    Biological and environmental rhythms in (dark) deep-sea hydrothermal ecosystems

    During 2011, two deep-sea observatories focusing on hydrothermal vent ecology were up and running in the Atlantic (Eiffel Tower, Lucky Strike vent field) and the Northeast Pacific Ocean (NEP) (Grotto, Main Endeavour Field). Both ecological modules recorded imagery and environmental variables jointly for a time span of 23 days (7–30 October 2011) and environmental variables for up to 9 months (October 2011–June 2012). Community dynamics were assessed based on imagery analysis and rhythms in temporal variation for both fauna and environment were revealed. Tidal rhythms were found to be at play in the two settings and were most visible in temperature and tubeworm appearances (at NEP). A â€‰âˆŒâ€‰â€Ż6 h lag in tidal rhythm occurrence was observed between Pacific and Atlantic hydrothermal vents, which corresponds to the geographical distance and time delay between the two sites.info:eu-repo/semantics/publishedVersio

    A dual sensor device to estimate fluid flow velocity at diffuse hydrothermal vents

In this study, we combine two methods to accurately estimate fluid flow velocities at diffuse flow areas. While the first method uses a hot film anemometer that performs high frequency measurements, the second allows a relatively rapid assessment of fluid flow velocity through video imagery and provides in situ data to calibrate the sensor. Measurements of flow velocities on hydrothermal diffuse flow areas were obtained on the Mid-Atlantic Ridge (MAR). They range from 1.1 to 4.9 mm/sec., at the substratum level, in low temperature (4.5 to 16.4°C) diffuse flow areas from the Tour Eiffel sulfide edifice. A strong correlation was observed between fluid flow velocities and temperature, supporting the possible use of temperature as a proxy to estimate flow rates in diffuse flow areas where such a simple linear flow/temperature relation is shown to dominate.The first part of this research was sponsored by a NOAA/NURP grant award #NA96RU0221 and NSF grant OCE-9901563 to MKT and JS.     Removal processes for tributyltin during municipal wastewater treatment

Copyright @ 2013 Springer.The fate and behaviour of tributyltin (TBT) at two wastewater treatment works was examined. Both sites had two inlet streams, and each utilised high rate biological filters (biofilters) on one the streams, before treatment of the combined flows on trickling filters, with one having additional tertiary processes, installed to remove ammonia and solids. The study was designed to determine if these processes enhanced the removal of TBT. Degradation of TBT was observed in one of the biofilters, possibly as a result of temperature and hydraulic loading. At the treatment works with tertiary processes, the mass flux showed the overall removal of TBT was 68 %, predominantly due to removal with solids in the primary settlement processes. However, overall removal of 95 % was observed in the conventional trickling filter works with 94 % of this due to biodegradation in the trickling filter. The two works both removed TBT, but at different treatment stages and by different processes. Differences in the form (solubility) of TBT in the influent may have attributed to this, although further understanding of factors controlling degradation would allow for a more complete assessment of the potential of biological processes to remove hazardous compounds from wastewaters.United Utilities PL

    Evidence of a recent magma dike intrusion at the slow spreading Lucky Strike segment, Mid-Atlantic Ridge

The definitive version was published in Journal of Geophysical Research 109 (2004): B12102, doi:10.1029/2004JB003141.Mid-ocean ridge volcanic activity is the fundamental process for creation of ocean crust, yet the dynamics of magma emplacement along the slow spreading Mid-Atlantic Ridge (MAR) are largely unknown. We present acoustical, seismological, and biological evidence of a magmatic dike intrusion at the Lucky Strike segment, the first detected from the deeper sections (>1500 m) of the MAR. The dike caused the largest teleseismic earthquake swarm recorded at Lucky Strike in >20 years of seismic monitoring, and one of the largest ever recorded on the northern MAR. Hydrophone records indicate that the rate of earthquake activity decays in a nontectonic manner and that the onset of the swarm was accompanied by 30 min of broadband (>3 Hz) intrusion tremor, suggesting a volcanic origin. Two submersible investigations of high-temperature vents located at the summit of Lucky Strike Seamount 3 months and 1 year after the swarm showed a significant increase in microbial activity and diffuse venting. This magmatic episode may represent one form of volcanism along the MAR, where highly focused pockets of magma are intruded sporadically into the shallow ocean crust beneath long-lived, discrete volcanic structures recharging preexisting seafloor hydrothermal vents and ecosystems.This study was made possible through the support of the U.S. National Science Foundation (grants OCE-9811575, OCE- 0137164, and OCE-0201692) and the NOAA Vents Program

    High-temperature batteries for geothermal and oil/gas borehole applications

    A literature survey and technical evaluation was carried out of past and present battery technologies with the goal of identifying appropriate candidates for use in geothermal borehole and, to a lesser extent, oil/gas boreholes. The various constraints that are posed by such an environment are discussed. The promise as well as the limitations of various candidate technologies are presented. Data for limited testing of a number of candidate systems are presented and the areas for additional future work are detailed. The use of low-temperature molten salts shows the most promise for such applications and includes those that are liquid at room temperature. The greatest challenges are to develop an appropriate electrochemical couple that is kinetically stable with the most promising electrolytes--both organic as well as inorganic--over the wide operating window that spans both borehole environments

    Towards the Establishment of a Porcine Model to Study Human Amebiasis

    BACKGROUND: Entamoeba histolytica is an important parasite of the human intestine. Its life cycle is monoxenous with two stages: (i) the trophozoite, growing in the intestine and (ii) the cyst corresponding to the dissemination stage. The trophozoite in the intestine can live as a commensal leading to asymptomatic infection or as a tissue invasive form producing mucosal ulcers and liver abscesses. There is no animal model mimicking the whole disease cycle. Most of the biological information on E. histolytica has been obtained from trophozoite adapted to axenic culture. The reproduction of intestinal amebiasis in an animal model is difficult while for liver amebiasis there are well-described rodent models. During this study, we worked on the assessment of pigs as a new potential model to study amebiasis. METHODOLOGY/PRINCIPAL FINDINGS: We first co-cultured trophozoites of E. histolytica with porcine colonic fragments and observed a disruption of the mucosal architecture. Then, we showed that outbred pigs can be used to reproduce some lesions associated with human amebiasis. A detailed analysis was performed using a washed closed-jejunal loops model. In loops inoculated with virulent amebas a severe acute ulcerative jejunitis was observed with large hemorrhagic lesions 14 days post-inoculation associated with the presence of the trophozoites in the depth of the mucosa in two out four animals. Furthermore, typical large sized hepatic abscesses were observed in the liver of one animal 7 days post-injection in the portal vein and the liver parenchyma. CONCLUSIONS: The pig model could help with simultaneously studying intestinal and extraintestinal lesion development

    It was reported previously that two capsular polysaccharides, types 5 and 8 (CP5 and CP8), account for 70 to 80% of Staphylococcus aureus strains isolated from human and animal sources. The capsular material has been shown to play a part in virulence and in resistance to phagocytosis. With a view to investigating the role that CP plays in pathogenicity or protection, relative measurement of cell-associated CP is desirable. Flow cytometry, which permits the analysis of individual bacteria, was used to that end. Thirty isolates expressing CP5, of human (n = 7) and animal (cow, n = 11; goat, n = 3; swine, n = 3; hen, n = 3; and rabbit, n = 3) origin, were cultivated on either brain heart infusion agar (BHI) or modified medium 110 (mod 110) agar. Staphylococci were incubated with a mouse anti-CP5 monoclonal antibody (an immunoglobulin M, which does not react with staphylococcal protein A) and then stained with a fluorescein-labeled anti-murine antibody. The bacteria were washed, sonicated, and analyzed by flow cytometry. Except for three isolates, the expression of cell-bound CP5 was higher when bacteria were cultivated on mod 110 than when they were cultivated on BHI. We found a wide intraisolate phenotypic heterogeneity in surface-exposed CP5 in many strains, which appeared as mixtures of stained and unstained bacteria. Four main patterns could be distinguished on the basis of the distribution of the fluorescence of individual bacteria within the strain population as a function of growth medium. Great variations in both percentages of stained bacteria and fluorescence intensity were recorded among strains regardless of their origin. Flow cytometry analysis provided information on both the relative amounts and the distribution patterns of the surface expression of CP. This information is potentially useful for the evaluation of the part played by the capsule in the interaction of bacteria with host cells or for the study of the activities of antibodies to this target antigen, such as opsonization or prevention of adherence
