopic and spectrometric methodology and analyte separation techniques such as liquid chromatography and gas chroma-tography. Additionally, other methodology including protein-based biomolecular techniques and analytical approaches like metabolomic, hhyperspectral imaging and chemometrics are discussed.The research leading to these results was supported by MICINN with the Ramón&Cajal grant for M.
