    Increasing the efficiency of utilization of soil and fertilizer phosphorus in the subtropical and tropical agricultural systems C

    As a rule, soils of the subtropical and tropical regions, in which rainfall is not limiting, are acidic, and low in phosphorus, and, to a less extent, in other macro and micronutrients as well, such a sulfur, boron and zinc. The establishment of a permanent agricultural prac. tice therefore, demands relatively high usage of liming and phosphatic fertilization, to begin with. Several approaches, not mutually exclusive, could be used in order to increase the efficiency of utilization of soil and fertilizer phosphorus so that, goal of diminishing costs of production is reached. The use of liming materials bringing up pH to 6.0-6.5 causes the conversion of iron and aluminum phosphates to more available calcium phosphates; on the other hand, by raising calcium saturation in the exchange complex, it improves the development and operation if the root system which allows c or a higher utilization of all soil nutrients, including phosphorus, and helps of stand water deficits which may occur. The role of mycorrhizal fungi should be considered as a way of increasing soil and fertilizer P utilization, as well as the limitations thereof. Screening of and breeding for varieties with higher efficiency of uptake and utilization of soil and fertilizer phosphorus leads to a reduction in cost of inputs and to higher benefit/cost ratios. Corrective fertilization using ground rock phosphate helps to saturate the fixation power of the soil thereby reducing, as a consequence, the need for phosphorus in the maintenance fertilization. Maintenance fertilization, in which soluble phos-phatic sources are used, could be improved by several means whose performance has been proved: limimg, granula tion, placement, use of magnesium salts. Last, cost of phosphate fertilization could be further reduced, without impairing yields, through impairing yields, through changes in technology designed to obtain products better adapted to local conditions and to the availability or raw materials and energy sources.A falta de P no solo é o fator que com mais freqüência limita a produção nas condições brasileiras, particularmente a das culturas anuais.     Adubacão foliar de mudas de café (Coffea arabica, L., var. Mundo Nôvo) com três fontes de nitrogênio - 15N

    This work was carried out with the aim of evaluating the effect of foliar application of nitrogen in young coffee plants (Coffea arabica L., var. Mundo- Novo). Three sources of nitrogen fertilizers were use: NaN0(3), (NH4)2SO4 and (NH2)2CO 0,7, 0,6 and 0,4 atom % N15 in excess, respectively. Three different methods of application for each source were used: 1) total application of the dosis to the soil; 2) total application of the dosis to the leaves; and 3) fractional application of the dosis, one half to the soil and one half by foliar application. The corresponding checks were also included. The experimental design used was complete randomized blocks, including 14 treatments replicated 3 times. Part of these treatments was in factorial arrangement. The 6 months old coffee plants were grown in a greenhouse during a period of 7 months. The plants were then separated in three fractions: root, stem and leaves. These fractions were oven dried, weighed, ground and total nitrogen and the isotopic relation of 14N/15N were determined. The dry matter yield, the amount of total nitrogen uptake, the quantity of this nutrient coming from the soil and fertilizer, for each treatment and fraction and for the whole plant, were the factors utilized to evaluate the effects of foliar application of the tested nitrogen sources. For the foliar application of nitrogen, a special system of spraying was tried. The most outstanding conclusions obtained are as follows : As a general rule, the effect of the three sources was always similar and the application to the soil followed by the application one half to the soil and one half foliar aplication were responsible for the increased dry matter yield. In relation to the total nitrogen uptake, a higher uptake was obtained with the application to the soil compared to foliar application, for the three nitrogen sources under study. In respect to the nitrogen in these young coffee plants coming from the fertilizes, the application to the soil was also superior to the foliar application. For foliar application, the efficiency of the nitrogen was 12.8%, 9.7% and 8.6% for sodium nitrate, urea and ammonium sulphate, respectively. In one half soil and one half forliar application, the efficiency was 33.5%, 23.3% and 22.7% for sodium nitrate, ammonium sulphate and urea, respectivaly. Nitrogen translocation from the top to the roots of the plant was evident when applied through the leaves. In the half to the soil half to the leaves treatment with ammonium sulphate, no similar results were obtained. It is difficult to explain the low absorption of nitrogen applied to the leaves for the three sources, sodium nitrate being superior to urea.O presente trabalho foi conduzido com a finalidade de se avaliar o efeito da aplicação foliar no nitrogênio em mudas de café (Coffea arabica L, var. Mundo Nôvo). Para isso, três fontes de fertilizantes nitrogenados - NaNO3, (NH4)2SO4 e (NH2)2CO - enriquecidos com 0,7, 0,6 e 0,4% de átomos de 15N em excesso, respectivamente, foram utilizados, aplicando-as de vários modos: 1) aplicação da dose total de cada fonte de nitrogênio ao solo; 2) aplicação da dose total de cada fonte exclusivamente foliar e 3) aplicação da metade desta dose total ao solo e da outra metade por via foliar, além das correspondentes testemunhas. O desenho experimental foi de blocos ao acaso, com 14 tratamentos e 3 repetições, com parte dos tratamentos em arranjo fatorial. As mudas de café, de 6 meses de idade foram cultivadas em casa de vegetação, durante um período de 7 meses. A seguir, as palantas foram separadas em frações raiz, caule e folha. Sobre estas frações, secas em estufa, pesadas e moídas, procedeu-se às análises químicas do nitrogênio e à determinação da relação isotópica 14N/15N. A produção de matéria seca, a quantidade de nitrogênio absorvido, aquela proveniente do solo e do fertilizante, por cada tratamento, em cada fração e na planta toda. foram utilizados para avaliar o efeito da adubação foliar ds fontes de nitrogênio. Pra a aplicação foliar do nitrogênio, foi construído um sistema espacial de pulverização. De acordo com os resultados obtidos são apresentadas, a continuação, as seguintes inferências. De maneira geral, a aplicação das três fontes de nitrogênio ao solo, seguida pela aplicação combinada ao solo e à folha, deram maior produção de matéria seca, não havendo diferença entre as fontes. Em relação a absorção do nitrogênio total, foi verificada uma maior absorção com a aplicação ao solo das três fontes de nitrogênio. No que diz respeito ao nitrogênio nas plantulas provenientes do fertilizante, a aplicação ao solo foi superior a aplicação foliar, para as três fontes. A porcentagem da eficiência do nitrogênio foi de 39%. Na aplicação foliar, a eficiência do nitrogênio foi 12,8%, 9,7% e 8,6% para nitrato de sódio, a uréia e o sulfato de amônio respectivamente. Na aplicação combinada, metade solo mais metade folha, a eficiência foi de 33,5%, 23,3% e 22,7% para o nitrato de sódio, o sulfato de amônio e a uréia, respectivamente. É difícil, no presente trabalho, explicar a pouca absorção do nitrogênio aplicada por pulverização foliar, destacando-se o nitrato de sódio em relação Ys outras fontes

    Efeito do inibidor de nitrificação «AM» em alguns solos do estado de São Paulo

    The nitrification inhibitor "AM" was tested in four different soils of São Paulo State, in order to determine the effective dose for nitrification inhibition for each soil. The doses of "AM" used (0, 12, 24, 36, 48 and 60 ppm) were not sufficient to act significantly in the Latossol Roxo and Podzolizado of Lins and Marília, var. Marilia soils; however, positive results were obtained in the Podzolizado of Piracicaba and Regossol Intergrade soils. For the first soil, doses of 48 to 60 ppm were sufficient to act for a period of 60 days and for the second, the "AM" was effective with 24 ppm. for a same period of time.Utilizou-se o inibidor de nitrificação "AM" em quatro solos diferentes do Estado de São Paulo, a fim de se determinar a dose efetiva de inibição da nitrificação em cada solo. As doses usadas (0,12, 24, 36, 48 e 60 ppm) de "AM" não foram suficientes para atuarem significativamente no Latossol Roxo e no Podzolizado de Lins e Miarília, var. Marília; porém resultados positivos foram obtidos com os solos Podzolizado de Piracicaba e Regossol Intergrade, para os quais,, doses de 24 a 60 ppm foram suficientes para atuarem por um período de 60 dias

    LRP1 Functions as an Atheroprotective Integrator of TGFβ and PDGF Signals in the Vascular Wall: Implications for Marfan Syndrome

    BACKGROUND: The multifunctional receptor LRP1 controls expression, activity and trafficking of the PDGF receptor-β in vascular smooth muscle cells (VSMC). LRP1 is also a receptor for TGFβ1 and is required for TGFβ mediated inhibition of cell proliferation. METHODS AND PRINCIPAL FINDINGS: We show that loss of LRP1 in VSMC (smLRP(−)) in vivo results in a Marfan-like syndrome with nuclear accumulation of phosphorylated Smad2/3, disruption of elastic layers, tortuous aorta, and increased expression of the TGFβ target genes thrombospondin-1 (TSP1) and PDGFRβ in the vascular wall. Treatment of smLRP1(−) animals with the PPARγ agonist rosiglitazone abolished nuclear pSmad accumulation, reversed the Marfan-like phenotype, and markedly reduced smooth muscle proliferation, fibrosis and atherosclerosis independent of plasma cholesterol levels. CONCLUSIONS AND SIGNIFICANCE: Our findings are consistent with an activation of TGFβ signals in the LRP1-deficient vascular wall. LRP1 may function as an integrator of proliferative and anti-proliferative signals that control physiological mechanisms common to the pathogenesis of Marfan syndrome and atherosclerosis, and this is essential for maintaining vascular wall integrity

    LTBP2 null mutations in an autosomal recessive ocular syndrome with megalocornea, spherophakia, and secondary glaucoma

    The latent TGFβ-binding proteins (LTBPs) and fibrillins are a superfamily of large, multidomain proteins with structural and TGFβ-signalling roles in the extracellular matrix. Their importance is underscored by fibrillin-1 mutations responsible for Marfan syndrome, but their respective roles are still incompletely understood. We report here on two families where children from healthy, consanguineous parents, presented with megalocornea and impaired vision associated with small, round, dislocated lenses (microspherophakia and ectopia lentis) and myopia, as well as a high-arched palate, and, in older children, tall stature with an abnormally large arm span over body height ratio, that is, associated features of Marfan syndrome. Glaucoma was not present at birth, but was diagnosed in older children. Whole genome homozygosity mapping followed by candidate gene analysis identified homozygous truncating mutations of LTBP2 gene in patients from both families. Fibroblast mRNA analysis was consistent with nonsense-mediated mRNA decay, with no evidence of mutated exon skipping. We conclude that biallelic null LTBP2 mutations cause the ocular phenotype in both families and could lead to Marfan-like features in older children. We suggest that intraocular pressures should be followed-up in young children with an ocular phenotype consisting of megalocornea, spherophakia and/or lens dislocation, and recommend LTBP2 gene analysis in these patients

    Hundreds of variants clustered in genomic loci and biological pathways affect human height

    Most common human traits and diseases have a polygenic pattern of inheritance: DNA sequence variants at many genetic loci influence the phenotype. Genome-wide association (GWA) studies have identified more than 600 variants associated with human traits, but these typically explain small fractions of phenotypic variation, raising questions about the use of further studies. Here, using 183,727 individuals, we show that hundreds of genetic variants, in at least 180 loci, influence adult height, a highly heritable and classic polygenic trait. The large number of loci reveals patterns with important implications for genetic studies of common human diseases and traits. First, the 180 loci are not random, but instead are enriched for genes that are connected in biological pathways (P = 0.016) and that underlie skeletal growth defects (P < 0.001). Second, the likely causal gene is often located near the most strongly associated variant: in 13 of 21 loci containing a known skeletal growth gene, that gene was closest to the associated variant. Third, at least 19 loci have multiple independently associated variants, suggesting that allelic heterogeneity is a frequent feature of polygenic traits, that comprehensive explorations of already-discovered loci should discover additional variants and that an appreciable fraction of associated loci may have been identified. Fourth, associated variants are enriched for likely functional effects on genes, being over-represented among variants that alter amino-acid structure of proteins and expression levels of nearby genes. Our data explain approximately 10% of the phenotypic variation in height, and we estimate that unidentified common variants of similar effect sizes would increase this figure to approximately 16% of phenotypic variation (approximately 20% of heritable variation). Although additional approaches are needed to dissect the genetic architecture of polygenic human traits fully, our findings indicate that GWA studies can identify large numbers of loci that implicate biologically relevant genes and pathways.

    Subcellular optogenetic inhibition of G proteins generates signaling gradients and cell migration

    Cells sense gradients of extracellular cues and generate polarized responses such as cell migration and neurite initiation. There is static information on the intracellular signaling molecules involved in these responses, but how they dynamically orchestrate polarized cell behaviors is not well understood. A limitation has been the lack of methods to exert spatial and temporal control over specific signaling molecules inside a living cell. Here we introduce optogenetic tools that act downstream of native G protein–coupled receptor (GPCRs) and provide direct control over the activity of endogenous heterotrimeric G protein subunits. Light-triggered recruitment of a truncated regulator of G protein signaling (RGS) protein or a Gβγ-sequestering domain to a selected region on the plasma membrane results in localized inhibition of G protein signaling. In immune cells exposed to spatially uniform chemoattractants, these optogenetic tools allow us to create reversible gradients of signaling activity. Migratory responses generated by this approach show that a gradient of active G protein αi and βγ subunits is sufficient to generate directed cell migration. They also provide the most direct evidence so for a global inhibition pathway triggered by Gi signaling in directional sensing and adaptation. These optogenetic tools can be applied to interrogate the mechanistic basis of other GPCR-modulated cellular functions

    Loss-of-function mutations in the X-linked biglycan gene cause a severe syndromic form of thoracic aortic aneurysms and dissections.

    Thoracic aortic aneurysm and dissection (TAAD) is typically inherited in an autosomal dominant manner, but rare X-linked families have been described. So far, the only known X-linked gene is FLNA, which is associated with the periventricular nodular heterotopia type of Ehlers-Danlos syndrome. However, mutations in this gene explain only a small number of X-linked TAAD families. We performed targeted resequencing of 368 candidate genes in a cohort of 11 molecularly unexplained Marfan probands. Subsequently, Sanger sequencing of BGN in 360 male and 155 female molecularly unexplained TAAD probands was performed. We found five individuals with loss-of-function mutations in BGN encoding the small leucine-rich proteoglycan biglycan. The clinical phenotype is characterized by early-onset aortic aneurysm and dissection. Other recurrent findings include hypertelorism, pectus deformity, joint hypermobility, contractures, and mild skeletal dysplasia. Fluorescent staining revealed an increase in TGF-β signaling, evidenced by an increase in nuclear pSMAD2 in the aortic wall. Our results are in line with those of prior reports demonstrating that Bgn-deficient male BALB/cA mice die from aortic rupture. In conclusion, BGN gene defects in humans cause an X-linked syndromic form of severe TAAD that is associated with preservation of elastic fibers and increased TGF-β signaling.Genet Med 19 4, 386-395

    Tai Chi and vestibular rehabilitation improve vestibulopathic gait via different neuromuscular mechanisms: Preliminary report

    BACKGROUND: Vestibular rehabilitation (VR) is a well-accepted exercise program intended to remedy balance impairment caused by damage to the peripheral vestibular system. Alternative therapies, such as Tai Chi (TC), have recently gained popularity as a treatment for balance impairment. Although VR and TC can benefit people with vestibulopathy, the degree to which gait improvements may be related to neuromuscular adaptations of the lower extremities for the two different therapies are unknown. METHODS: We examined the relationship between lower extremity neuromuscular function and trunk control in 36 older adults with vestibulopathy, randomized to 10 weeks of either VR or TC exercise. Time-distance measures (gait speed, step length, stance duration and step width), lower extremity sagittal plane mechanical energy expenditures (MEE), and trunk sagittal and frontal plane kinematics (peak and range of linear and angular velocity), were measured. RESULTS: Although gait time-distance measures were improved in both groups following treatment, no significant between-groups differences were observed for the MEE and trunk kinematic measures. Significant within groups changes, however, were observed. The TC group significantly increased ankle MEE contribution and decreased hip MEE contribution to total leg MEE, while no significant changes were found within the VR group. The TC group exhibited a positive relationship between change in leg MEE and change in trunk velocity peak and range, while the VR group exhibited a negative relationship. CONCLUSION: Gait function improved in both groups consistent with expectations of the interventions. Differences in each group's response to therapy appear to suggest that improved gait function may be due to different neuromuscular adaptations resulting from the different interventions. The TC group's improvements were associated with reorganized lower extremity neuromuscular patterns, which appear to promote a faster gait and reduced excessive hip compensation. The VR group's improvements, however, were not the result of lower extremity neuromuscular pattern changes. Lower-extremity MEE increases corresponded to attenuated forward trunk linear and angular movement in the VR group, suggesting better control of upper body motion to minimize loss of balance. These data support a growing body of evidence that Tai Chi may be a valuable complementary treatment for vestibular disorders
