    Multiscale entropy-based analyses of soil transect data

    A deeper understanding of the spatial variability of soil properties and the relationships between them is needed to scale up measured soil properties and to model soil processes. The object of this study was to describe the spatial scaling properties of a set of soil physical properties measured on a common 1024-m transect across arable fields at Silsoe in Bedfordshire, east-central England. Properties studied were volumetric water content ({theta}), total porosity ({pi}), pH, and N2O flux. We applied entropy as a means of quantifying the scaling behavior of each transect. Finally, we examined the spatial intrascaling behavior of the correlations between {theta} and the other soil variables. Relative entropies and increments in relative entropy calculated for {theta}, {pi}, and pH showed maximum structure at the 128-m scale, while N2O flux presented a more complex scale dependency at large and small scales. The intrascale-dependent correlation between {theta} and {pi} was negative at small scales up to 8 m. The rest of the intrascale-dependent correlation functions between {theta} with N2O fluxes and pH were in agreement with previous studies. These techniques allow research on scale effects localized in scale and provide the information that is complementary to the information about scale dependencies found across a range of scale

    The Child Brain Computes and Utilizes Internalized Maternal Choices

    As children grow, they gradually learn how to make decisions independently. However, decisions like choosing healthy but less-tasty foods can be challenging for children whose self-regulation and executive cognitive functions are still maturing. We propose a computational decision-making process in which children estimate their mother's choices for them as well as their individual food preferences. By employing functional magnetic resonance imaging during real food choices, we find that the ventromedial prefrontal cortex (vmPFC) encodes children's own preferences and the left dorsolateral prefrontal cortex (dlPFC) encodes the projected mom's choices for them at the time of children's choice. Also, the left dlPFC region shows an inhibitory functional connectivity with the vmPFC at the time of children's own choice. Our study suggests that in part, children utilize their perceived caregiver's choices when making choices for themselves, which may serve as an external regulator of decision-making, leading to optimal healthy decisions

    A novel bocavirus in canine liver

    Background: Bocaviruses are classified as a genus within the Parvoviridae family of single-stranded DNA viruses and are pathogenic in some mammalian species. Two species have been previously reported in dogs, minute virus of canines (MVC), associated with neonatal diseases and fertility disorders; and Canine bocavirus (CBoV), associated with respiratory disease. Findings: In this study using deep sequencing of enriched viral particles from the liver of a dog with severe hemorrhagic gastroenteritis, necrotizing vasculitis, granulomatous lymphadenitis and anuric renal failure, we identified and characterized a novel bocavirus we named Canine bocavirus 3 (CnBoV3). The three major ORFs of CnBoV3 (NS1, NP1 and VP1) shared less than 60% aa identity with those of other bocaviruses qualifying it as a novel species based on ICTV criteria. Inverse PCR showed the presence of concatemerized or circular forms of the genome in liver. Conclusions: We genetically characterized a bocavirus in a dog liver that is highly distinct from prior canine bocaviruses found in respiratory and fecal samples.     Enhanced mitochondrial superoxide scavenging does not Improve muscle insulin action in the high fat-fed mouse

    Improving mitochondrial oxidant scavenging may be a viable strategy for the treatment of insulin resistance and diabetes. Mice overexpressing the mitochondrial matrix isoform of superoxide dismutase (sod2(tg) mice) and/or transgenically expressing catalase within the mitochondrial matrix (mcat(tg) mice) have increased scavenging of O2(˙-) and H2O2, respectively. Furthermore, muscle insulin action is partially preserved in high fat (HF)-fed mcat(tg) mice. The goal of the current study was to test the hypothesis that increased O2(˙-) scavenging alone or in combination with increased H2O2 scavenging (mtAO mice) enhances in vivo muscle insulin action in the HF-fed mouse. Insulin action was examined in conscious, unrestrained and unstressed wild type (WT), sod2(tg), mcat(tg) and mtAO mice using hyperinsulinemic-euglycemic clamps (insulin clamps) combined with radioactive glucose tracers following sixteen weeks of normal chow or HF (60% calories from fat) feeding. Glucose infusion rates, whole body glucose disappearance, and muscle glucose uptake during the insulin clamp were similar in chow- and HF-fed WT and sod2(tg) mice. Consistent with our previous work, HF-fed mcat(tg) mice had improved muscle insulin action, however, an additive effect was not seen in mtAO mice. Insulin-stimulated Akt phosphorylation in muscle from clamped mice was consistent with glucose flux measurements. These results demonstrate that increased O2(˙-) scavenging does not improve muscle insulin action in the HF-fed mouse alone or when coupled to increased H2O2 scavenging

    UPGro Hidden Crisis Research Consortium : unravelling past failures for future success in Rural Water Supply. Survey 1 Results, Country Report Ethiopia

    Statistics on the functionality of water points from the Hidden Crisis project in Ethiopia are presented. The survey, undertaken in 2016, was focussed on boreholes equipped with handpumps (HPBs) within igneous volcanic rocks in the Ethiopian Highlands (covering approximately 400 Woredas). A stratified two-stage sampling strategy was adopted, and a tiered definition of functionality developed which enabled more nuanced definitions to be reported. The results from the survey indicate: • 82% of HPBs were working on the day of the survey (similar to national statistics) • 59% of HPBs passed the design yield of 10 litres per minute • 45% passed the design yield and also experienced < 1 month downtime within a year. • 28% of HPB’s which passed the design yield and reliability, also passed WHO standards of water quality indicators (TTCs and inorganic chemistry). The results of the survey indicate the utility of carrying out more detailed assessments of functionality to help unpack national statistics. A linked survey of the performance of the water management arrangements at water points showed that for 85% of the sites water management arrangements were judged to be functional or highly functional. The Hidden Crisis project is a 4 year (2015-19) research project aimed at developing a robust evidence base and understanding of the complex and multi-faceted causes which underlie the current high failure rates of many new groundwater supplies in Africa, so that future WASH investments can be more sustainable. The project is being undertaken by an interdisciplinary team of established researchers in physical and social sciences from the UK, Ethiopia, Uganda, Malawi and Australia, led by the British Geological Survey

    Miscarriage and stillbirth following maternal Zika virus infection in nonhuman primates.

    Zika virus (ZIKV) infection is associated with congenital defects and pregnancy loss. Here, we found that 26% of nonhuman primates infected with Asian/American ZIKV in early gestation experienced fetal demise later in pregnancy despite showing few clinical signs of infection. Pregnancy loss due to asymptomatic ZIKV infection may therefore be a common but under-recognized adverse outcome related to maternal ZIKV infection

    Ethanol production of semi-simultaneous saccharification and fermentation from mixture of cotton gin waste and recycled paper sludge

    Ethanol production from the steam-exploded mixture of 75% cotton gin waste and 25% recycled paper sludge in various conditions was investigated by semi-simultaneous saccharification and fermentation (SSSF) consisting of a pre-hydrolysis and a simultaneous saccharification and fermentation (SSF). Four cases were studied: 24-h pre-hydrolysis + 48-h SSF (SSSF 24), 12-h pre-hydrolysis + 60-h SSF (SSSF 12), 72-h SSF, and 48-h hydrolysis + 24-h fermentation (SHF). The ethanol concentration, yield, and productivity of SSSF 24 were higher than those of the other operations. A model of SSF was used to simulate the data for four components in SSF. The analysis of the reaction rates of cellobiose, glucose, cell, and ethanol using the model and the parameters from the experiments showed that there was a transition point of the rate-controlling step at which the cell growth control in the initial 2 h was changed to the cellobiose reaction control in later period during ethanol production of SSF from the mixture

    Targeting adenosine monophosphate-activated protein kinase (AMPK) in preclinical models reveals a potential mechanism for the treatment of neuropathic pain

    Neuropathic pain is a debilitating clinical condition with few efficacious treatments, warranting development of novel therapeutics. We hypothesized that dysregulated translation regulation pathways may underlie neuropathic pain. Peripheral nerve injury induced reorganization of translation machinery in the peripheral nervous system of rats and mice, including enhanced mTOR and ERK activity, increased phosphorylation of mTOR and ERK downstream targets, augmented eIF4F complex formation and enhanced nascent protein synthesis. The AMP activated protein kinase (AMPK) activators, metformin and A769662, inhibited translation regulation signaling pathways, eIF4F complex formation, nascent protein synthesis in injured nerves and sodium channel-dependent excitability of sensory neurons resulting in a resolution of neuropathic allodynia. Therefore, injury-induced dysregulation of translation control underlies pathology leading to neuropathic pain and reveals AMPK as a novel therapeutic target for the potential treatment of neuropathic pain
