    Dextran-curcumin nanoparticles as a methotrexate delivery vehicle: A step forward in breast cancer combination therapy

    With the aim to effectively deliver methotrexate (MTX) to breast cancer cells, we designed a nanocarrier system (DC) derived from the self-assembly of a dextran-curcumin conjugate prepared via enzyme chemistry with immobilized laccase acting as a solid biocatalyst. Nanoparticles consisted of homogeneously dispersed nanospheres with a mean diameter of 290 nm, as characterized by combined transmission electron microscopy and dynamic light scattering investigations. DC was able to control the MTX release overtime (t1/2 value of 310 min), with cell internalization studies proving its presence inside MCF-7 cytoplasm. Finally, improved MTX efficacy was obtained in viability assays, and attributed to the synergy of curcumin moieties and loaded MTX as underlined by a combination index (CI) < 1

    Reactive crystallisation process for magnesium recovery from concentrated brines

    Seawater brines, generated either by natural or anthropic processes, often cause significant environmental issues related to their disposal. A clear example is the case of brines from desalination plants, which can have severe environmental impacts on the receiving water body. On the other side, brines can represent a rich and appealing source of raw materials, especially when they are very concentrated, as it happens with bitterns (i.e. exhausted brines) produced in saltworks. In particular, magnesium concentration can reach values up to 30-40 kg/m3 of brine, which is 20-30 times that of typical seawater. An experimental campaign has been carried out in the present work for assessing the potentials for magnesium recovery from concentrated brines. Real brines were collected from the final basins of the saltworks operating in the district of Trapani (Sicily - Italy). Experiments were performed both in a semi-batch and in a continuous 5 litre crystalliser operating by a reactive precipitation process. NaOH solutions were adopted as standard alkaline reactant in order to assess the influence of all operating parameters and reactor configuration on the recovery efficiency and purity of the Mg(OH)2 powder produced. Results have highlighted a very promising strategy for the recovery of Mg from concentrated brines, which could be scaled-up and applied to a number of different scenarios, including existing saltworks and newly designed integrated cycles for Zero Liquid Discharge desalination

    Fishery-Induced Selection for Slow Somatic Growth in European Eel

    Both theoretical and experimental studies have shown that fishing mortality can induce adaptive responses in body growth rates of fishes in the opposite direction of natural selection. We compared body growth rates in European eel (Anguilla anguilla) from three Mediterranean stocks subject to different fishing pressure. Results are consistent with the hypotheses that i) fast-growing individuals are more likely to survive until sexual maturity than slow-growing ones under natural conditions (no fishing) and ii) fishing can select for slow-growing individuals by removing fast-growing ones. Although the possibility of human-induced evolution seems remote for a panmictic species like such as the European eel, further research is desirable to assess the implications of the intensive exploitation on this critically endangered fish

    Simple gene transfer technique based on I-SceI meganuclease and cytoplasmic injection in IVF bovine embryos

    Although transgenic methods in mammals are inefficient, an easy and highly efficient transgenesis systemusing I-SceI meganuclease (intron-encoded endonuclease fromS. cerevisiae)was recently described in Xenopus. The method consisted of injection into fertilized eggs of an I-SceI reaction mixture with a plasmid DNA carrying the transgene, flanked by the meganuclease recognition sites (pIS). In the present study, the effects of I-SceI on gene transfer were tested apparently for the first time in mammals, in particular, in cattle. Various conditions were evaluated, including three concentrations of the plasmid pIS Pax6egfp, carrying I-SceI recognition sites flanking egfp under Pax6 promoter and two injection times (before IVM and after IVF) of pIS CAGegfp, carrying I-SceI sites fanking egfp under CAG promoter. In addition, the quantity of transgenewasmeasured using quantitative polymerase chain reaction, and presence of transgene signals was evaluated using fluorescence in situ hybridization analysis. Transgene expression rateswere higher (P< 0.05) for groups treated after IVF (79.1%, 91/115 and 63.0%, 75/ 119) than before IVM (32.6%, 31/95 and 34.7%, 33/95), with and without I-SceI, respectively. Interestingly, injectionwith pIS plus I-SceI after IVF increased frequency (P<0.05) of nonmosaic transgene-expressing embryos (58.3%, 42/72 vs. 29.7%, 25/84) for pIS plus I-SceI and pIS alone. Based on fluorescence in situ hybridization analysis, injectionwith I-SceI increased (P<0.05) the proportion of embryos with transgene signals in all blastomeres compared with pIS alone (44.0%,11/25 vs. 6.9%, 2/29) for pIS plus I-SceI and pIS alone. In addition, transgene copy number was numerically higher for the group treated with pIS plus I-SceI compared with pIS alone.     Trace elements and isotopes analyses on historical samples of white sharks from the Mediterranean Sea

    The white shark Carcharodon carcharias has been present in the Mediterranean Sea since 3.2 million years ago. Nevertheless, the current population shows a low genetic variability suggesting an endangered small population, on which there is scarce information regarding ecotoxicology or trophic ecology. Given that white shark's sightings are rare in the Mediterranean and the possibility of obtaining samples is highly limited, the aim of this research was to provide general information regarding the concentration of trace elements and stable isotopes (delta N-15 and delta C-13). Laboratory analyses were performed on 18 and 12 subsamples from two different white sharks' vertebrae obtained from two adult specimens caught in 1987, in Favignana Island, Italy. Perforations were made along the vertebrae to describe both trace elements and stable isotopes at different life stages. A total of 38 trace elements were analysed, in which the highest concentrations were found in Fe, Sr, U, Pb, and Zn. The fluctuations of these elements during the ontogeny of both individuals could have been related to changes in diet and environment, although the specific origin remains unknown. Regarding stable isotopes, the vertebrae from the male showed an isotopic range from 9.6 parts per thousand to 10.8 parts per thousand (delta N-15) and from -16.5 parts per thousand to -13.0 parts per thousand (delta C-13) with a mean +/- SD value of 10.3 +/- 0.4 parts per thousand for delta N-15 and -14.6 +/- 1.3 parts per thousand for delta C-13; whereas the female vertebrae had an isotopic range from 9.8 parts per thousand to 11.1 parts per thousand (delta N-15) and from -16.9 parts per thousand to -15.0 parts per thousand (delta C-13), with a mean +/- SD value of 10.8 +/- 0.6 parts per thousand for delta N-15 and -15.8 +/- 0.8 parts per thousand for delta C-13. There were no significant delta N-15 differences (U = 6, p = 0.07346) between the two individuals. However, there were just significant differences in delta C-13 (t = -1.8, p = 0.049256), which could suggest sexual segregation in terms of habitat use and feeding habits

    Predicting Head Pose in Dyadic Conversation

    Natural movement plays a significant role in realistic speech animation. Numerous studies have demonstrated the contribution visual cues make to the degree we, as human observers, find an animation acceptable. Rigid head motion is one visual mode that universally co-occurs with speech, and so it is a reasonable strategy to seek features from the speech mode to predict the head pose. Several previous authors have shown that prediction is possible, but experiments are typically confined to rigidly produced dialogue. Expressive, emotive and prosodic speech exhibit motion patterns that are far more difficult to predict with considerable variation in expected head pose. People involved in dyadic conversation adapt speech and head motion in response to the others’ speech and head motion. Using Deep Bi-Directional Long Short Term Memory (BLSTM) neural networks, we demonstrate that it is possible to predict not just the head motion of the speaker, but also the head motion of the listener from the speech signal

    A Database of Full Body Virtual Interactions Annotated with Expressivity Scores

    Abstract Recent technologies enable the exploitation of full body expressions in applications such as interactive arts but are still limited in terms of dyadic subtle interaction patterns. Our project aims at full body expressive interactions between a user and an autonomous virtual agent. The currently available databases do not contain full body expressivity and interaction patterns via avatars. In this paper, we describe a protocol defined to collect a database to study expressive full-body dyadic interactions. We detail the coding scheme for manually annotating the collected videos. Reliability measures for global annotations of expressivity and interaction are also provided

    The Impact of Lockdown on Couples' Sex Lives

    Background: the aim of this study was to perform an Italian telematics survey analysis on the changes in couples' sex lives during the coronavirus disease 2019 (COVID-19) lockdown. Methods: a multicenter cross sectional study was conducted on people sexually active and in stable relationships for at least 6 months. To evaluate male and female sexual dysfunctions, we used the international index of erectile function (IIEF-15) and the female sexual function index (FSFI), respectively; marital quality and stability were evaluated by the marital adjustment test (items 10-15); to evaluate the severity of anxiety symptoms, we used the Hamilton Anxiety Rating Scale. The effects of the quarantine on couples' relationships was assessed with questions created in-house. Results: we included 2149 participants. The sex lives improved for 49% of participants, particularly those in cohabitation; for 29% it deteriorated, while for 22% of participants it did not change. Women who responded that their sex lives deteriorated had no sexual dysfunction, but they had anxiety, tension, fear, and insomnia. Contrarily, men who reported deteriorating sex lives had erectile dysfunctions and orgasmic disorders. In both genders, being unemployed or smart working, or having sons were risk factors for worsening the couples' sex lives. Conclusion: this study should encourage evaluation of the long-term effects of COVID-19 on the sex lives of couples
