    L-asparaginase (EC 3.5.1.1), a therapeutic enzyme used in the treatment of childhood acute lymphoblastic leukemia (ALL). Hence, the goal of this work is study the expression and evaluation of hydrolysis activity of native sequence (X12746) encoding for L-asparaginase from Erwinia chrysanthemi NCPBB1125 in the popular expression system Pichia pastoris. The sequence of asn encoded for mature protein was expressed in P. pastoris SMD1168 and X33. SDS-PAGE analysis showed recombinant L-asparaginase was secreted efficiently. Stable and high hydrolysis activity of extracellular L-asparaginase in P. pastoris SMD1168 making it a potential candidate to produce recombinant protein. After purification, a specific band whose appearance approximately 45 kDa indicating the glycosylated protein with specific activity by 6.251 Umg-1 and about 3 folds purifications.L-asparaginase (EC 3.5.1.1), một loại enzyme được sử dụng trong điều trị bệng ung thư bạch cầu mãn tính ở trẻ em. Mục tiêu của nghiên cứu này là biểu hiện và đánh giá hoạt tính thủy phân của L-asparaginase mã hóa bởi đoạn gene (X12746) tương ứng từ Erwinia chrysanthemi NCPBB1125 được biểu hiện trong nấm men Pichia pastoris. Gene đã được cắt signal peptide và biểu hiện trong P. pastoris SMD1168 and X33. Qua phân tích kết quả điện di SDS-PAGE của môi trường sau lên men, L-asparaginase tái tổ hợp được tìm thấy trong dịch ngoại bào của P. pastoris. Với khả năng sản xuất protein có hoạt tính cao hơn so với chủng P. pastoris X33, SMD1168 được lựa chọn để biểu hiện L-asparaginase tái tổ hợp. Sau khi tinh sạch, sự xuất hiện của một băng có kích khối lượng phân tử xấp xỉ 45 kDa trên điện di SDS-PAGE cho thấy protein tái tổ hợp đã bị glycosyl hóa với hoạt tính riêng 6.251 Umg-1 và đạt độ sạch 3.471 lần

    The magnesium and magnesium alloys has applied widely in different industrial aspects in Vietnam in the modern life. Especially, the products from magnesium alloys implementing in the automotive have increased rapidly since the car elements tend to be generated by the light alloys in order to save the fuel. However, in the current time, Vietnam has no factories to produce the magnesium to adapt the domestic demand although it owns an enrich resource of raw materials. This research indicates the possibility of using the dolomite ore in Thanhhoa – Vietnam to make the magnesium as well as evaluate the primary factors like recovering temperature, reducing agent rate, recovering time having effect on the reduction efficiency of Thanhhoa dolomite by metallothermic method in vacuum (Pidgeon Process). This is basic process, low investment and suitable for the small and medium scales in Vietnam. The experiment includes heating, indicating the chemical ingredients and recovering experiment on the dolomite after calcination (dolime) by using ferrosilicon. The thermodynamic model is created to estimate the recovering efficiency in the Pidgeon. The result shows that the CaO/MgO molar ratio of calcination dolomite in Thanhhoa is nearly 1.5 which is suitable to produce magnesium in the case of highly-required efficiency and pureness. Besides, the result from the furnace of the experiment is lower than the one in the model. The samples are set up to check the influence of the rate of ferrosilicon in the compound. The result indicates that the ideal efficiency reaches 85 % with 30 % ferrosilicon. Moreover, the study confirms that the optimal operating conditions in this process are recovering during three hours at 1200 °C and 100 Pa pressure. This result proves the potential application of Thanhhoa dolomite in the industry suitable with the current condition in Vietna

    An efficient protocol for in vitro regeneration of peanut (Arachis hypogaea L.) cultivar L14

    The present work aims to establish an efficient protocol for in vitro regeneration of peanut (Arachis hypogaea) cultivar L14. The study showed that de-embryonated cotyledon was a suitable explant for shoot multiplication on MS medium containing 4 mg/L BAP. The highest number of shoots per explant obtained after 4 weeks of culture was up to 6.8 shoots. Shoots in vitro were able to produce a large number of approximately 11 roots on MS medium supplemented with 0.5 mg/L NAA. These results will be very useful in establishing an in vitro regeneration protocol for peanut cultivar L14 during gene transfer in the next studies to improve their disease resistance

    Development of a one-step multiplex (RT)-PCR for simultaneous detection of virus-induced respiratory disease complex in dogs in Vietnam

    Canine infectious respiratory disease (CIRD) viruses, particularly canine distemper virus (CDV), canine adenovirus type 2 (CAV-2), and canine parainfluenza virus (CPIV), are the primary agents responsible for respiratory diseases in dogs. A dog can be infected with a single or multiple viruses with similar clinical signs. Although CIRD is globally prevalent in dogs, investigation into the causative viruses in Vietnam remains limited. This study aims to identify the presence of CIRD virus(es) in dogs via molecular detection. Three primer sets were newly designed and applied into (RT)-PCR reaction to detect viral genes of CDV, CAV-2, and CPIV in a commercial vaccine (Vanguard® Plus 5/L, Zoetis, USA). Both simplex and multiplex (RT)-PCR reactions using the three primer sets could detect the presence of CAV-2, CDV, and CPIV in the vaccine sample. Subsequently, the applicability of the one-step multiplex (RT)-PCR was demonstrated to test for 27 clinical nasal swab samples collected from dogs suspected of having CIRD. The results showed that our (RT)-PCR could detect CIRD virus(es) in all tested clinical samples. PCR amplicons for each representative CAV-2, CDV, and CPIV were selected for sequencing and showed high genetic similarity to respective field viruses. In conclusion, this study successfully developed a one-step multiplex (RT)-PCR reaction to detect and differentiate causative viruses of CIRD in dogs

    An archetypal metal-organic framework, UiO-6,6, synthesized with the solvo-thermal method, has a large specific surface area of 1071 m2·g–1. This material was modified with NiCl2 in the direct (Nix(tt)/UiO-66) and indirect (Nix(gt)/UiO-66) methods. The two modified materials were characterized by using powder X-ray diffraction, scanning electron microscopy, and infrared spectroscopy. Their crystalline and chemical structure is not much different. However, the thermal and porous properties differ from each other.UiO-66 là họ vật liệu khung hữu cơ kim loại tổng hợp bằng phương pháp nhiệt dung môi có diện tích bề mặt riêng lớn (1071 m2·g–1).     Stem cell-based therapy for human diseases

    Recent advancements in stem cell technology open a new door for patients suffering from diseases and disorders that have yet to be treated. Stem cell-based therapy, including human pluripotent stem cells (hPSCs) and multipotent mesenchymal stem cells (MSCs), has recently emerged as a key player in regenerative medicine. hPSCs are defined as self-renewable cell types conferring the ability to differentiate into various cellular phenotypes of the human body, including three germ layers. MSCs are multipotent progenitor cells possessing self-renewal ability (limited in vitro) and differentiation potential into mesenchymal lineages, according to the International Society for Cell and Gene Therapy (ISCT). This review provides an update on recent clinical applications using either hPSCs or MSCs derived from bone marrow (BM), adipose tissue (AT), or the umbilical cord (UC) for the treatment of human diseases, including neurological disorders, pulmonary dysfunctions, metabolic/endocrine-related diseases, reproductive disorders, skin burns, and cardiovascular conditions. Moreover, we discuss our own clinical trial experiences on targeted therapies using MSCs in a clinical setting, and we propose and discuss the MSC tissue origin concept and how MSC origin may contribute to the role of MSCs in downstream applications, with the ultimate objective of facilitating translational research in regenerative medicine into clinical applications. The mechanisms discussed here support the proposed hypothesis that BM-MSCs are potentially good candidates for brain and spinal cord injury treatment, AT-MSCs are potentially good candidates for reproductive disorder treatment and skin regeneration, and UC-MSCs are potentially good candidates for pulmonary disease and acute respiratory distress syndrome treatment

    The global response: How cities and provinces around the globe tackled Covid-19 outbreaks in 2021

    Background: Tackling the spread of COVID-19 remains a crucial part of ending the pandemic. Its highly contagious nature and constant evolution coupled with a relative lack of immunity make the virus difficult to control. For this, various strategies have been proposed and adopted including limiting contact, social isolation, vaccination, contact tracing, etc. However, given the heterogeneity in the enforcement of these strategies and constant fluctuations in the strictness levels of these strategies, it becomes challenging to assess the true impact of these strategies in controlling the spread of COVID-19.Methods: In the present study, we evaluated various transmission control measures that were imposed in 10 global urban cities and provinces in 2021 Bangkok, Gauteng, Ho Chi Minh City, Jakarta, London, Manila City, New Delhi, New York City, Singapore, and Tokyo.Findings: Based on our analysis, we herein propose the population-level Swiss cheese model for the failures and pit-falls in various strategies that each of these cities and provinces had. Furthermore, whilst all the evaluated cities and provinces took a different personalized approach to managing the pandemic, what remained common was dynamic enforcement and monitoring of breaches of each barrier of protection. The measures taken to reinforce the barriers were adjusted continuously based on the evolving epidemiological situation.Interpretation: How an individual city or province handled the pandemic profoundly affected and determined how the entire country handled the pandemic since the chain of transmission needs to be broken at the very grassroot level to achieve nationwide control

    Burden of cancer in the Eastern Mediterranean Region, 2005-2015: findings from the Global Burden of Disease 2015 Study

INTERNATIONAL JOURNAL OF PUBLIC HEALTH. 2018;63(Suppl. 1):151-164.To estimate incidence, mortality, and disability-adjusted life years (DALYs) caused by cancer in the Eastern Mediterranean Region (EMR) between 2005 and 2015. Vital registration system and cancer registry data from the EMR region were analyzed for 29 cancer groups in 22 EMR countries using the Global Burden of Disease Study 2015 methodology. In 2015, cancer was responsible for 9.4% of all deaths and 5.1% of all DALYs. It accounted for 722,646 new cases, 379,093 deaths, and 11.7 million DALYs. Between 2005 and 2015, incident cases increased by 46%, deaths by 33%, and DALYs by 31%. The increase in cancer incidence was largely driven by population growth and population aging. Breast cancer, lung cancer, and leukemia were the most common cancers, while lung, breast, and stomach cancers caused most cancer deaths. Cancer is responsible for a substantial disease burden in the EMR, which is increasing. There is an urgent need to expand cancer prevention, screening, and awareness programs in EMR countries as well as to improve diagnosis, treatment, and palliative care services
