Efficient Algorithms for Optimal Control of Quantum Dynamics: The "Krotov'' Method unencumbered
Efficient algorithms for the discovery of optimal control designs for
coherent control of quantum processes are of fundamental importance. One
important class of algorithms are sequential update algorithms generally
attributed to Krotov. Although widely and often successfully used, the
associated theory is often involved and leaves many crucial questions
unanswered, from the monotonicity and convergence of the algorithm to
discretization effects, leading to the introduction of ad-hoc penalty terms and
suboptimal update schemes detrimental to the performance of the algorithm. We
present a general framework for sequential update algorithms including specific
prescriptions for efficient update rules with inexpensive dynamic search length
control, taking into account discretization effects and eliminating the need
for ad-hoc penalty terms. The latter, while necessary to regularize the problem
in the limit of infinite time resolution, i.e., the continuum limit, are shown
to be undesirable and unnecessary in the practically relevant case of finite
time resolution. Numerical examples show that the ideas underlying many of
these results extend even beyond what can be rigorously proved.Comment: 19 pages, many figure
Universities and community-based research in developing countries: community voice and educational provision in rural Tanzania
The main focus of recent research on the community engagement role of universities has been in developed countries, generally in towns and cities and usually conducted from the perspectives of universities rather than the communities with which they engage. The purpose of this paper is to investigate the community engagement role of universities in the rural areas of developing countries, and its potential for strengthening the voice of rural communities. The particular focus is on the provision of primary and secondary education. The paper is based on the assumption that in order for community members to have both the capacity and the confidence to engage in political discourse for improving educational capacity and quality, they need the opportunity to become involved and well-versed in the options available, beyond their own experience. Particular attention is given in the paper to community-based research (CBR). CBR is explored from the perspectives of community members and local leaders in the government-community partnerships which have responsibility for the provision of primary and secondary education in rural Tanzania. The historical and policy background of the partnerships, together with findings from two case studies, provide the context for the paper
Global analyses of TetR family transcriptional regulators in mycobacteria indicates conservation across species and diversity in regulated functions
BACKGROUND: Mycobacteria inhabit diverse niches and display high metabolic versatility. They can colonise both humans and animals and are also able to survive in the environment. In order to succeed, response to environmental cues via transcriptional regulation is required. In this study we focused on the TetR family of transcriptional regulators (TFTRs) in mycobacteria. RESULTS: We used InterPro to classify the entire complement of transcriptional regulators in 10 mycobacterial species and these analyses showed that TFTRs are the most abundant family of regulators in all species. We identified those TFTRs that are conserved across all species analysed and those that are unique to the pathogens included in the analysis. We examined genomic contexts of 663 of the conserved TFTRs and observed that the majority of TFTRs are separated by 200 bp or less from divergently oriented genes. Analyses of divergent genes indicated that the TFTRs control diverse biochemical functions not limited to efflux pumps. TFTRs typically bind to palindromic motifs and we identified 11 highly significant novel motifs in the upstream regions of divergently oriented TFTRs. The C-terminal ligand binding domain from the TFTR complement in M. tuberculosis showed great diversity in amino acid sequence but with an overall architecture common to other TFTRs. CONCLUSION: This study suggests that mycobacteria depend on TFTRs for the transcriptional control of a number of metabolic functions yet the physiological role of the majority of these regulators remain unknown. Hepatitis C Virus Infection May Lead to Slower Emergence of P. falciparum in Blood
International audienceBACKGROUND: Areas endemic for Plasmodium falciparum, hepatitis B virus (HBV) and hepatitis C virus (HCV) overlap in many parts of sub-Saharan Africa. HBV and HCV infections develop in the liver, where takes place the first development stage of P. falciparum before its further spread in blood. The complex mechanisms involved in the development of hepatitis may potentially influence the development of the liver stage of malaria parasites. Understanding the molecular mechanisms of these interactions could provide new pathophysiological insights for treatment strategies in Malaria. METHODOLOGY: We studied a cohort of 319 individuals living in a village where the three infections are prevalent. The patients were initially given a curative antimalarial treatment and were then monitored for the emergence of asexual P. falciparum forms in blood, fortnightly for one year, by microscopy and polymerase chain reaction. PRINCIPAL FINDINGS: At inclusion, 65 (20.4%) subjects had detectable malaria parasites in blood, 36 (11.3%) were HBV chronic carriers, and 61 (18.9%) were HCV chronic carriers. During follow-up, asexual P. falciparum forms were detected in the blood of 203 patients. The median time to P. falciparum emergence in blood was respectively 140 and 120 days in HBV- and HBV+ individuals, and 135 and 224 days in HCV- and HCV+ individuals. HCV carriage was associated with delayed emergence of asexual P. falciparum forms in blood relative to patients without HCV infection. CONCLUSIONS: This pilot study represents first tentative evidence of a potential epidemiological interaction between HBV, HCV and P. falciparum infections. Age is an important confounding factor in this setting however multivariate analysis points to an interaction between P. falciparum and HCV at the hepatic level with a slower emergence of P. falciparum in HCV chronic carriers. More in depth analysis are necessary to unravel the basis of hepatic interactions between these two pathogens, which could help in identifying new therapeutic approaches against malaria
Safety and immunogenicity of radiation-attenuated PfSPZ vaccine in equatoguinean infants, children, and adults
The radiation-attenuated Plasmodium falciparum sporozoites (PfSPZ) Vaccine has demonstrated safety and immunogenicity in 5-month-old to 50-year-old Africans in multiple trials. Except for one, each trial has restricted enrollment to either infants and children or adults /= 0.17). There were no significant differences between vaccinees and controls with respect to the rates or severity of unsolicited AEs or laboratory abnormalities. Development of antibodies to P. falciparum circumsporozoite protein occurred in 67/69 vaccinees (97%) and 0/15 controls. Median antibody levels were highest in infants and 1-5-year-olds and declined progressively with age. Antibody responses in children were greater than in adults protected against controlled human malaria infection. Robust immunogenicity, combined with a benign AE profile, indicates children are an ideal target for immunization with PfSPZ Vaccine
MycoRRdb: A Database of Computationally Identified Regulatory Regions within Intergenic Sequences in Mycobacterial Genomes
The identification of regulatory regions for a gene is an important step towards deciphering the gene regulation. Regulatory regions tend to be conserved under evolution that facilitates the application of comparative genomics to identify such regions. The present study is an attempt to make use of this attribute to identify regulatory regions in the Mycobacterium species followed by the development of a database, MycoRRdb. It consist the regulatory regions identified within the intergenic distances of 25 mycobacterial species. MycoRRdb allows to retrieve the identified intergenic regulatory elements in the mycobacterial genomes. In addition to the predicted motifs, it also allows user to retrieve the Reciprocal Best BLAST Hits across the mycobacterial genomes. It is a useful resource to understand the transcriptional regulatory mechanism of mycobacterial species. Assessing Africa-wide pangolin exploitation by scaling local data
Overexploitation is one of the main pressures driving wildlife closer to extinction, yet broad-scale data to evaluate species’ declines are limited. Using African pangolins (Family: Pholidota) as a case study, we demonstrate that collating local-scale data can provide crucial information on regional trends in exploitation of threatened species to inform conservation actions and policy. We estimate that 0.4-2.7 million pangolins are hunted annually in Central African forests. The number of pangolins hunted has increased by ~150% and the proportion of pangolins of all vertebrates hunted increased from 0.04% to 1.83% over the past four decades. However, there were no trends in pangolins observed at markets, suggesting use of alternative supply chains. We found evidence that the price of giant (Smutsia gigantea) and arboreal (Phataginus sp.) pangolins in urban markets has increased, mirroring trends in Asian pangolins. Efforts and resources are needed to increase law enforcement and population monitoring, and investigate linkages between subsistence hunting and illegal wildlife trade
Interaction of Temperature and Light in the Development of Freezing Tolerance in Plants
Abstract Freezing tolerance is the result of a wide range
of physical and biochemical processes, such as the induction
of antifreeze proteins, changes in membrane composition,
the accumulation of osmoprotectants, and changes
in the redox status, which allow plants to function at low
temperatures. Even in frost-tolerant species, a certain period
of growth at low but nonfreezing temperatures, known
as frost or cold hardening, is required for the development
of a high level of frost hardiness. It has long been known
that frost hardening at low temperature under low light
intensity is much less effective than under normal light
conditions; it has also been shown that elevated light
intensity at normal temperatures may partly replace the
cold-hardening period. Earlier results indicated that cold
acclimation reflects a response to a chloroplastic redox
signal while the effects of excitation pressure extend
beyond photosynthetic acclimation, influencing plant
morphology and the expression of certain nuclear genes
involved in cold acclimation. Recent results have shown
that not only are parameters closely linked to the photosynthetic
electron transport processes affected by light
during hardening at low temperature, but light may also
have an influence on the expression level of several other
cold-related genes; several cold-acclimation processes can
function efficiently only in the presence of light. The
present review provides an overview of mechanisms that
may explain how light improves the freezing tolerance of
plants during the cold-hardening period
