    Study of membrane ageing and grafting mechanisms using electron paramagnetic resonance

    An important setback for a wider use of membrane processes in industry is fouling, caused by aggregation of biomolecules at membrane surface and pores. Two important approaches to reduce this effect are the use of chemical cleaning procedures and the functionalisation of the membrane surface. However, both processes may lead to membrane degradation and structure alteration due to free radical formation or radical interaction with membrane polymer chains. In this work, electron paramagnetic resonance (EPR) was used to evaluate and quantify radical formation in both chemical cleaning and membrane functionalisation by UV grafting, allowing for a better understanding of free radical formation processes and their influence on membrane characteristics. Studies under different cleaning and grafting conditions, such as, cleaning agent concentration and pH, light intensity and irradiation were also performed showing the potential of EPR as a technique for monitoring both procedures. The information provided by EPR may contribute significantly to the development of new cleaning strategies which minimise the effect of membrane ageing and to the implementation of new and more efficient grafting procedure

    Análise Numérica do Transporte de Poluentes Emitidos por Incêndios na Amazônia

    Este trabalho teve como objetivo apresentar uma análise do transporte atmosférico de gases e partículas inertes emitidos por incêndios que ocorreram durante o período de outubro de 2009, incluindo com a situação sinótica atuante na exportação destes poluentes. Para simulação numérica do transporte do monóxido de carbono (CO) e do material particulado (PM2.5) emitido foi utilizado o modelo CATT-BRAMS (Coupled Aerosol and Tracers Transport model to the Brazilian Regional Atmospheric Modeling System). O modelo foi inicializado e ajustado com as análises do modelo global AVN/NCEP (Aviation run of the National Center for Environmental Prediction Global Spectral Model), configurado com 3 grades com resolução horizontal de 120, 30 e 10 km, centradas em 01º26’S; 48º26’W sendo que a terceira grade, de maior resolução, cobre o estado do Pará, no qual foi analisado a concentração e transporte dos poluentes, uma vez que, o Pará e Mato Grosso contribuíram com cerca de 70% do desmatamento em toda a Amazônia Legal em 2008/2009. Os resultados indicaram que a simulação apresentou uma boa performance em caracterizar as condições atmosféricas, tendo como padrão de exportação predominante para o interior do continente brasileiro, atingindo Amazonas e Mato Gross

    Growth of melon cultivated under saline stress and potassium doses

    O Estado do Rio Grande do Norte, em destaque a região da Chapada do Apodi, se destaca na produção e exportação de melão no País, em regime de irrigação, devido à distribuição pluvial baixa e irregular. A Chapada do Apodi possui dois aqüíferos subterrâneos; a do lençol menos profundo de alta salinidade, porém com menor custo de bombeamento, ocorrendo o contrário com o de maior profundidade. Objetivou-se, ante o exposto, avaliar o efeito de duas águas de salinidades diferentes (0,52 e 2,41 dS m-1), combinadas com cinco doses de K2O (218, 273, 328, 383 e 438 kg ha-1) sobre o crescimento do meloeiro (Cucumis melo L.), cultivar Goldex, utilizando-se do delineamento de blocos ao acaso, com parcelas subdivididas; amostras de planta foram coletadas aos 21, 28, 35, 49 e 63 dias após a semeadura, determinando-se a fitomassa seca das plantas, estas separadas em ramos (caule + folhas), flores e frutos; avaliou-se, também, a taxa de crescimento absoluto e relativo e a produção de frutos. Em geral, o crescimento do melão foi favorecido com o uso de água mais salina; a taxa de crescimento absoluto foi máxima entre 35 e 49 dias após a semeadura. Obteve-se maior produção de fitomassa total com 438 kg ha-1 de K2O e uso de água mais salina, ao final do ciclo.The region of 'Chapada do Apodi', in the State of Rio Grande do Norte, stands out in Brazilian production and exportation of melon. This region possesses two aquiffers, one of lower exploration cost, though of high salinity, another of low salinity, with higher cost and limited use. This work was carried out with the objective to evaluate the effect of waters of different salinities combined with five doses of K in the dry matter accumulation and productivity of the melon (Cucumis melo L.) cultivar Goldex. The experimental design adopted was split plot in completely randomized blocks. The melon crop was irrigated with low (0.52 dS m-1) and high (2.41 dS m-1) salinity water combined with five doses of K2O (218, 273, 328, 383 and 438 kg ha-1). Plants were collected at 21, 28, 35, 49 and 63 days after sowing, and were separated in branches (shoot + leaves), flowers and fruits, and the total dry matter of the aerial parts obtained by summation. The absolute and relative growth rates and the production of fruits were also evaluated. The absolute growth rate of the plant was maximum between 35 and 49 days.     Brazilian gamma detection device for sentinel lymph node biopsy

    OBJECTIVE: To test the effectiveness of an intra-operative gamma detection Brazilian device (IPEN) on sentinel lymph node biopsy (SLNB) procedures. METHODS: Forty melanoma or breast cancer patients with indication for undergoing SLNB were studied. Lymphoscintigraphy was done 2 to 24 hours prior to surgery. Lymphatic mapping with vital dye and gamma detection were performed intraoperatively. For gamma detection Neoprobe ® 1500 was used followed by IPEN (equipment under test) in the first 20 patients and for the remaining half IPEN was used first to verify its ability to locate the sentinel node (SN). Measurements were taken from the radiopharmaceutical product injection site, from SN (in vivo and ex vivo) and from background. It was recorded if the SN was stained or not and if it was found easily by surgeon. RESULTS: There were 33 (82.5%) breast cancer and 7 (17.5%) melanoma patients. Ages varied from 21 to 68 year-old (median age of 46). Sex distribution was 35 (87.5%) women and 5 (12.5%) men. Sentinel node was found in all but one patient. There was no statistical difference between the reasons ex vivo/ background obtained with the measures of both equipments (p=0, 2583-ns). The SN was easily found by the surgeon with both devices. CONCLUSION: The SLNB was successfully performed using either equipment. It was possible to do SLNB with the Brazilian device developed by IPEN without prejudice for the patient.OBJETIVO: Testar a eficácia de equipamento de detecção gama intra-operatória (DGI) desenvolvido pelo IPEN (Brasil), em procedimentos de biópsia de linfonodo sentinela (BLS) no melanoma e no câncer de mama. MÉTODOS: Foram estudados 40 pacientes portadores de melanoma ou câncer de mama com indicação para realização de BLS.Todos pacientes foram submetidos à linfocintilografia e a BLS ocorreu entre 2 a 24 horas após a mesma. Concomitantemente à DGI, realizou-se o mapeamento linfático com corante vital. Foram feitas leituras com o equipamento convencional Neoprobe® 1500 e com o equipamento em teste (IPEN) dos valores de captação do sítio de injeção do radiofármaco, do LS in vivo e ex vivo e da captação de fundo. Foi registrado se o LS estava corado e se o cirurgião teve facilidade para encontrá-lo. Nos primeiros 20 pacientes utilizou-se o equipamento convencional e depois o de teste; nos outros 20, utilizou-se primeiro o equipamento em teste, com objetivo de verificar se o mesmo identificava primariamente o LS. RESULTADOS: Dos quarenta pacientes, 33 eram portadores de tumor de mama e sete de melanoma cutâneo; variação da idade: 21 a 68 anos (mediana= 46 anos); 35 mulheres e 5 homens. Em apenas um paciente o LS não foi encontrado, nem pela DGI nem pelo corante vital. Não houve diferença estatística entre as razões ex vivo/fundo obtidas com os dois equipamentos (p=0, 2583-ns).     Towards a Low-SWaP 1024-beam Digital Array: A 32-beam Sub-system at 5.8 GHz

    Millimeter wave communications require multibeam beamforming in order to utilize wireless channels that suffer from obstructions, path loss, and multi-path effects. Digital multibeam beamforming has maximum degrees of freedom compared to analog phased arrays. However, circuit complexity and power consumption are important constraints for digital multibeam systems. A low-complexity digital computing architecture is proposed for a multiplication-free 32-point linear transform that approximates multiple simultaneous RF beams similar to a discrete Fourier transform (DFT). Arithmetic complexity due to multiplication is reduced from the FFT complexity of O(NlogN)\mathcal{O}(N\: \log N) for DFT realizations, down to zero, thus yielding a 46% and 55% reduction in chip area and dynamic power consumption, respectively, for the N=32N=32 case considered. The paper describes the proposed 32-point DFT approximation targeting a 1024-beams using a 2D array, and shows the multiplierless approximation and its mapping to a 32-beam sub-system consisting of 5.8 GHz antennas that can be used for generating 1024 digital beams without multiplications. Real-time beam computation is achieved using a Xilinx FPGA at 120 MHz bandwidth per beam.     Proteome of the phytopathogen Xanthomonas citri subsp. citri: a global expression profile

    <p>Abstract</p> <p>Background</p> <p>Citrus canker is a disease caused by <it>Xantomonas citri </it>subsp.<it>citri (Xac)</it>, and has emerged as one of the major threats to the worldwide citrus crop because it affects all commercial citrus varieties, decreases the production and quality of the fruits and can spread rapidly in citrus growing areas. In this work, the first proteome of <it>Xac </it>was analyzed using two methodologies, two-dimensional liquid chromatography (2D LC) and tandem mass spectrometry (MS/MS).</p> <p>Results</p> <p>In order to gain insight into the metabolism of <it>Xac</it>, cells were grown on two different media (<b>NB </b>- Nutrient Broth and <b>TSE </b>- Tryptone Sucrose broth enriched with glutamic acid), and proteins were proteolyzed with trypsin and examined by 2D LC-MS/MS. Approximately 39% of all predicted proteins by annotation of <it>Xac </it>were identified with their component peptides unambiguously assigned to tandem mass spectra. The proteins, about 1,100, were distributed in all annotated functional categories.</p> <p>Conclusions</p> <p>This is the first proteomic reference map for the most aggressive strain of <it>Xanthomonas </it>pathogen of all orange varieties. The compilation of metabolic pathways involved with bacterial growth showed that <it>Xac </it>expresses a complete central and intermediary metabolism, replication, transcription and translation machineries and regulation factors, distinct membrane transporters (ABC, MFS and pumps) and receptors (MCP, TonB dependent and metabolites acquisition), two-component systems (sensor and regulatory components) and response regulators. These data corroborate the growth curve <it>in vitro </it>and are the first reports indicating that many of these genome annotated genes are translated into operative in <it>Xac</it>. This proteomic analysis also provided information regarding the influence of culture medium on growth and protein expression of <it>Xac</it>.</p

    Anticipating the Climate Change Impacts on Madeira’s Agriculture: The Characterization and Monitoring of a Vine Agrosystem

    Climate—Madeira Strategy (CMS) foresees two models to describe the climate scenarios for the Madeira region in 2050 and 2070. These scenarios anticipate an average temperature rise of 1.4 to 3.7 ◦C and a decrease in precipitation by 30 to 40%. Consequently, Madeira’s agriculture will suffer the impacts of climate change. To understand these impacts, a baseline of major agrosystem components needs to be established, with the ultimate goal to monitor its consequences in its functioning. CASBio project used the 1961–1991 and 2010–2020 meteorological data series to modulate climate conditions and characterize and monitor six agrosystems for 2 years. One of them was a vineyard, Quinta das Vinhas, representing a typical agrosystem in the Mediterranean climate. The annual and seasonal variation in climatic parameters, soil conditions, microbiological communities, floristic and insect diversity, and crop production was assessed, using a total of 50 parameters. The results were used to establish a baseline of the agrosystem components and their seasonal and annual variation. The major findings are: (i) winter and summer extreme events show a trend in temperature and precipitation supporting a fast change in climate; (ii) a critical imbalance between nitrogen fixing and denitrifying bacteria was identified, especially in summer, that could be determined by the rise in temperature and drought; (iii) among floristic diversity, the therophytes and geophytes confirm to be the most suitable indicators for the rise in temperature and reduction in precipitation in the agrosystems; (iv) an imbalance in favor of C. capitata plague was observed, associated with the summer rise in temperature and decrease in precipitation; (v) despite an increase in most of the grape varieties production, the Madeiran wine local varieties were shown to be less stable in productivity under observed climate conditions. The agrosystem baseline is a starting point for long term monitoring and allows for further quantifying the influence of climate change on agrosystem productivity, resilience, and sustainability.info:eu-repo/semantics/publishedVersio
