    The effect of prior victory or defeat in the same site as that of subsequent encounter on the determination of dyadic dominance in the domestic hen

    We examined the effect of prior victory or defeat in the same site as that of a subsequent encounter on the outcome of dyadic encounter of domestic hens by placing them in two situations: In the first set of dyads, two unacquainted hens having experienced prior victory were introduced in the site where one had experienced victory. In the second set, two unacquainted hens having experienced defeat were introduced in the site where one had recently lost. Results indicate that victories are equally shared between individuals with prior victory experiences, while familiarity with the meeting site did not give any advantage. However, hens having previously lost were disadvantaged when the encounter occurred in the same site as that of their prior defeat. This demonstrates that previous social experience in a site is more important on the outcome of subsequent encounters for losers than winners. Losers seem to associate the site with the stressful effect of losing or being more easily dominated

    This study simultaneously varied experiences of recent victory or defeat, 2-hour familiarity with the meeting place, and hen weight in order to understand their combined effects on the establishment of dyadic dominance relationships between hens not previously acquainted with each other. Three kinds of encounters were arranged: (i) a previous winner unfamiliar with the meeting place met a previous loser familiar with the meeting place (n =28 dyads); (ii) a previous winner met a previous loser, both unfamiliar with the meeting place (n=27); (iii) a previous winner familiar with the meeting place encountered a previous loser unfamiliar with the meeting place (n=28). The weight asymmetry was combined with these three types of encounters by selecting hens showing various weight differences, in favour of the recent loser in 54 dyads and of the recent winner in 29 dyads. Results indicate that recent victory or defeat experience significantly affected the outcome. Even an important weight asymmetry, or familiarity with the meeting place were not sufficient for a hen recently defeated to overcome an opponent that was previously victorious. A 2-hour period of familiarization with the meeting place did not provide any significant advantage over unfamiliarity. Although a significant relationship was found to exist between comb and wattles areas and the initial and final statuses, examination of partial correlations indicates that the influence was from initial status to final status, rather than from comb and wattles to final status. These results suggest that more importance should be attributed to recent social experience in comparison to intrinsic factors in determining dyadic dominance in the hen

    Recent victory or defeat experiences and 2-hour familiarity with the meeting place were combined with size differences in order to better understand their effects on the behaviour leading to the establishment of dyadic dominance relationships between hens not previously acquainted with each other. Three kinds of encounters were videotaped: (i) a previous winner unfamiliar with the meeting place met a previous loser familiar for 2 hours with the meeting place (n = 12 dyads); (ii) as in (i) but both were unfamiliar with the meeting place (n=12); (iii) as in (i) but the previous winner was familiar with the meeting place while the previous loser was unfamiliar (n=13). The weight asymmetry was combined with these three types of encounters by selecting hens of various weight differences: in 29 dyads the recent loser was heavier than the recent winner and in 8 dyads it was the reverse. Recent experience had a major influence upon both agonistic behaviour and dominance outcome. Hens that were familiar with the meeting site initiated attacks more frequently than their unfamiliar opponent but did not win significantly more often. Recent experience and site familiarity could be used to identify 80% of future initiators. Once the first aggressive behaviour had been initiated, it led to victory of its initiator in 92% of cases. Weight was not found to influence agonistic behaviour nor dominance outcome. However, hens with superior comb and wattles areas won significantly more initial meetings than opponents with smaller ones. In the final encounters, victory also went more frequently to the bird showing larger comb and wattles, which happened also to be the previous dominant in a majority of cases. The use of higher-order partial correlations as an ex post facto control for comb and wattles indicates that they were not influential upon agonistic behaviour nor on dominance outcome, but were simply co-selected with the selection of victorious and defeated birds in the first phase of the experiment designed to let hens acquire recent victory/defeat experience

    Coherent use of information by hens observing their former dominant defeating or being defeated by a stranger.

    This study examines the role of observation during the formation of triads in female domestic hens. Results indicate that during hierarchy formation, a hen observing agonistic interactions and conflict settlement between its former dominant and a stranger uses this information when in turn confronted by the latter. Under a first condition (E, N=15 triads), bystanders witnessed their prior dominant being defeated by a stranger before being introduced to them. In a second condition (C1, N=16 triads), bystanders witnessed the victory of their prior dominant over a stranger. In a third condition (C2, N=15 triads), bystanders witnessed two strangers establishing a dominance relationship before being introduced to their prior dominant and to a stranger the former had just defeated. The behavioural strategies of bystanders depended on the issue of the conflict they had witnessed. Bystanders of the E condition behaved as having no chance of defeating the stranger. They never initiated an attack against it, and upon being attacked, readily submitted in turn to the stranger. On the contrary, bystanders of the C1 condition behaved as having some chances against the stranger. They initiated attacks in 50% of cases, and won 50% of conflicts against the stranger. Under condition C2, bystanders first initiated contact with the strangers in only 27% of cases, which approximates the average of their chances for defeating the stranger. However, bystanders finally defeated the strangers in 40% of cases. These results suggest that bystanders of conditions E and C1 gained some information on the relationship existing between their prior dominant and the stranger and that they used it coherently, perhaps through transitive inference, thus contributing to the existence of transitive relationships within the triads. This methods paper details the first attempt at monitoring bank erosion, flow and suspended sediment at a site during flooding on the Mekong River induced by the passage of tropical cyclones. We deployed integrated mobile laser scanning (MLS) and multibeam echo sounding (MBES), alongside acoustic Doppler current profiling (aDcp), to directly measure changes in river bank and bed at high (~0.05 m) spatial resolution, in conjunction with measurements of flow and suspended sediment dynamics. We outline the methodological steps used to collect and process this complex point cloud data, and detail the procedures used to process and calibrate the aDcp flow and sediment flux data. A comparison with conventional remote sensing methods of estimating bank erosion, using aerial images and Landsat imagery, reveals that traditional techniques are error prone at the high temporal resolutions required to quantify the patterns and volumes of bank erosion induced by the passage of individual flood events. Our analysis reveals the importance of cyclone-driven flood events in causing high rates of erosion and suspended sediment transport, with a c. twofold increase in bank erosion volumes and a fourfold increase in suspended sediment volumes in the cyclone-affected wet season.     Coastal cliff ground motions and response to extreme storm waves

    Coastal cliff erosion from storm waves is observed worldwide, but the processes are notoriously difficult to measure during extreme storm wave conditions when most erosion normally occurs, limiting our understanding of cliff processes. Over January–February 2014, during the largest Atlantic storms in at least 60 years with deepwater significant wave heights of 6–8 m, cliff-top ground motions showed vertical ground displacements in excess of 50–100 ”m; an order of magnitude larger than observations made previously. Repeat terrestrial laser scanner surveys over a 2 week period encompassing the extreme storms gave a cliff face volume loss of 2 orders of magnitude larger than the long-term erosion rate. The results imply that erosion of coastal cliffs exposed to extreme storm waves is highly episodic and that long-term rates of cliff erosion will depend on the frequency and severity of extreme storm wave impacts

    Testing fluvial erosion models using the transient response of bedrock rivers to tectonic forcing in the Apennines, Italy

    The transient response of bedrock rivers to a drop in base level can be used to discriminate between competing fluvial erosion models. However, some recent studies of bedrock erosion conclude that transient river long profiles can be approximately characterized by a transport‐limited erosion model, while other authors suggest that a detachment‐limited model best explains their field data. The difference is thought to be due to the relative volume of sediment being fluxed through the fluvial system. Using a pragmatic approach, we address this debate by testing the ability of end‐member fluvial erosion models to reproduce the well‐documented evolution of three catchments in the central Apennines (Italy) which have been perturbed to various extents by an independently constrained increase in relative uplift rate. The transport‐limited model is unable to account for the catchments’response to the increase in uplift rate, consistent with the observed low rates of sediment supply to the channels. Instead, a detachment‐limited model with a threshold corresponding to the field‐derived median grain size of the sediment plus a slope‐dependent channel width satisfactorily reproduces the overall convex long profiles along the studied rivers. Importantly, we find that the prefactor in the hydraulic scaling relationship is uplift dependent, leading to landscapes responding faster the higher the uplift rate, consistent with field observations. We conclude that a slope‐ dependent channel width and an entrainment/erosion threshold are necessary ingredients when modeling landscape evolution or mapping the distribution of fluvial erosion rates in areas where the rate of sediment supply to channels is low

    Control of breathing and respiratory gas exchange in high-altitude ducks native to the Andes

    We examined the control of breathing and respiratory gas exchange in six species of high-altitude duck that independently colonized the high Andes. We compared ducks from high-altitude populations in Peru (Lake Titicaca at ∌3800 m above sea level; Chancay River at ∌3000–4100 m) with closely related populations or species from low altitude. Hypoxic ventilatory responses were measured shortly after capture at the native altitude. In general, ducks responded to acute hypoxia with robust increases in total ventilation and pulmonary O2 extraction. O2 consumption rates were maintained or increased slightly in acute hypoxia, despite ∌1–2°C reductions in body temperature in most species. Two high-altitude taxa – yellow-billed pintail and torrent duck – exhibited higher total ventilation than their low-altitude counterparts, and yellow-billed pintail exhibited greater increases in pulmonary O2 extraction in severe hypoxia. In contrast, three other high-altitude taxa – Andean ruddy duck, Andean cinnamon teal and speckled teal – had similar or slightly reduced total ventilation and pulmonary O2 extraction compared with low-altitude relatives. Arterial O2 saturation (SaO2) was elevated in yellow-billed pintails at moderate levels of hypoxia, but there were no differences in SaO2 in other high-altitude taxa compared with their close relatives. This finding suggests that improvements in SaO2 in hypoxia can require increases in both breathing and haemoglobin–O2 affinity, because the yellow-billed pintail was the only high-altitude duck with concurrent increases in both traits compared with its low-altitude relative. Overall, our results suggest that distinct physiological strategies for coping with hypoxia can exist across different high-altitude lineages, even among those inhabiting very similar high-altitude habitats

    Fusing simulation and experiment: The effect of mutations on the structure and activity of the influenza fusion peptide

    During the infection process, the influenza fusion peptide (FP) inserts into the host membrane, playing a crucial role in the fusion process between the viral and host membranes. In this work we used a combination of simulation and experimental techniques to analyse the molecular details of this process, which are largely unknown. Although the FP structure has been obtained by NMR in detergent micelles, there is no atomic structure information in membranes. To answer this question, we performed bias-exchange metadynamics (BE-META) simulations, which showed that the lowest energy states of the membrane-inserted FP correspond to helical-hairpin conformations similar to that observed in micelles. BE-META simulations of the G1V, W14A, G12A/G13A and G4A/G8A/G16A/G20A mutants revealed that all the mutations affect the peptide's free energy landscape. A FRET-based analysis showed that all the mutants had a reduced fusogenic activity relative to the WT, in particular the mutants G12A/G13A and G4A/G8A/G16A/G20A. According to our results, one of the major causes of the lower activity of these mutants is their lower membrane affinity, which results in a lower concentration of peptide in the bilayer. These findings contribute to a better understanding of the influenza fusion process and open new routes for future studies

    Atomic-Resolution Simulations Predict a Transition State for Vesicle Fusion Defined by Contact of a Few Lipid Tails

    Membrane fusion is essential to both cellular vesicle trafficking and infection by enveloped viruses. While the fusion protein assemblies that catalyze fusion are readily identifiable, the specific activities of the proteins involved and nature of the membrane changes they induce remain unknown. Here, we use many atomic-resolution simulations of vesicle fusion to examine the molecular mechanisms for fusion in detail. We employ committor analysis for these million-atom vesicle fusion simulations to identify a transition state for fusion stalk formation. In our simulations, this transition state occurs when the bulk properties of each lipid bilayer remain in a lamellar state but a few hydrophobic tails bulge into the hydrophilic interface layer and make contact to nucleate a stalk. Additional simulations of influenza fusion peptides in lipid bilayers show that the peptides promote similar local protrusion of lipid tails. Comparing these two sets of simulations, we obtain a common set of structural changes between the transition state for stalk formation and the local environment of peptides known to catalyze fusion. Our results thus suggest that the specific molecular properties of individual lipids are highly important to vesicle fusion and yield an explicit structural model that could help explain the mechanism of catalysis by fusion proteins
