    Dopamine transporter and transmission of psychopathological risk. A review of gene-environment interplay

    Research underlines that intergenerational transmission of psychopathological risk results from a complex interplay of genetic and environmental risk factors which predispose child to develop emotionalbehavioral problems. Mechanisms of transmission are poorly understood, but few studies have focused on the role played by dopamine transporter (DAT) gene. This review aims to examine mediating mechanism of DAT genotype-environmental interaction (GxE), DAT genotype-environmental correlation (rGE), and methylation status involved in transmission of psychopathological risk. The review of literature was made through researches in university libraries on paper material, and telematics systems research. Studies have evidenced that DAT is implicated in intergenerational transmission of psychopathological risk. Results are mixed regarding its genetic variants, but mechanisms through which this gene can affect both quality of parenting and child development are partially established. Only few studies have examined methylation mechanisms that can be implicated. Findings suggest to involve an improved focus on DAT genotypes, methylation status associated, and their relationship with environment to better understanding child’s vulnerability and resilience following exposure to contextual risk factors associated with parental psychopathological symptoms

    Background: The effect of gene polymorphisms and promoter methylation, associated with maladaptive developmental outcomes, vary depending on environmental factors (e.g., parental psychopathology). Most studies have focused on 0- to 5-year-old children, adolescents, or adults, whereas there is dearth of research on school-age youths and pre-adolescents. Methods: In a sample of 21 families recruited at schools, we addressed parents' psychopathological symptoms (through SCL-90-R); offspring emotional-behavioral functioning (through CBCL-6-18); dopamine transporter gene (DAT1) for epigenetic status of the 5'-untranslated region (UTR) and for genotype, i.e., variable number of tandem repeats polymorphism at the 3'-UTR. Possible associations were explored between bio-genetic and psychological characteristics within the same individual and between triplets of children, mothers, and fathers. Results: DAT methylation of CpG at positions M1, M6, and M7 in mothers was correlated with maternal (phobic) anxiety, whereas in fathers' position M6 was related to paternal depression, anxiety, hostility, psychoticism, and higher Global Severity Index (GSI). No significant correlations were found between maternal and offspring DAT methylation. Significant correlations were found between fathers' methylation at CpG M1 and children's methylation at CpG M6. Linear regressions showed that mothers and fathers' GSI predicted children's methylation at CpG sites M2, M3, and M6, whereas fathers' GSI predicted children's methylation at CpG sites, particularly M1, M2, and M6. Moreover, offspring methylation of DAT at CpG M2 predicted somatic complaint, internalizing and attention problems; methylation of DAT at CpG M6 predicted withdraw. Conclusion: This study may have important clinical implication for the prevention and treatment of emotional-behavioral difficulties in children, as it adds to previous knowledge about the role of genetic and environmental factors in predicting psychopathological symptoms within non-clinical population

    Associations among internet addiction, genetic polymorphisms, family functioning, and psychopathological risk. Cross-sectional exploratory study

    Background: International research has emphasized that youths are at higher risk for the onset of internet addiction (IA), but studies investigating biological, psychological, and social factors associated with this condition are limited. Objective: This study aims to investigate the possible association between IA and genetic polymorphisms in monoamine oxidase A (MAO-A), serotonin-transporter (5-HTTPR), dopamine receptor (DRD4), and dopamine transporter (DAT1) genes by considering the role played by the perception of young adults in their family functioning and their depression, anxiety, and avoidant personality problems. Methods: In a sample of 104 male and female young adults aged between 19 and 23 years (mean age 21.87, SD 2.29 years) recruited from universities in the central southern part of Italy, we addressed the presence of IA using the Young criteria of the IA test. Moreover, the perception of young adults of their family functioning and their psychopathological symptoms were assessed through the Family Assessment Device (FAD) and the Adult Self-Report, respectively. Results: We found no significant association between IA and any genetic polymorphisms, neither among males or females. Young adults with IA reported significantly higher scores in the subscale of FAD affective responsiveness (AR; P=.01) and in depressive problems (P=.02), anxiety problems (P=.009), and avoidant personality problems (P=.003) than those in the control group. Results of mediation analyses showed a mediation role played by depressive symptoms (B=0.99; 95% CI 0.22 to 1.97) and avoidant personality problems (B=1.09; 95% CI 0.32 to 2.05) of young adults on the relationship between the FAD, AR, and IA. Finally, this relationship was moderated by the genotype of the 5-HTTLPR (P<.001), DAT1 (P<.001), and MAO-A (P<.001) genes in young adults. Conclusions: This exploratory study supports the recent evidence on the mutual relationship among biological, individual, and social risk factors associated with IA in young adulthood. Our findings may have important clinical implications for the development of prevention and treatment programs

    Causes and Solutions for High Direct Care Staff Turnover

    This quantitative research project explores the reasons and solutions for the high rates of direct care staff turnover. Emails were sent out to social service agency supervisors asking for their approval to allow their employees to participate in an online survey about direct care staff turnover. Agencies that agreed to participate were then emailed a script and a consent form with instructions to email both the script and the consent form to their employees. The ten question online survey explored the direct care staff\u27s opinions on topics such as compensation, support and training. Additionally, there was a qualitative question at the end of the survey asking for direct care staff\u27s input as to possible solutions to reduce direct care staff turnover. Twenty-six individuals participated in the survey. Answers were analyzed and entered into SPSS in order to find correlations in the data. Themes were identified amongst the responses to the qualitative question. A majority of the respondents did not feel they received adequate support from their supervisor or adequate compensation for the work that they do. Answers showed that direct care staff who participated in the survey attributed inadequate compensation as the largest contributor to the high rates of direct care staff turnover. The answers revealed no statistically significant data however, some correlations approached statistical significance. Results from this quantitative research project were consistent with pre-existing literature

    Virtual enactment effect on memory in young and aged populations: a systematic review

    Background: Spatial cognition is a critical aspect of episodic memory, as it provides the scaffold for events and enables successful retrieval. Virtual enactment (sensorimotor and cognitive interaction) by means of input devices within virtual environments provides an excellent opportunity to enhance encoding and to support memory retrieval with useful traces in the brain compared to passive observation. Methods: We conducted a systematic review with Preferred Reporting Items for Systematic Reviews and Meta-Analysis (PRISMA) guidelines concerning the virtual enactment effect on spatial and episodic memory in young and aged populations. We aim at giving guidelines for virtual enactment studies, especially in the context of aging, where spatial and episodic memory decline. Results: Our findings reveal a positive effect on spatial and episodic memory in the young population and promising outcomes in aging. Several cognitive factors (e.g., executive function, decision-making, and visual components) mediate memory performances. Findings should be taken into account for future interventions in aging. Conclusions: The present review sheds light on the key role of the sensorimotor and cognitive systems for memory rehabilitation by means of a more ecological tool such as virtual reality and stresses the importance of the body for cognition, endorsing the view of an embodied mind

    Linking primary and secondary care after psychiatric hospitalisation: comparison between transitional case management setting and routine care for common mental disorders

    Objectives. To improve engagement with care and prevent psychiatric readmission, a transitional case management intervention has been established to link with primary and secondary care. The intervention begins during hospitalisation and ends one month after discharge. The goal of this study was to assess the effectiveness of this short intervention in terms of the level of engagement with outpatient care and the rate of readmissions during one year after discharge. Methods. Individuals hospitalised with common mental disorders were randomly assigned to be discharged to routine follow up by private psychiatrists or general practitioners with (n=51) or without (n=51) the addition of a transitional case management intervention. Main outcome measures were number of contacts with outpatient care and rate of readmission during twelve months after discharge. Results. Transitional case management patients reported more contacts with care service in the period between 1 to 3 month after discharge (p = .004). Later after discharge (3-12 month), no significant differences of number of contacts remained. The transitional case management intervention had no statistically significant beneficial impact on the rate of readmission (Hazard ratio = 0.585, p = .114). Conclusions. The focus on follow-up after discharge during hospitalisation leads to an increased short term rate of engagement with ambulatory care despite no differences between the two groups after 3 month of follow-up. This short transitional intervention did however not significantly reduce the rate of readmissions during the first year following discharge. Trial registration number.     Abstract The use of bioeffectors, formally known as plant biostimulants, has become common practice in agriculture and provides a number of benefits in stimulating growth and protecting against stress. A biostimulant is loosely defined as an organic material and/or microorganism that is applied to enhance nutrient uptake, stimulate growth, enhance stress tolerance or crop quality. This review is intended to provide a broad overview of known effects of biostimulants and their ability to improve tolerance to abiotic stresses. Inoculation or application of extracts from algae or other plants have beneficial effects on growth and stress adaptation. Algal extracts, protein hydrolysates, humic and fulvic acids, and other compounded mixtures have properties beyond basic nutrition, often enhancing growth and stress tolerance. Non-pathogenic bacteria capable of colonizing roots and the rhizosphere also have a number of positive effects. These effects include higher yield, enhanced nutrient uptake and utilization, increased photosynthetic activity, and resistance to both biotic and abiotic stresses. While most biostimulants have numerous and diverse effects on plant growth, this review focuses on the bioprotective effects against abiotic stress. Agricultural biostimulants may contribute to make agriculture more sustainable and resilient and offer an alternative to synthetic protectants which have increasingly falling out of favour with consumers. An extensive review of the literature shows a clear role for a diverse number of biostimulants that have protective effects against abiotic stress but also reveals the urgent need to address the underlying mechanisms responsible for these effects.     A case of serious bleeding

    Acquired haemophilia A (AHA) is a rare disorder with a high mortality rate. It occurs due to autoantibodies against coagulation factor VIII (FVIII) which neutralise its procoagulant function resulting in severe bleeding. This disease may be associated with autoimmune diseases, malignancies, infections or medications and occurs most commonly in the elderly. Diagnosis is based on the isolated prolongation of aPTT which does not normalise after the addition of normal plasma along with reduced FVIII levels. Treatment involves eradication of antibodies and maintaining effective haemostasis during bleeding. We report a case of a 76-year-old patient with a history of haemorrhage with severe anaemia.
