    Detection of individual sub-pixel features in Edge-Illumination X-Ray Phase Contrast Imaging by means of the dark-field channel

    We report on a direct comparison in the detectability of individual sub-pixel-size features between the three complementary contrast channels provided by edge-illumination x-ray phase contrast imaging at constant exposure time and spatial sampling pitch. The dark-field (or ultra-small-angle x-ray scattering) image is known to provide information on sample micro-structure at length scales that are smaller than the system's spatial resolution, averaged over its length. By using a custom-built groove sample, we show how this can also be exploited to detect individual, isolated features. While these are highlighted in the dark-field image, they remain invisible to the phase and attenuation contrast channels. Finally, we show images of a memory SD card as an indication towards potential applications

    A profitability comparison between a “one-way” car sharing service and a "modified one-way" car sharing service

    The research looks at comparing the different performance offered by two types of one-way car sharing services. In particular, we compare the “traditional” service in which users can return vehicles to a pre-determined permitted parking space to the “modified” service in which the decision of where to return the vehicle to is made at the end of its usage and vehicles can be returned also outside the permitted parking areas. The comparison is based on common and given demand/offer assumptions. The mathematical modelling uses state of the art algorithms that allow us to determine for both types of service the optimal number of personnel to re-position the vehicles in order to maximise profit. In particular, the attractiveness of the two services herewith compared, has been analysed both in terms of overall profitability as well as in terms of maximum number of users. The results show and quantify how the “modified” service, whilst allowing a greater degree of flexibility to users in terms of return locations, causes lower economic returns for the service company and lowers the number of users that can be served. Finally, the model allows us to calculate the required tariff increase necessary to transform a “traditional” service into a “modified” service assuming an inelastic demand curve as well as constant profits for the service company

    Methotrexate and Cyclosporine Treatments Modify the Activities of Dipeptidyl Peptidase IV and Prolyl Oligopeptidase in Murine Macrophages

    Analysis of the effects of cyclosporine A (25–28 mgkg−1) and/or methotrexate (0.1 mgkg−1) treatments on dipeptidyl peptidase IV (DPPIV) and prolyl oligopeptidase (POP) activities and on algesic response in two distinct status of murine macrophages (Mφs) was undertaken. In resident Mφs, DPPIV and POP were affected by neither individual nor combined treatments. In thioglycolate-elicited Mφs, methotrexate increased DPPIV (99–110%) and POP (60%), while cyclosporine inhibited POP (21%). Combined treatment with both drugs promoted a rise (51–84%) of both enzyme activities. Only cyclosporine decreased (42%) the tolerance to algesic stimulus. Methotrexate was revealed to exert prevalent action over that of cyclosporine on proinflammatory Mφ POP. The opposite effects of methotrexate and cyclosporine on POP activity might influence the availability of the nociceptive mediators bradykinin and substance P in proinflammatory Mφs. The exacerbated response to thermally induced algesia observed in cyclosporine-treated animals could be related to upregulation of those mediators

    Real-time evaluation of the on-board comfort of standing passenger in bus transit services

    Bus on-board comfort may be intended as a multidimensional concept declined according to several attributes (e.g., vibrations, the load factor, the driving style, etc.) and represents a key factor of service quality. Thus, its measurement is crucial for public transport companies as it can support for the monitoring, evaluation and implementation of specific actions to improve their services. The objective of this paper is to build a numerical scale for real-time measurement of bus on-board comfort. This is pursued integrating subjective measurements of the driving style provided by the passengers with objective ones of longitudinal and transversal accelerations data collected by Intelligent Transportation System tools. The results are very useful because they represent a contribution to establish a comfort scale in a real operational environment, as a tool to regulate the driver's behavior: each driver is in the position of real-time monitoring the quality of a bus ride regarding on-board comfort level

    Migraine and gastrointestinal disorders in middle and old age: A UK Biobank study

    Introduction: Migraine is a prevalent condition causing a substantial level of disability worldwide. Despite this, the pathophysiological mechanisms are not fully understood. Migraine often co-occurs with gastrointestinal disorders, but the direction of a potential causal link is unclear. The aim of this project was to investigate the associations between migraine and several gastrointestinal disorders in the same cohort in order to determine the relative strengths of these associations. Methods: This cross-sectional study examined whether migraine is associated with irritable bowel syndrome (IBS), peptic ulcers, Helicobacter pylori (HP) infections, celiac disease, Crohn's disease and ulcerative colitis. Baseline data covering 489,753 UK Biobank participants (migraine group: n = 14,180) were analyzed using Pearson's chi-square tests and adjusted binary logistic regression models. Results: Migraine was significantly associated with IBS (odds ratio [OR] 2.24, 95% confidence interval [CI] 2.08–2.40, p <.001) and peptic ulcers (OR 1.55, 95% CI 1.35–1.77, p <.001). Migraine was not associated with HP infection (OR 1.34, 95% CI 1.04–1.73, p =.024), celiac disease (OR 1.29, 95% CI 1.04–1.60, p =.023), Crohn's disease (OR 1.08, 95% CI 0.80–1.45, p =.617) or ulcerative colitis (OR 1.00, 95% CI 0.79–1.27, p =.979) after adjusting for multiple testing. Conclusions: Migraine was associated with IBS and peptic ulcers in this large population-based cohort. The associations with HP infection, celiac disease, Crohn's disease, and ulcerative colitis did not reach significance, suggesting a weaker link between migraine and autoimmune gastrointestinal conditions or HP infection

    X-ray phase contrast tomography; proof of principle for post-mortem imaging

    Objectives: To demonstrate the feasibility of using X-ray phase contrast tomography to assess internal organs in a post-mortem piglet model, as a possible non-invasive imaging autopsy technique. Methods: Tomographic images of a new-born piglet were obtained using a Free Space Propagation (FSP) X-ray phase contrast imaging setup at a synchrotron (European Synchrotron Radiation Facility, Grenoble, France). A monochromatic X-ray beam (52 keV) was used in combination with a detector pixel size of 46x46 μm2. A phase-retrieval algorithm was applied to all projections, which were then reconstructed into tomograms using the filtered-back projection algorithm. Images were assessed for diagnostic quality. Results: Images obtained with the FSP setup presented high soft tissue contrast and sufficient resolution for resolving organ structure. All of the main body organs (heart, lungs, kidneys, liver and intestines) were easily identified and adequately visualised. In addition, grey/white matter differentiation in the cerebellum while still contained within the skull was shown. Conclusions: The feasibility of using X-ray phase contrast tomography as a post-mortem imaging technique in an animal model has been demonstrated. Future studies will focus on translating this experiment to a laboratory-based setup

    Maternal age effect and severe germ-line bottleneck in the inheritance of human mitochondrial DNA

    The manifestation of mitochondrial DNA (mtDNA) diseases depends on the frequency of heteroplasmy (the presence of several alleles in an individual), yet its transmission across generations cannot be readily predicted owing to a lack of data on the size of the mtDNA bottleneck during oogenesis. For deleterious heteroplasmies, a severe bottleneck may abruptly transform a benign (low) frequency in a mother into a disease-causing (high) frequency in her child. Here we present a high-resolution study of heteroplasmy transmission conducted on blood and buccal mtDNA of 39 healthy mother–child pairs of European ancestry (a total of 156 samples, each sequenced at ∼20,000× per site). On average, each individual carried one heteroplasmy, and one in eight individuals carried a disease-associated heteroplasmy, with minor allele frequency ≥1%. We observed frequent drastic heteroplasmy frequency shifts between generations and estimated the effective size of the germ-line mtDNA bottleneck at only ∼30–35 (interquartile range from 9 to 141). Accounting for heteroplasmies, we estimated the mtDNA germ-line mutation rate at 1.3 × 10−8 (interquartile range from 4.2 × 10−9 to 4.1 × 10−8) mutations per site per year, an order of magnitude higher than for nuclear DNA. Notably, we found a positive association between the number of heteroplasmies in a child and maternal age at fertilization, likely attributable to oocyte aging. This study also took advantage of droplet digital PCR (ddPCR) to validate heteroplasmies and confirm a de novo mutation. Our results can be used to predict the transmission of disease-causing mtDNA variants and illuminate evolutionary dynamics of the mitochondrial genome

    Conservation and Variability of Dengue Virus Proteins: Implications for Vaccine Design

    Dengue viruses (DENVs) circulate in nature as a population of 4 distinct types, each with multiple genotypes and variants, and represent an increasing global public health issue with no prophylactic and therapeutic formulations currently available. Viral genomes contain sites that are evolutionarily stable and therefore highly conserved, presumably because changes in these sites have deleterious effects on viral fitness and survival. The identification and characterization of the historical dynamics of these sites in DENV have relevance to several applications such as diagnosis and drug and vaccine development. In this study, we have identified sequence fragments that were conserved across the majority of available DENV sequences, analyzed their historical dynamics, and evaluated their relevance as candidate vaccine targets, using various bioinformatics-based methods and immune assay in human leukocyte antigen (HLA) transgenic mice. This approach provides a framework for large-scale and systematic analysis of other human pathogens
