    Becoming Asustado (Scared): An Ethnographic Contribution to a Transdisciplinary Approach to Children’s Health and Development

    We characterized and analyzed women’s narrative around the idea of becoming asustado (scared) as a cultural way of understanding why children get sick repeatedly or develop illnesses that become increasingly severe, as part of a study carried out in rural communities from the Molinos District, in the North-West of Argentina. We analyze and discuss the implications of becoming asustado for everyday child-rearing and children's health, sociability, and performance in different community endeavors from ethnographic data. We intentionally selected 15 cases elaborated based on 55 semi-structured interviews with 15 women, between 25 and 55 years old, all caregivers of children under 6 years old. Our results show that susto (fright) serves as an explanation for those people who do not fit with cultural expectations about their phenotype and social performance.     Navigational style influences eye movement pattern during exploration and learning of an environmental map

    During navigation people may adopt three different spatial styles (i.e., Landmark, Route, and Survey). Landmark style (LS) people are able to recall familiar landmarks but cannot combine them with directional information; Route style (RS) people connect landmarks to each other using egocentric information about direction; Survey style (SS) people use a map-like representation of the environment. SS individuals generally navigate better than LS and RS people. Fifty-one college students (20 LS; 17 RS, and 14 SS) took part in the experiment. The spatial cognitive style (SCS) was assessed by means of the SCS test; participants then had to learn a schematic map of a city, and after 5 min had to recall the path depicted on it. During the learning and delayed recall phases, eye-movements were recorded. Our intent was to investigate whether there is a peculiar way to explore an environmental map related to the individual's spatial style. Results support the presence of differences in the strategy used by the three spatial styles for learning the path and its delayed recall. Specifically, LS individuals produced a greater number of fixations of short duration, while the opposite eye movement pattern characterized SS individuals. Moreover, SS individuals showed a more spread and comprehensive explorative pattern of the map, while LS individuals focused their exploration on the path and related targets. RS individuals showed a pattern of exploration at a level of proficiency between LS and SS individuals. We discuss the clinical and anatomical implications of our data

    Women outperform men in remembering to remember

    The study of gender differences in prospective memory (i.e., remembering to remember) has received modest attention in the literature. The few reported studies investigating either subjective or objective evaluations of prospective memory have shown inconsistent data. In this study, we aimed to verify the presence of gender differences during the performance of an objective prospective memory test by considering the weight of specific variables such as length of delay, type of response, and type of cue. We submitted a sample of 100 healthy Italian participants (50 men and 50 women) to a test expressly developed to assess prospective memory: The Memory for Intentions Screening Test. Women performed better than men in remembering to do an event-based task (i.e., prompted by an external event) and when the task required a physical response modality. We discuss the behavioural differences that emerged by considering the possible role of sociological, biological, neuroanatomical, and methodological variables

    Ethnography and Rural Health: Trajectories of Investigation in the CalchaquĂ­es Valleys, Argentina

    En este artículo nos proponemos aportar a la construcción del panorama regional acerca de la diversidad de investigaciones contemporáneas en Antropología de la Salud en Latinoamérica, a partir del análisis de nuestra trayectoria como equipo en el estudio etnográfico de procesos de salud-enfermedad-atención (PSEA) en comunidades rurales de Molinos (Salta, Argentina). Se describen los antecedentes en el abordaje clásico de la salud y la enfermedad desde el «folklore médico» y las sucesivas innovaciones teórico-metodológicas que resultaron en transformaciones en nuestro objeto de investigación y en la adopción de un enfoque transdisciplinario y transectorial. Esto condujo a articular metodologías y datos (etnográficos, relacionales, geoespaciales, epidemiológicos, demográficos) en un encuadre de métodos mixtos. Junto a ello, reflexionamos sobre el modo en que nuestras investigaciones integraron demandas e intereses de instituciones y actores comunitarios que condujeron a diseñar proyectos colaborativos para desarrollar innovaciones tecnológicas y sociales que optimicen intervenciones de salud pública.In this article, we contribute to the construction of a regional overview of Anthropology of Health research in Latin America through the analysis of our team’s trajectory, dedicated to the ethnographic study of health-illness-attention processes in rural communities from Molinos Department (Salta, Argentina). We describe antecedents regarding classical approaches in the study of health and illness from a «medical folklore» perspective and the theoretical and methodological innovations that lead to successive transformations in our research object and the adoption of a transdisciplinary and transectoral approach. These innovations drive us to articulate several methodologies and data (ethnographic, relational, geospatial, epidemiological and demographic) in a mixed-methods design. Moreover, we reflect on how our research integrates local actors’ and institutions’ demands and interests which led us to design collaborative projects to develop technological and social innovations oriented to optimize public health interventions.Fil: Remorini, Carolina.     Ghrelin induces apoptosis in colon adenocarcinoma cells via proteasome inhibition and autophagy induction.

    Ghrelin is a metabolism-regulating hormone recently investigated for its role in cancer survival and progression. Controversially, ghrelin may act as either anti-apoptotic or pro-apoptotic factor in different cancer cells, suggesting that the effects are cell type dependent. Limited data are currently available on the effects exerted by ghrelin on intracellular proteolytic pathways in cancer. Both the lysosomal and the proteasomal systems are fundamental in cellular proliferation and apoptosis regulation. With the aim of exploring if the proteasome and autophagy may be possible targets of ghrelin in cancer, we exposed human colorectal adenocarcinoma cells to ghrelin. Preliminary in vitro fluorimetric assays evidenced for the first time a direct inhibition of 20S proteasomes by ghrelin, particularly evident for the trypsin-like activity. Moreover, 1 ÎĽM ghrelin induced apoptosis in colorectal adenocarcinoma cells by inhibiting the ubiquitin-proteasome system and by activating autophagy, with p53 having an "interactive" role

    Multifunctional graphene oxide/biopolymer composite aerogels for microcontaminants removal from drinking water

    Due to water depletion and increasing level of pollution from standard and emerging contaminants, the development of more efficient purification materials and technology for drinking water treatment is a crucial challenge to be addressed in the near future. Graphene oxide (GO) has been pointed as one of the most promising materials to build structure and devices for new adsorbents and filtration systems. Here, we analyzed two types of GO doped 3D chitosan-gelatin aerogels with GO sheets embedded in the bulk or deposited on the surface. Through combined structural characterization and adsorption tests on selected proxies of drinking water micropollutants, we compared both GO-embedded and GO-coated materials and established the best architecture for achieving enhanced removal efficiency toward con- taminants in water. To evaluate the best configuration, we studied the adsorption capacity of both systems on two organic molecules (i.e., fluoroquinolonic antibiotics ofloxacin and ciprofloxacin) and a heavy metal (lead Pb2\ufe) of great environmental relevance and with already proved high affinity for GO. The Pb monolayer maximum adsorption capacity qmax was 11.1 mg/g for embedded GO aerogels and 1.5 mg/g in coated GO-ones. Only minor differences were found for organic contaminants between coating and embedding approaches with an adsorption capacity of 5e8 mg/g and no adsorption was found for chitosan-gelatin control aerogels without GO. Finally, potential antimicrobial effects were found particularly for the GO-coated aerogels materials, thus corroborating the multifunctionality of the newly developed porous structures

    Oral contraceptives modify DNA methylation and monocyte-derived macrophage function

    Background: Fertile women may be encouraged to use contraception during clinical trials to avoid potential drug effects on fetuses. However, hormonal contraception interferes with pharmacokinetics and pharmacodynamics and modifies internal milieus. Macrophages depend on the milieu to which they are exposed. Therefore, we assessed whether macrophage function would be affected by the use of combined oral contraceptives (OCs) and if this influence depended on the androgenic or nonandrogenic properties of progestin. Methods: Healthy adult women were enrolled and stratified into two groups: women who did not use OCs (Fs) and women treated with OCs (FOCs). FOCs were further stratified as a function of androgenic (FOCA+) and non-androgenic (FOCA-) properties of progestins. Routine hematological, biochemical, inflammatory and endothelial dysfunction parameters were measured. Monocyte-derived macrophages (MDMs) were evaluated for the expression and activity of estrogen receptors and androgen receptors, and release of tumor necrosis factor α (TNFα) was measured from unstimulated and lipopolysaccharide-stimulated cells. Results: As is already known, the use of OCs changed numerous parameters: the number of lymphocytes, iron levels, total iron-binding capacity of transferrin, triglycerides, high-density lipoprotein, total cholesterol, and C-reactive protein increased, while prothrombin time and alkaline phosphatase decreased. Hormonal levels also varied: cortisol was higher in FOCs, while luteinizing hormone, folliclestimulating hormone, and testosterone were lower in FOCs. Asymmetric dimethylarginine, an index of endothelial function, was lower in FOC than in Fs, as were cysteine and bilirubin. The androgenic properties of progestins affected the activity of OCs: in particular, white blood cell count, hemoglobin, high-density lipoprotein and calcium were higher in FOCA- than in FOCA+, whereas percentage oxygen saturation and γ-glutamyl transpeptidase were lower in FOCA- than in FOCA+. Importantly, FOCs had a lower global DNA methylation, indicating that OC may have epigenetic effects on gene expression. OC did not modify the expression of androgen receptor but increased estrogen receptor α expression, more considerably in FOCA+, and decreased estrogen receptor β, more considerably in FOCA-. Importantly, the activation state of estrogen receptor β in FOCs was decreased, while estrogen receptor α was not active in either Fs or FOCs. Unstimulated MDMs obtained from FOCs showed higher release of TNFα in comparison with Fs. After lipopolysaccharide stimulation, the release of TNFα was significantly higher in Fs than in FOCs. Conclusions: OC use induced many changes in hematological and plasmatic markers, modifying hormonal levels, endothelial function, inflammation index and some redox state parameters, producing a perturbation of the internal milieu that impacted macrophagic function. In fact, different levels of estrogen receptor expression and release of TNFα were observed in macrophages derived from OC users. Some of the above activities were linked to the androgenic properties of progestin. Even though it is not known whether these effects are reversible, the results indicate that to avoid potential skewing of results only a single type of OC should be used during a single clinical trial.</br

    Amino acid-driven adsorption of emerging contaminants in water by modified graphene oxide nanosheets

    Graphene oxide nanosheets have shown promising adsorption properties toward emerging organic contaminants in drinking water. Here, we report a family of graphene oxide nanosheets covalently modified with amino acids and the study on their adsorption properties toward a mixture of selected contaminants, including pharmaceuticals, additives, and dyes. Graphene oxides modified with l-glutamic acid and l-methionine (GO-Glu and GO-Met) were synthesized and purified with a scalable and fast synthetic and purification procedure, and their structure was studied by combined X-ray photoelectron spectroscopy and elemental analysis. An amino acid loading of about 5% and a slight reduction (from 27% down to 14-20% oxygen) were found and associated with the adsorption selectivity. They were compared to unmodified GO, reduced GO (rGO), GO-lysine, and to the reference sample GO-NaOH. Each type of modified GO possesses a higher adsorption capacity toward bisphenol A (BPA), benzophenone-4 (BP4), and carbamazepine (CBZ) than standard GO and rGO, and the adsorption occurred within the first hour of contact time. The maximum adsorption capacity (estimated from the adsorption isotherms) was strictly related to the amino acid loading. Accordingly, molecular dynamics simulations highlighted higher interaction energies for the modified GOs than unmodified GO, as a result of higher van der Waals and hydrophobic interactions between the contaminants and the amino acid side chains on the nanosheet surface

    Chemical Tailoring of β-Cyclodextrin-Graphene Oxide for Enhanced Per- and Polyfluoroalkyl Substances (PFAS) Adsorption from Drinking Water

    We report on the synthesis of β-cyclodextrin (βCD) modified graphene oxide (GO) nanosheets, having different sized alkyl linkers (GO-Cn-βCD) and their exploitation as sorbent of per- and polyfluoroalkyl substances (PFAS) from drinking water. βCD were functionalized with a pending amino group, and the resulting precursors grafted to GO nanosheets by epoxide ring opening reaction. Loading of βCD units in the range 12 %–36 % was estimated by combined XPS and elemental analysis. Adsorption tests on perfluorobutanoic acid (PFBA), a particularly persistent PFAS selected as case study, revealed a strong influence of the alkyl linker length on the adsorption efficiency, with the hexyl linker derivative GO-C6-βCD outperforming both pristine GO and granular activated carbon (GAC), the standard sorbent benchmark. Molecular dynamic simulations ascribed this evidence to the favorable orientation of the βCD unit on the surface of GO which enables a strong contaminant molecules retention
