    Toxoplasma gondii infection in native village chickens (Gallus domesticus) in Selangor and Melaka, Malaysia

    Toxoplasmosis is a worldwide zoonosis caused by the protozoa Toxoplasma gondii which affects human and animals. Village chickens Gallus domesticus) most commonly known as Ayam Kampung or free-range chickens, have been suggested to play a role in the epidemiology of toxoplasmosis. This study determines the presence of T. gondii in the villagechicken populations in two states of Malaysia. A total of 50 serum samples from the chickens from Selangor (n=20) and Melaka (n=30) were collected and analysed using commercial serological kits. T. gondii antigen was detected in 20% (Selangor 30%; Melaka 13%) samplesusing ELISA test and anti-T. gondii antibody was detected in all positive ELISA samples using the indirect haemagglutination test (IHAT). Histopathological examination revealed tissue changes such as inflammation and degeneration in brain and liver of seropositive chickens.This is the first report of T. gondii infection in the village chickens in Malaysia

    Determination of fibernolytic enzyme activities of white rot fungi isolated from oil palm fronds

    Oil palm fronds (OPF) is widely used as the source of roughage for the farm animals. However, the full potential of OPF as animal feed is limited by their high lignin content which limits the rumen microbe’s access to the cellulose and hemicellulose. White rot fungi (WRF) are a group of fungi belonging to basidiomycete phylum and are commonly found in decaying woody plant. They possess the ability to degrade lignin. This experiment aims to identify the phylum of the best lignin decaying fungi based on their enzymes activity. In this experiment, 11 fungi species were isolated from decaying oil palm fronds. They are labelled as WR1, WR 2, WR3, WR4, WR5, WR6, WR7, WR8, WR9, WR10 and WR11. Their fibernolytic enzyme activities which includes laccase, manganese peroxidase, lignin peroxidase, avicelase, carboxylmethylcellulase and xylanase are analysed using the solid state fermentation method. It is was found that 5 fungi species which are the WR1, WR2, WR4, WR7 and WR10 produced the highest ratio of lignin degrading enzyme to cellulose and hemicellulose degrading enzyme. The fungi are then analysed under microscope to determine the phylum of the fungi. From the observation, the fungi are identified to belong to the phylum basidiomycetes due to presence of clamp connection

    The aim of this research was to determine the effects of Momordica charantia (MC) fruit aqueous extract on pancreatic histopathological changes in neonatal STZ-induced type-II diabetic rats. Diabetes mellitus was induced in one day Sprague-Dawley neonatal rats using a single intrapretoneal injection of streptozotocin (STZ) (85 mg/kg body weight) and monitored for 12 weeks thereafter. The diabetic rats were separated into three groups, as follows: the diabetic control group (i.e. nSTZ), the diabetic group (i.e. nSTZ/M) - which was orally given 20 mg/kg of MC fruit extract, and the diabetic group (i.e. nSTZ/G) - that was treated with glibenclamide, 0.1 mg/kg for a period of four weeks. At the end of treatment, the animals were sacrificed and blood samples were collected from the saphenous vein to measure the blood glucose and serum insulin level. The pancreatic specimens were removed and processed for light microscopy, electron microscopy examination and immunohistochemical study. The results of this study showed that MC fruit aqueous extract reduced the blood glucose level as well as glibenclamide and increased the serum insulin level in the treated diabetic rats (P<0.05). The fruit extract of MC alleviated pancreatic damage and increased the number of ß-cells in the diabetic treated rats (P<0.05). Our results suggest that oral feeding of MC fruit extract may have a significant role in the renewal of pancreatic ß-cells in the nSTZ rats

    Efficacy of Nigella sativa in alleviating benzo[a]pyrene-induced immunotoxicity in broilers

    The immune response of broiler chickens exposed to intra-tracheal (i.t.) administration of benzo[a]pyrene (BaP) with and without Nigella sativa (Ns) supplementation was investigated. A total of 120 day-old chicks were divided into four groups comprising 30 birds each, into a control, Ns, BaP, and BaP+Ns group. Immune responses to Newcastle disease (ND) were evaluated by haemagglutination inhibition (HI), phytohaemagglutinin (PHA) skin test and carbon clearance assay (CCA). In most instances, there was a significant increase (p<0.05) in the ND-HI antibody titers, PHA skin-swelling response and phagocytic activity in the BaP + Ns group compared to that of the BaP group. Likewise, organ weight and indices of the spleen, bursa of Fabricius and thymus of birds from the BaP + Ns group were also higher (p<0.05) than that of the BaP group from day 1 until day 21. It is concluded that exposure to BaP may exert adverse effects on the immune system of broilers which may increase their susceptibility to disease, and Ns supplementation significantly reduces these alteration

    Crude extracts of epidermal mucus and epidermis of climbing perch Anabas testudineus and its antibacterial and hemolytic activities

    Epidermal mucus of fish contains a number of biologically active constituents including antimicrobial peptides that are continuously expressed and also engaged in provision of protection to the fish against injurious substances as well as potential pathogenic microbes. This study seeks to determine the hemolytic and antibacterial properties of crude extract from epidermal mucus of climbing perch. Epidermal mucus and epidermis were obtained from 15 climbing perches after hypothermic stress stimulation for extraction of crude by aqueous and acidic methods. Antibacterial activity by well diffusion agar was applied for both extracts against 11 strains belonging to 7 different bacterial species using well agar diffusion test assay. In addition, haemolytic activity was investigated using horse RBCs. Antibacterial activity was performed for acidified crude extract only and it was found to be significant (P < 0.05) against P. aeruginosa while the least reactivity was observed against Methicillin Resistant Staphylococcus aureus. Total soluble protein concentration was 1.2 mg/ml with 13 bands of proteins (245–11) kDa distinguished. There was absence of haemolytic activity of acidified crude extract using horse RBCs. The finding of this study indicates that proteins in bioactive crude extracted from climbing perch have potential therapeutic application.
