    Functional analysis of cytosolic tryparedoxin peroxidase in antimony-resistant and –susceptible Leishmania braziliensis and Leishmania infantum lines

    BACKGROUND: Tryparedoxin peroxidase (TXNPx) participates in defence against oxidative stress as an antioxidant by metabolizing hydrogen peroxide into water molecules. Reports suggest that drug-resistant parasites may increase the levels of TXNPx and other enzymes, thereby protecting them against oxidative stress. METHODS: In this study, the gene encoding cytosolic TXNPx (cTXNPx) was characterized in lines of Leishmania (Viannia) braziliensis and Leishmania (Leishmania) infantum that are susceptible and resistant to potassium antimony tartrate (Sb(III)). We investigated the levels of mRNA and genomic organization of the cTXNPx gene. In addition, we transfected the Leishmania lines with the cTXNPx gene and analysed the susceptibility of transfected parasites to Sb(III) and to hydrogen peroxide (H2O2). RESULTS: Northern blot and real-time reverse transcriptase polymerase chain reaction analyses revealed that the level of TXNPx mRNA was approximately 2.5-fold higher in the Sb(III)-resistant L. braziliensis line than in the parental line. In contrast, no significant difference in cTXNPx mRNA levels between the L. infantum lines was observed. Southern blot analyses revealed that the cTXNPx gene is not amplified in the genome of the Sb(III)-resistant Leishmania lines analysed. Functional analysis of cTXNPx was performed to determine whether overexpression of the enzyme in L. braziliensis and L. infantum lines would change their susceptibility to Sb(III). Western blotting analysis showed that the level of cTXNPx was 2 to 4-fold higher in transfected clones compared to non-transfected cells. Antimony susceptibility test (EC50 assay) revealed that L. braziliensis lines overexpressing cTXNPx had a 2-fold increase in resistance to Sb(III) when compared to the untransfected parental line. In addition, these clones are more tolerant to exogenous H2O2 than the untransfected parental line. In contrast, no difference in Sb(III) susceptibility and a moderate index of resistance to H2O2 was observed in L. infantum clones overexpressing cTXNPx. CONCLUSION: Our functional analysis revealed that cTXNPx is involved in the antimony-resistance phenotype in L. braziliensis

    The scalar spin chirality is a three-body physical observable that plays an outstanding role both in classical magnetism, characterizing non-co-planar spin textures, and in quantum magnetism, as an order parameter for chiral spin liquids. In quantum information, the scalar spin chirality is a witness of genuine tripartite entanglement. Here we propose an indirect measurement scheme, based on the Hadamard test, to estimate the scalar spin chirality for general quantum states. We apply our method to study chirality in two types of quantum states: generic one-magnon states of a ferromagnet, and the ground state of a model with competing symmetric and antisymmetric exchange. We show a single-shot determination of the scalar chirality is possible for chirality eigenstates, via quantum phase estimation with a single auxiliary qutrit.     Analysis of blink dynamics in patients with blepharoptosis

    Owing to the rapid movements of the human upper eyelid, a high-speed camera was used to record and characterize voluntary blinking and the blink dynamics of blepharoptosis patients were compared to a control group. Twenty-six blepharoptosis patients prior to surgery and 45 control subjects were studied and the vertical height of the palpebral aperture (PA) was measured manually at 2 ms intervals during each blink cycle. The PA and blinking speed were plotted with respect to time and a predictive model was generated. The blink dynamic was analysed in closing and opening phases, and revealed a reduced speed of the initial opening phase in ptotic patients, suggesting intrinsic muscle function change in ptosis pathogenesis. The PA versus time curve for each subject was reconstructed using custom-built parameters; however, there were significant differences between the two groups. Those parameters used included the rate of closure, the delay between opening and closing, rate of initial opening, rate of slow opening (nonlinear function) and the 'switch point' between those two rates of opening. The model was tested against a new group of subjects and was able to discriminate ptosis patients from controls with 80% accuracy

    Exploring legal and policy aspects of urban sanitation and hygiene

    During 2012-2014, SNV did four country reviews of legal arrangements for urban sanitation and hygiene (Nepal, Bhutan, Bangladesh and Indonesia). Based on this experience, this document was developed to provide guidance on how to undertake a legal scan for urban sanitation. Over time, the ultimate objective of this work and related activities is that WASH professionals will be able to undertake a high-level assessment of legal arrangements for urban sanitation in order to both: 1. use the frameworks and tools offered by legal and policy arrangements to improve urban sanitation and hygiene outcomes; and 2. advocate for improvements in legal, policy and institutional arrangements to facilitate sustainable sanitation and hygiene outcomes for all

    TWO RECIPES FROM PORTUGUESE TRADITION OF GILDING ON WOODEN SUPPORT BETWEEN LABORATORY REPRODUCTION AND ANALYTICAL INVESTIGATION

    This paper has the main purpose to compile and highlight the first data obtained from experimental studies on docμmented reconstructions of gilded composites performed within a research project on gilding materials and techniques in Portugal (www.gilt-teller.pt) funded by FCT. Two water gilding recipes were appropriately chosen from the treatises written by the Portuguese Filipe Nunes (1615) and José Lopes Baptista de Almada (1749) as being representative for Baroque époque. Based on these recipes, the production of raw materials - “gesso grosso”, “gesso fino”, bole, animal glue (from lamb and goat skins) - was made as faithful as possible. Their application was then performed in laboratory following the indications given by these authors or by treatises of previous époques (e.g. the Bolognese treatise for thawing leather), on plane and curved wooden supports (pine and oak species) using three types of leaf: gold (Au/Ag/Cu) of 22 and 23.75 karat respectively and silver. After the completion of gilding, the samples’ surfaces were divided into areas and on each different finishing layers (wax, animal glue size) and decorations (punching, “esgrafitado”, “estofado”) were applied. An analytical campaign using optical microscopy (OM) and scanning electron microscopy (SEM) on surfaces and cross sections, X-ray diffraction (XRD), microcomputerized tomography (microCT) and colorimetry (CIEL*a*b*) was undertaken in order to characterize the gilded composites and to assess the faithfulness of the reproductions in the laboratory. Correlations between the information given by the recipes and the composition and stratigraphical patterns of the reconstructions can be established. Furthermore, the study aims to highlight the difficulties encountered in analyzing real samples and comparing the results with those from reproductions as the number and types of layered materials are not always reproducible. A critical approach is needed and criteria for faithful reproduction of ancient recipes are suggested

    Aging effect on the integrity of the traditional portuguese timber roof structures

    Traditional buildings basically include natural and autochthonous construction materials and are built with sustainable construction techniques. The knowledge of these materials’s state and construction techniques is required for a proper conservation/renovation work. It is well known that the lack of maintenance is one of the main causes that contribute to the high degree of degradation that the Portuguese traditional buildings unfortunately have. That degradation may even compromise the overall structural integrity of the building thus reducing its lifetime`s expectation. Meanwhile, a partial or even full demolition of a building is still an often occurrence in the Portuguese context. From a sustainable and building heritage perspectives that practise may be inadequate. Taking into account that generally the degradation problems are linked with roof`s leaking anomalies and that the structural components of the roof tend to be the first ones to be affected, this timber structural roof components of Portuguese dwellings were the object of the research work here presented

    Quantitative evaluation of visual function 12 months after bilateral implantation of a diffractive trifocal IOL

    PURPOSE: To quantitatively evaluate visual function 12 months after bilateral implantation of the Physiol FineVision® trifocal intraocular lens (IOL) and to compare these results with those obtained in the first postoperative month. METHODS: In this prospective case series, 20 eyes of 10 consecutive patients were included. Monocular and binocular, uncorrected and corrected visual acuities (distance, near, and intermediate) were measured. Metrovision® was used to test contrast sensitivity under static and dynamic conditions, both in photopic and low-mesopic settings. The same software was used for pupillometry and glare evaluation. Motion, achromatic, and chromatic contrast discrimination were tested using 2 innovative psychophysical tests. A complete ophthalmologic examination was performed preoperatively and at 1, 3, 6, and 12 months postoperatively. Psychophysical tests were performed 1 month after surgery and repeated 12 months postoperatively. RESULTS: Final distance uncorrected visual acuity (VA) was 0.00 ± 0.08 and distance corrected VA was 0.00 ± 0.05 logMAR. Distance corrected near VA was 0.00 ± 0.09 and distance corrected intermediate VA was 0.00 ± 0.06 logMAR. Glare testing, pupillometry, contrast sensitivity, motion, and chromatic and achromatic contrast discrimination did not differ significantly between the first and last visit (p>0.05) or when compared to an age-matched control group (p>0.05). CONCLUSIONS: The Physiol FineVision® trifocal IOL provided satisfactory full range of vision and quality of vision parameters 12 months after surgery. Visual acuity and psychophysical tests did not vary significantly between the first and last visit

    Videographic Analysis of Blink Dynamics following Upper Eyelid Blepharoplasty and Its Association with Dry Eye

    Background: This study was undertaken to characterize the effects of upper eyelid blepharoplasty on blink dynamics and to evaluate the hypothesis that changes in blink dynamics following blepharoplasty are associated with postoperative dry eye. // Methods: The voluntary blink of 14 eyes of 7 patients with dermatochalasis undergoing upper eyelid blepharoplasty was recorded with a high-speed camera preoperatively and 6–8 months postoperatively, alongside a group of 11 controls. The images were analyzed for palpebral aperture, blink duration, and maximum velocity during opening and closing phases. Patients undergoing blepharoplasty were assessed for dry eye symptoms pre- and postoperatively at 6–8 months using the ocular surface disease index score. // Results: Despite intraoperative orbicularis oculi resection, there was no significant compromise of blink duration or maximum velocity of eyelid opening or closure post-blepharoplasty. Postoperatively, patients had an increase in palpebral aperture compared with both preoperatively (8.71 versus 7.85mm; P = 0.013) and control groups (8.71 versus 7.87mm; P = 0.04). Postoperatively at 6–8 months, there was an increase in dry eye symptoms in 6 of 7 patients compared with preoperatively (ocular surface disease index, 16.6 versus 12.5; P < 0.05). There was no positive correlation between the increase in palpebral aperture and the increase in dry eye symptoms (r = –0.4; P = 0.30). // Conclusions: Using modern videographic technology, this study demonstrates that upper eyelid blepharoplasty results in an increase in resting palpebral aperture but has no effect on dynamic blink parameters. Changes in palpebral aperture or blink dynamics are unlikely to be the cause of dry eye syndrome following blepharoplasty

    Analysis of the Clinicopathological Characteristics of Patients with Upper Urinary Tract Transitional Cell Carcinoma

    OBJECTIVE: To describe the clinicopathological characteristics of patients with upper urinary tract transitional cell carcinomas who are treated surgically and to analyze the occurrence of bladder tumors as well as the development of metastases outside the urinary tract. MATERIALS AND METHODS: The study comprised a retrospective analysis of 25 patients treated between February 1994 and August 2006. The variables analyzed were: patient age, gender, and clinical presentation; diagnostic methods; pathologic characteristics at the primary site of the tumor (pelvis or ureter); tumor stage and grade; and presence of carcinoma in situ, microvascular invasion and squamous differentiation. The Kaplan-Meier method and the Log-Rank test were used for statistical analysis of bladder recurrence-free survival. RESULTS: Eighty-four percent of patients were male, and macroscopic hematuria was the most common clinical presentation. The majority of cases (56%) were infiltrative (T2-T3) and high-grade (76%) tumors. Synchronous or metachronous bladder tumors were found in 72% of cases. Five (20%) patients had a history of bladder tumor before the diagnosis of upper urinary tract transitional cell carcinomas. The mean follow-up period was 36 months (range: 1.5 to 156). During the follow-up period, eleven (44%) patients developed bladder tumors. After five years, the probability of being free of bladder tumor recurrence was 40%. No pathological variable was predictive for bladder tumor recurrence. Four patients presented disease recurrence outside the urinary tract. CONCLUSIONS: The presence of metachronous bladder tumors is more often observed after the diagnosis of upper urinary tract transitional cell carcinomas. All of these patients should undergo rigorous follow-up during the postoperative period. Only patients with infiltrative and high-grade tumors developed metastases outside the urinary tract
