    Possible attenuation of the G2 DNA damage cell cycle checkpoint in HeLa cells by extremely low frequency (ELF) electromagnetic fields

    BACKGROUND: The issue remains unresolved as to whether low frequency magnetic fields can affect cell behaviour, with the possibility that they may be in part responsible for the increased incidence of leukaemia in parts of the population exposed to them. METHODS: Combined treatment of HeLa cells with gamma-irradiation (1, 3 and 5 Grays) and extra low frequency magnetic fields of ~50 Hz was carried out under rigorously controlled conditions. RESULTS: Synchronised cells progressing from S-phase arrived at mitosis on average marginally ahead of irradiation controls not exposed to ELF. In no instance out of a total of twenty separate experiments did this "double-insult" further delay entry of cells into mitosis, as had been anticipated. CONCLUSION: This apparently "non-genotoxic" agent (ELF) appears to be capable of affecting cells that would normally arrest for longer in G2, suggesting a weakening of the stringency of the late cycle (G2) checkpoint

    Automated analysis of 15N and 14C in biological samples

    Includes bibliographical references (page 947).An automated method for the simultaneous analysis of total N, total C, 15N and 14C in small plant and soil samples is described. A commercial C-N analyser - continuous flow isotope ratio mass spectrometer (ANCA-MS) has been extended to also measure CO2 and collect 14CO2 produced by sample combustion. Samples containing 20 - 200 μg N and up to 5 mg C can be analysed directly with no sample preparation other than drying and fine grinding. The precision of total elemental analysis is comparable to that by conventional methods. The average standard deviation of 15N analyses of plant material at natural abundance was ±1 ‰. This is accurate enough for all 15N studies except those using natural abundance and possibly long term studies of soil organic matter. Recovery of 14C in test samples was 100%. The instrument can be operated by graduate students under supervision and operating costs are primarily for sample cups, combustion catalyst and quartz tubes

    Upper Extremity Biomechanical Model for Evaluation of Pediatric Joint Demands during Wheelchair Mobility

    Current methods for evaluating upper extremity (UE) dynamics during pediatric wheelchair use are limited. We propose a new model to characterize UE joint kinematics and kinetics during pediatric wheelchair mobility. The bilateral model is comprised of the thorax, clavicle, scapula, upper arm, forearm, and hand segments. The modeled joints include: sternoclavicular, acromioclavicular, glenohumeral, elbow and wrist. The model is complete and is currently undergoing pilot studies for clinical application. Results may provide considerable quantitative insight into pediatric UE joint dynamics to improve wheelchair prescription, training and long term care of children with orthopaedic disabilities

    The importance of scale in spatially varying coefficient modeling

    While spatially varying coefficient (SVC) models have attracted considerable attention in applied science, they have been criticized as being unstable. The objective of this study is to show that capturing the "spatial scale" of each data relationship is crucially important to make SVC modeling more stable, and in doing so, adds flexibility. Here, the analytical properties of six SVC models are summarized in terms of their characterization of scale. Models are examined through a series of Monte Carlo simulation experiments to assess the extent to which spatial scale influences model stability and the accuracy of their SVC estimates. The following models are studied: (i) geographically weighted regression (GWR) with a fixed distance or (ii) an adaptive distance bandwidth (GWRa), (iii) flexible bandwidth GWR (FB-GWR) with fixed distance or (iv) adaptive distance bandwidths (FB-GWRa), (v) eigenvector spatial filtering (ESF), and (vi) random effects ESF (RE-ESF). Results reveal that the SVC models designed to capture scale dependencies in local relationships (FB-GWR, FB-GWRa and RE-ESF) most accurately estimate the simulated SVCs, where RE-ESF is the most computationally efficient. Conversely GWR and ESF, where SVC estimates are naively assumed to operate at the same spatial scale for each relationship, perform poorly. Results also confirm that the adaptive bandwidth GWR models (GWRa and FB-GWRa) are superior to their fixed bandwidth counterparts (GWR and FB-GWR)

    A Framework for Establishing Restoration Goals for Contaminated Ecosystems

    This article represents 1 of 6 articles in the special series “Restoration of Impaired Ecosystems: An Ounce of Prevention or a Pound of Cure?” The articles result from a Technical Workshop organized by SETAC and the Society for Ecological Restoration, held June 2014 in Jackson, Wyoming, that focused on advancing the practice of restoring ecosystems that have been contaminated or impaired from industrial activities.As natural resources become increasingly limited, the value of restoring contaminated sites, both terrestrial and aquatic, becomes increasingly apparent. Traditionally, goals for remediation have been set before any consideration of goals for ecological restoration. The goals for remediation have focused on removing or limiting contamination whereas restoration goals have targeted the ultimate end use. Here, we present a framework for developing a comprehensive set of achievable goals for ecological restoration of contaminated sites to be used in concert with determining goals for remediation. This framework was developed during a Society of Environmental Toxicology and Chemistry (SETAC) and Society of Ecological Restoration (SER) cosponsored workshop that brought together experts from multiple countries. Although most members were from North America, this framework is designed for use internationally. We discuss the integration of establishing goals for both contaminant remediation and overall restoration, and the need to include both the restoration of ecological and socio-cultural-economic value in the context of contaminated sites. Although recognizing that in some countries there may be regulatory issues associated with contaminants and clean up, landscape setting and social drivers can inform the restoration goals. We provide a decision tree support tool to guide the establishment of restoration goals for contaminated ecosystems. The overall intent of this decision tree is to provide a framework for goal setting and to identify outcomes achievable given the contamination present at a site.     Endocytic Function, Glycosaminoglycan Specificity, and Antibody Sensitivity of the Recombinant Human 190-kDa Hyaluronan Receptor for Endocytosis (HARE)

    The human hyaluronan receptor for endocytosis (hHARE) mediates the endocytic clearance of hyaluronan (HA) and chondroitin sulfate from lymph fluid and blood. Two hHARE isoforms (190 and 315 kDa) are present in sinusoidal endothelial cells of liver, spleen, and lymph nodes (Zhou, B., McGary, C. T., Weigel, J. A., Saxena, A., and Weigel, P. H. (2003) Glycobiology 13, 339–349). Here we report the specificity and function of the 190-kDa HARE, expressed without the larger isoform, in Flp-In 293 cell lines (190hHARE cells). Like the native protein, recombinant hHARE contains ~25 kDa of N-linked oligosaccharides, binds HA in a ligand blot assay, cross-reacts with three anti-rat HARE monoclonal antibodies, and is inactivated by reduction. The 190hHARE cell lines mediated rapid, continuous 125I-HA endocytosis and degradation for \u3e1 day. About 30–50% of the total cellular receptors were on the cell surface, and their recycling time for reutilization was ~8.5 min. The average Kd for the binding of HA to the 190-kDa hHARE at 4 °C was 7 nm with 118,000 total HA binding sites per cell. Competition studies at 37 °C indicated that the 190- kDa hHARE binds HA and chondroitin better than dermatan sulfate and chondroitin sulfates A, C, D, and E, but it does not bind to heparin, heparan sulfate, or keratan sulfate. Although competition was observed at 37 °C, none of the glycosaminoglycans tested, except HA, competed for 125I-HA binding by 190hHARE cells at 4 °C. Anti-HARE monoclonal antibodies #30 and #154, which do not inhibit 125I-HA uptake mediated by the 175-kDa rat HARE, partially blocked HA endocytosis by the 190-kDa hHARE. We conclude that the 190-kDa hHARE can function independently of other hHARE isoforms to mediate the endocytosis of multiple glycosaminoglycans. Furthermore, the rat and human small HARE isoforms have different glycosaminoglycan specificities and sensitivities to inhibition by cross-reacting antibodies

    The hyaluronic acid receptor for endocytosis (HARE; also designated Stabilin-2) mediates systemic clearance of hyaluronan and chondroitin sulfates from the vascular and lymphatic circulations. The internalized glycosaminoglycans are degraded in lysosomes, thus completing their normal turnover process. Sinusoidal endothelial cells of human liver, lymph node, and spleen express two HARE isoforms of 315 and 190 kDa. Here we report that the 190- and 315-kDa HARE isoforms, expressed stably either in Flp-In 293 cell lines or as soluble ectodomains, specifically bind heparin (Hep). The Kd for Hep binding to purified 190- and 315-kDa HARE ectodomains was 17.2 ± 4.9 and 23.4 ± 5.3 nm, respectively. Cells expressing HARE readily and specifically internalized 125I-streptavidin-biotin-Hep complexes, which was inhibited \u3e70% by hyperosmolar conditions, confirming that uptake is mediated by the clathrin-coated pit pathway. Internalization of Hep occurred for many hours with an estimated HARE recycling time of ~12 min. Internalized fluorescent streptavidin-biotin-Hep was present in a typical endocytic vesicular pattern and was delivered to lysosomes. We conclude that HARE in the sinusoidal endothelial cells of lymph nodes and liver likely mediates the efficient systemic clearance of Hep and many different Hep-binding protein complexes from the lymphatic and vascular circulations

    Continuous flow isotope ratio mass spectrometry of carbon dioxide trapped as strontium carbonate

    Includes bibliographical references (pages 756-757).The isotopic signal provided by differential discrimination against atmospheric carbon dioxide (13CO2) by C3 and C4 plant photosynthetic pathways is being widely used to study the processes of carbon (C) fixation, soil organic matter formation, and mineralization in nature. These studies have been facilitated by the availability of automated C and nitrogen (N) combustion analyzers (ANCA) combined with continuous flow isotope ratio mass spectrometers (CFIRMS). Analysis of 13CO2 in these instruments requires consistent sample mass for best precision, a requirement that is easily satisfied for soil and tissue samples by adjusting sample weight. Consistent CO2 sample size is much more difficult to achieve using gas handling systems for samples of headspace gases when CO2 concentrations vary widely. Long storage of gaseous samples also is difficult. Extended respiration studies are most easily conducted by trapping CO2 in alkali and conversion to an insoluble carbonate. Thermal decomposition of the carbonate in an on-line ANCA allows consistent and optimal CO2 sample mass to be obtained. The use of precipitated carbonates also facilitates storage of samples and enables full automation of sample analysis using an ANCA interfaced to a CFIRMS. Calcium (Ca), strontium (Sr), and barium (Ba) carbonates were tested. Strontium carbonate (SrCO3) with the addition of vanadium pentoxide (V2O5) as a combustion catalyst was found most suitable

    Abstract Background Agricultural workers may be exposed to potential carcinogens including pesticides, sensitizing agents and solar radiation. Previous studies indicate increased risks of hematopoietic cancers and decreased risks at other sites, possibly due to differences in lifestyle or risk behaviours. We present findings from CanCHEC (Canadian Census Health and Environment Cohort), the largest national population-based cohort of agricultural workers. Methods Statistics Canada created the cohort using deterministic and probabilistic linkage of the 1991 Canadian Long Form Census to National Cancer Registry records for 1992–2010. Self-reported occupations were coded using the Standard Occupational Classification (1991) system. Analyses were restricted to employed persons aged 25–74 years at baseline (N = 2,051,315), with follow-up until December 31, 2010. Hazard ratios (HR) and 95% confidence intervals (CI) were modeled using Cox proportional hazards for all workers in agricultural occupations (n = 70,570; 70.8% male), stratified by sex, and adjusted for age at cohort entry, province of residence, and highest level of education. Results A total of 9515 incident cancer cases (7295 in males) occurred in agricultural workers. Among men, increased risks were observed for non-Hodgkin lymphoma (HR = 1.10, 95% CI = 1.00–1.21), prostate (HR = 1.11, 95% CI = 1.06–1.16), melanoma (HR = 1.15, 95% CI = 1.02–1.31), and lip cancer (HR = 2.14, 95% CI = 1.70–2.70). Decreased risks in males were observed for lung, larynx, and liver cancers. Among female agricultural workers there was an increased risk of pancreatic cancer (HR = 1.36, 95% CI = 1.07–1.72). Increased risks of melanoma (HR = 1.79, 95% CI = 1.17–2.73), leukemia (HR = 2.01, 95% CI = 1.24–3.25) and multiple myeloma (HR = 2.25, 95% CI = 1.16–4.37) were observed in a subset of female crop farmers. Conclusions Exposure to pesticides may have contributed to increased risks of hematopoietic cancers, while increased risks of lip cancer and melanoma may be attributed to sun exposure. The array of decreased risks suggests reduced smoking and alcohol consumption in this occupational group compared to the general population
