    Study of In-vitro Antioxidant and Antibacterial activity of leaf extract of Azadirachta indica, and Ocimum sanctum in different organic solvents and Cow urine

    Background: Neem (Azadirachta indica), Tulsi (Ocimum sanctum) and Cow urine (Bos indicus) has been widely used as a therapeutic potential or additive agent in different ethnomedicinal practices as well as modern science also proved its therapeutic potential to treat various diseases and balance the body conditions. Aims: Alone study on Neem, Tulsi and Cow urine (CU) has been widely performed but this study aims to evaluate the individual as well as synergistic antioxidant and antibacterial activity of Neem, and Tulsi’s leaves extract in different organic solvents and Cow urine. Materials and Methods: Antioxidant activity was carried out by using 2,2-Diphenyl1-picrylhydrazyl (DPPH) radical scavenging activity and the antibacterial activity was tested by the agar disc diffusion method against gram-positive (Staphylococcus aureus)  and gram-negative (Escherichia coli, Proteus vulgaris and Klebsiella species) bacteria. Results: The antioxidant and antibacterial activity is concentration-dependent. The neem extract in chloroform shows the highest ZOI against S. aureus (14±2). Similarly, the neem extract in cow urine (8±0.86) and tulsi extract in methanol (8±1.04) shows the highest ZOI against E. coli. Against K. species cow urine is most effective (9±0.29) followed by neem extract in chloroform and cow urine (8±0.77). In P. vulgaris almost all sample shows ZOI around 7 mm. In synergistic antibacterial study tulsi and neem extract in cow urine shows the highest ZOI against E. coli (12±1.75) and P. vulgaris (10±1.44). The tulsi and neem extract in hexane is not effective against S. aureus and E. coli but effective in P. vulgaris and K. species (ZOI 7 mm). Tulsi and neem extract in chloroform, methanol, and cow urine show similar ZOI (9 mm). In DPPH free radical scavenging activity the methanolic extract of tulsi shows the highest inhibition of free radicle (96.35±0.001), followed by methanolic extract of neem (84.85±0.042). The fresh cow urine inhibits (25.19±0.037) followed by neem extract in chloroform (24.53±0.003), neem extract in cow urine (21.8±0.012), tulsi extract in chloroform (20.86±0.047), tulsi extract in hexane (19.5±0.031), tulsi extract in cow urine (18.33±0.023) and the least inhibition by neem extract in hexane (6.74±0.042)

    Energy from Waste: Poterioochromonas malhamensis Used for Managing Dairy Effluent and Producing Valuable Microalgal Lipid

    Currently, microalgae have become a marvelous and resource-friendly alternative source of advantageous bioproducts, such as lipids, carbohydrates, proteins, or other bioactive compounds. Because of the richness of microalgae in these high-value-added metabolites, still, it is an underdeveloped source of sustainable energy and food. There are some hurdles to profitable production, such as culture contamination and costly harvesting techniques. In the current work, a chrysophyte was isolated from dairy wastewater, identified as Poterioochromonas malhamensis based on its morphology and partial 18S rRNA gene sequences. This isolate was used to remediate dairy waste water (DWW) and to obtain neutral lipids (fatty acids) from microalgae. Microalgal growth was optimized by using different concentrations of DWW, supplemented with all the nutritive requirements for better progression and flourishment. Maximum biomass yield 1.478 g L−1 was achieved by optimized cultural conditions (different concentrations of DWW with BBM media). This strain showed high nitrate and phosphate removal efficiency (87.45% and 88.96%), respectively in 15 days. The experimental results highlighted that the lipid content and the chemical oxygen demand (COD) removal were 31.60% and 88.84%, respectively, and the lipid profile of isolated microalga was C16:0, C16:1, C18:0, C18:1, and C18:2 fatty acids. For growth and treatment purposes, 75% DWW with Bold’s Basal Medium (BBM) media showed better results. This is the first report of DWW treatment using the microalga Poterioochromonas malhamensis, as far as we are aware. Its cultivation prevented the spread of pollution of freshwater sources, remedied the DWW, and generated important lipids for industry

    An educational intervention’s effect on healthcare professionals’ attitudes towards pharmacovigilance

    BackgroundPharmacovigilance concerns the detection, assessment, understanding, and prevention of adverse effects or any other drug-related problems. Consumer pharmacovigilance is the involvement of consumers in adverse drug reaction (ADR) reporting. Assessing healthcare professionals’ (HCPs) knowledge of and attitude towards pharmacovigilance and consumer pharmacovigilance is integral to strengthening adverse drug reaction reporting systems. AimsTo study knowledge of and attitude towards pharmacovigilance and consumer pharmacovigilance among HCPs from KIST Medical College, Lalitpur, Nepal, and to plan an appropriate educational intervention to address deficiencies noted.Methods The study was conducted from February 2013 to December 2013 at KIST Medical College using a self-administered, pretested, structured questionnaire. The maximum possible scores for knowledge, attitude, and total were 100, 95, and 195, respectively. Baseline knowledge and attitude were studied. Two months after the questionnaire, an intervention that used a combination of methods about pharmacovigilance and consumer pharmacovigilance was undertaken. Knowledge and attitudes were studied immediately after the intervention. Scores before and after the intervention were compared using Wilcoxon signed-rank test.Results A total of 105 HCPs participated. The median (interquartile range) knowledge, attitude, and total scores before the intervention were 56 (7), 72 (9), and 127 (16), respectively.     Regenerative callus induction and biochemical analysis of Stevia rebaudiana Bertoni

    Stevia Leaves are the principal source of stevioside, which is estimated to be 100-300 times sweeter than table sugar. Stevioside has clinical significance as they are reported to maintain glucose levels in human blood. Owing to the difficulties in propagation of stevia through seeds and vegetative methods, callus culture has been an efficient alternative for generation of stevioside. The aim of this study is to develop an efficient and standardized protocol for maximum induction and multiplication of callus from a leaf. Callus culture was established from leaves in MS basal media fortified with various combinations (BAP, NAA, 2,4-D, KN, IBA) and concentrations of phytohormones. The best callusing (100%) was recorded in MS media supplemented with (2,4-D 1.0mg/l + NAA 1.0mg/l). The callus was harvested after 4 weeks and screened for the presence of various bioactive compounds. The qualitative results showed that the extracts of callus contained bioactive compounds like flavonoids, glycosides, phenol, tannins, sterols and saponins thereby making callus one of the sources for extraction of various secondary metabolites

    Microduplications of 16p11.2 are associated with schizophrenia

    Recurrent microdeletions and microduplications of a 600-kb genomic region of chromosome 16p11.2 have been implicated in childhood-onset developmental disorders1,2,3. We report the association of 16p11.2 microduplications with schizophrenia in two large cohorts. The microduplication was detected in 12/1,906 (0.63%) cases and 1/3,971 (0.03%) controls (P = 1.2 × 10−5, OR = 25.8) from the initial cohort, and in 9/2,645 (0.34%) cases and 1/2,420 (0.04%) controls (P = 0.022, OR = 8.3) of the replication cohort. The 16p11.2 microduplication was associated with a 14.5-fold increased risk of schizophrenia (95% CI (3.3, 62)) in the combined sample. A meta-analysis of datasets for multiple psychiatric disorders showed a significant association of the microduplication with schizophrenia (P = 4.8 × 10−7), bipolar disorder (P = 0.017) and autism (P = 1.9 × 10−7). In contrast, the reciprocal microdeletion was associated only with autism and developmental disorders (P = 2.3 × 10−13). Head circumference was larger in patients with the microdeletion than in patients with the microduplication (P = 0.0007)

    Prevalence, associated factors and outcomes of pressure injuries in adult intensive care unit patients: the DecubICUs study

Epidemiologic data is however unavailable. We aimed to provide an international picture of the extent of pressure injuries and factors associated with ICU-acquired pressure injuries in adult ICU patients. Methods: International 1-day point-prevalence study; follow-up for outcome assessment until hospital discharge (maximum 12 weeks). Factors associated with ICU-acquired pressure injury and hospital mortality were assessed by generalised linear mixed-effects regression analysis. Results: Data from 13,254 patients in 1117 ICUs (90 countries) revealed 6747 pressure injuries; 3997 (59.2%) were ICU-acquired. Overall prevalence was 26.6% (95% confidence interval [CI] 25.9–27.3). ICU-acquired prevalence was 16.2% (95% CI 15.6–16.8). Sacrum (37%) and heels (19.5%) were most affected. Factors independently associated with ICU-acquired pressure injuries were older age, male sex, being underweight, emergency surgery, higher Simplified Acute Physiology Score II, Braden score 3 days, comorbidities (chronic obstructive pulmonary disease, immunodeficiency), organ support (renal replacement, mechanical ventilation on ICU admission), and being in a low or lower-middle income-economy. Gradually increasing associations with mortality were identified for increasing severity of pressure injury: stage I (odds ratio [OR] 1.5; 95% CI 1.2–1.8), stage II (OR 1.6; 95% CI 1.4–1.9), and stage III or worse (OR 2.8; 95% CI 2.3–3.3). Conclusion: Pressure injuries are common in adult ICU patients. ICU-acquired pressure injuries are associated with mainly intrinsic factors and mortality. Optimal care standards, increased awareness, appropriate resource allocation, and further research into optimal prevention are pivotal to tackle this important patient safety threat
