    Relationship study of the verified human epidermal growth factor receptor 2 amplification with other tumor markers and clinicohistopathological characteristics in patients with invasive breast cancer, using chromogenic in situ hybridization

    Objective: Human epidermal growth factor receptor 2 (HER-2), as a crucial factor involved in about 20 of breast cancer cases, is one of the most reliable tumor markers to determine prognosis and therapeutic trend of this disease. This marker is generally assessed by immunohistochemistry (IHC) technique. In the cases that result of IHC test cast doubt (+2), the test should be repeated or validated by applying in situ hybridization techniques, like chromogenic in situ hybridization (CISH). In this regard, the goal of current study was to figure out the link between different clinicopathological characteristics of patients suffering from invasive breast cancer, using tumor markers, hormone receptor (HR) and HER-2. Comparing IHC and CISH techniques for evaluating diagnostic value and usefulness of HER-2 were also the other objective of this study. Materials and Methods: Based on this retrospective study, histological markers of 113 individuals suffering from invasive breast cancer -such as estrogen receptor (ER), progesterone receptor, HER-2 receptor, E-cadherin, CK5/6, vimentin and Ki67 were examined by IHC technique. HER-2 amplification of all patients was also evaluated by CISH. Clinicopathological information of the patients was also extracted from medical documents and their associations with tumor markers were statistically evaluated. Results: There is a significant relationship between tumor size, CK5/6 and tumor grade with HR status. Similar relationship was observed between HER-2 status and HR status, as well as vascular invasion (P<0.05). The comparison of HER-2 amplification showed no complete concordance of the result obtained from these two techniques, with score +3. Conclusion: Since the status of HER-2 is very important in decision making of the treatment process, CISH technique is recommended in the malignant conditions as the primary test, instead of IHC. In this study, we also determined that HER-2 expression is greatly correlated with ER- and PR- status. This might propose a better prognosis for HER-2+ patients. © 2019 Royan Institute (ACECR).     A vaccine using Anaplasma marginale subdominant type IV secretion system recombinant proteins was not protective against a virulent challenge

    Anaplasma marginale is the most prevalent tick-borne livestock pathogen with worldwide distribution. Bovine anaplasmosis is a significant threat to cattle industry. Anaplasmosis outbreaks in endemic areas are prevented via vaccination with live A. centrale produced in splenectomized calves. Since A. centrale live vaccine can carry other pathogens and cause disease in adult cattle, research efforts are directed to develop safe recombinant subunit vaccines. Previous work found that the subdominant proteins of A. marginale type IV secretion system (T4SS) and the subdominant elongation factor-Tu (Ef-Tu) were involved in the protective immunity against the experimental challenge in cattle immunized with the A. marginale outer membrane (OM). This study evaluated the immunogenicity and protection conferred by recombinant VirB9.1, VirB9.2, VirB10, VirB11, and Ef-Tu proteins cloned and expressed in E. coli. Twenty steers were randomly clustered into four groups (G) of five animals each. Cattle from G1 and G2 were immunized with a mixture of 50 μg of each recombinant protein with Quil A® or Montanide™ adjuvants, respectively. Cattle from G3 and G4 (controls) were immunized with Quil A and Montanide adjuvants, respectively. Cattle received four immunizations at three-week intervals and were challenged with 107 A. marginale-parasitized erythrocytes 42 days after the fourth immunization. After challenge, all cattle showed clinical signs, with a significant drop of packed cell volume and a significant increase of parasitized erythrocytes (p<0.05), requiring treatment with oxytetracycline to prevent death. The levels of IgG2 induced in the immunized groups did not correlate with the observed lack of protection. Additional strategies are required to evaluate the role of these proteins and their potential utility in the development of effective vaccines.EEA RafaelaFil: Sarli, Macarena.     Development of a pig mammary epithelial cell culture model as a non‐clinical tool for studying epithelial barrier— a contribution from the imi‐conception project

    The ConcePTION project aims at generating further knowledge about the risks related to the use of medication during breastfeeding, as this information is lacking for most commonly used drugs. Taking into consideration multiple aspects, the pig model has been considered by the consortium as the most appropriate choice. The present research was planned to develop an efficient method for the isolation and culture of porcine Mammary Epithelial Cells (pMECs) to study the mammary epithelial barrier in vitro. Mammary gland tissues were collected at a local slaughterhouse, dissociated and the selected cellular population was cultured, expanded and characterized by morphology, cell cycle analysis and immunophenotyping. Their ability to create a barrier was tested by TEER measurement and sodium fluorescein transport activity. Expression of 84 genes related to drug transporters was evaluated by a PCR array. Our results show that primary cells express epithelial cell markers: CKs, CK18, E‐Cad and tight junctions molecules ZO‐1 and OCL. All the three pMEC cellular lines were able to create a tight barrier, although with different strengths and kinetics, and express the main ABC and SLC drug transporters. In conclusion, in the present paper we have reported an efficient method to obtain primary pMEC lines to study epithelial barrier function in the pig model

    Porcine Lawsonia intracellularis Ileitis in Italy and Its Association with Porcine Circovirus Type 2 (PCV2) Infection

    The objective of this study was to employ a diagnostic algorithm, which involves detecting positive farms by stool PCR followed by PCR and histology/immunohistochemistry on ileum samples, for diagnosing Lawsonia intracellularis proliferative enteritis in Northern Italy. The primary aim was to examine the relationship between the gold standard of L. intracellularis diagnostics, namely histology and immunohistochemistry, and PCR in acute and chronic cases of L. intracellularis enteritides. An additional goal was to investigate the coinfection of L. intracellularis with porcine circovirus type 2 (PCV2). Twenty-eight ileum samples, including four from acute cases and 24 from chronic cases, were collected. PCR yielded positive results in 19 cases (four acute and 15 chronic cases). In comparison, immunohistochemistry was positive in 16 cases (four acute and 12 chronic cases), with an observed agreement of 89%. The findings suggest that performing the two tests in series can increase the specificity of the causal diagnosis. PCR may be used as a screening tool to identify the presence of the microorganism, and only positive cases will be examined by histology and immunohistochemistry to confirm the causative role of L. intracellularis. Co-infection with PCV2 was demonstrate in two out of four acute cases and in two out of 24 chronic cases, providing further evidence to support the hypothesis that when the infection starts with ubiquitous pathogens such as L. intracellularis, it may boost the possibility of PCV2 replication, especially in acute cases. As a result, this may trigger a transition from subclinical to clinical forms of PCV2 disease

    Bionic for training: Smart framework design for multisensor mechatronic platform validation

    Home monitoring supports the continuous improvement of the therapy by sharing data with healthcare professionals. It is required when life-threatening events can still occur after hospital discharge such as neonatal apnea. However, multiple sources of external noise could affect data quality and/or increase the misdetection rate. In this study, we developed a mechatronic platform for sensor characterizations and a framework to manage data in the context of neonatal apnea. The platform can simulate the movement of the abdomen in different plausible newborn positions by merging data acquired simultaneously from three-axis accelerometers and infrared sensors. We simulated nine apnea conditions combining three different linear displacements and body postures in the presence of self-generated external noise, showing how it is possible to reduce errors near to zero in phenomena detection. Finally, the development of a smart 8Ws-based software and a customizable mobile application were proposed to facilitate data management and interpretation, classifying the alerts to guarantee the correct information sharing without specialized skills

    Reproductive patterns in the Baluchistan gerbil, Gerbillus nanus (Rodentia: Muridae), from western Saudi Arabia : the role of rainfall and temperature

    There is little information pertaining to the reproductive biology of the Baluchistan gerbil (Gerbillus nanus) despite a broad distribution throughout the Middle East. In the current study, body mass, reproductive–tract morphometrics plus gonadal histology and endocrine profiles of males and females were studied over 12 consecutive months in a field population of Baluchistan gerbils from the western region of Saudi Arabia in an attempt to gain insights into a) the environmental zeitgeber(s) that correlate with reproduction as well as b) to assess if reproduction is seasonal or aseasonal in this species. With the exception of testosterone all male reproductive indicators varied seasonally and were lower during winter than the rest of the year and most increased with rainfall. In contrast, ovarian mass and volume as well as immature follicle counts showed no seasonal patterns and were independent of rainfall or temperature. First pregnancies were observed in late spring coinciding with the increased progesterone concentrations and peaked in summer. This was accompanied by seasonal peaks in the number of Graafian follicles and corpora lutea in females and these parameters were significantly correlated with temperature, but not rainfall In the Arabian Peninsula male, but not female Baluchistan gerbils show seasonal recrudescence and this appears to be correlated largely to rainfall.The Deanship of Scientific Research at the King Saud University through the research group project (No. RGP_VPP_020)http://www.elsevier.com/locate/jaridenvhj201

    Seasonal reproduction in the Arabian spiny mouse, Acomys dimidiatus (Rodentia : Muridae) from Saudi Arabia : the role of rainfall and temperature

    In deserts where unpredictable rainfall arises and a large variation in temperature occurs, this poses severe energetic and water related constraints on reproduction in small mammals and makes the use of photoperiod as cue for reproduction unreliable. In the present study, the gonad morphometrics, gonadal histology and endocrine profiles of the Arabian spiny mouse (Acomys dimidiatus) were studied over 12 consecutive months in a field population from western Saudi Arabia in an attempt to assess the seasonality of reproduction and gain insights into the environmental cues that may trigger reproduction in this species. Although sperm was present throughout the year, most reproductive parameters exhibited a seasonal pattern with a minimum in winter in both sexes and pregnancies were observed from spring to autumn. Whereas testes volume, the number of corpora lutea and progesterone levels were correlated with temperature, all other reproductive parameters were correlated with rainfall. Thus, it appears that A. dimidiatus ceases to breed during winter and uses rainfall as an environmental cue to initiate reproduction in the study area.     Mejora de la memoria gracias al entrenamiento musical en niños en edad preescolar

    El objetivo de este trabajo es examinar el efecto del entrenamiento musical activo y receptivo por un periodo de 4 o 10 semanas sobre la memoria emocional de niños de 4 y 5 años de edad. Se espera que los chicos recuerden más imágenes emocionales que neutras (efecto de arousal). Además, que recuerden más las imágenes negativas que las positivas (efecto de valencia). Por otro lado, se predice que, debido a los patrones esperables de desarrollo, los nenes de mayor edad rindan mejor que los niños más pequeños. Finalmente, se espera que el patrón de recuerdo se vea afectado por el entrenamiento musical, siendo más pronunciadas las diferencias con el entrenamiento activo y que cuanto más tiempo de entrenamiento tenga el niño, mayores sean las diferencias (4 vs 10 semanas).Facultad de ArtesLaboratorio para el Estudio de la Experiencia Musica
