    Using impact response surfaces to analyse the likelihood of impacts on crop yield under probabilistic climate change

    Conventional methods of modelling impacts of future climate change on crop yields often rely on a limited selection of projections for representing uncertainties in future climate. However, large ensembles of climate projections offer an opportunity to estimate yield responses probabilistically. This study demonstrates an approach to probabilistic yield estimation using impact response surfaces (IRSs). These are constructed from a set of sensitivity simulations that explore yield responses to a wide range of changes in temperature and precipitation. Options for adaptation and different levels of future atmospheric carbon dioxide concentration [CO2] defined by representative concentration pathways (RCP4.5 and RCP8.5) were also considered. Model-based IRSs were combined with probabilistic climate projections to estimate impact likelihoods for yields of spring barley (Hordeum vulgare L.) in Finland during the 21st century. Probabilistic projections of climate for the same RCPs were overlaid on IRSs for corresponding [CO2] levels throughout the century and likelihoods of yield shortfall calculated with respect to a threshold mean yield for the baseline (1981–2010). Results suggest that cultivars combining short pre- and long post-anthesis phases together with earlier sowing dates produce the highest yields and smallest likelihoods of yield shortfall under future scenarios. Higher [CO2] levels generally compensate for yield losses due to warming under the RCPs. Yet, this does not happen fully under the more moderate warming of RCP4.5 with a weaker rise in [CO2], where there is a chance of yield shortfall throughout the century. Under the stronger warming but more rapid [CO2] increase of RCP8.5, the likelihood of yield shortfall drops to zero from mid-century onwards. Whilst the incremental IRS-based approach simplifies the temporal and cross-variable complexities of projected climate, it was found to offer a close approximation of evolving future likelihoods of yield impacts in comparison to a more conventional scenario-based approach. The IRS approach is scenario-neutral and existing plots can be used in combination with any new scenario that falls within the sensitivity range without the need to perform new runs with the impact model. A single crop model is used for demonstration, but an ensemble IRS approach could additionally capture impact model uncertainties.peerReviewe

    Neonatal Intestinal Failure Is Independently Associated With Impaired Cognitive Development Later in Childhood

    Objective: The impact of pediatric intestinal failure (IF) on neurodevelopment beyond infancy has not been systematically studied. Our aim was to evaluate cognitive and motor impairment and to identify risk factors for adverse outcomes among children with IF. Methods: We conducted a cross-sectional single-center study at the Helsinki University Children's Hospital. Patients with IF with >60 days of parental nutrition (PN) dependency aged between 3 and 16 years (n = 40) were invited to participate. The cognitive and motor skills were evaluated using validated tests: Wechsler Preschool and Primary Scale of Intelligence, 3rd edition, Wechsler Intelligence Scale for Children, 4th edition, and Movement Assessment Battery for Children, 2nd edition. Results: All the patients attending the study tests (n = 30, males = 24) were included. Their median age, gestational age, and birth weight was 7.5 (range 3-16) years, 35 (interquartile range [IQR] 28-38) weeks and 2238 (IQR 1040-3288) grams, respectively. Median duration of PN was 13 (IQR 5-37) months and 9 patients were currently on PN. Median intelligence quotient was 78 (IQR 65-91) and 10 (35%) patients had an intelligence quotient under 70 (-2 standard deviation). Significant motor impairment was detected in 10 patients (36%) and milder difficulties in 8 (28%). Adverse cognitive outcome was associated with neonatal short bowel syndrome, number of interventions under general anesthesia, and length of inpatient status, whereas adverse motor outcome was associated with prematurity. Conclusion: Clinically significant cognitive and motor impairments are alarmingly common among neonatal patients with IF. We recommend early neurodevelopmental follow-up for all children with IF.Peer reviewe

    Serum matrix metalloproteinase 8 and tissue inhibitor of metalloproteinase 1 : Potential markers for malignant transformation of recurrent respiratory papillomatosis and for prognosis of laryngeal cancer

    Background Biomarkers that could predict malignant transformation of recurrent respiratory papillomatosis (RRP) would be useful in patient follow-up. We investigated whether serum matrix metalloproteinase 8 (MMP-8) and tissue inhibitor of metalloproteinase 1 (TIMP-1) could predict malignant transformation of RRP and whether they associate with survival in laryngeal squamous cell carcinoma (LSCC) without preexisting RRP. Methods We analyzed serum MMP-8 (S-MMP-8) and serum TIMP-1 (s-TIMP-1) in 114 patients: 55 were treated for RRP and 59 for LSCC without preexisting RRP. Five patients with RRP developed LSCC during follow-up. Results Elevated S-MMP-8 level in RRP was associated with malignant transformation (P = .01). Compared to patients with RRP, S-MMP-8 in patients with LSCC was significantly higher (P <.001). Increased S-TIMP-1 level in LSCC was associated with poor overall survival (P = .02) and recurrence-free survival (P = .05). Conclusion In RRP, high S-MMP-8 may predict malignant transformation. In LSCC, elevated S-TIMP-1 is connected to poor survival.Peer reviewe

    Binge drinking and sexual assault among women in Jos Metropolis, Nigeria

    Background: Alcohol related sexual assault is a growing epidemic world wide that affects mainly women. There is urgent need to empower women to identify behaviors and situations that may predispose them to sexual victimization.Objectives: The study was carried out to determine the relationship between binge drinking and sociodemographic factors. It also assessed the relationship between binge drinking and sexual assault.Methods: The cross-sectional descriptive study was carried out in Jos North Local Government Area from March to July, 2017. Multistage sampling technique was employed to select 272 participants aged 18 years and above who consented after obtaining ethical approval.Results: The total numbers of participants were 272 females with an age range of 18-60 years. The mean age was 28.8 ± 8.6 years. The socio-demographic variables significantly associated with binge drinking were marital status (X2 = 9.847, DF = 2, p = 0.007), educational status (X2 = 10.684, DF = 3, p = 0.014) and employment status (X2= 5.122, DF = 1, p = 0.024). Binge drinking was significantly associated with sexual assault (X2 = 10.732, DF = 1, p = 0.001). Previously married were significantly more likely to binge drink compared with never married and married. Those with no formal education were more likely to binge drink compared to those with tertiary education while the unemployed were less likely to binge drink compared with the employed. The sexually assaulted (P = 0.01, OR = 2.429, CI = 1.419-4.157) were 2 times more likely to binge drink.Conclusion: There was a significant relationship between binge drinking with marital status, employment, lower level of education and sexual assault. Women should be provided with information about the safe level of alcohol consumption and the many consequences of heavy drinking including sexual assault.Keywords: Binge drinking, women, sexual assault, socio-demographic, Jo

    Rare germline copy number variants (CNVs) and breast cancer risk

    Germline copy number variants (CNVs) are pervasive in the human genome but potential disease associations with rare CNVs have not been comprehensively assessed in large datasets. We analysed rare CNVs in genes and non-coding regions for 86,788 breast cancer cases and 76,122 controls of European ancestry with genome-wide array data. Gene burden tests detected the strongest association for deletions in BRCA1 (P = 3.7E-18). Nine other genes were associated with a p-value < 0.01 including known susceptibility genes CHEK2 (P = 0.0008), ATM (P = 0.002) and BRCA2 (P = 0.008). Outside the known genes we detected associations with p-values < 0.001 for either overall or subtype-specific breast cancer at nine deletion regions and four duplication regions. Three of the deletion regions were in established common susceptibility loci. To the best of our knowledge, this is the first genome-wide analysis of rare CNVs in a large breast cancer case-control dataset. We detected associations with exonic deletions in established breast cancer susceptibility genes. We also detected suggestive associations with non-coding CNVs in known and novel loci with large effects sizes. Larger sample sizes will be required to reach robust levels of statistical significance. Dennis et al. investigate potential breast cancer associations with rare germline copy number variants (CNVs) by conducting a genome-wide analysis in a large breast cancer case-control dataset.     Pregabalin effects on neural response to emotional faces

    Pregabalin has shown promise in the treatment of anxiety disorders. Previous functional magnetic resonance imaging (fMRI) studies indicate agents used to treat anxiety, e.g., SSRIs and benzodiazepines, attenuate amygdala, insula, and medial prefrontal cortex (mPFC) activation during emotional processing. Our prior study has shown that during anticipation of an emotional stimulus, pregabalin attenuates amygdala and insula activation but increases medial PFC activation. In this study, we examined whether, similar to SSRIs and benzodiazepines, pregabalin attenuates amygdala, insula, and medial PFC during emotional face processing. Sixteen healthy volunteers underwent a double-blind within-subjects fMRI study investigating effects of placebo, 50 mg, and 200 mg pregabalin on neural activation during an emotional face-matching task. Linear mixed model analysis revealed that pregabalin dose-dependently attenuated left amygdala activation during fearful face-matching and left anterior insula activation during angry face-matching. The 50 mg dose exhibited more robust effects than the 200 mg dose in the right anterior insula and ventral ACC. Thus, pregabalin shares some similarity to SSRIs and benzodiazepines in attenuating anger and fear-related insula and amygdala activation during emotional face processing. However, there is evidence that a subclinical 50 mg dose of pregabalin produced more robust and widespread effects on neural responses in this paradigm than the more clinically relevant 200 mg dose. Taken together, pregabalin has a slightly different effect on brain activation as it relates to anticipation and emotional face processing, which may account for its unique characteristic as an agent for the treatment of anxiety disorders

    Post-release monitoring of diet profile and diet quality of reintroduced African buffalo (Syncerus caffer) in Umfurudzi Park, Zimbabwe

    Post-release monitoring of wildlife is essential to the success of ecological restoration initiatives. Translocation of wildlife to new ecosystems is associated with changes in diet profiles of individuals and ultimately animal performance, since productivity of rangelands varies in time and space. The population decline and local extinction of buffalo and other species in Umfurudzi Park, Zimbabwe, in the late 1980s led to temporary suspension of hunting activities. Recently, efforts have been made to resuscitate Umfurudzi Park through wildlife reintroductions and active ecosystem management. We assessed the diet profile and diet quality of the African buffalo (Syncerus caffer) between 2011 and 2012. A total of 42 grass species constituted the diet spectrum of buffalo. Heteropogon contortus, Eragrostis racemosa, Steroechlaena tenuifolia andThemeda triandra contributed significantly to the buffalo diet during the four seasons of the study period. Although a wide spectrum of grass species was selected in the wet season, the diversity of grass species selected was not significantly different among the seasons. Grass species crude protein deteriorated from 4.5% in the wet season to 3.5% in the dry season. Crude protein and phosphorus levels in buffalo faecal samples were within the recommended nutrient scale for southern Africa large herbivores. We conclude that feed availability and quality may not be limiting the persistence of the reintroduced buffalo. There is a need to closely monitor grass availability, dietary shifts, and forage quality over time, as well as resource partitioning with other feeding guilds

    Genome-Wide Association Study Implicates Atrial Natriuretic Peptide Rather Than B-Type Natriuretic Peptide in the Regulation of Blood Pressure in the General Population

    Background Cardiomyocytes secrete atrial natriuretic peptide (ANP) and B-type natriuretic peptide (BNP) in response to mechanical stretching, making them useful clinical biomarkers of cardiac stress. Both human and animal studies indicate a role for ANP as a regulator of blood pressure with conflicting results for BNP. Methods and Results We used genome-wide association analysis (n=6296) to study the effects of genetic variants on circulating natriuretic peptide concentrations and compared the impact of natriuretic peptide-associated genetic variants on blood pressure (n=27059). Eight independent genetic variants in 2 known (NPPA-NPPB and POC1B-GALNT4) and 1 novel locus (PPP3CC) associated with midregional proANP (MR-proANP), BNP, aminoterminal proBNP (NT-proBNP), or BNP:NT-proBNP ratio. The NPPA-NPPB locus containing the adjacent genes encoding ANP and BNP harbored 4 independent cis variants with effects specific to either midregional proANP or BNP and a rare missense single nucleotide polymorphism in NT-proBNP seriously altering its measurement. Variants near the calcineurin catalytic subunit gamma gene PPP3CC and the polypeptide N-acetylgalactosaminyltransferase 4 gene GALNT4 associated with BNP:NT-proBNP ratio but not with BNP or midregional proANP, suggesting effects on the post-translational regulation of proBNP. Out of the 8 individual variants, only those correlated with midregional proANP had a statistically significant albeit weak impact on blood pressure. The combined effect of these 3 single nucleotide polymorphisms also associated with hypertension risk (P=8.2x10(-4)). Conclusions Common genetic differences affecting the circulating concentration of ANP associated with blood pressure, whereas those affecting BNP did not, highlighting the blood pressure-lowering effect of ANP in the general population.Peer reviewe
