    Differential acquisition of amino acid and peptide enantiomers within the soil microbial community and its implications for carbon and nitrogen cycling in soil

    l-isomeric amino acids and oligopeptides are thought to represent a key nitrogen (N) source for plants and soil microorganisms, bypassing the need to take up inorganic N, whilst self-cycling of d-enantiomers within peptidoglycan-containing bacteria may provide a further short circuit within the N cycle. Here we use stable isotope profiling (SIP) to identify the fate of organic N within soil microbial communities. We followed the incorporation of 13C-labelled d- or l-labelled amino acids/peptides into phospholipid fatty acids (PLFAs). l-alanine and its peptides were taken up more rapidly than d-enantiomers by Gram-positive bacteria with 13C incorporation being predominantly into anteiso- and iso-fatty acids typically associated with Gram-positive bacteria. d-enantiomer uptake was found not to differ significantly between the microbial groups, providing little support for the view that soil bacteria may self-cycle d-forms of amino acids and peptides. There was no consistent association between peptide chain length and incorporation. The concentrations of l- and d-isomeric amino acids in soil solution were 866 nM and 72 nM, respectively. We conclude that Gram-positive bacteria appear to be the primary competitors for l-enantiomeric forms of amino acids and their peptides, but that both d- and l-enantiomers are available N and C sources for bacteria and fungi

    Effect of statins on atrial fibrillation: collaborative meta-analysis of published and unpublished evidence from randomised controlled trials

    Objective To examine whether statins can reduce the risk of atrial fibrillation. Design Meta-analysis of published and unpublished results from larger scale statin trials, with comparison of the findings against the published results from smaller scale or shorter duration studies. Data sources Medline, Embase, and Cochrane's CENTRAL up to October 2010. Unpublished data from longer term trials were obtained through contact with investigators. Study selection Randomised controlled trials comparing statin with no statin or comparing high dose versus standard dose statin; all longer term trials had at least 100 participants and at least six months' follow-up. Results In published data from 13 short term trials (4414 randomised patients, 659 events), statin treatment seemed to reduce the odds of an episode of atrial fibrillation by 39% (odds ratio 0.61, 95% confidence interval 0.51 to 0.74; P<0.001), but there was significant heterogeneity (P<0.001) between the trials. In contrast, among 22 longer term and mostly larger trials of statin versus control (105 791 randomised patients, 2535 events), statin treatment was not associated with a significant reduction in atrial fibrillation (0.95, 0.88 to 1.03; P=0.24) (P<0.001 for test of difference between the two sets of trials). Seven longer term trials of more intensive versus standard statin regimens (28 964 randomised patients and 1419 events) also showed no evidence of a reduction in the risk of atrial fibrillation (1.00, 0.90 to 1.12; P=0.99). Conclusions The suggested beneficial effect of statins on atrial fibrillation from published shorter term studies is not supported by a comprehensive review of published and unpublished evidence from larger scale trials

    A Multiwavelength Photometric Census of AGN and Star Formation Activity in the Brightest Cluster Galaxies of X-ray Selected Clusters

    Despite their reputation as being ‘red and dead’, the unique environment inhabited by brightest cluster galaxies (BCGs) can often lead to a self-regulated feedback cycle between radiatively cooling intracluster gas and star formation and active galactic nucleus (AGN) activity in the BCG. However the prevalence of ‘active’ BCGs, and details of the feedback involved, are still uncertain. We have performed an optical, UV and mid-IR photometric analysis of the BCGs in 981 clusters at 0.03 < z < 0.5, selected from the ROSAT All Sky Survey. Using Pan-STARRS PS1 3π, GALEX and WISE survey data we look for BCGs with photometric colours which deviate from that of the bulk population of passive BCGs – indicative of AGN and/or star formation activity within the BCG. We find that whilst the majority of BCGs are consistent with being passive, at least 14 per cent of our BCGs show a significant colour offset from passivity in at least one colour index. And, where available, supplementary spectroscopy reveals the majority of these particular BCGs show strong optical emission lines. On comparing BCG ‘activity’ with the X-ray luminosity of the host cluster, we find that BCGs showing a colour offset are preferentially found in the more X-ray luminous clusters, indicative of the connection between BCG ‘activity’ and the intracluster medium

    Are markers of inflammation more strongly associated with risk for fatal than for nonfatal vascular events?

    &lt;p&gt;&lt;b&gt;Background:&lt;/b&gt; Circulating inflammatory markers may more strongly relate to risk of fatal versus nonfatal cardiovascular disease (CVD) events, but robust prospective evidence is lacking. We tested whether interleukin (IL)-6, C-reactive protein (CRP), and fibrinogen more strongly associate with fatal compared to nonfatal myocardial infarction (MI) and stroke.&lt;/p&gt; &lt;p&gt;&lt;b&gt;Methods and Findings:&lt;/b&gt; In the Prospective Study of Pravastatin in the Elderly at Risk (PROSPER), baseline inflammatory markers in up to 5,680 men and women aged 70-82 y were related to risk for endpoints; nonfatal CVD (i.e., nonfatal MI and nonfatal stroke [n = 672]), fatal CVD (n = 190), death from other CV causes (n = 38), and non-CVD mortality (n = 300), over 3.2-y follow-up. Elevations in baseline IL-6 levels were significantly (p = 0.0009; competing risks model analysis) more strongly associated with fatal CVD (hazard ratio [HR] for 1 log unit increase in IL-6 1.75, 95% confidence interval [CI] 1.44-2.12) than with risk of nonfatal CVD (1.17, 95% CI 1.04-1.31), in analyses adjusted for treatment allocation. The findings were consistent in a fully adjusted model. These broad trends were similar for CRP and, to a lesser extent, for fibrinogen. The results were also similar in placebo and statin recipients (i.e., no interaction). The C-statistic for fatal CVD using traditional risk factors was significantly (+0.017; p&lt;0.0001) improved by inclusion of IL-6 but not so for nonfatal CVD events (p = 0.20).&lt;/p&gt; &lt;p&gt;&lt;b&gt;Conclusions:&lt;/b&gt; In PROSPER, inflammatory markers, in particular IL-6 and CRP, are more strongly associated with risk of fatal vascular events than nonfatal vascular events. These novel observations may have important implications for better understanding aetiology of CVD mortality, and have potential clinical relevance.&lt;/p&gt

    To wet or not to wet: that is the question

    Wetting transitions have been predicted and observed to occur for various combinations of fluids and surfaces. This paper describes the origin of such transitions, for liquid films on solid surfaces, in terms of the gas-surface interaction potentials V(r), which depend on the specific adsorption system. The transitions of light inert gases and H2 molecules on alkali metal surfaces have been explored extensively and are relatively well understood in terms of the least attractive adsorption interactions in nature. Much less thoroughly investigated are wetting transitions of Hg, water, heavy inert gases and other molecular films. The basic idea is that nonwetting occurs, for energetic reasons, if the adsorption potential's well-depth D is smaller than, or comparable to, the well-depth of the adsorbate-adsorbate mutual interaction. At the wetting temperature, Tw, the transition to wetting occurs, for entropic reasons, when the liquid's surface tension is sufficiently small that the free energy cost in forming a thick film is sufficiently compensated by the fluid- surface interaction energy. Guidelines useful for exploring wetting transitions of other systems are analyzed, in terms of generic criteria involving the "simple model", which yields results in terms of gas-surface interaction parameters and thermodynamic properties of the bulk adsorbate.Comment: Article accepted for publication in J.     Genome-wide meta-analyses reveal novel loci for verbal short-term memory and learning

    Understanding the genomic basis of memory processes may help in combating neurodegenerative disorders. Hence, we examined the associations of common genetic variants with verbal short-term memory and verbal learning in adults without dementia or stroke (N = 53,637). We identified novel loci in the intronic region of CDH18, and at 13q21 and 3p21.1, as well as an expected signal in the APOE/APOC1/TOMM40 region. These results replicated in an independent sample. Functional and bioinformatic analyses supported many of these loci and further implicated POC1. We showed that polygenic score for verbal learning associated with brain activation in right parieto-occipital region during working memory task. Finally, we showed genetic correlations of these memory traits with several neurocognitive and health outcomes. Our findings suggest a role of several genomic loci in verbal memory processes

    Trans-ancestry genome-wide association study identifies 12 genetic loci influencing blood pressure and implicates a role for DNA methylation

    We carried out a trans-ancestry genome-wide association and replication study of blood pressure phenotypes among up to 320,251 individuals of East Asian, European and South Asian ancestry. We find genetic variants at 12 new loci to be associated with blood pressure (P = 3.9 &times; 10-11 to 5.0 &times; 10-21). The sentinel blood pressure SNPs are enriched for association with DNA methylation at multiple nearby CpG sites, suggesting that, at some of the loci identified, DNA methylation may lie on the regulatory pathway linking sequence variation to blood pressure. The sentinel SNPs at the 12 new loci point to genes involved in vascular smooth muscle (IGFBP3, KCNK3, PDE3A and PRDM6) and renal (ARHGAP24, OSR1, SLC22A7 and TBX2) function. The new and known genetic variants predict increased left ventricular mass, circulating levels of NT-proBNP, and cardiovascular and all-cause mortality (P = 0.04 to 8.6 &times; 10-6). Our results provide new evidence for the role of DNA methylation in blood pressure regulation
