Effect of Water Vapor Amount in a Hydrogenous Atmosphere on Structure and Properties of Nickel-Zirconia Anode Materials for Solid Oxide Fuel Cells
Nickel-zirconia anode ceramics of YSZ-NiO system for solid oxide fuel cells (SOFCs) has been investigated. A series of specimens were singly reduced in hydrogenous atmosphere (the Ar-5 vol%H2 mixture) at 600°C under the pressure of 0.15 MPa or subjected to reduction-oxidation (redox) cyclic treatment at 600°C. Influence of water vapor concentration in hydrogenous atmosphere on structure and properties of the materials was studied. Based on structural changes in the as-received material it was revealed that a small amount of water vapor in Ar-5 vol% H2 mixture (water vapor pressure below 0.03 MPa) accelerates a reduction of the nickel phase at 600°C with formation of nanopores on tiny Ni particles. A higher concentration of water vapor (the pressure above 0.03-0.05 MPa) causes a converse change in the reduction kinetics. For as-received material, such an amount of water vapor in the mixture is an obstacle for its reduction. For the material treated by redox cycling, better physical and mechanical properties were revealed after dwelling at 600°C in a water depleted gas mixture. Based on the SEM microscopy and the data on the conductivity and strength, the dual effect of water vapor on durability of a nickel-zirconia anode is discussed

Biosynthesis of landomycin E deoxysugar part in Streptomyces globisporus 1912: sequencing and analysis of lndZ1 and lndZ3 genes
DNA fragment of landomycin E biosynthesis gene cluster 1.5 kb in size has been completely sequenced and two open reading frames were identified. Gene lndZ1 resembles NDP-hexose-3,5-epimerase and lndZ3 is similar to NDP-hexose-4-ketoreductases. Heterologous expression reveals ancient properties of Tei3 - A VanS ortholog from the teicoplanin producer Actinoplanes teichomyceticus
Glycopeptide antibiotics (GPAs) are among the most clinically successful antimicrobials.
GPAs inhibit cell-wall biosynthesis in Gram-positive bacteria via binding to lipid II. Natural GPAs are produced by various actinobacteria. Being themselves Gram-positives, the GPA producers evolved sophisticated mechanisms of self-resistance to avoid suicide during antibiotic production. These self-resistance genes are considered the primary source of GPA resistance genes actually spreading among pathogenic enterococci and staphylococci. The GPA-resistance mechanism in Actinoplanes teichomyceticus—the producer of the last-resort-drug teicoplanin—has been intensively studied in recent years, posing relevant questions about the role of Tei3 sensor histidine kinase. In
the current work, the molecular properties of Tei3 were investigated. The setup of a GPA-responsive assay system in the model Streptomyces coelicolor allowed us to demonstrate that Tei3 functions as a non-inducible kinase, conferring high levels of GPA resistance in A. teichomyceticus. The expression of different truncated versions of tei3 in S. coelicolor indicated that both the transmembrane helices of Tei3 are crucial for proper functioning. Finally, a hybrid gene was constructed, coding for a chimera protein
ombining the Tei3 sensor domain with the kinase domain of VanS, with the latter
being the inducible Tei3 ortholog from S. coelicolor. Surprisingly, such a chimera did not respond to teicoplanin, but indeed to the related GPA A40926. Coupling these experimental results with a further in silico analysis, a novel scenario on GPA-resistance and biosynthetic genes co-evolution in A teichomyceticus was hereby proposed

SEQUENCING AND ANALYSIS OF PUTATIVE 3D-4H RING CYCLASE GENE lndF OF STREPTOMYCES GLOBISPORUS 1912 LANDOMYCIN E BIOSYNTHETIC GENE CLUSTER
DNA fragment of landomycin E biosynthesis gene cluster 700 bp in size has been completely sequenced. 3?-end of lndE (oxygenase) was identified, 5?-end of lndA (ketosynthase) and entire ORF for previously not sequenced lanF homologue, lndF. Analysis of lndF putative translation product revealed that it is highly conservative in comparison with other known cyclases from antibiotic biosynthesis gene clusters. Unlike urdamycin, jadomycin biosynthetic clusters, lndF and lndA are uncoupled, as well as genes lanF and lanA. Genes lndF and lndA are not preceded by direct or inverted repeats, putative sites for binding of transcriptional activator LndI. In contrast, lanF is flanked at it's 5?-end by three direct repeats, possible target for regulatory protein LanI.L. Castro is acknowledged for help in sequencing.

Electrochemical diagnostics of the structural-phase state of pipelanes steels at durable operation
Показано, що внаслідок неоднорідного напружено-деформованого стану згинів парогонів зі сталей 12Х1МФ та 15Х1М1Ф у різних зонах (розтягнута, середня, стиснена) процеси трансформації структури і накопичення пошкоджуваності в процесі експлуатації проходить з різною інтенсивністю. Встановлено, що стандартні механічні характеристики, передбачені діючими нормативними документами, виявляють слабку чутливість (знижуються в 1.03-1.24 раза) до процесів зміни мікроструктури та накопичення пошкоджень за умови повзучості сталей 12Х1МФ і 15Х1М1Ф. З іншого боку, зміни фазового складу та мікроструктури парогінних сталей суттєво впливають на їх електрохімічну поведінку, зокрема, на швидкість репасивації свіжоутвореної поверхні Vr.It is shown that the processes of structure transformation and damage evolution under service conditions passes with different intensity as a result of nonuniform stress-strain state in different zones (tensile, middle, compressed) of the bends of 12Х1МФ or 15Х1М1Ф steel pipelines. It is established that standard mechanical properties, provided by current normative documents, exhibit weak sensitivity (decrease in 1.03–1.24 times) to the processes of the microstructure change and damage accumulation under creep conditions of 12Х1МФ and 15Х1М1Ф steels. Preconditioning of the YSZ-NiO Fuel Cell Anode in Hydrogenous Atmospheres Containing Water Vapor
Heterologous cross-expression of oxygenase and glycosyltransferase genes in streptomycetes, producing angucyclic antibiotics
The heterologous expression of oxygenase genes urdM, ovmOIII, lanM2, lndZ5 and glycosyltransferase gene urdGT2 in different angucycline producers of genus Streptomyces has been carried out. The introduction of genes urdM, lanM2 and urdGT2 results in the accumulation of new glycosylated compounds in several strains under investigation. A number of processed recombinant strains display streptomycin sensitivity and decrease in total antibacterial activity. The obtained data is an evidence of changes in antibiotics production, resulting from post polyketide synthase (postPKS) tailoring gene expression across streptomycete strains. Our study demonstrates the potential of post PKS tailoring genes for generation of novel bioactive metabolites.Здійснено гетерологічну експресію генів оксигеназ urdM, ovmOIII, lanM2, lndZ5 та глікозилтрансферази urdGT2 у різних видів продуцентів ангуциклінів з роду Streptomyces. Введення генів urdM, lanM2 та urdGT2 у деякі з досліджуваних штамів зумовлює накопичення ними нових глікозильованих сполук. Виявлено виникнення чутливості до стрептоміцину та різке зниження загальної антибактерійної активності у деяких із одержаних рекомбінантних штамів. Отримані дані є свідченням модифікації продукції антибіотиків, яка є наслідком експресії генів постполікетидного синтезу (пост-ПКС) у штамах стрептоміцетів. Наші дослідження демонструють потенціал генів пост-ПКС у створенні нових біологічно активних метаболітів.Проведена гетерологическая экспрессия генов оксигеназ urdM, ovmOIII, lanM2, lndZ5 и гликозилтрансферазы urdGT2 в разных видах продуцентов ангуциклинов из рода Streptomyces. Введение генов urdM, lanM2 и urdGT2 в некоторые из исследуемых штаммов обусловливает накопление ими новых гликозилированных соединений. Обнаружено появление чувствительности к стрептомицину и резкое снижение общей антибактериальной активности у некоторых из полученных рекомбинантных штаммов. Представленные данные свидетельствуют о модификации продукции антибиотиков, которая является следствием экспрессии генов пост-поликетидного синтеза (пост-ПКС) в штаммах стрептомицетов.
