Teff (Eragrostis tef (Zucc.) Trotter) fodder yield and quality as affected by cutting frequency
Teff (Eragrostis tef (Zucc.) Trotter) is a well-adapted, fast-growing crop with
competitive forage quality as its nutritive value for livestock fodder is similar to other grasses
utilized as hay or ensiled feeds. Two field experiments were conducted from May to October
2017 in order to determine the effect of cutting frequency on yield and quality of teff (Eragrostis
tef) as fodder crop under Mediterranean climatic conditions. The agronomic performance and
nutritive value of teff was analyzed in order to define alternatives to local forages for animal
feeding in the Mediterranean region. The experiments conducted at two sites (Western and
Central Greece) were laid out in a completely randomized design with three replicates and three
cutting frequencies (10, 20, 30 days’ interval between cuttings - F10, F20, and F30, respectively).
The results of this study demonstrate that the cutting interval has a marked effect on the fodder
yield and quality. The highest total dry matter yield (6,322–6,778 kg ha-1) was found in F10
treatment. In terms of qualitative characteristics, the highest fat levels and lowest ADF levels was
found in F30 treatment, the highest protein levels and the lowest NDF levels in F10 treatment,
and the highest levels of fibrous substances, ADF and NDF, in F20 treatment. Data suggest that
teff could be successfully integrated into Mediterannean grasslands with the prospect for
improving their nutritional quality and the possibility for increasing protein yield through the
application of frequent cuttings
Seismic retrofitting of RC frames with RC infilling
The effectiveness of seismic retrofitting of multi-storey multi-bay RC-frame buildings, by converting selected bays into new walls through infilling with RC, was studied experimentally at the ELSA facility at JRC, Ispra, and the results are reported here. The full-scale model tested with the pseudo-dynamic method consisted of two parallel frames, linked through 0.15m slabs, having three bays each (8.5m long), with the central bay (2.5m) infilled with RC wall, and being four storeys tall (12m). The frames were designed and detailed for gravity loads only and are typical of similar frames built in Cyprus in the 1970's. Different connection details and reinforcement percentages for the two infilled frames were used in order to study the effects of these parameters. The results of the pseudo-dynamic and cyclic testing performed are presented and conclusions are drawn
Conserved structured domains in plant non-coding RNA enod40, their evolution and recruitment of sequences from transposable elements
Plant long noncoding RNA enod40 is involved in the regulation of symbiotic associations with bacteria, in particular, in nitrogen-fixing root nodules of legumes, and with fungi in phosphate-acquiring arbuscular mycorrhizae formed by various plants. The presence of enod40 genes in plants that do not form such symbioses indicates its other roles in cell physiology. The molecular mechanisms of enod40 RNA function are poorly understood. Enod40 RNAs form several structured domains, conserved to different extents. Due to relatively low sequence similarity, identification of enod40 sequences in plant genomes is not straightforward, and many enod40 genes remain unannotated even in complete genomes. Here, we used comparative structure analysis and sequence similarity searches in order to locate enod40 genes and determine enod40 RNA structures in nitrogen-fixing clade plants and in grasses. The structures combine conserved features with considerable diversity of structural elements, including insertions of structured domain modules originating from transposable elements. Remarkably, these insertions contain sequences similar to tandem repeats and several stem-loops are homologous to microRNA precursors.</p
Fatty Acid Methyl Esters as Biosolvents of Epoxy Resins: A Physicochemical Study
The C8 to C18 fatty acid methyl esters (FAME) have been compared as solvents for two epoxy resin pre-polymers, bisphenol A diglycidyl ether (DGEBA) and triglycidyl paminophenol ether (TGPA). It was found that the solubilization limits vary according to the ester and that methyl caprylate is the best solvent of both resins. To explain these solubility performances, physical and chemical properties of FAME were studied, such as the Hansen parameters, viscosity, binary diffusion coefficient and vaporization enthalpy. Determination of the physicochemical parameters of FAME was carried out by laboratory experimentations and by calculation from bibliographic data. The Hansen parameters of FAME and epoxy resins pre-polymers were theoretically and experimentally determined. The FAME chain length showed a long dependence on the binary diffusion parameters and kinematic viscosity, which are mass and momentum transport properties. Moreover, the vaporization enthalpy of these compounds was directly correlated with the solubilization limits
Lotus japonicus contains two distinct ENOD40 genes that are expressed in symbiotic, nonsymbiotic, and embryonic tissues
ENOD40, an early nodulin gene, has been postulated to play a significant role in legume root nodule ontogenesis, We have isolated two distinct ENOD40 genes from Lotus japonicus, The transcribed regions of the two ENOD40 genes share 65% homology, while the two promoters showed no significant homology, Both transcripts encode a putative dodecapeptide similar to that identified in other legumes forming determinate nodules, Both ENOD40 genes are coordinately expressed following inoculation of roots with Mesorhizobium loti or treatment with purified Nod factors. In the former case, mRNA accumulation could be detected up to 10 days following inoculation while in the latter case the accumulation was transient. High levels of both ENOD40 gene transcripts were found in nonsymbiotic tissues such as stems, fully developed flowers, green seed pods, and hypocotyls, A relatively lower level of both transcripts was observed in leaves, roots, and cotyledons. In situ hybridization studies revealed that, in mature nodules, transcripts of both ENOD40 genes accumulate in the nodule vascular system; additionally, in young seed pods strong signal is observed in the ovule, particularly In the phloem and epithelium, as well as in globular stage embryos.Animal science
Reversed-phase high-performance liquid chromatography–fluorescence detection for the analysis of glutathione and its precursor γ-glutamyl cysteine in wines and model wines supplemented with oenological inactive dry yeast preparations
El pdf del artículo es la versión pre-print.A reversed-phase high-performance liquid chromatography-fluorescence detection methodology involving a pre-column derivatization procedure using 2,3-naphtalenedialdehyde in the presence of 5 and 0. 5 mM of dithiothreitol to determine total and reduced glutathione (GSH) and γ-glutamyl-cysteine (γ-glu-cys) in musts and wines has been set up and validated. The proposed method showed good linearity (R 2 >99% for reduced and total GSH, and R 2 >98% for γ-glu-cys) in synthetic wines, over a wide range of concentration (0-10 mg L -1). The limits of detection for reduced GSH in synthetic and real wines were almost the same (0. 13 and 0. 15 mg L -1, respectively) and slightly higher for γ-glu-cys (0. 24 mg L -1). The application of the method allowed knowing, for the first time, the amount of total and reduced GSH and γ-glu-cys released into synthetic wines by oenological preparations of commercial inactive dry yeast (IDY). In addition, the evolution of these three compounds during the winemaking and shelf life (0-9 months) of an industrially manufactured rosé wine supplemented with a GSH-enriched IDY showed that although GSH is effectively released from IDY, it is rapidly oxidized during alcoholic fermentation, contributing to the higher total GSH content determined in wines supplemented with GSH-enriched IDYs compared to control wines. © 2011 Springer Science+Business Media, LLC.IAO and JJRB acknowledge CAM and CSIC for their respective research grants. Natural orifice surgery: initial clinical experience
Natural orifice translumenal endoscopic surgery (NOTES) has moved quickly from preclinical investigation to clinical implementation. However, several major technical problems limit clinical NOTES including safe access, retraction and dissection of the gallbladder, and clipping of key structures. This study aimed to identify challenges and develop solutions for NOTES during the initial clinical experience.
Under an Institutional Review Board (IRB)-approved protocol, patients consented to a natural orifice operation for removal of either the gallbladder or the appendix via either the vagina or the stomach using a single umbilical trocar for safety and assistance.
Nine transvaginal cholecystectomies, one transgastric appendectomy, and one transvaginal appendectomy have been completed to date. All but one patient were discharged on postoperative day 1 as per protocol. No complications occurred.
The limited initial evidence from this study demonstrates that NOTES is feasible and safe. The addition of an umbilical trocar is a bridge allowing safe performance of NOTES procedures until better instruments become available. The addition of a flexible long grasper through the vagina and a flexible operating platform through the stomach has enabled the performance of NOTES in a safe and easily reproducible manner. The use of a uterine manipulator has facilitated visualization of the cul de sac in women with a uterus to allow for safe transvaginal access
Sexuality and Body Image After Uterine Artery Embolization and Hysterectomy in the Treatment of Uterine Fibroids: A Randomized Comparison
In this paper the effect of uterine artery embolization (UAE) on sexual functioning and body image is investigated in a randomized comparison to hysterectomy for symptomatic uterine fibroids. The EMbolization versus hysterectoMY (EMMY) trial is a randomized controlled study, conducted at 28 Dutch hospitals. Patients were allocated hysterectomy (n = 89) or UAE (n = 88). Two validated questionnaires (the Sexual Activity Questionnaire [SAQ] and the Body Image Scale [BIS]) were completed by all patients at baseline, 6 weeks, and 6, 12, 18, and 24 months after treatment. Repeated measurements on SAQ scores revealed no differences between the groups. There was a trend toward improved sexual function in both groups at 2 years, although this failed to reach statistical significance except for the dimensions discomfort and habit in the UAE arm. Overall quality of sexual life deteriorated in a minority of cases at all time points, with no significant differences between the groups (at 24 months: UAE, 29.3%, versus hysterectomy, 23.5%; p = 0.32). At 24 months the BIS score had improved in both groups compared to baseline, but the change was only significant in the UAE group (p = 0.009). In conclusion, at 24 months no differences in sexuality and body image were observed between the UAE and the hysterectomy group. On average, both after UAE and hysterectomy sexual functioning and body image scores improved, but significantly so only after UAE
Identification of conserved secondary structures and expansion segments in enod40 RNAs reveals new enod40 homologues in plants
enod40 is a plant gene that participates in the regulation of symbiotic interaction between leguminous plants and bacteria or fungi. Furthermore, it has been suggested to play a general role in non-symbiotic plant development. Although enod40 seems to have multiple functions, being present in many land plants, the molecular mechanisms of its activity are unclear; they may be determined though, by short peptides and/or RNA structures encoded in the enod40 genes. We utilized conserved RNA structures in enod40 sequences to search nucleotide sequence databases and identified a number of new enod40 homologues in plant species that belong to known, but also, to yet unknown enod40-containing plant families. RNA secondary structure predictions and comparative sequence analysis of enod40 RNAs allowed us to determine the most conserved structural features, present in all known enod40 genes. Remarkably, the topology and evolution of one of the conserved structural domains are similar to those of the expansion segments found in structural RNAs such as rRNAs, RNase P and SRP RNAs. Surprisingly, the enod40 RNA structural elements are much more stronger conserved than the encoded peptides. This finding suggests that some general functions of enod40 gene could be determined by the encoded RNA structure, whereas short peptides may be responsible for more diverse functions found only in certain plant families
