Analysis of Mono-, Di- and Oligosaccharides by CE Using a Two-Stage Derivatization Method and LIF Detection.
A sensitive CE with LIF method has been developed for quantitative analysis of small carbohydrates. In this work, 17 carbohydrates including mono-, di- and oligosaccharides
were simultaneously derivatized with 4-fluoro 7-nitrobenzo furazane (NBD-F) via a twostep reaction involving reductive amination with ammonia followed by condensation with NBD-F. Under the optimized derivatization conditions all carbo-hydrates were successfully derivatized within 2.5 h and separated within 15 min using borate buffer (90 mmol/L, pH 9.2). For sugar standards LODs were in the range of 49.7 to
243.6 nmol/L. Migration time and peak area reproducibility were better than RSD 0.1 and 3%, respectively. The method was applied to measure sugars in nanoliter volume samples of phloem sap obtained by stylectomy from wheat and to honeydew samples obtained from aphids feeding from wheat and willow
Fundamental noise limitations to supercontinuum generation in microstructure fiber
Broadband noise on supercontinuum spectra generated in microstructure fiber
is shown to lead to amplitude fluctuations as large as 50 % for certain input
laser pulse parameters. We study this noise using both experimental
measurements and numerical simulations with a generalized stochastic nonlinear
Schroedinger equation, finding good quantitative agreement over a range of
input pulse energies and chirp values. This noise is shown to arise from
nonlinear amplification of two quantum noise inputs: the input pulse shot noise
and the spontaneous Raman scattering down the fiber.Comment: 16 pages with 6 figure
The Genetics of Language Acquisition
This chapter focuses on the understanding of the role of genetics in language and explores how genetics contribute to language, and shows how new genetic techniques can offer inroads into the molecular basis of language acquisition. It discusses some of the key findings of gene x environment studies and provides a snapshot of the understanding in the field, considering some of the limitations of the type of study design. The chapter describes the field of play in the genetics of language acquisition and explains the heritability of language and the role of family and twin studies in the understanding of language. It also explores the inheritance mechanisms that are implicated in language development. The chapter considers how modern DNA sequencing approaches are revolutionizing the field of language genetics. Heritability studies have provided many key insights into the genetics of both language acquisition and language disorders. Insights into mechanisms can also come from the opposite end of the language ability spectrum
Full phase stabilization of a Yb:fiber femtosecond frequency comb via high-bandwidth transducers
We present full phase stabilization of an amplified Yb:fiber femtosecond
frequency comb using an intra-cavity electro-optic modulator and an
acousto-optic modulator. These transducers provide high servo bandwidths of 580
kHz and 250 kHz for frep and fceo, producing a robust and low phase noise fiber
frequency comb. The comb was self-referenced with an f - 2f interferometer and
phase locked to an ultra-stable optical reference used for the JILA Sr optical
clock at 698 nm, exhibiting 0.21 rad and 0.47 rad of integrated phase errors
(over 1 mHz - 1 MHz) respectively. Alternatively, the comb was locked to two
optical references at 698 nm and 1064 nm, obtaining 0.43 rad and 0.14 rad of
integrated phase errors respectively
A mutation in amino acid permease AAP6 reduces the amino acid content of the Arabidopsis sieve elements but leaves aphid herbivores unaffected
The aim of this study was to investigate the role of the amino acid permease gene AAP6 in regulating phloem amino acid composition and then to determine the effects of this altered diet on aphid performance. A genotype of Arabidopsis thaliana (L.) was produced in which the function of the amino acid permease gene AAP6 (At5g49630) was abolished. Plants homozygous for the insertionally inactivated AAP6 gene had a significantly larger mean rosette width than the wild type and a greater number of cauline leaves. Seeds from the aap6 mutant were also significantly larger than those from the wild-type plants. Sieve element (SE) sap was collected by aphid stylectomy and the amino acids derivatized, separated, and quantified using Capillary Electrophoresis with Laser Induced Fluorescence (CE-LIF). In spite of the large variation across samples, the total amino acid concentration of SE sap of the aap6 mutant plants was significantly lower than that of the wild-type plants. The concentrations of lysine, phenylalanine, leucine, and aspartic acid were all significantly lower in concentration in the aap6 mutant plants compared with wild-type plants. This is the first direct demonstration of a physiological role for an amino acid transporter in regulating SE composition in vivo. The amino acid availability in sieve element sap is thought to be the major limiting factor for aphid growth and reproduction. Despite the changes in their diet, the aphid Myzus persicae (Sulzer) displayed only small changes in feeding behaviour on mutant plants when measured using the Electronic Penetration Graph (EPG) technique. Salivation by the aphid into the SE (E1 phase) was increased on mutant plants but there was no significant effect on other feeding EPG behaviours, or in the rate of honeydew production. Consistent with the small effect on aphid feeding behaviour, there was only a small effect of reduced sieve element amino acid concentration on aphid reproduction. The data are discussed in relation to the regulation of phloem composition and the role of phloem amino acids in regulating aphid performance
The Dynamic Use of the Kawa Model: A Scoping Review
Background: The Kawa model, a framework to guide culturally relevant occupational therapy, has gained recognition and become more widely used in practice. Research on the model thus far, while still relatively sparse, provides guidance for the model’s use, including its strengths and facets that require further exploration to support its use and effectiveness in dynamic ways.
Method: A scoping review was completed to gather, organize, appraise, and synthesize the current research evidence on use of the model.
Results: Findings support the Kawa model’s culturally flexible application and its capacity to garner client-centered qualitative information, as well as to build therapeutic relationships in a variety of settings. Challenges to the model’s use include therapists’ inexperience limiting effectiveness and the need for additional quantitative assessment measures to supplement the qualitative findings gathered during use of the Kawa. Limitations to this review include author preconceptions, homogeneity among the authors, and inclusion of non-peer-reviewed theses.
Conclusion: The Kawa model is an adaptable tool to examine and enhance well-being. It may be most effective when used by experienced therapists and in conjunction with other relevant tools. Further research is recommended to continue to evaluate its dynamic use
Generalized Structured Component Analysis in candidate gene association studies: Applications and limitations [version 2; peer review: 3 approved]
Background: Generalized Structured Component Analysis (GSCA) is a component-based alternative to traditional covariance-based structural equation modelling. This method has previously been applied to test for association between candidate genes and clinical phenotypes, contrasting with traditional genetic association analyses that adopt univariate testing of many individual single nucleotide polymorphisms (SNPs) with correction for multiple testing. Methods: We first evaluate the ability of the GSCA method to replicate two previous findings from a genetics association study of developmental language disorders. We then present the results of a simulation study to test the validity of the GSCA method under more restrictive data conditions, using smaller sample sizes and larger numbers of SNPs than have previously been investigated. Finally, we compare GSCA performance against univariate association analysis conducted using PLINK v1.9. Results: Results from simulations show that power to detect effects depends not just on sample size, but also on the ratio of SNPs with effect to number of SNPs tested within a gene. Inclusion of many SNPs in a model dilutes true effects. Conclusions: We propose that GSCA is a useful method for replication studies, when candidate SNPs have been identified, but should not be used for exploratory analysis
Perceptions of body weight that vary by body mass index: Clear associations with perceptions based on personal control and responsibility.
This project aimed to identify the perceptions of body weight that vary by body mass index. First, a qualitative study explored body weight perceptions in 17 individuals with overweight. Second, a questionnaire was developed and completed by a UK sample with body mass index from 16.6 to 59.7 kg/m2 (N = 328). A higher body mass index was associated with perceptions of less personal control and responsibility. Body mass index in females was also associated with three other questionnaire factors and body mass index in males with illness/medication. Thus, body mass index was associated with different perceptions of body weight. Focussing on personal control and responsibility may be useful for treatment and prevention
