    Individually Modified Saliva Delivery Changes the Perceived Intensity of Saltiness and Sourness

    Individuals vary largely in their salivary flow and composition, and given the importance of saliva on perception of taste, this might influence how the tastant stimuli are perceived. We therefore hypothesise that altering the individual salivary flow rates has an impact on the perceived taste intensity. In this study, we investigated the role of saliva amount on the perceived taste intensity by excluding parotid saliva and adding artificial saliva close to the parotid duct at preset flow rates. Significant decreases in perception with increasing salivary flow rates were observed for citric acid and sodium chloride. This can partially be explained by a dilution effect which is in line with previous studies on detectable concentration differences. However, since the bitterness and sweetness remained unaffected by the salivary flow conditions and the dilution effect was comparable to that of saltiness, further explanation is needed. Furthermore, we investigated whether the suppression of taste intensity in binary mixtures (taste–taste interactions) could possibly be caused by the increased salivary flow rate induced by an additional taste attribute. The results show, however, that suppression of taste intensity in binary mixtures was not affected by the rate of salivation. This was more likely to be explained by psychophysics

    Utilisation of an operative difficulty grading scale for laparoscopic cholecystectomy

    Background A reliable system for grading operative difficulty of laparoscopic cholecystectomy would standardise description of findings and reporting of outcomes. The aim of this study was to validate a difficulty grading system (Nassar scale), testing its applicability and consistency in two large prospective datasets. Methods Patient and disease-related variables and 30-day outcomes were identified in two prospective cholecystectomy databases: the multi-centre prospective cohort of 8820 patients from the recent CholeS Study and the single-surgeon series containing 4089 patients. Operative data and patient outcomes were correlated with Nassar operative difficultly scale, using Kendall’s tau for dichotomous variables, or Jonckheere–Terpstra tests for continuous variables. A ROC curve analysis was performed, to quantify the predictive accuracy of the scale for each outcome, with continuous outcomes dichotomised, prior to analysis. Results A higher operative difficulty grade was consistently associated with worse outcomes for the patients in both the reference and CholeS cohorts. The median length of stay increased from 0 to 4 days, and the 30-day complication rate from 7.6 to 24.4% as the difficulty grade increased from 1 to 4/5 (both p < 0.001). In the CholeS cohort, a higher difficulty grade was found to be most strongly associated with conversion to open and 30-day mortality (AUROC = 0.903, 0.822, respectively). On multivariable analysis, the Nassar operative difficultly scale was found to be a significant independent predictor of operative duration, conversion to open surgery, 30-day complications and 30-day reintervention (all p < 0.001). Conclusion We have shown that an operative difficulty scale can standardise the description of operative findings by multiple grades of surgeons to facilitate audit, training assessment and research. It provides a tool for reporting operative findings, disease severity and technical difficulty and can be utilised in future research to reliably compare outcomes according to case mix and intra-operative difficulty

    Tracking the Atlantic Multidecadal Oscillation through the last 8,000 years

    Understanding the internal ocean variability and its influence on climate is imperative for society. A key aspect concerns the enigmatic Atlantic Multidecadal Oscillation (AMO), a feature defined by a 60- to 90-year variability in North Atlantic sea-surface temperatures. The nature and origin of the AMO is uncertain, and it remains unknown whether it represents a persistent periodic driver in the climate system, or merely a transient feature. Here, we show that distinct, ∼55- to 70-year oscillations characterized the North Atlantic ocean-atmosphere variability over the past 8,000 years. We test and reject the hypothesis that this climate oscillation was directly forced by periodic changes in solar activity. We therefore conjecture that a quasi-persistent ∼55- to 70-year AMO, linked to internal ocean-atmosphere variability, existed during large parts of the Holocene. Our analyses further suggest that the coupling from the AMO to regional climate conditions was modulated by orbitally induced shifts in large-scale ocean-atmosphere circulation

    DNA sequence variation and methylation in an arsenic tolerant earthworm population

    Evidence is emerging that earthworms can evolve tolerance to trace element enriched soils. However, few studies have sought to establish whether such tolerance is determined through adaption or plasticity. Here we report results from a combined analysis of mitochondrial (cytochrome oxidase II, COII), nuclear (amplified fragment length polymorphism, AFLP) variation and DNA methylation in populations of the earthworm Lumbricus rubellus from sites across an abandoned arsenic and copper mine. Earthworms from the mine site population demonstrated clear arsenic tolerance in comparison to a naïve strain. COII and AFLP results suggest that L. rubellus from the unexposed and the adapted populations comprises two cryptic lineages (Linages A and B) each of which was present across all of the sites. AFLP analysis by lineage highlighted variations associated with soil metal/metalloid concentrations (most clearly for Lineage A) suggesting a genetic component to the observed tolerance. The methylation sensitive AFLP (Me-AFLP) identified a high genome methylation content (average 13.5%) in both lineages. For Lineage A, Me-AFLP analysis did not identify a strong association with soil arsenic levels. For Lineage B, however, a clear association of methylation patterns with soil arsenic concentrations was found. This suggests that Lineage B earthworms utilise epigenetic mechanisms to adapt to the presence of contamination. These fundamentally different genetic adjustments in the two clades indicate that the two lineages employ distinct adaptive strategies (genetic or epigenetic) in response to arsenic exposure. Mechanisms driving this variation may be founded within the colonisation histories of the lineages
