    Altered Innate Immune Responses in Neutrophils from Patients with Well- and Suboptimally Controlled Asthma

Respiratory infections are a major cause of asthma exacerbations where neutrophilic inflammation dominates and is associated with steroid refractory asthma. Structural airway cells in asthma differ from nonasthmatics; however it is unknown if neutrophils differ. We investigated neutrophil immune responses in patients who have good (AGood) and suboptimal (ASubopt) asthma symptom control. Methods. Peripheral blood neutrophils from AGood (ACQ 0.75, n=7), and healthy controls (HC) (n=9) were stimulated with bacterial (LPS (1 g/mL), fMLF (100 nM)), and viral (imiquimod (3 g/mL), R848 (1.5 g/mL), and poly I:C (10 g/mL)) surrogates or live rhinovirus (RV) 16 (MOI1). Cell-free supernatant was collected after 1 h for neutrophil elastase (NE) and matrix metalloproteinase- (MMP-) 9 measurements or after 24 h for CXCL8 release. Results. Constitutive NE was enhanced in AGood neutrophils compared to HC. fMLF stimulated neutrophils from ASubopt but not AGood produced 50% of HC levels. fMLF induced MMP-9 was impaired in ASubopt and AGood compared to HC. fMLF stimulated CXCL8 but not MMP-9 was positively correlated with FEV1 and FEV1/FVC. ASubopt and AGood responded similarly to other stimuli. Conclusions. Circulating neutrophils are different in asthma; however, this is likely to be related to airflow limitation rather than asthma control

    Modelling impacts of seasonal wastewater treatment plant effluent permits and biosolid substitution for phosphorus management in catchments and river systems

    The issues of diffuse and point source phosphorus (P) pollution in river systems are presented using a catchment model to assess nutrient behaviour, seasonal effluent standards and biosolid substitution. A process-based, dynamic water quality model (INCA-P) has been applied to four UK catchments, namely, the Rivers Tywi, Wensum, Lunan and Hampshire Avon, to simulate water fluxes, sediments, total phosphorus and soluble reactive phosphorus (SRP) concentrations. The model has been used to assess impacts of both agricultural runoff and point P sources from wastewater treatment plants (WWTPs) on water quality. With increasing costs for P fertilizer and P reduction at WWTPs, a strategy of recycling P from WWTPs as biosolids to substitute for fertilizers in vulnerable catchments has been investigated. Significant reductions in P concentrations are achieved if this substitution were implemented on a large scale. Reductions in SRP of between 6% and 41% can be achieved using this strategy. The effects of implementing new WWTP standards are shown to reduce SRP by 30%. Seasonal consent standards applied in only summer months could reduce SRP by 53% and achieve a substantial reduction in treatment costs year round

    Regulating, Measuring, and Modeling the Viscoelasticity of Bacterial Biofilms

    Biofilms occur in a broad range of environments under heterogeneous physicochemical conditions, such as in bioremediation plants, on surfaces of biomedical implants, and in the lungs of cystic fibrosis patients. In these scenarios, biofilms are subjected to shear forces, but the mechanical integrity of these aggregates often prevents their disruption or dispersal. Biofilms' physical robustness is the result of the multiple biopolymers secreted by constituent microbial cells which are also responsible for numerous biological functions. A better understanding of the role of these biopolymers and their response to dynamic forces is therefore crucial for understanding the interplay between biofilm structure and function. In this paper, we review experimental techniques in rheology, which help quantify the viscoelasticity of biofilms, and modeling approaches from soft matter physics that can assist our understanding of the rheological properties. We describe how these methods could be combined with synthetic biology approaches to control and investigate the effects of secreted polymers on the physical properties of biofilms. We argue that without an integrated approach of the three disciplines, the links between genetics, composition, and interaction of matrix biopolymers and the viscoelastic properties of biofilms will be much harder to uncover

    Prospective memory functioning among ecstasy/polydrug users: evidence from the Cambridge Prospective Memory Test (CAMPROMPT)

    Rationale: Prospective memory (PM) deficits in recreational drug users have been documented in recent years. However, the assessment of PM has largely been restricted to self-reported measures that fail to capture the distinction between event-based and time-based PM. The aim of the present study is to address this limitation. Objectives: Extending our previous research, we augmented the range laboratory measures of PM by employing the CAMPROMPT test battery to investigate the impact of illicit drug use on prospective remembering in a sample of cannabis only, ecstasy/polydrug and non-users of illicit drugs, separating event and time-based PM performance. We also administered measures of executive function and retrospective memory in order to establish whether ecstasy/polydrug deficits in PM were mediated by group differences in these processes. Results: Ecstasy/polydrug users performed significantly worse on both event and time-based prospective memory tasks in comparison to both cannabis only and non-user groups. Furthermore, it was found that across the whole sample, better retrospective memory and executive functioning was associated with superior PM performance. Nevertheless, this association did not mediate the drug-related effects that were observed. Consistent with our previous study, recreational use of cocaine was linked to PM deficits. Conclusions: PM deficits have again been found among ecstasy/polydrug users, which appear to be unrelated to group differences in executive function and retrospective memory. However, the possibility that these are attributable to cocaine use cannot be excluded

    Soft branes in supersymmetry-breaking backgrounds

    We revisit the analysis of effective field theories resulting from non-supersymmetric perturbations to supersymmetric flux compactifications of the type-IIB superstring with an eye towards those resulting from the backreaction of a small number of anti-D3-branes. Independently of the background, we show that the low-energy Lagrangian describing the fluctuations of a stack of probe D3-branes exhibits soft supersymmetry breaking, despite perturbations to marginal operators that were not fully considered in some previous treatments. We take this as an indication that the breaking of supersymmetry by anti-D3-branes or other sources may be spontaneous rather than explicit. In support of this, we consider the action of an anti-D3-brane probing an otherwise supersymmetric configuration and identify a candidate for the corresponding goldstino.Comment: 36+5 pages.     Non-perturbative Vacuum Destabilization and D-brane Dynamics

    We analyze the process of string vacuum destabilization due to instanton induced superpotential couplings which depend linearly on charged fields. These non-perturbative instabilities result in potentials for the D-brane moduli and lead to processes of D-brane recombination, motion and partial moduli stabilization at the non-perturbative vacuum. By using techniques of D-brane instanton calculus, we explicitly compute this scalar potential in toroidal orbifold compactifications with magnetized D-branes by summing over the possible discrete instanton configurations. We illustrate explicitly the resulting dynamics in globally consistent models. These instabilities can have phenomenological applications to breaking hidden sector gauge groups, open string moduli stabilization and supersymmetry breaking.     Wavefunctions and the Point of E8 in F-theory

    In F-theory GUTs interactions between fields are typically localised at points of enhanced symmetry in the internal dimensions implying that the coefficient of the associated operator can be studied using a local wavefunctions overlap calculation. Some F-theory SU(5) GUT theories may exhibit a maximum symmetry enhancement at a point to E8, and in this case all the operators of the theory can be associated to the same point. We take initial steps towards the study of operators in such theories. We calculate wavefunctions and their overlaps around a general point of enhancement and establish constraints on the local form of the fluxes. We then apply the general results to a simple model at a point of E8 enhancement and calculate some example operators such as Yukawa couplings and dimension-five couplings that can lead to proton decay.Comment: 46 page
