Candida spp. biotypes in the oral cavity of school children from different socioeconomic categories in Piracicaba - SP, Brazil
Duzentos e trinta e nove crianças brasileiras foram estudadas, distribuĂdas em cinco categorias socioeconĂ´micas distintas (A a E). As amostras de saliva foram analisadas avaliando-se o fluxo salivar, pH, capacidade tampĂŁo e parâmetros microbianos. Os resultados mostraram Candida em 47,3% da amostra total. A espĂ©cie mais encontrada foi C. albicans em todas as categorias socioeconĂ´micas seguidas por C. tropicalis, C. krusei e C. parapsilosis. A saliva e os parâmetros de investigacĂŁo clĂnicos como a proporcĂŁo de secreção, capacidade tampĂŁo e CFU/ml de Candida nĂŁo mostraram nenhuma correlação estatĂstica entre estes parâmetros. A freqĂĽĂŞncia de Candida nĂŁo diferiu substancialmente entre os grupos; no entanto elas foram mais acentuadas entre as categorias B e C. Entre todas as espĂ©cies, C. albicans foi a mais prevalente. Somente 5% das amostras mostraram mais de uma espĂ©cie e revelaram a presença de C. albicans associada com C. tropicalis, C. Parapsilosis ou C. krusei. Foi possĂvel detectar uma correlação significante entre os Ăndices de cárie e as categorias socioeconĂ´micas. As categorias A e E mostraram Ăndices de cárie aumentados; no entanto, a amostra da população foi considerada de risco de cárie baixo. NĂŁo havia nenhuma correlação positiva entre Candida e risco de cárie.Two hundred and thirty-nine (239) Brazilian children, distributed into five distinct socioeconomic categories (A to E) were studied. Saliva samples were analyzed as to flow rate, pH, buffer capacity and microbial parameters. The results revealed the presence of Candida spp. in 47.3% of the samples. The most commonly isolated species was C. albicans, in all socioeconomic categories, followed by C. tropicalis, C. krusei and C. parapsilosis. There was no statistical correlation between secretion rate, buffer capacity and Candida spp. CFU/ml. The prevalence of Candida spp. did not differ substantially among the groups; however the microorganisms were more detected in categories B and C. Among all species, C. albicans was the most prevalent. Only 5% of the sample presented more than one species - C. albicans associated with C. tropicalis, C. parapsilosis or C. krusei. It was possible to detect a significant correlation between caries indices and the socioeconomic categories. All categories presented increased caries indices; however the studied population was considered of low caries risk. There was no positive correlation between the presence of Candida and caries risk in the analyzed population
Influence of disinfectant solutions on test materials used for the determination of masticatory performance

The objective of this study was to evaluate the reliability of four different test materials (Optosil, Optocal, Zetapuls, and Perfil) and five disinfection protocols by aspersion and immersion (no disinfection, 2% glutaraldehyde, 2% chlorhexidine, 5.25% sodium hypochlorite, and 70% alcohol) on the MP, determined at three moments (24 hours, 15 and 60 days) after storing the fragmented blocks. MP was evaluated by calculating X50 through the sieving technique and the Rosim-Ramler equation. The weight and microbiologic count (colony forming units, CFUs) of chewed blocks were measured to identify any variations that would make MP determination unfeasible. Differences in MP were observed among the materials (p 0.05). The time and disinfection type had no influence on MP (p > 0.05). The number of CFUs differed between the nondisinfected group and all other disinfection groups at all time points (p < 0.01). No other significant difference in CFU count between disinfection groups was observed. In conclusion, disinfection did not alter the reliability of the test materials for the MP calculation for up to 60 days
Canagliflozin and renal outcomes in type 2 diabetes and nephropathy
BACKGROUND Type 2 diabetes mellitus is the leading cause of kidney failure worldwide, but few effective long-term treatments are available. In cardiovascular trials of inhibitors of sodium–glucose cotransporter 2 (SGLT2), exploratory results have suggested that such drugs may improve renal outcomes in patients with type 2 diabetes. METHODS In this double-blind, randomized trial, we assigned patients with type 2 diabetes and albuminuric chronic kidney disease to receive canagliflozin, an oral SGLT2 inhibitor, at a dose of 100 mg daily or placebo. All the patients had an estimated glomerular filtration rate (GFR) of 30 to <90 ml per minute per 1.73 m2 of body-surface area and albuminuria (ratio of albumin [mg] to creatinine [g], >300 to 5000) and were treated with renin–angiotensin system blockade. The primary outcome was a composite of end-stage kidney disease (dialysis, transplantation, or a sustained estimated GFR of <15 ml per minute per 1.73 m2), a doubling of the serum creatinine level, or death from renal or cardiovascular causes. Prespecified secondary outcomes were tested hierarchically. RESULTS The trial was stopped early after a planned interim analysis on the recommendation of the data and safety monitoring committee. At that time, 4401 patients had undergone randomization, with a median follow-up of 2.62 years. The relative risk of the primary outcome was 30% lower in the canagliflozin group than in the placebo group, with event rates of 43.2 and 61.2 per 1000 patient-years, respectively (hazard ratio, 0.70; 95% confidence interval [CI], 0.59 to 0.82; P=0.00001). The relative risk of the renal-specific composite of end-stage kidney disease, a doubling of the creatinine level, or death from renal causes was lower by 34% (hazard ratio, 0.66; 95% CI, 0.53 to 0.81; P<0.001), and the relative risk of end-stage kidney disease was lower by 32% (hazard ratio, 0.68; 95% CI, 0.54 to 0.86; P=0.002). The canagliflozin group also had a lower risk of cardiovascular death, myocardial infarction, or stroke (hazard ratio, 0.80; 95% CI, 0.67 to 0.95; P=0.01) and hospitalization for heart failure (hazard ratio, 0.61; 95% CI, 0.47 to 0.80; P<0.001). There were no significant differences in rates of amputation or fracture. CONCLUSIONS In patients with type 2 diabetes and kidney disease, the risk of kidney failure and cardiovascular events was lower in the canagliflozin group than in the placebo group at a median follow-up of 2.62 years
ATLANTIC EPIPHYTES: a data set of vascular and non-vascular epiphyte plants and lichens from the Atlantic Forest
Epiphytes are hyper-diverse and one of the frequently undervalued life forms in plant surveys and biodiversity inventories. Epiphytes of the Atlantic Forest, one of the most endangered ecosystems in the world, have high endemism and radiated recently in the Pliocene. We aimed to (1) compile an extensive Atlantic Forest data set on vascular, non-vascular plants (including hemiepiphytes), and lichen epiphyte species occurrence and abundance; (2) describe the epiphyte distribution in the Atlantic Forest, in order to indicate future sampling efforts. Our work presents the first epiphyte data set with information on abundance and occurrence of epiphyte phorophyte species. All data compiled here come from three main sources provided by the authors: published sources (comprising peer-reviewed articles, books, and theses), unpublished data, and herbarium data. We compiled a data set composed of 2,095 species, from 89,270 holo/hemiepiphyte records, in the Atlantic Forest of Brazil, Argentina, Paraguay, and Uruguay, recorded from 1824 to early 2018. Most of the records were from qualitative data (occurrence only, 88%), well distributed throughout the Atlantic Forest. For quantitative records, the most common sampling method was individual trees (71%), followed by plot sampling (19%), and transect sampling (10%). Angiosperms (81%) were the most frequently registered group, and Bromeliaceae and Orchidaceae were the families with the greatest number of records (27,272 and 21,945, respectively). Ferns and Lycophytes presented fewer records than Angiosperms, and Polypodiaceae were the most recorded family, and more concentrated in the Southern and Southeastern regions. Data on non-vascular plants and lichens were scarce, with a few disjunct records concentrated in the Northeastern region of the Atlantic Forest. For all non-vascular plant records, Lejeuneaceae, a family of liverworts, was the most recorded family. We hope that our effort to organize scattered epiphyte data help advance the knowledge of epiphyte ecology, as well as our understanding of macroecological and biogeographical patterns in the Atlantic Forest. In vitro bacterial plaque suppression and recolonization by S. mutans and S. sobrinus Supressão e recolonização de placa bacteriana por S. mutans e S. sobrinus in vitro
The in vitro study of the interactions between S. mutans and S. sobrinus is important to determine the role of these microorganisms in the formation of biofilms on dental structures and their potential to induce carious lesions. The objective of this research was to study the suppression of bacterial plaque formation and its recolonization by rifampycin-resistant S.mutans and streptomycin-resistant S. sobrinus. To study the competitive relationship between these species, previously standardized strains were incubated in media containing different fermentable carbohydrates. At determined time intervals, samples were collected from mixed cultures of S. mutans and S. sobrinus, diluted and plated on BHI-agar containing rifampycin or streptomycin to determine the number of viable cells of each species by counting colony-forming units. In order to study the bacterial colonization process and in vitro recolonization of bacterial plaque, three experiments were performed: I - co-cultivation of S. mutans and S. sobrinus; II - inoculation of bacterial plaque pre formed by S. sobrinus with S. mutans; and III - bacterial plaque pre formed by S. mutans dispersed and plated on BHI- agar containing streptomycin or rifampicin to determine the number of viable cells for each species. The results indicated a predominance of S. mutans in relation to S. sobrinus, demonstrating the capacity of S. mutans to inhibit plaque formation by S. sobrinus and recolonize the surfaces.<br>O estudo in vitro das interações entre S. mutans e S. sobrinus pode ser importante na determinação do papel desses microrganismos na formação de biofilmes nas estruturas dentais e seu potencial em induzir lesões cariosas. O objetivo da presente pesquisa foi estudar a supressĂŁo da formação da placa dental e sua recolonização por S. mutans rifampicina-resistentes e S. sobrinus estreptomicina-resistentes in vitro. Para avaliar as relações de competitividade entre essas espĂ©cies, cepas que foram previamente padronizadas foram incubadas em meio de cultura contendo diferentes carboidratos fermentáveis. Em intervalos de tempo determinados, amostras de S. mutans e S. sobrinus foram coletadas a partir de culturas mistas, diluĂdas e semeadas em placas com meio BHI-ágar contendo rifampicina ou estreptomicina para determinação do nĂşmero de cĂ©lulas viáveis de cada espĂ©cie por contagem de unidades formadoras de colĂ´nia. Para a avaliação da colonização bacteriana e recolonização da placa bacteriana in vitro, trĂŞs experimentos foram realizados: I - co-cultivo de S. mutans e S. sobrinus; II - inoculação de S. mutans em placa bacteriana prĂ©-formada por S. sobrinus; e III - placa bacteriana prĂ©-formada por S. mutans dispersada e plaqueada em meio BHI-ágar contendo estreptomicina ou rifampicina para determinação do nĂşmero de cĂ©lulas viáveis para cada espĂ©cie. Os resultados indicaram uma predominância de S. mutans em relação ao S. sobrinus, demonstrando a capacidade do S. mutans em inibir a formação de placa por S. sobrinus e recolonizar a superfĂcie dentária
Caries prevalence, level of mutans streptococci, salivary flow rate, and buffering capacity in subjects with Down syndrome
The aim of this study was to correlate caries experience and
physiological and microbiological profiles. The study group comprised
60 individuals with Down syndrome, both genders, aged from one to 48
years. The prevalence of caries was analyzed by DMFT/DMFS and dmft/dmfs
indexes. Physiological factors such as flow rate, and buffer capacity
and microbiological factor such as mutans streptococci counts were
observed. The average DMFT and DMFS were, respectively, 4.53 and 6.85,
whereas the mean dmft and dmfs values were 1.55 and 2.55, respectively.
Ninety-four percent of 18 individuals that saliva was possible to
collected presented low flow rate and only 6% of them had normal flow
rate; 44% percent had low buffer capacity, 39% had limited buffer
capacity and 16% had normal buffer capacity. Sixty percent of
individuals had high values of CFU/mL (>1.000.000 S. mutans); while
40% presented low values of microorganisms (<100.000 S. mutans).
Data of clinical, physiological and microbiological characterization
were statistically analyzed through Pearson’s correlation and
Chi-square test. A p-value d” 0.05 was considered significant.
DMFT/DMTS and dmft/dmfs indexes increased with age. Pearson’s
correlation showed significant values to DMFT/DMFS x age (r= 0.80 and
r= 0.82; p < 0.01). Flow rate and buffering capacity were low.
Individuals had high mutans streptococci counts (CFU/mL). DMFT/DMFS did
not present significant correlation with flow rate, buffering capacity
and mutans streptococci counts and no association with gender. The
prevalence of dental caries increased with age at individuals with Down
syndrome. As caries is a multifactor disease, other factors, which were
not evaluated in the present study, such as diet, host and oral hygiene
might be influencing the development of dental caries in these
individuals
Salivary biotypes of mutans Streptococci levels in schoolchildren aging 6-8 year old having a socioeconomic base
There is still little information on the mutans Streptococci biotypes
in South America, specifically in Brazil, including regional and local
information, associated to different socioeconomic categories. In this
paper, the Streptococcus mutans levels and biotypes in Brazilian
children aged 6-8 year old having a socioeconomic base was
investigated. The sampling consisted of 200 selected children belonging
to five different socioeconomic categories. Bacterial examination was
made using children saliva diluted and cultivated in Bacitracin sucrose
agar (SB-20). The plates were incubated by 48 hours/37°C at 10%
CO2 in anaerobiose and after colonies growth samples identification was
performed by biochemical tests. Results showed that 78% of the 491
tested samples harbored S. mutans, followed by S. sobrinus (11.6%), S.
rattus (4.69%), S. mutans V (2.65%), S. cricetus (1.83%) and S. ferus
(1.22%). The prevalence of Streptococcus mutans was found in every
socioeconomic level. Among children harboring multiple species, the
most frequent association detected was S. mutans/ S. sobrinus (17%)
