    Utilization of Oil Palm Empty Bunches Waste as Biochar-microbes for Improving Availability of Soil Nutrients

    There are about 23% waste oil palm empty fruit bunches (OPEFB) of total waste generated from the production of crude palm oil in oil palm plantations. Pyrolysis technology can be used to convert waste into biochar and further can be utilized for the improvement of soil. Biochar-microbes of OPEFB are biochar from OPEFB biomass that enriched with soil microbes. Biochar-microbes is expected to be used for the improvement of the soil and plants. Therefore the purpose of this research was to study the ability of biochar-microbes OPEFB to increase availability of the nutrients in sandy soils. The process of making biochar done by using slow pyrolysis technology by heating 300oC and 400oC for 2 and 3 hours, and with sizes 40 and 80 mesh, as well as indigenous microbial Bulkhorderia nodosa G.52.Rif1 and Trichoderma sp.) added. The biochar production and research were conducted in the Department of Forestry Laboratory and in the Department of Agronomy Laboratory, Faculty of Agriculture, Palangka Raya University. In general, the study showed that biochar-microbes could maintain the soil pH value and tends to increase the soil pH, increasing the holding capacity of sandy soil to the elements of P and K as well as increasing the availability of nutrients N, P and K. Furthermore, this study showed that the biochar process by 400oC heating for 3 hours and 40 mesh with microbes or without microbes were the best effect on the improvement of the quality of holding capacity and the nutrients supply in sandy soils

    Carcinoma-associated fucosylated antigens are markers of the epithelial state and can contribute to cell adhesion through CLEC17A (Prolectin)

    International audienceTerminal fucosylated motifs of glycoproteins and glycolipid chains are often altered in cancer cells. We investigated the link between fucosylation changes and critical steps in cancer progression: epithelial-to-mesenchymal transition (EMT) and lymph node metastasis. Using mammary cell lines, we demonstrate that during EMT, expression of some fucosylated antigens (e.g.: Lewis Y) is decreased as a result of repression of the fucosyltransferase genes FUT1 and FUT3. Moreover, we identify the fucose-binding bacterial lectin BC2L-C-Nt as a specific probe for the epithelial state. Prolectin (CLEC17A), a human lectin found on lymph node B cells, shares ligand specificities with BC2L-C-Nt. It binds preferentially to epithelial rather than to mesenchymal cells, and microfluidic experiments showed that prolectin behaves as a cell adhesion molecule for epithelial cells. Comparison of paired primary tumors/ lymph node metastases revealed an increase of prolectin staining in metastasis and high FUT1 and FUT3 mRNA expression was associated with poor prognosis. Our data suggest that tumor cells invading the lymph nodes and expressing fucosylated motifs associated with the epithelial state could use prolectin as a colonization factor

    Dayak tribe in Central Kalimantan Province has long time ago applied the local wisdom of healthy life and environmental friendly by consuming more local vegetables that grow wild. The research was conducted from July to September 2018, by conducting survey observations and interviewing respondents in Kapuas district, Pulang Pisau district, Katingan district, Gunung Mas district, and Palangka Raya city. There are a total of 14 species found from swamp areas, namely Singkah enyuh (Cocos nucifera), Singkah undus (Elaeis guineensis), Singkah hambie (Metroxylon sagu), singkah uwei (Calamus sp), Taya (Nauclea sp), Ujau (Bambusa sp), Bajei (Diplazium esculentum), Bakung (Crinum asiaticum), Kalakai (Stenochlaena palustris), Kujang (Colocasia esculenta), Uru mahamen (Neptunia olearecea), Pucuk teratai (Nymphae sp), Genjer (Limnocharis flava), dan Kangkung danum (Ipomoea aquatica). The benefits of local vegetables aside from being a food source that has the potential as a drug, pesticides, bioethanol and various other industrial materials. The majority of respondents aged over 40 years prefer vegetables (local) and food types were soup, while middle age (16-40 years) and young (under 16 years) can adapt to vegetables from outside and a new menu from outside Kalimantan. Factors for choosing the types of vegetables to be consumed are more influenced by taste reasons (54-86%), followed by habits (32-47%) and benefits for health (39-40%)

    Do women prefer caesarean sections? A qualitative evidence synthesis of their views and experiences

    Background: Caesarean sections (CS) continue to increase worldwide. Multiple and complex factors are contributing to the increase, including non-clinical factors related to individual women, families and their interactions with health providers. This global qualitative evidence synthesis explores women’s preferences for mode of birth and factors underlying preferences for CS. Methods: Systematic database searches (MEDLINE, EMBASE, CINAHL, PsycINFO) were conducted in December 2016 and updated in May 2019 and February 2021. Studies conducted across all resource settings were eligible for inclusion, except those from China and Taiwan which have been reported in a companion publication. Phenomena of interest were opinions, views and perspectives of women regarding preferences for mode of birth, attributes of CS, societal and cultural beliefs about modes of birth, and right to choose mode of birth. Thematic synthesis of data was conducted. Confidence in findings was assessed using GRADE-CERQual. Results: We included 52 studies, from 28 countries, encompassing the views and perspectives of pregnant women, non-pregnant women, women with previous CS, postpartum women, and women’s partners. Most of the studies were conducted in high-income countries and published between 2011 and 2021. Factors underlying women preferences for CS had to do mainly with strong fear of pain and injuries to the mother and child during labour or birth (High confidence), uncertainty regarding vaginal birth (High confidence), and positive views or perceived advantages of CS (High confidence). Women who preferred CS expressed resoluteness about it, but there were also many women who had a clear preference for vaginal birth and those who even developed strategies to keep their birth plans in environments that were not supportive of vaginal births (High confidence). The findings also identified that social, cultural and personal factors as well as attributes related to health systems impact on the reasons underlying women preferences for various modes of birth (High confidence). Conclusions: A wide variety of factors underlie women’s preferences for CS in the absence of medical indications. Major factors contributing to perceptions of CS as preferable include fear of pain, uncertainty with vaginal birth and positive views on CS. Interventions need to address these factors to reduce unnecessary CS

    Converse Smith-Martin cell cycle kinetics by transformed B lymphocytes

    Recent studies using direct live cell imaging have reported that individual B lymphocytes have correlated transit times between their G1 and S/G2/M phases. This finding is in contradiction with the influential model of Smith and Martin that assumed the bulk of the total cell cycle time variation arises in the G1 phase of the cell cycle with little contributed by the S/G2/M phase. Here we extend these studies to examine the relation between cell cycle phase lengths in two B lymphoma cell lines. We report that transformed B lymphoma cells undergo a short G1 period that displays little correlation with the time taken for the subsequent S/G2/M phase. Consequently, the bulk of the variation noted for total division times within a population is found in the S/G2/M phases and not the G1 phase. Models that reverse the expected source of variation and assume a single deterministic time in G1 followed by a lag + exponential distribution for S/G2/M fit the data well. These models can be improved further by adopting two sequential distributions or by using the stretched lognormal model developed for primary lymphocytes. We propose that shortening of G1 transit times and uncoupling from other cell cycle phases may be a hallmark of lymphocyte transformation that could serve as an observable phenotypic marker of cancer evolution.K.     Characterization of Cyclin E Expression in Multiple Myeloma and Its Functional Role in Seliciclib-Induced Apoptotic Cell Death

    Multiple Myeloma (MM) is a lymphatic neoplasm characterized by clonal proliferation of malignant plasma cell that eventually develops resistance to chemotherapy. Drug resistance, differentiation block and increased survival of the MM tumor cells result from high genomic instability. Chromosomal translocations, the most common genomic alterations in MM, lead to dysregulation of cyclin D, a regulatory protein that governs the activation of key cell cycle regulator – cyclin dependent kinase (CDK). Genomic instability was reported to be affected by over expression of another CDK regulator - cyclin E (CCNE). This occurs early in tumorigenesis in various lymphatic malignancies including CLL, NHL and HL. We therefore sought to investigate the role of cyclin E in MM. CCNE1 expression was found to be heterogeneous in various MM cell lines (hMMCLs). Incubation of hMMCLs with seliciclib, a selective CDK-inhibitor, results in apoptosis which is accompanied by down regulation of MCL1 and p27. Ectopic over expression of CCNE1 resulted in reduced sensitivity of the MM tumor cells in comparison to the paternal cell line, whereas CCNE1 silencing with siRNA increased the cell sensitivity to seliciclib. Adhesion to FN of hMMCLs was prevented by seliciclib, eliminating adhesion–mediated drug resistance of MM cells. Combination of seliciclib with flavopiridol effectively reduced CCNE1 and CCND1 protein levels, increased subG1 apoptotic fraction and promoted MM cell death in BMSCs co-culture conditions, therefore over-coming stroma-mediated protection. We suggest that seliciclib may be considered as essential component of modern anti MM drug combination therapy
