    A helium-3 refrigerator employing capillary confinement of liquid cryogen

    A condensation refrigerator suitable for operation in a zero gravity space environment was constructed. The condensed liquid refrigerant is confined by surface tension inside a porous metal matrix. Helium-4 and helium-3 gases were condensed and held in a copper matrix. Evaporative cooling of confined liquid helium-4 resulted in a temperature of 1.4K. Using a zeolite adsorption pump external to the cryostat, a temperature of 0.6 K was achieved through evaporative cooling of liquid helium-3. The amount of time required for complete evaporation of a controlled mass of liquid helium-4 contained in the copper matrix was measured as a function of the applied background power. For heating powers below 18 mW the measured times are consistent with the normal boiling of the confined volume of liquid refrigerant. At background powers above 18 mW the rapid rise in the temperature of the copper matrix the signature of the absence of confined liquid occurs in a time a factor of two shorter than that expected on the basis of an extrapolation of the low power data

    Whole-cell pertussis vaccine protects against Bordetella pertussis exacerbation of allergic asthma

    The prevalence of asthma and allergic disease has increased in many countries and there has been speculation that immunization promotes allergic sensitization. Bordetella pertussis infection exacerbates allergic asthmatic responses. We investigated whether whole-cell pertussis vaccine (Pw) enhanced or prevented B.pertussis induced exacerbation of allergic asthma. Groups of mice were immunized with Pw, infected with B.pertussis and/or sensitized to ovalbumin. Immunological, pathological and physiological changes were measured to assess the impact of Pw immunization on immune deviation and airway function. Pw immunization modulated ovalbumin-specific serum IgE production, and reduced local and systemic IL-13 and other cytokine responses to sensitizing allergen. Histopathological examination revealed Pw immunization reduced the severity of airway pathology and decreased bronchial hyperreactivity to methacholine exposure. Pw does not enhance airway IL-13 and consequently does not enhance but protects against the exacerbation of allergic responses. We find no evidence of Pw contributing to allergic asthma, but rather provide evidence of a mechanism whereby whole-cell pertussis vaccination has a protective role

    Prior Bordetella pertussis infection modulates allergen priming and the severity of airway pathology in a murine model of allergic asthma

    Background It has been proposed that T helper (Th)2-driven immune deviation in early life can be countered by Th1 inducing childhood infections and that such counter-regulation can protect against allergic asthma. Objective To test whether Th1-inducing infection with Bordetella pertussis protects against allergic asthma using well-characterized murine models. Methods Groups of mice were sensitized to ovalbumin (OVA) in the presence or absence of B. pertussis, a well-characterized Th1 inducing respiratory infection. Immunological, pathological and physiological parameters were measured to assess the impact of infection on immune deviation and airway function. Results We demonstrate that OVA sensitization does not affect the development of B. pertussisspecific immune responses dominated by IgG2a and IFN-g and does not impair Th1-mediated clearance of airway infection. In contrast, B. pertussis infection at the time of sensitization modulated the response to OVA and significantly reduced total serum and OVA-specific IgE. The pattern of cytokine responses, in particular OVA-specific IL-5 responses in the spleen was also modulated. However, B. pertussis did not cause global suppression as IL-10 and IL-13 levels were enhanced in OVA-stimulated spleen cell cultures and in lavage fluid from infected co-sensitized mice. Histopathological examination revealed that B. pertussis infection prior to OVA sensitization resulted in increased inflammation of bronchiolar walls with accompanying hyperplasia and mucous metaplasia of lining epithelia. These pathological changes were accompanied by increased bronchial hyper-reactivity to methacholine exposure. Conclusion Contrary to the above premise, a Th1 response induced by a common childhood infection does not protect against bronchial hyper-reactivity, but rather exacerbates the allergic asthmatic response, despite modulation of immune mediators

    Expecting a boomtown? Exploring potential housing-related impacts of large scale resource developments in Darwin

    Darwin is a city in the Northern Territory of Australia expecting a ‘boomtown’ scenario due to significant natural resource developments in the Greater Darwin area. The experience of ‘booming’ has a range of impacts upon communities. Housing is a key area of impact, particularly for the most vulnerable members of a population, who may not reap the benefits of the ‘boom’. In Darwin, new resource developments will begin in the context of record high house prices, high rents and high homelessness rates. This literature review explores what is known about the housing-related impacts of boomtowns and considers the likely housing-related impacts of a boomtown scenario in Darwin. While the city’s diverse economy and population size may provide some insulation from severe boomtown impacts, housing availability and affordability is likely to be negatively impacted. The implications of this for the most vulnerable members of the greater Darwin population require careful consideration

    Sintered Bottom and Vitrified Silica Ashes Derived from Incinerated Municipal Solid Waste as Circular Economy-Friendly Partial Replacements for Cement in Mortars

    Sintered bottom ash (SBA) and vitrified ash (VA) derived from the incineration of municipal solid waste (MSW) at 1200 °C were used in this study as replacements for Type 2 Portland cement (CEM-II) based mortars. This approach negates the need to send them to landfill, benefits the circular economy and contributes towards the decarbonisation of cementitious construction materials in response to international net zero carbon emission agendas. The material (physico-chemical) characteristics of VA and SBA were analysed before being used as partial replacements for CEM-II in mortars, whereby compressive strength (CS) was the primary criterion for assessing engineering performance. VA and SBA replaced CEM-II at dosages of 10%, 25% and 50% based on their high inorganic and pozzolanic contents; whereby the 10% and 25% replacements did not compromise mortar strength. The alkalinity and pozzolanic properties of SBA collectively indicated it has greater potential as a cementitious material over VA, which possessed a neutral pH. The 28-day CS recorded for mixtures containing 25% VA and 10% SBA were 13.74 MPa and 11.77 MPa, respectively compared with 17.06 MPa for CEM-II control samples. The use of 2% additional water in 25% SBA mortar designs improved strength further, indicating that SBA’s water retention properties permitted further hydration and strength development with curing. Microstructural, mineralogical and infrared spectroscopy analyses indicated that these strengths were owed to the formation of silicate-based hydration products. The outcomes from this study highlight that SBA has potential for replacing CEM-II and VA as a filler in cementitious mortar. Graphical Abstract: [Figure not available: see fulltext.]</p

    In Vivo Assessment of Water Content, Trans-Epidermial Water Loss and Thickness in Human Facial Skin

    Mapping facial skin in terms of its biophysical properties plays a fundamental role in many practical applications, including, among others, forensics, medical and beauty treatments, and cosmetic and restorative surgery. In this paper we present an in vivo evaluation of the water content, trans-epidermial water loss and skin thickness in six areas of the human face: cheeks, chin, forehead, lips, neck and nose. The experiments were performed on a population of healthy subjects through innovative sensing devices which enable fast yet accurate evaluations of the above parameters. A statistical analysis was carried out to determine significant differences between the facial areas investigated and clusters of statistically-indistinguishable areas. We found that water content was higher in the cheeks and neck and lower in the lips, whereas trans-epidermal water loss had higher values for the lips and lower ones for the neck. In terms of thickness the dermis exhibited three clusters, which, from thickest to thinnest were: chin and nose, cheek and forehead and lips and neck. The epidermis showed the same three clusters too, but with a different ordering in term of thickness. Finally, the stratum corneum presented two clusters: the thickest, formed by lips and neck, and the thinnest, formed by all the remaining areas. The results of this investigation can provide valuable guidelines for the evaluation of skin moisturisers and other cosmetic products, and can help guide choices in re-constructive/cosmetic surgery

    Skin Characterizations by Using Contact Capacitive Imaging and High-Resolution Ultrasound Imaging with Machine Learning Algorithms

    We present our latest research on skin characterizations by using Contact Capacitive Imaging and High-Resolution Ultrasound Imaging with Machine Learning algorithms. Contact Capacitive Imaging is a novel imaging technology based on the dielectric constant measurement principle, with which we have studied the skin water content of different skin sites and performed image classification by using pre-trained Deep Learning Neural Networks through Transfer Learning. The results show lips and nose have the lowest water content, whilst cheek, eye corner and under-eye have the highest water content. The classification yields up to 83.8% accuracy. High-Resolution Ultrasound Imaging is a state-of-the-art ultrasound technology, and can produce high-resolution images of the skin and superficial soft tissue to a vertical resolution of about 40 microns, with which we have studied the thickness of different skin layers, such as stratum corneum, epidermis and dermis, around different locations on the face and around different body parts. The results show the chin has the highest stratum corneum thickness, and the arm has the lowest stratum corneum thickness. We have also developed two feature-based image classification methods which yield promising results. The outcomes of this study could provide valuable guidelines for cosmetic/medical research, and methods developed in this study can also be extended for studying damaged skin or skin diseases. The combination of Contact Capacitive Imaging and High-Resolution Ultrasound Imaging could be a powerful tool for skin studies
