Analisis Perbedaan Pada Uji Kualitas Air Sumur Di Kelurahan Madidir Ure Kota Bitung Berdasarkan Parameter Fisika
: Limited availability of raw water is one of the problems encountered in the provision of water services in Indonesia. The purpose of this study is to determine the difference in the quality of well water within 0-100 meters and within 101-200 meters from the beach by measuring physical parameters, i.e.unclearness. The number of samples in this study were 65 wells owned by residents in the Village of Madidir Ure and from those wells there are 25 with a distance of 0-100 meters and 40 with a distance of 101-200 meters from the beach.The parameters were observed referring to the Regulation of the Minister of Health of Indonesia Number 479/Menkes/Per/IV/2010 about the Terms and Water Quality Monitoring, including the physical parameters such as unclearness measured by using turbidity. Data collection methods used in this study is a cross sectional and the laboratory analysis was done in “Badan Teknik Kesehatan Lingkungan (BTKL)” Manado. The results, show water quality from the well within distance of 0-100 meters exceeds a set of maximum levels of more than 5 NTU, while the result of the water quality within 101-200 meters does not exceed the maximum of 5 NTU. Conclusion: there is a difference in the quality of water in the Village of Madidir Bitung City Ure taken from the well within a distance of 0-100 meters and the well within 101-200 meters from the beach
Jumping-based asymmetries are negatively associated with jump, change of direction, and repeated sprint performance, but not linear speed, in adolescent handball athletes
The aim of the present study was to determine the association of multi-directional jumping asymmetries with measures of physical performance. Forty-two youth handball athletes (age: 16.0 ± 1.3 years; body height: 174.11 ± 7.3 cm; body mass: 70.49 ± 13.3 kg) performed a mid-season fitness test battery consisting of single leg countermovement, lateral and broad jump tests, two change of direction speed (CODS) tests, an 8 x 10 m repeated sprint test, and a 20 m sprint. The Kappa coefficient showed only ‘slight’ levels of agreement (K range = -0.05 to 0.15), indicating that asymmetries rarely favoured the same side during each of the jump tests. The single leg countermovement jump showed significantly (p = 0.006) larger asymmetries (11.2 ± 8.4) than the broad jump (6.4 ± 4.6) and significant correlations were present between jumping asymmetries and jump (r = -0.32 to -0.52), CODS (r = 0.31 to 0.32) and repeated sprint (r = 0.35 to 0.40) performance. The findings of the present study highlight the independent nature of jumping asymmetries and associations with measures of physical performance. Practitioners are encouraged to use multiple tests to detect existing side differences and consider appropriate training interventions for the reduction of inter-limb asymmetries
The validity and reliability of a novel app for the measurement of change of direction performance
The aim of the present investigation was to analyze the validity and reliability of a novel iPhone app (CODTimer) for the measurement of total time and interlimb asymmetry in the 5 + 5 change of direction test (COD). To do so, twenty physically active adolescent athletes (age = 13.85 ± 1.34 years) performed six repetitions in the COD test while being measured with a pair of timing gates and CODTimer. A total of 120 COD times measured both with the timing gates and the app were then compared for validity and reliability purposes. There was an almost perfect correlation between the timing gates and the CODTimer app for the measurement of total time (r = 0.964; 95% Confidence interval (CI) = 0.95-1.00; Standard error of the estimate = 0.03 s.; p 0.05). Similar levels of reliability were observed between the timing gates and the app for the measurement of the 6 different trials of each participant (Timing gates: Intraclass correlation coefficient (ICC) = 0.651-0.747, Coefficient of variation (CV) = 2.6-3.5%; CODTimer: ICC = 0.671-0.840, CV = 2.2-3.2%). The results of the present study show that change of direction performance can be measured in a valid, reliable way using a novel iPhone app
Virus Replication as a Phenotypic Version of Polynucleotide Evolution
In this paper we revisit and adapt to viral evolution an approach based on
the theory of branching process advanced by Demetrius, Schuster and Sigmund
("Polynucleotide evolution and branching processes", Bull. Math. Biol. 46
(1985) 239-262), in their study of polynucleotide evolution. By taking into
account beneficial effects we obtain a non-trivial multivariate generalization
of their single-type branching process model. Perturbative techniques allows us
to obtain analytical asymptotic expressions for the main global parameters of
the model which lead to the following rigorous results: (i) a new criterion for
"no sure extinction", (ii) a generalization and proof, for this particular
class of models, of the lethal mutagenesis criterion proposed by Bull,
Sanju\'an and Wilke ("Theory of lethal mutagenesis for viruses", J. Virology 18
(2007) 2930-2939), (iii) a new proposal for the notion of relaxation time with
a quantitative prescription for its evaluation, (iv) the quantitative
description of the evolution of the expected values in in four distinct
"stages": extinction threshold, lethal mutagenesis, stationary "equilibrium"
and transient. Finally, based on these quantitative results we are able to draw

Predictability of evolutionary trajectories in fitness landscapes
Experimental studies on enzyme evolution show that only a small fraction of
all possible mutation trajectories are accessible to evolution. However, these
experiments deal with individual enzymes and explore a tiny part of the fitness
landscape. We report an exhaustive analysis of fitness landscapes constructed
with an off-lattice model of protein folding where fitness is equated with
robustness to misfolding. This model mimics the essential features of the
interactions between amino acids, is consistent with the key paradigms of
protein folding and reproduces the universal distribution of evolutionary rates
among orthologous proteins. We introduce mean path divergence as a quantitative
measure of the degree to which the starting and ending points determine the
path of evolution in fitness landscapes. Global measures of landscape roughness
are good predictors of path divergence in all studied landscapes: the mean path
divergence is greater in smooth landscapes than in rough ones. The
model-derived and experimental landscapes are significantly smoother than
random landscapes and resemble additive landscapes perturbed with moderate
amounts of noise; thus, these landscapes are substantially robust to mutation.
The model landscapes show a deficit of suboptimal peaks even compared with
noisy additive landscapes with similar overall roughness. We suggest that
smoothness and the substantial deficit of peaks in the fitness landscapes of
protein evolution are fundamental consequences of the physics of protein

Extreme genetic fragility of the HIV-1 capsid
Genetic robustness, or fragility, is defined as the ability, or lack thereof, of a biological entity to maintain function in the face of mutations. Viruses that replicate via RNA intermediates exhibit high mutation rates, and robustness should be particularly advantageous to them. The capsid (CA) domain of the HIV-1 Gag protein is under strong pressure to conserve functional roles in viral assembly, maturation, uncoating, and nuclear import. However, CA is also under strong immunological pressure to diversify. Therefore, it would be particularly advantageous for CA to evolve genetic robustness. To measure the genetic robustness of HIV-1 CA, we generated a library of single amino acid substitution mutants, encompassing almost half the residues in CA. Strikingly, we found HIV-1 CA to be the most genetically fragile protein that has been analyzed using such an approach, with 70% of mutations yielding replication-defective viruses. Although CA participates in several steps in HIV-1 replication, analysis of conditionally (temperature sensitive) and constitutively non-viable mutants revealed that the biological basis for its genetic fragility was primarily the need to coordinate the accurate and efficient assembly of mature virions. All mutations that exist in naturally occurring HIV-1 subtype B populations at a frequency >3%, and were also present in the mutant library, had fitness levels that were >40% of WT. However, a substantial fraction of mutations with high fitness did not occur in natural populations, suggesting another form of selection pressure limiting variation in vivo. Additionally, known protective CTL epitopes occurred preferentially in domains of the HIV-1 CA that were even more genetically fragile than HIV-1 CA as a whole. The extreme genetic fragility of HIV-1 CA may be one reason why cell-mediated immune responses to Gag correlate with better prognosis in HIV-1 infection, and suggests that CA is a good target for therapy and vaccination strategies

Low nadir CD4+ T-cell counts predict gut dysbiosis in HIV-1 infection
Human immunodeficiency virus (HIV)-1 infection causes severe gut and systemic immune damage, but its effects on the gut microbiome remain unclear. Previous shotgun metagenomic studies in HIV-negative subjects linked low-microbial gene counts (LGC) to gut dysbiosis in diseases featuring intestinal inflammation. Using a similar approach in 156 subjects with different HIV-1 phenotypes, we found a strong, independent, dose-effect association between nadir CD4+ T-cell counts and LGC. As in other diseases involving intestinal inflammation, the gut microbiomes of subjects with LGC were enriched in gram-negative Bacteroides, acetogenic bacteria and Proteobacteria, which are able to metabolize reactive oxygen and nitrogen species; and were depleted in oxygen-sensitive methanogenic archaea and sulfate-reducing bacteria. Interestingly, subjects with LGC also showed increased butyrate levels in direct fecal measurements, consistent with enrichment in Roseburia intestinalis despite reductions in other butyrate producers. The microbiomes of subjects with LGC were also enriched in bacterial virulence factors, as well as in genes associated with beta-lactam, lincosamide, tetracycline, and macrolide resistance. Thus, low nadir CD4+ T-cell counts, rather than HIV-1 serostatus per se, predict the presence of gut dysbiosis in HIV-1 infected subjects. Such dysbiosis does not display obvious HIV-specific features; instead, it shares many similarities with other diseases featuring gut inflammation
