    Two different approaches to dry-land training do not improve the water performance of swimmers

    International Journal of Exercise Science 16(6): 770-790, 2023. Literature diverges about the performance improvement after dry-land training. Thus, the objective of the present study was to compare the effect of two models of dry-land training. Twenty-nine swimmers were divided into three groups, combined strength and power training (PTG), only strength training (STG), and a control group (CG). Measurements were taken for six weeks, before dry-land exposure (M1), after four weeks of specific training with exposure to dry-land training by two groups (M2), and after two weeks of taper without exposure to dry-land training (M3). Strength in specific exercises, jumping tests, and 50, 100, and 200m freestyle performance were evaluated on M1 and M3, while hematological and strength parameters in tethered swimming were measured in M1, M2, and M3.     Tannins possessing bacteriostatic effect impair pseudomonas aeruginosa adhesion and biofilm formation

    Plants produce many compounds that are biologically active, either as part of their normal program of growth and development or in response to pathogen attack or stress. Traditionally, Anadenanthera colubrina, Commiphora leptophloeos and Myracrodruon urundeuva have been used by communities in the Brazilian Caatinga to treat several infectious diseases. The ability to impair bacterial adhesion represents an ideal strategy to combat bacterial pathogenesis, because of its importance in the early stages of the infectious process; thus, the search for anti-adherent compounds in plants is a very promising alternative. This study investigated the ability of stem-bark extracts from these three species to control the growth and prevent biofilm formation of Pseudomonas aeruginosa, an important opportunistic pathogen that adheres to surfaces and forms protective biofilms. A kinetic study (0–72 h) demonstrated that the growth of extract-treated bacteria was inhibited up to 9 h after incubation, suggesting a bacteriostatic activity. Transmission electron microscopy and fluorescence microscopy showed both viable and nonviable cells, indicating bacterial membrane damage; crystal violet assay and scanning electron microscopy demonstrated that treatment strongly inhibited biofilm formation during 6 and 24 h and that matrix production remained impaired even after growth was restored, at 24 and 48 h of incubation. Herein, we propose that the identified (condensed and hydrolyzable) tannins are able to inhibit biofilm formation via bacteriostatic properties, damaging the bacterial membrane and hindering matrix production. Our findings demonstrate the importance of this abundant class of Natural Products in higher plants against one of the most challenging issues in the hospital setting: biofilm resilience

    Potent and Broad-Spectrum Antimicrobial Activity of Analogs from the Scorpion Peptide Stigmurin

    Scorpion venom constitutes a rich source of biologically active compounds with high potential for therapeutic and biotechnological applications that can be used as prototypes for the design of new drugs. The aim of this study was to characterize the structural conformation, evaluate the antimicrobial activity, and gain insight into the possible action mechanism underlying it, for two new analog peptides of the scorpion peptide Stigmurin, named StigA25 and StigA31. The amino acid substitutions in the native sequence for lysine residues resulted in peptides with higher positive net charge and hydrophobicity, with an increase in the theoretical helical content. StigA25 and StigA31 showed the capacity to modify their structural conformation according to the environment, and were stable to pH and temperature variation-results similar to the native peptide. Both analog peptides demonstrated broad-spectrum antimicrobial activity in vitro, showing an effect superior to that of the native peptide, being non-hemolytic at the biologically active concentrations. Therefore, this study demonstrates the therapeutic potential of the analog peptides from Stigmurin and the promising approach of rational drug design based on scorpion venom peptide to obtain new anti-infective agents.publishersversionpublishe

    Validity of a specific protocol for neuromuscular fatigue analysis of the elbow extensor musculature.

    A utilização da eletroestimulação muscular é uma estratégia utilizada para investigação da fadiga neuromuscular. Entretanto, esta técnica é pouco aplicada na musculatura extensora do cotovelo em função de limitações biomecâncias (atuação de músculos sinergistas), neurais (influência de reflexo neural) e fisiológicas (facilidade em atingir estados de potencialização pós-ativação e fadiga). Portanto, o objetivo deste estudo foi padronizar procedimentos para avaliação neuromuscular da musculatura extensora do cotovelo. Para isso, foram realizados três estudos que envolveram a adequação da técnica de eletroestimulação e um \"estudo principal\" para validar um novo protocolo de avaliação neuromuscular. Após resultados positivos em estudos complementares, a presente dissertação propôs a validação de um protocolo para análise neuromuscular com contrações submáximas para a musculatura extensora do cotovelo. Vinte participantes com idade de 25,1 ± 6,8 anos foram submetidos, em dias diferentes, a dois esforços, um a fim de promover fadiga central e fadiga periférica respectivamente. Antes e após os esforços envolvendo fadiga (periférica ou central) foram realizadas avaliações de fadiga neuromuscular para musculatura extensora do joelho (protocolo já validado) e musculatura extensora do cotovelo (protocolo proposto). Verificou-se que: (i.) o modelo de análise proposto não promove nenhum tipo de fadiga ou potencialização pós-ativação; (ii) a resposta de um pulso controle no início e no fim do modelo de análise proposto promovem respostas diferentes; (iii) o modelo proposto foi capaz de identificar redução da resposta neuromuscular; (iv) o esforço envolvendo fadiga central não promoveu fadiga central significante; (v) a utilização de um modelo matemático para investigação de fadiga neuromuscular (coeficientes resultantes de uma regressão linear) com os valores de estimulação em contrações submáximas foi sensível a fadiga mas não foi capaz de discriminar fadiga central e periférica. É possível concluir que o modelo de análise proposto para a musculatura extensora do cotovelo apresentou sensibilidade para verificar a fadiga neuromuscular. Entretanto, este não foi capaz de discriminar fadiga central e periférica. A utilização da regressão linear parece ser uma boa estratégia para diferenciar os dois modelos de fadiga e deve ser estudada com mais profundidade futuramente.Muscular electrostimulation is a strategy used to investigate neuromuscular fatigue. However, this technique is poorly applied in the elbow extensor muscles due to the limitations of biomechanics (acting of synergistic muscles), neural (influence of neural reflex) and physiological (easiness to achieve post-activation and fatigue states of potentiation). Therefore, the purpose of this study was to standardize a neuromuscular evaluation strategy for the elbow extensor musculature. Thus, three complementary studies aimed at adjusting the electrostimulation technique and one \"main study\" to validate a new neuromuscular assessment protocol were realized. After positive results in complementary studies, the present dissertation proposed a protocol validation for neuromuscular analysis with submaximal contractions on the elbow extensor musculature. Twenty participants with 25.1 ± 6.8 years old performed, on different days, two efforts, one to promote central fatigue and peripheral fatigue respectively. Before and after the exercise involving fatigue (peripheral or central) there were realized neuromuscular fatigue evaluations for knee extensor muscles (protocol already validated) and elbow extensor musculature (protocol). The results indicate that: (i) the proposed analysis model does not promote any type of fatigue or post-activation potentiation; (ii) the response of a control pulse at the beginning and end of the proposed analysis model promotes different responses; (iii) the proposed model was able to identify reduction of neuromuscular response; (iv) the effort involving central fatigue did not promote significant central fatigue; (v) the use of a mathematical model to investigate neuromuscular fatigue (coefficients resulting from a linear regression) with the values of stimulation in submaximal contractions was sensitive after fatigue but was not able to discriminate central and peripheral fatigue. It is possible to conclude that the proposed analysis model for the elbow extensor musculature presented sensitivity to verify the neuromuscular fatigue. However, it was not able to discriminate central and peripheral fatigue. The use of linear regression seems to be a good strategy to differentiate the two models of fatigue and should be studied in more depth in the future

    The development of the National Curriculum Parameters for High School (PCN) is established from a logic in which the disciplines are integrated and coordinated in order to promote interdisciplinarity. However, there are few didactic-pedagogic activities that enable teachers in practice to reach this goal. In this paper, we propose an experimental activity to be used with High School students, addressing the concept of viscosity from an interdisciplinary perspective between the disciplines of Physics and Chemistry. For that purpose, flow times of sucrose solutions of different concentrations will be measured in a glass Ford and these data will be used to construct a calibration curve, which will be used to determine the concentration of sucrose in artificial flavor syrup of gooseberry.Os Parâmetros Curriculares Nacionais para o Ensino Médio propõem que o desenvolvimento curricular se estabeleça a partir de uma lógica na qual as disciplinas estejam integradas e articuladas de modo a favorecer a interdisciplinaridade. Contudo, são poucas as atividades de caráter didático-pedagógicas que possibilitam aos professores, na prática, a concretizarem esse objetivo.     Simulation of diets for dairy goats and growing doelings using nonlinear optimization procedures

    ABSTRACT The objective of this study was to simulate total dry matter intake and cost of diets optimized by nonlinear programming to meet the nutritional requirements of dairy does and growing doelings. The mathematical model was programmed in a Microsoft Excel(r) spreadsheet. Increasing values of body mass and average daily weight gain for growing doelings and increasing body mass values and milk yield for dairy does were used as inputs for optimizations. Three objective functions were considered: minimization of the dietary cost, dry matter intake maximization, and maximization of the efficiency of use of the ingested crude protein. To solve the proposed problems we used the Excel(r) Solver(r) algorithm. The Excel(r) Solver(r) was able to balance diets containing different objective functions and provided different spaces of feasible solutions. The best solutions are obtained by least-cost formulations; the other two objective functions, namely maximize dry matter intake and maximize crude protein use, do not produce favorable diets in terms of costs
