ABSTRACTBackground: Nigella sativa, a widely used medicinal plant throughout the world belongs to family ranunculaceae. Its Seeds and oil have a long history of folklore usage in various traditional systems of medicines and food. This study was performed to evaluate the protective effect of Nigella sativa oil (NSO) on kidney when simultaneously given with colistin sulfate (CS) which induces tubular damage in rats. Methods: Animals were treated for 7 days: Group I (n=6) with normal saline and CMC, Group II, III and IV with 300.000IU/kg/day of CS (n=6). Group III and Group IV with NSO at the dose of 1 and 2ml/kg per orum prior to CS administration. All the animals were sacrificed on 8th day. Afterwards, the plasma creatinine (pCr), blood urea, renal tissue level of malondialdehyde (MDA), reduced glutathione (GSH) and renal histology were performed. Results: Colistin sulfate induced tubular damage, increased the plasma creatinine (pCr), blood urea and MDA levels and decreased the reduced glutathione (GSH). However, simultaneous treatment with Nigella sativa oil at the dose of 1ml/kg and 2ml/kg for one week produced dose dependant improvement in tubular damage and reduced the biochemical alteration. Conclusions: It could be concluded that, Colistin sulfate induced nephrotoxicity is ameliorated by NS oil especially in higher dose of (2ml/kg). This nephroprotective effect is ascribed to free radical scavenging and potent antioxidant activity in Nigella sativa

Piscicidal effects of several solvent extarcts (distilled water, 50% ethanol, absolute ethanol and 80% methanol) of Terminalia arjuna leaves, barks and fruits were studied against a common fresh water predatory fish, Heteropneustes fossilis under laboratory conditions in terms of Behavior and mortality of fishes after 24 h. Fishes exposed to plant extracts showed agitating movement with quick surfacing, loss of balance, mucus secretion, and finally died. The LC50 values of distilled water, 50% ethanol, absolute ethanol and 80% methanol extracts were found to be 311.726, 236.141, 183.541, 478.794 ppm for leaves, 117.894, 96.998, 38.990, 304.193 ppm for barks and 1400.033, 949.209, 555.201, 875.158 ppm for fruits, respectively. Chi-square values were found to be insignificant at P0.01. Based on LC50 values, order of piscicidal activity followed the pattern, bark > leaf > fruit extracts. Order of piscicidal activity for the extracts was like absolute ethanol > 50% ethanol > distilled water > 80% methanol for leaf and bark, while for fruit extracts, trend was like absolute ethanol > 80% methanol > 50% ethanol > distilled water

The present study was undertaken to fix the suitable concentration of 6-benzyl aminopurine on shoot development from meristem for producing virus free potato plantlet. The experiment consisted of five potato cultivars namely Diamant, Heera, Dheera, Granula and Cardinal for meristem culture and four 6-benzyl aminopurine levels namely 0, 1.0, 1.5, 2.0 mg/l. As a whole, twenty treatments were allotted in complete randomized design with three replications. Resulted in vitro regenerated plantlets were used as treatment for double antibody sandwich (DAS-), enzyme linked immunosorbent assay (ELISA) test, while, the infected plant parts were used as positive control. In virus elimination through meristem culture, Cardinal with 1 mg l-1 6-benzyl aminopurine gave maximum number of shoots (2.43 cm) explant-1, whereas, Dheera with 1.5 mg l-1 6-benzyl aminopurine gave the tallest plantlet (5.23 cm). On the other hand, explants of Dheera and Cardinal with 1.5 mg l-1 6-benzyl aminopurine and explants of Diamant, Heera, Granula and Cardinal with 2.0 mg l-1 6-benzyl aminopurine produced no roots. Finally, after DAS-ELISA test, the infected field samples developed yellow color but in vitro regenerated plantlets of all the varieties under study showed 100% virus freeness. Keywords: Virus elimination, meristem culture, virus detection, seed potatoAfrican Journal of Biotechnology Vol. 12(18), pp. 2406-241
