    Content validation and inter-rater reliability of a protocol for the precision assessment of boccia players

    Purpose: To verify a protocol's content validation and reliability for assessing the precision in the throw of Paralympic boccia in two different steps. Methods: The study was divided into two steps: In step 1, the perception of 15 boccia coaches was evaluated using a questionnaire containing 6 questions about the pertinence of the protocol on a Likert scale (1 to 5). In step 2, reliability was evaluated by two researchers, applying the protocol with two targets (0.5 and 1.0, targets) to verify the short precision (SP), average precision (AP), long precision (LP), and total precision (TP) of 23 boccia athletes (BC1 = 5; BC2 = 7; BC3 = 1; BC4 = 10) in tournaments of the modality. The Content Validity Index (CVI) calculation was applied for the electronic questionnaire, the Intraclass Correlation Coefficient (ICC) for the agreement between evaluators, and the T-Test for the difference between means. p < .05 was adopted. Results: In the result of the CVI, reliability was noticed through the experts' evaluation (Question1 = 1.0; Q2 = 0.93; Q3 = 0.80; Q4 = 0.80; Q5 = 0.93; Q6 = 0.93). There was an agreement between the evaluators by the ICC in the 0.5 targets for SP (p < .01), AP (p < .01), LP (p < .01), and TP (p < .01), and in the 1.0 target for SP (p < .01), AP (p < .01), LP (p < .01), and TP (p < .01). No differences were found between the means in the t-test. Conclusion: It was demonstrated that the protocol meets the established reliability and content validity criteria, allowing its practical use to evaluate the precision in the boccia

    Essential role of the czc determinant for cadmium, cobalt and zinc resistance in Gluconacetobacter diazotrophicus PAl 5

    The mechanisms of cadmium, cobalt and zinc resistance were characterized in the plant-growth-promoting bacterium Gluconacetobacter diazotrophicus PAl 5. The resistance level of the wild-type strain was evaluated through the establishment of minimum inhibitory concentrations (MIC) of the soluble compounds CdCl2&middot;H2O, CoCl2&middot;6H2O and ZnCl2. Gluconacetobacter diazotrophicus PAl 5 was resistant to high concentrations of Cd, Co and Zn, with MICs of 1.2, 20 and 20 mM, respectively. Screening of an insertion library from transposon EZ-Tn5&lt;R6Kyori/KAN-2&gt; in the presence of ZnO revealed that the mutant GDP30H3 was unable to grow in the presence of the compound. This mutant was also highly sensitive to CdCl2&middot;H2O, CoCl2&middot;6H2O and ZnCl2. Molecular characterization established that the mutation affected the czcA gene, which encodes a protein involved in metal efflux. In silico analysis showed that czcA is a component of the czcCBARS operon together with four other genes. This work provides evidence of the high tolerance of G. diazotrophicus PAl 5 to heavy metalsand that czc is a determinant for metal resistance in this bacterium. [Int Microbiol 2012; 15(2):69-78

    Contact endoscopy (microstomatoscopy) in oral lesions: evaluation of the method

    OBJETIVO: Descrever as dificuldades da endoscopia de contato aplicada à boca e ao lábio (microestomatoscopia de contato) e aplicar nas lesões do lábio os critérios usados na endoscopia de contato de outras topografias. MÉTODO: Estudo prospectivo utilizando o endoscópio de contato Storz 8715A e o corante azul de toluidina. Vinte exames foram feitos para aprendizado da técnica e depois mais 31 (21 lesões do lábio e 10 da boca), todos exames in vivo. Os critérios avaliados foram: Celularidade, Arquitetura, Estratificação e Morfologia. RESULTADOS: A falta de nitidez, os tremores finos e o deslizamento do aparelho dificultaram o procedimento em 100% dos casos, mas permitiram um exame adequado. Os 13 diagnósticos de malignidade das lesões do lábio apresentaram todos os critérios alterados. CONCLUSÃO: as dificuldades encontradas foram o contato do aparelho com a superfície anatômica, o deslizamento do aparelho, os tremores finos e a falta de nitidez da imagem, que não inviabilizaram a realização e a interpretação do exame. Os critérios são aplicáveis às lesões da boca e do lábio inferior.BACKGROUND: 1- To describe difficulties of contact endoscopy related to mouth and lip (contact microstomatoscopy) and 2- apply the criteria used in contact endoscopy of other topographies when lip lesions are diagnosed. METHODS: A prospective study was performed with the contact endoscope Storz 8715A and toluidine blue was used. Twenty examinations were accomplished in order to learn the technique; after that, other 31 were included (21 lip lesions and 10 oral lesions), all in vivo examinations. The criteria assessed were: cellularity, architecture, stratification and morphology. RESULTS: Lack of image clearness, slight tremors and sliding of the equipment made the procedure difficult in 100% of the cases; even though, accomplishment of proper examination was obtained. The 13 cases of lip malignancy lesions had the criteria altered as a whole. CoNCLUSION: Difficulties found were the contact of the equipment with anatomic surface, the sliding of the equipment, the slight tremors, and the lack of image clearness. Nevertheless, such elements did not interrupt the accomplishment and the examination analysis. The criteria are applied to oral and lower lip lesions

    Dengue Infection Increases the Locomotor Activity of Aedes aegypti Females

    BACKGROUND: Aedes aegypti is the main vector of the virus causing Dengue fever, a disease that has increased dramatically in importance in recent decades, affecting many tropical and sub-tropical areas of the globe. It is known that viruses and other parasites can potentially alter vector behavior. We investigated whether infection with Dengue virus modifies the behavior of Aedes aegypti females with respect to their activity level. METHODS/PRINCIPAL FINDINGS: We carried out intrathoracic Dengue 2 virus (DENV-2) infections in Aedes aegypti females and recorded their locomotor activity behavior. We observed an increase of up to ∼50% in the activity of infected mosquitoes compared to the uninfected controls. CONCLUSIONS: Dengue infection alters mosquito locomotor activity behavior. We speculate that the higher levels of activity observed in infected Aedes aegypti females might involve the circadian clock. Further studies are needed to assess whether this behavioral change could have implications for the dynamics of Dengue virus transmission

    Alternative respiratory chain enzymes: Therapeutic potential and possible pitfalls

    The alternative respiratory chain (aRC), comprising the alternative NADH dehydrogenases (NDX) and quinone oxidases (AOX), is found in microbes, fungi and plants, where it buffers stresses arising from restrictions on electron flow in the oxidative phosphorylation system. The aRC enzymes are also found in species belonging to most metazoan phyla, including some chordates and arthropods species, although not in vertebrates or in Drosophila. We postulated that the aRC enzymes might be deployed to alleviate pathological stresses arising from mitochondrial dysfunction in a wide variety of disease states. However, before such therapies can be contemplated, it is essential to understand the effects of aRC enzymes on cell metabolism and organismal physiology. Here we report and discuss new findings that shed light on the functions of the aRC enzymes in animals, and the unexpected benefits and detriments that they confer on model organisms. In Ciona intestinalis, the aRC is induced by hypoxia and by sulfide, but is unresponsive to other environmental stressors. When expressed in Drosophila, AOX results in impaired survival under restricted nutrition, in addition to the previously reported male reproductive anomalies. In contrast, it confers cold resistance to developing and adult flies, and counteracts cell signaling defects that underlie developmental dysmorphologies. The aRC enzymes may also influence lifespan and stress resistance more generally, by eliciting or interfering with hormetic mechanisms. In sum, their judicious use may lead to major benefits in medicine, but this will require a thorough characterization of their properties and physiological effects.Peer reviewe
