    Spatiotemporal Analysis of Hydration Mechanism in Sodium Alginate Matrix Tablets

    Methods of spatiotemporal characterization of nonequilibrated polymer based matrices are still immature and imperfect. The purpose of the study was to develop the methodology for the spatiotemporal characterization of water transport and properties in alginate tablets under hydration. The regions of low water content were spatially and temporally sampled using Karl Fisher and Differential Scanning Callorimetry (spatial distribution of freezing/nonfreezing water) with spatial resolution of 1 mm. In the regions of high water content, where sampling was infeasible due to gel/sol consistency, magnetic resonance imaging (MRI) enabled characterization with an order of magnitude higher spatial resolution. The minimally hydrated layer (MHL), infiltration layer (IL) and fully hydrated layer (FHL) were identified in the unilaterally hydrated matrices. The MHL gained water from the first hour of incubation (5-10% w/w) and at 4 h total water content was 29-39% with nonfreezing pool of 28-29%. The water content in the IL was 45-47% and at 4 h it reached ~50% with the nonfreezing pool of 28% and T2 relaxation time < 10 ms. The FHL consisted of gel and sol layer with water content of 85-86% with a nonfreezing pool of 11% at 4 h and T2 in the range 20-200 ms. Hybrid destructive/nondestructive analysis of alginate matrices under hydration was proposed. It allowed assessing the temporal changes of water distribution, its mobility and interaction with matrices in identified layers

    Interleukin-17 (IL-17) is a proinflammatory cytokine produced, although not exclusively, by T helper 17 recently identified as a distinct T helper lineage mediating tissue inflammation. IL-17 is known to be involved in a number of chronic disorders although the mechanisms regulating its production in inflammatory disease are still unclear. The beneficial properties of the polyphenolic compound resveratrol including its anti-inflammatory, antioxidant, and antitumor effects, its role in the aging process and in the prevention of heart and neurodegenerative diseases are well-known. In addition, derivatives of resveratrol, including glucosylated molecules as polydatin have been linked to similar beneficial effects. We have investigated the effects of resveratrol and polydatin on the in vitro production of IL-17 in a model of inflammation in vitro. The results obtained by activated human peripheral blood mononuclear cells, stimulated with anti-CD3/anti-CD28 monoclonal antibodies and treated with these polyphenolic compounds at different concentrations show that both decrease IL-17 production in a concentration-dependent manner. This study confirms the anti-inflammatory activity of resveratrol and its derivatives and suggests a potential clinical relevance in the therapy of inflammatory diseases

    Are dietary supplements dangerous? Assessing how society perceives the usage of dietary supplements - results of the preliminary analysis of nationwide survey data

    Background: Dietary supplements (DSs) are concentrated sources of nutrients and/or other substances, whose purpose is to supplement the normal diet. Their consumption in Poland has been on the rise, leading to numerous concerns. However, current legislation does not provide any background for a relevant pharmacovigilance system. Therefore, little is known about their adverse effects (AEs) or patterns of use in Poland. To shed some light on this, we conducted a nationwide survey. Aim of the study: To assess the prevalence of, and the reasons for, the use of dietary supplements in Poland via national survey with a collection and an analyse of supposed adverse effects of dietary supplements. Material and methods: The study was conducted using an online survey based on the original questionnaire. In this report, we present the results of the analysis of the first 200 responses. Results: As many as 55.5% (111) of respondents were ‘definitely sure’ or ‘rather sure’ about DSs safety. However, 70.5% (141) believed that they could have negative effects. Over 3/4 (153) of respondents used DSs themselves, and 61.0% (122) used them within last 12 months. Of those using DSs, 11.1% (17) reported diverse Aes, with GI tract irritation and mental disturbances being the most common (experienced by 35.3% (6), and 17.6% (3) of those reporting AE s, respectively). Conclusions: Respondents had mixed feelings about DS safety. Despite that, they used these products frequently. Of a note is high incidence of adverse effects experienced by DS users. This undoubtedly points to the need for a relevant pharmacovigilance system

    Hydration Patterns in Sodium Alginate Polymeric Matrix Tablets The Result of Drug Substance Incorporation

    The purpose was to show, using destructive/nondestructive methods, that the interplay between water, tablet structure, and composition determine the unique spatiotemporal hydration pattern of polymer-based matrices. The tablets containing a 1:1 w/w mixture of sodium alginate with salicylic acid (ALG/SA) or sodium salicylate (ALG/SNA) were studied using Karl Fischer titration, differential scanning calorimetry, x-ray microtomography, and magnetic resonance imaging. As the principal results, matrix specific features were detected, e.g., 'locking' of the internal part of the matrix (ALG/SA); existence of lamellar region associated with detection of free/freezing water (ALG/SA); existence of water penetrating the matrix forming specific region preceding infiltration layer (ALG/SNA); switch in the onset temperature of endothermic water peak associated with an increase in the fraction of non-freezing water weight per dry matrix weight in the infiltration layer (ALG/SNA). The existence of complicated spatiotemporal hydration patterns influenced by matrix composition and molecular properties of constituents has been demonstrated

    Analizamos la evolución de las publicaciones, incluidas en MEDLINE, LILACS y Sociological Abstracts, sobre las Reformas de Salud en el mundo y las influencias que determinan su orientación y distribución en el período de 1990-2004. En total se seleccionaron 8.729 publicaciones. Los principios de “Sostenibilidad” y “Calidad y Efectividad” son los más atendidos, existiendo distintos patrones de atención, dependiendo de regiones y países. De 199 países, el 61% tiene referencia sobre sus procesos de reforma, siendo Estados Unidos y Gran Bretaña los que agrupan la mayor cantidad. Se observó que existen fuertes influencias para el estudio de las Reformas de Salud, provenientes de los patrones Británico y Norteamericano de atención a los Principios de Reforma. Esto puede estar limitando la visibilidad científica de cuestiones como la equidad, participación y eficiencia.The authors analyze the evolution in publications indexed in MEDLINE, LILACS, and Sociological Abstracts concerning health reforms around the world and the determinants of their orientation and distribution from 1990 to 2004. Of 199 countries, 61% included references as to their health reform processes, with the largest numbers in the United States and the Great Britain. The British and U.S. standards for attention to health reform principles displayed strong influences on the study of health reforms elsewhere. This may limit the scientific visibility of issues like equity, participation, and efficiency
