    Cdc42, dynein, and dynactin regulate MTOC reorientation independent of Rho-regulated microtubule stabilization

    AbstractIn migrating adherent cells such as fibroblasts and endothelial cells, the microtubule-organizing center (MTOC) reorients toward the leading edge [1–3]. MTOC reorientation repositions the Golgi toward the front of the cell [1] and contributes to directional migration [4]. The mechanism of MTOC reorientation and its relation to the formation of stabilized microtubules (MTs) in the leading edge, which occurs concomitantly with MTOC reorientation [3], is unknown. We show that serum and the serum lipid, lysophosphatidic acid (LPA), increased Cdc42 GTP levels and triggered MTOC reorientation in serum-starved wounded monolayers of 3T3 fibroblasts. Cdc42, but not Rho or Rac, was both sufficient and necessary for LPA-stimulated MTOC reorientation. MTOC reorientation was independent of Cdc42-induced changes in actin and was not blocked by cytochalasin D. Inhibition of dynein or dynactin blocked LPA- and Cdc42-stimulated MTOC reorientation. LPA also stimulates a Rho/mDia pathway that selectively stabilizes MTs in the leading edge [5, 6]; however, activators and inhibitors of MTOC reorientation and MT stabilization showed that each response was regulated independently. These results establish an LPA/Cdc42 signaling pathway that regulates MTOC reorientation in a dynein-dependent manner. MTOC reorientation and MT stabilization both act to polarize the MT array in migrating cells, yet these processes act independently and are regulated by separate Rho family GTPase-signaling pathways

    Stand Establishment and Persistence of Perennial Cool-Season Grasses in the Intermountain West and the Central and Northern Great Plains

    The choice of plant materials is an important component of revegetation following disturbance. To determine the utility and effectiveness of various perennial grass species for revegetation on varied landscapes, a meta analysis was used to evaluate the stand establishment and persistence of 18 perennial cool-season grass species in 34 field studies in the Intermountain and Great Plains regions of the United States under monoculture conditions. Combined across the 34 studies, stand establishment values ranged from 79% to 43% and stand persistence values ranged from 70% to 0%. Intermediate wheatgrass (Thinopyrum intermedium [Host] Barkworth & D. R. Dewey), tall wheatgrass (Thinopyrum ponticum [Podp.] Z.-W. Liu & R.-C. Wang), crested wheatgrass (Agropyron spp.), Siberian wheatgrass (Agropyron fragile [Roth] P. Candargy), and meadow brome (Bromus riparius Rehmann) possessed the highest stand establishment (≄69%). There were no significant differences among the 12 species with the largest stand persistence values. Basin wildrye (Leymus cinereus (Scribn. & Merr.) A. Love), Altai wildrye (Leymus angustus [Trin.] Pilg.), slender wheatgrass (Elymus trachycaulus [Link] Gould ex Shinners), squirreltail (Elymus spp.), and Indian ricegrass (Achnatherum hymenoides [Roem. & Schult.] Barkworth) possessed lower stand persistence (≀32%) than the majority of the other species, and Indian ricegrass (0%) possessed the lowest stand persistence of any of the species. Correlations between environmental conditions and stand establishment and persistence showed mean annual study precipitation to have the most consistent, although moderate effect (r=~0.40) for establishment and persistence. This relationship was shown by the relatively poor stand establishment and persistence of most species at sites receiving less than 310 mm of annual precipitation. These results will be a tool for land managers to make decisions concerning the importance of stand establishment, stand persistence, and annual precipitation for revegetation projects on disturbed sites

    Risk factors for postoperative complications after adrenalectomy for phaeochromocytoma: multicentre cohort study

    Background: To determine the incidence and risk factors for postoperative complications and prolonged hospital stay after adrenalectomy for phaeochromocytoma. Methods: Demographics, perioperative outcomes and complications were evaluated for consecutive patients who underwent adrenalectomy for phaeochromocytoma from 2012 to 2020 in nine high-volume UK centres. Odds ratios were calculated using multivariable models. The primary outcome was postoperative complications according to the Clavien–­­Dindo classification and secondary outcome was duration of hospital stay. Results: Data were available for 406 patients (female n = 221, 54.4 per cent). Two patients (0.5 per cent) had perioperative death, whilst 148 complications were recorded in 109 (26.8 per cent) patients.
