    Owner perceptions of radiotherapy treatment for veterinary patients with cancer

    Veterinary clients may have trepidation about treating their pet with radiotherapy due to concerns about radiation side effects or repeated anaesthetics. The purpose of this study is to assess whether owners’ attitudes towards veterinary radiotherapy, including concerns over side effects, change during the course of treatment, and whether radiotherapy was perceived to affect pets’ quality of life. A prospective cohort study of clients from 2012-2015 was performed. Pets received palliative or definitive radiotherapy for various tumours. Clients completed questionnaires before, during and after radiotherapy. Questions assessed owner preconceptions before treatment, including side effect expectations, actual side effects experienced, and overall satisfaction with the process. In addition, at each time point the owners assessed their pet’s quality of life using a simple numerical scale. 49 patients were included. After completing treatment, owners were significantly less concerned about potential side effects of radiotherapy (P<0.001), side effects associated with repeat anaesthetics (P<0.001), and about radiotherapy in general (P<0.001). Quality of life did not show a significant change at any point during or after treatment. Following treatment, 94% reported the experience was better than expected and 100% supported the use of radiotherapy in pets. This is the first prospective study evaluating client attitudes and satisfaction before and after radiotherapy treatment in pets. The results indicate that radiotherapy is well tolerated, and the anxiety associated with radiotherapy is significantly alleviated after experiencing the process. These results will help veterinarians allay client concerns, and will hopefully lead to an increase in clients pursuing radiotherapy in pets

    Variation de la densité et la couleur du bois de Pterocarpus erinaceus (Poir) en fonction des conditions environnementales en Afrique de l’Ouest

    National audiencePterocarpus erinaceus (Poir.) is currently spontaneous species of Guinea-Sudanese and Sudano-Sahelian areas overexploited and threatened in West Africa. This paper analyzes the density and color of the wood of the species in the agro-ecological zones of three countries in West Africa (Togo, Burkina Faso and Niger). The results of 452 samples collected from 95 trees from the three countries show significant differences in values of basic density and color parameters (P <0.05) according to agro-ecological zones (Sudan, Guinea and Sahel). At scale intra-tree, even if there is an increase in density with the number of rings counted from the pith, this increase is very small (R²≤ 0.18). This reflects the fact that age has little influence on the trees density. In addition, a significant correlation was observed between density and color parameters.Pterocarpus erinaceus (Poir.) est actuellement l’espèce spontanée des zones guinéo-soudaniennes et soudano-sahéliennes très exploitée et menacée en Afrique de l’Ouest. Le présent travail analyse la densité et la couleur du bois de l’espèce dans les zones agroécologiques de trois pays de l’Afrique de l’Ouest (Togo, Burkina Faso et Niger). Les résultats obtenus sur 452 éprouvettes prélevées de 95 arbres issus des 3 pays indiquent des différences significatives des valeurs de l’infradensité et des paramètres de couleur (P<0,05) en fonction des zones agro-écologiques (soudanienne, guinéenne et sahélienne).     Polar bears are inefficient predators of seabird eggs

    Climate-mediated sea-ice loss is disrupting the foraging ecology of polar bears (Ursus maritimus) across much of their range. As a result, there have been increased reports of polar bears foraging on seabird eggs across parts of their range. Given that polar bears have evolved to hunt seals on ice, they may not be efficient predators of seabird eggs. We investigated polar bears\u27 foraging performance on common eider (Somateria mollissima) eggs on Mitivik Island, Nunavut, Canada to test whether bear decision-making heuristics are consistent with expectations of optimal foraging theory. Using aerial-drones, we recorded multiple foraging bouts over 11 days, and found that as clutches were depleted to completion, bears did not exhibit foraging behaviours matched to resource density. As the season progressed, bears visited fewer nests overall, but marginally increased their visitation to nests that were already empty. Bears did not display different movement modes related to nest density, but became less selective in their choice of clutches to consume. Lastly, bears that capitalized on visual cues of flushing eider hens significantly increased the number of clutches they consumed; however, they did not use this strategy consistently or universally. The foraging behaviours exhibited by polar bears in this study suggest they are inefficient predators of seabird eggs, particularly in the context of matching behaviours to resource density

In these models the lepton number and CP violating interactions of the triplet higgs scalars can give rise to lepton number asymmetry through non-equilibrium decays of the SU(2)LSU(2)_L triplet higgs and the right handed neutrinos.     Cell-free protein synthesis as a tool to study RXFP3- Relaxin-3 protein interactions

    With the discovery of the relaxin family peptide receptors there is interest in obtaining a clearer understanding of the structure of these proteins and the molecular mechanism of receptor-ligand interaction. As G-protein coupled receptors, obtaining milligram quantities for structural investigations is hampered by the inherent instability of these integral membrane proteins. In the current context, understanding of GPCR structural biology has increased dramatically with crystal structures of several inactive and now active forms solved. In addition, the first nuclear magnetic resonance structure of a GPCR was obtained which is of crucial importance to studying these receptors in a more “biologically relevant” setting. However despite this expansion in the field, most structures have been solved on modified systems so as to increase stability and are not necessarily representative of the native receptors. In relation to the relaxin family peptide receptors, we chose to investigate relaxin-family peptide receptor-3 expressed by cellfree protein synthesis. In contrast to in-vivo expression, cell-free was capable of producing large amounts of native receptor which makes it amenable to demanding structural studies

    Biased allosteric modulation at the CaS receptor engendered by structurally diverse calcimimetics

    Background and Purpose Clinical use of cinacalcet in hyperparathyroidism is complicated by its tendency to induce hypocalcaemia, arising partly from activation of calcium-sensing receptors (CaS receptors) in the thyroid and stimulation of calcitonin release. CaS receptor allosteric modulators that selectively bias signalling towards pathways that mediate desired effects [e.g. parathyroid hormone (PTH) suppression] rather than those mediating undesirable effects (e.g. elevated serum calcitonin), may offer better therapies. Experimental Approach We characterized the ligand-biased profile of novel calcimimetics in HEK293 cells stably expressing human CaS receptors, by monitoring intracellular calcium (Ca2+i) mobilization, inositol phosphate (IP)1 accumulation, ERK1/2 phosphorylation (pERK1/2) and receptor expression. Key Results Phenylalkylamine calcimimetics were biased towards allosteric modulation of Ca2+i mobilization and IP1 accumulation. S,R-calcimimetic B was biased only towards IP1 accumulation. R,R-calcimimetic B and AC-265347 were biased towards IP1 accumulation and pERK1/2. Nor-calcimimetic B was unbiased. In contrast to phenylalkylamines and calcimimetic B analogues, AC-265347 did not promote trafficking of a loss-of-expression, naturally occurring, CaS receptor mutation (G670E). Conclusions and Implications The ability of R,R-calcimimetic B and AC-265347 to bias signalling towards pERK1/2 and IP1 accumulation may explain their suppression of PTH levels in vivo at concentrations that have no effect on serum calcitonin levels. The demonstration that AC-265347 promotes CaS receptor receptor signalling, but not trafficking reveals a novel profile of ligand-biased modulation at CaS receptors The identification of allosteric modulators that bias CaS receptor signalling towards distinct intracellular pathways provides an opportunity to develop desirable biased signalling profiles in vivo for mediating selective physiological responses
