A platform for screening abiotic/biotic interactions using indicator displacement assays
This paper describes novel adaptations of optically sectioned planar format assays to screen compounds for their affinities to materials surfaces. The novel platform, which we name Optical sectioned Indicator Displacement Assays (O-IDA), makes use of displaceable dyes in a format adaptable to high-throughput multi-well plate technologies. We describe two approaches; in the first the dye exhibits fluorescence in both the surface-bound and unbound state. In the second, fluorescence is lost upon displacement of the dye from the surface. Half maximal inhibitory concentration (IC50), binding affinity (Ki), and binding free energy (ΔGads) values can be extracted from the raw data. Representative biomolecules were tested for interactions with silica in aqueous environment and ZnO (0001)-Zn and (10-10) facets in a non-aqueous environment. We provide the first experimental values for both the binding of small molecules to silica and the facet-dependent ZnO binding affinity of key amino acids associated with ZnO-specific oligopeptides. The specific data will be invaluable to those studying interactions at interfaces both experimentally and computationally. O-IDA provides a general framework for the high-throughput screening of molecules binding to materials surfaces, which has important applications in drug delivery, (bio-) catalysis, biosensing and biomaterials engineering
The nature of the silicaphilic fluorescence of PDMPO
PDMPO (2-(4-pyridyl)-5-((4-(2-dimethylaminoethylaminocarbamoyl)methoxy)phenyl)oxazole), has unique silica specific fluorescence and is used in biology to understand biosilicification. This ‘silicaphilic’ fluorescence is not well understood nor is the response to local environmental variables like solvent and pH. We investigated PDMPO in a range of environments: using UV-vis and fluorescence spectroscopy supported by computational data, (SPARC, molecular dynamics simulations, density functional theory calculations), dynamic light scattering and zeta potential measurements to understand the PDMPO–silica interaction. From absorption data, PDMPO exhibited a pKa of 4.20 for PDMPOH22+ to PDMPOH+ . Fluorescence emission measurements revealed large shifts in excited state pKa* values with different behaviour when bound to silica (pKa* of 10.4). PDMPO bound to silica particles is located in the Stern layer with the dye exhibiting pH dependent depolarising motion. In aqueous solution, PDMPO showed strong chromaticity with correlation between the maximum emission wavelength for PDMPOH+* and dielectric constant (4.8–80). Additional chromatic effects were attributed to changes in solvent accessible surface area. Chromatic effects were also observed for silica bound dye which allow its use as a direct probe of bulk pH over a range far in excess of what is possible for the dye alone (3–5.2). The unique combination of chromaticity and excited state dynamics allows PDMPO to monitor pH from 3 to 13 while also reporting on surface environment opening a new frontier in the quantitative understanding of (bio)silicification
Size-Tailored Physicochemical Properties of Monodisperse Polystyrene Nanoparticles and the Nanocomposites Made Thereof
The latex monodisperse polystyrene (PS) colloids are important for different advanced applications (e.g. in coating, biotechnology etc.). However, the size dependency of their structural properties that impacts the characteristics of the nanocomposites composed thereof is largely unknown. Here, monodisperse PS nanoparticles (MPNPs) are synthesized via emulsion polymerization in five sizes (50, 150, 300, 350, and 450 nm). The size of the PS MPNPs is tailored by controlling the reaction time, temperature, and amount of surfactant and initiator. The correlation between the particle size and structural properties of the PS MPNPs is established by different thermomechanical and optical characterizations. The smaller particles (50 and 150 nm) show a lower glass transition (Tg) and thermal decomposition temperature and a lower Raman peak intensity. Yet, they trigger a higher IR absorption, thanks to a larger surface area. When incorporated in a polyvinyl alcohol (PVA) matrix, the smaller particles impart the resulting nanocomposite a higher tensile strength, and elastic and storage moduli. Whereas, they decline the elongation and loss factor. The very few examples of the MPNPs incorporated polymeric nanocomposites have been unstudied from this perspective. Thus, these tangible knowledge can profit scalable production of this kind of nanocomposite materials for different applications in a cost/energy efficient manner.Peer reviewe
Health systems strengthening to arrest the global disability burden: Empirical development of prioritised components for a global strategy for improving musculoskeletal health
Introduction Despite the profound burden of disease, a strategic global response to optimise musculoskeletal (MSK) health and guide national-level health systems strengthening priorities remains absent. Auspiced by the Global Alliance for Musculoskeletal Health (G-MUSC), we aimed to empirically derive requisite priorities and components of a strategic response to guide global and national-level action on MSK health. Methods Design: mixed-methods, three-phase design. Phase 1: qualitative study with international key informants (KIs), including patient representatives and people with lived experience. KIs characterised the contemporary landscape for MSK health and priorities for a global strategic response. Phase 2: scoping review of national health policies to identify contemporary MSK policy trends and foci. Phase 3: informed by phases 1-2, was a global eDelphi where multisectoral panellists rated and iterated a framework of priorities and detailed components/actions. Results Phase 1: 31 KIs representing 25 organisations were sampled from 20 countries (40% low and middle income (LMIC)). Inductively derived themes were used to construct a logic model to underpin latter phases, consisting of five guiding principles, eight strategic priority areas and seven accelerators for action. Phase 2: of the 165 documents identified, 41 (24.8%) from 22 countries (88% high-income countries) and 2 regions met the inclusion criteria. Eight overarching policy themes, supported by 47 subthemes, were derived, aligning closely with the logic model. Phase 3: 674 panellists from 72 countries (46% LMICs) participated in round 1 and 439 (65%) in round 2 of the eDelphi. Fifty-nine components were retained with 10 (17%) identified as essential for health systems. 97.6% and 94.8% agreed or strongly agreed the framework was valuable and credible, respectively, for health systems strengthening. Conclusion An empirically derived framework, co-designed and strongly supported by multisectoral stakeholders, can now be used as a blueprint for global and country-level responses to improve MSK health and prioritise system strengthening initiatives
Synthesis of scaffold-free, three dimensional, osteogenic constructs following culture of skeletal osteoprogenitor cells on glass surfaces
Background: efficient differentiation of stem cells into three-dimensional (3D) osteogenic construct is still an unmet challenge. These constructs can be crucial for patients with bone defects due to congenital or traumatic reasons. The modulation of cell fate and function as a consequence of interaction with the physical and chemical properties of materials is well known.Methods: the current study has examined the osteogenic differentiation potential of human skeletal populations following culture on glass surfaces, as a monolayer, or in glass tubes as a pellet culture. The 3D prosperities were assessed morphometrically and the differentiation was evaluated through molecular characterization as well as matrix formation.Results: early temporal expression of alkaline phosphatase expression of skeletal populations was observed following culture on glass surfaces. Skeletal populations seeded on glass tubes, adhered as a monolayer to the tube base and subsequently formed 3D pellets at the air -media interface. The pellets cultured on glass displayed 4.9 ± 1.3 times the weight and 2.9 ± 0.1 the diameter of their counterpart cultured in plastic tubes and displayed enhanced production of osteogenic matrix proteins, such a collagen I and osteonectin. The size and weight of the pellets correlated with surface area in contrast to cell numbers seeded. Global DNA methylation level was decreased in pellets cultured on glass. In contrast, gene expression analysis confirmed upregulation extracellular matrix proteins and osteogenesis-related growth factors.Conclusion: this simple approach to the culture of skeletal cells on glass tubes provides a scaffold-free, 3D construct platform for generating pellets enabling analysis and evaluation of tissue development and integration of multiple constructs with implications for tissue repair and regenerative application on scale-up
Execution of mantle field with multileaf collimator: A simple approach
Background: Until very recently mantle field radiotherapy remained the
gold standard for the treatment of favorable early-stage
Hodgkin\u2032s lymphoma. The classic mantle includes all the major
lymph nodes above the diaphragm and extends from the inferior portion
of the mandible to the level of the insertion of the diaphragm. Aims:
To describe a simple technique that has been devised to treat the
mantle field with the help of multileaf collimator and using computed
tomography (CT)-based treatment planning. Materials and Methods: CT
scan was performed with the patient in the supine position and the
datasets were transferred to the Eclipse\u2122 treatment planning
system. Elekta Precise\u2122 linear accelerator equipped with 40 pairs
of multileaf collimator (MLC) was used for the execution of the mantle
field. The MLC\u2032s shapes were designed to take the shape of the
conventional customized blocks used for treatment of mantle field. The
anterior mantle field was divided into three separate MLC segments with
the collimator kept at 0\ub0. The first MLC segment was shaped to
cover the neck, clavicular regions, and mediastinum. The second and the
third MLC segments covered the right and left axilla, respectively. The
posterior fields were opposed to the anterior subfields in a similar
fashion. The dose was prescribed at the midplane, using reference
points. Results and Conclusion: The technique described in this study
is very simple, easy to implement, and avoids unnecessary delay in the
execution of the mantle field. The mantle field can be easily shaped
with the multileaf collimators, without any collimator rotation
