    Fluorescence Sensing Using DNA Aptamers in Cancer Research and Clinical Diagnostics

    Among the various advantages of aptamers over antibodies, remarkable is their ability to tolerate a large number of chemical modifications within their backbone or at the termini without losing significant activity. Indeed, aptamers can be easily equipped with a wide variety of reporter groups or coupled to different carriers, nanoparticles, or other biomolecules, thus producing valuable molecular recognition tools effective for diagnostic and therapeutic purposes. This review reports an updated overview on fluorescent DNA aptamers, designed to recognize significant cancer biomarkers both in soluble or membrane-bound form. In many examples, the aptamer secondary structure switches induced by target recognition are suitably translated in a detectable fluorescent signal using either fluorescently-labelled or label-free aptamers. The fluorescence emission changes, producing an enhancement ("signal-on") or a quenching ("signal-off") effect, directly reflect the extent of the binding, thereby allowing for quantitative determination of the target in bioanalytical assays. Furthermore, several aptamers conjugated to fluorescent probes proved to be effective for applications in tumour diagnosis and intraoperative surgery, producing tumour-type specific, non-invasive in vivo imaging tools for cancer pre- and post-treatment assessment

    Anticancer Ruthenium(III) Complexes and Ru(III)-Containing Nanoformulations: An Update on the Mechanism of Action and Biological Activity

    The great advances in the studies on metal complexes for the treatment of different cancer forms, starting from the pioneering works on platinum derivatives, have fostered an increasingly growing interest in their properties and biomedical applications. Among the various metal-containing drugs investigated thus far, ruthenium(III) complexes have emerged for their selective cytotoxic activity in vitro and promising anticancer properties in vivo, also leading to a few candidates in advanced clinical trials. Aiming at addressing the solubility, stability and cellular uptake issues of low molecular weight Ru(III)-based compounds, some research groups have proposed the development of suitable drug delivery systems (e.g., taking advantage of nanoparticles, liposomes, etc.) able to enhance their activity compared to the naked drugs. This review highlights the unique role of Ru(III) complexes in the current panorama of anticancer agents, with particular emphasis on Ru-containing nanoformulations based on the incorporation of the Ru(III) complexes into suitable nanocarriers in order to enhance their bioavailability and pharmacokinetic properties. Preclinical evaluation of these nanoaggregates is discussed with a special focus on the investigation of their mechanism of action at a molecular level, highlighting their pharmacological potential in tumour disease models and value for biomedical applications

    Vulnerability of demersal fish assemblages to trawling activities: a traits-based index

    Reducing the impact on vulnerable species through changes in fishing practices, such as the spatial or temporal avoidance of certain areas, is key to increase the ecological sustainability of fisheries. However, it is often hampered by the availability of sufficiently detailed data and robust indicators. Existing trawl surveys are a cost-effective data source to assess the vulnerability of fishing areas based on the quantities of vulnerable species caught. We developed a biological traits-based approach to the vulnerability of demersal assemblages using commercial trawl catch data. An expert-based approach identified a set of biological traits that are expected to condition the species' response to trawling impact and are combined to produce the vulnerability index ranked into four levels (low, moderate, high, and very high vulnerability). The approach was tested in four southern European fishing grounds showing evidence of over-exploitation, through catches being dominated by species of relatively low vulnerability to fishing impacts. The general distribution of species' biomass amongst vulnerability groups was highly homogenous across case studies, despite local differences in fishing fleet structure, target species and fishing depths. Within all areas the species with moderate vulnerability dominated and, in most instances, species of "very high" vulnerability were not recorded. Nevertheless, differences emerged when comparing the proportions of highly vulnerable species in the catches. Variability in vulnerability level of the catch was also observed at small spatial scales, which was principally explained by differences in habitat type and depth and, secondarily, by fishing effort. In fine mud in the shallower areas there was a higher presence of low vulnerable fauna. Furthermore, vulnerable organisms decreased in their presence in sandier substrates on the continental shelf.     Characterization of Maltese pottery of the Late Neolithic, Bronze Age and Punic Period by neutron activation analysis

    A set of 41 samples from Tas-Silg, Malta, has been analysed by neutron activation. It contained nine ware groups formed by visual examination covering the Late Neolithic, Bronze Age and Punic Periods (c. 3000–218 BC). Despite this diversity and long time range, seven of these ware groups, including the ‘Thermi Ware’, all have a similar chemical composition and, therefore, have been made from the same clay. This points most probably to a local origin. One group from the Punic Period, containing only Bricky Red cooking ware, is chemically separate and represents a second distinct pattern probably assignable to a local production. Five amphora sherds also from the Punic Period, and consisting of a micaceous fabric, all have different chemical characteristics and are probably imports from overseas production sites of unknown location.peer-reviewe

    Identification and Biological Characterization of the Pyrazolo[3,4-d]pyrimidine Derivative SI388 Active as Src Inhibitor

    Src is a non-receptor tyrosine kinase (TK) whose involvement in cancer, including glioblastoma (GBM), has been extensively demonstrated. In this context, we started from our in-house library of pyrazolo[3,4-d]pyrimidines that are active as Src and/or Bcr-Abl TK inhibitors and performed a lead optimization study to discover a new generation derivative that is suitable for Src kinase targeting. We synthesized a library of 19 compounds, 2a-s. Among these, compound 2a (SI388) was identified as the most potent Src inhibitor. Based on the cell-free results, we investigated the effect of SI388 in 2D and 3D GBM cellular models. Interestingly, SI388 significantly inhibits Src kinase, and therefore affects cell viability, tumorigenicity and enhances cancer cell sensitivity to ionizing radiation

    Essential oils: pharmaceutical applications and encapsulation strategies into lipid-based delivery systems

    Essential oils are being studied for more than 60 years, but a growing interest has emerged in the recent decades due to a desire for a rediscovery of natural remedies. Essential oils are known for millennia and, already in prehistoric times, they were used for medicinal and ritual purposes due to their therapeutic properties. Using a variety of methods refined over the centuries, essential oils are extracted from plant raw materials: the choice of the extraction method is decisive, since it determines the type, quantity, and stereochemical structure of the essential oil molecules. To these components belong all properties that make essential oils so interesting for pharmaceutical uses; the most investigated ones are antioxidant, anti-inflammatory, antimicrobial, wound-healing, and anxiolytic activities. However, the main limitations to their use are their hydrophobicity, instability, high volatility, and risk of toxicity. A successful strategy to overcome these limitations is the encapsulation within delivery systems, which enable the increase of essential oils bioavailability and improve their chemical stability, while reducing their volatility and toxicity.     Reducción de los descartes en la pesca con trasmallo: resultados experimentales utilizando trasmallo con “faldón” en la pesca artesanal del camarón, Penaeus kerathurus, en el mar Ligur (Mediterráneo Occidental)

    This study aimed to test the effectiveness of a “guarding net”, a device placed at the bottom of a trammel net, for reducing unwanted catches in the caramote prawn trammel net fishery of the Ligurian Sea. This specialized and profitable fishery is affected by unwanted catches that generate high discard rates and damage to the nets, with environmental impacts and costs for fishermen. The experimental study consisted in comparing the catches of a standard trammel net (STN) with those of two “experimental” trammel nets, e.g. STNs provided with a guarding net of 19 cm (TGN20) and 24 cm height (TGN25), respectively. The guarding net, a strip of gillnet placed at the bottom of the net, can be considered a by-catch reducer device (BRD). Some fishermen of the investigated fishery have been using this device for several years. The results of the 15 experimental fishing trials performed from June to July 2016 indicate that the guarding nets significantly reduce discards (e.g. crabs and other invertebrates); the biomass of the unwanted species caught was 75% lower than that produced by the STN. The catch rates of the target species obtained with TGN20 and TGN25 were also significantly lower than those of the STN, though of a lesser amount. Nonetheless, this economic loss can be compensated by the decrease in sorting time and material and labour costs that can be achieved using the guarding net.El objetivo de este trabajo fue testar los efectos de un “faldón”, una red colocada en la parte inferior de un trasmallo, para reducir los descartes en la pesquería del camarón del mar Ligur. Se trata de una pesquería especializada y rentable, afectada por capturas no deseadas, que generan descartes y daños a las redes, con impacto ambiental y costes para los pescadores. Se llevaron a cabo pescas experimentales, para comparar la captura de un trasmallo estándar (STN) con la de dos trasmallos “experimentales”, construidos a partir de un trasmallo estándar, con el ajuste de un faldón de 19 cm de altura (TGN20), y de un faldón de 24 cm (TGN25). Este faldón, una banda de red de enmalle, se puede considerar como un dispositivo reductor de capturas accesorias (BRD). Algunos pescadores de la pesquería investigada ya utilizan este dispositivo desde hace algunos años. Los resultados de las quince pruebas experimentales, realizadas de junio a julio 2016, muestran que el faldón de red de enmalle contribuye significativamente a reducir los descartes (cangrejos y otros invertebrados), con una reducción de la biomasa de las especies descartadas hasta el 75%, respecto al trasmallo estándar. Al mismo tiempo, también las tasas de captura de las especies objetivo obtenidas con TGN20 y TGN25 fueron significativamente más bajas que las del STN, aunque de menor magnitud. Sin embargo, esta pérdida económica puede ser compensada por la disminución del tiempo de trabajo, de los costes del material y de la mano de obra, que se pueden lograr utilizando un trasmallo con “faldón”
