    Mesenchymal chondroprogenitor cell origin and therapeutic potential

    Mesenchymal progenitor cells, a multipotent adult stem cell population, have the ability to differentiate into cells of connective tissue lineages, including fat, cartilage, bone and muscle, and therefore generate a great deal of interest for their potential use in regenerative medicine. During development, endochondral bone is formed from a template of cartilage that transforms into bone; however, mature articular cartilage remains in the articulating joints, where its principal role is reducing friction and dispersing mechanical load. Articular cartilage is prone to damage from sports injuries or ageing, which regularly progresses to more serious joint disorders, such as osteoarthritis. Osteoarthritis is a degenerative joint disease characterized by the thinning and eventual wearing of articular cartilage, and affects millions of people worldwide. Due to low chondrocyte motility and proliferative rates, and complicated by the absence of blood vessels, cartilage has a limited ability to self-repair. Current pharmaceutical and surgical interventions fail to generate repair tissue with the mechanical and cellular properties of native host cartilage. The long-term success of cartilage repair will therefore depend on regenerative methodologies resulting in the restoration of articular cartilage that closely duplicates the native tissue. For cell-based therapies, the optimal cell source must be readily accessible with easily isolated, abundant cells capable of collagen type II and sulfated proteoglycan production in appropriate proportions. Although a cell source with these therapeutic properties remains elusive, mesenchymal chondroprogenitors retain their expansion capacity with the promise of reproducing the structural or biomechanical properties of healthy articular cartilage. As current knowledge regarding chondroprogenitors is relatively limited, this review will focus on their origin and therapeutic application

    The safety climate in primary care (SAP-C) study: study protocol for a randomised controlled feasibility study

    BACKGROUND: Research on patient safety has focused largely on secondary care settings, and there is a dearth of knowledge relating to safety culture or climate, and safety climate improvement strategies, in the context of primary care. This is problematic given the high rates of usage of primary care services and the myriad of opportunities for clinical errors daily. The current research programme aimed to assess the effectiveness of an intervention derived from the Scottish Patient Safety Programme in Primary Care. The intervention consists of safety climate measurement and feedback and patient chart audit using the trigger review method. The purpose of this paper is to describe the background to this research and to present the methodology of this feasibility study in preparation for a future definitive RCT. METHODS: The SAP-C study is a feasibility study employing a randomised controlled pretest-posttest design that will be conducted in 10 general practices in the Republic of Ireland and Northern Ireland. Five practices will receive the safety climate intervention over a 9-month period. The five practices in the control group will continue care as usual but will complete the GP-SafeQuest safety climate questionnaire at baseline (month 1) and at the terminus of the intervention (month 9). The outcomes of the study include process evaluation metrics (i.e. rates of participant recruitment and retention, rates of completion of safety climate measures, qualitative data regarding participants’ perceptions of the intervention’s potential efficacy, acceptability, and sustainability), patient safety culture in intervention and control group practices at posttest, and instances of undetected patient harm identified through patient chart audit using the trigger review method. DISCUSSION: The planned study investigates an intervention to improve safety climate in Irish primary care settings. The resulting data may inform our knowledge of the frequency of undetected patient safety incidents in primary care, may contribute to improved patient safety practices in primary care settings, and may inform future research on patient safety improvement initiatives.     Growth and break-up of methanogenic granules suggests mechanisms for biofilm and community development

    Methanogenic sludge granules are densely packed, small, spherical biofilms found in anaerobic digesters used to treat industrial wastewaters, where they underpin efficient organic waste conversion and biogas production. Each granule theoretically houses representative microorganisms from all of the trophic groups implicated in the successive and interdependent reactions of the anaerobic digestion (AD) process. Information on exactly how methanogenic granules develop, and their eventual fate will be important for precision management of environmental biotechnologies. Granules from a full-scale bioreactor were size-separated into small (0.6–1 mm), medium (1– 1.4 mm), and large (1.4–1.8 mm) size fractions. Twelve laboratory-scale bioreactors were operated using either small, medium, or large granules, or unfractionated sludge. After >50 days of operation, the granule size distribution in each of the small, medium, and large bioreactor sets had diversified beyond—to both bigger and smaller than—the size fraction used for inoculation. Interestingly, extra-small (XS; <0.6 mm) granules were observed, and retained in all of the bioreactors, suggesting the continuous nature of granulation, and/or the breakage of larger granules into XS bits. Moreover, evidence suggested that even granules with small diameters could break. “New” granules from each emerging size were analyzed by studying community structure based on high-throughput 16S rRNA gene sequencing. Methanobacterium, Aminobacterium, Propionibacteriaceae, and Desulfovibrio represented the majority of the community in new granules. H2-using, and not acetoclastic, methanogens appeared more important, and were associated with abundant syntrophic bacteria. Multivariate integration (MINT) analyses identified distinct discriminant taxa responsible for shaping the microbial communities in different-sized granules

    Spatio-Temporal Variability of Harbor Porpoise Life History Parameters in the North-East Atlantic

    Harbor porpoises exhibit early maturation, relatively short gestation/lactation periods and a faster rate of reproduction as compared to other cetacean species. Intrinsic and extrinsic factors can influence both population vital rates and population structure, which ultimately cause changes in dynamics within and between populations. Here, we undertook a retrospective analysis of mortality data collected over a 24-year period for assessing life history traits of the North-east Atlantic harbor porpoise population. We use time-period specific models for key life history relationships that considered cause of death of individuals (as a proxy for health status), sex and management unit (MU). Sexual variation in asymptotic length, asymptotic age, average length at 50% maturity (L50) and average age at 50% maturity (A50) were observed, with females attaining a larger asymptotic length, larger L50, and delaying attainment of both sexual and physical maturity, compared to males. While females are constrained in their minimum body size due to giving birth to proportionally larger offspring, males exhibited more plasticity in size at sexual maturity, enabling re-allocation of available energy resources toward reproduction. Data were then used to compare biological parameters among two porpoise MUs in United Kingdom waters, both of which in the current study exhibited reduced reproductive rates compared to other geographic regions. In both MUs, females significantly increased their A50 and males significantly declined in their L50. An increase in the age at asymptotic length was also observed in both sexes, along with a significant decline in the Gompertz growth rate parameter that was more apparent in the female data. While availability of suitable prey resources may be a limiting factor, a combination of other factors cannot be ruled out. Porpoises in the Celtic and Irish Seas MU were significantly larger in their maximum length, asymptotic length and L50 compared to porpoises in the North Sea MU throughout the study period, suggesting limited gene flow between these two MUs. These results justify the maintenance of these harbor porpoise MUs or assessment units, as two separate units, within the range of the North-east Atlantic population, and for indicator assessments under the EU’s Marine Strategy Framework Directive

    ‘We practise every day’: Parents’ attitudes towards early science learning and education among a sample of urban families in Ireland

    Educational policies increasingly emphasise early childhood science engagement. As key influencers in children’s early learning, parents (n=85) attending a science workshop in three urban schools in Ireland were surveyed regarding their attitudes towards science. Seventy per cent of parents believed that science education should begin in the pre-school years, before the age of four. Despite high levels of education, at least half of the parents expressed some lack of confidence in talking about, and in doing science with, their young children. Parents who reported less confidence in doing science activities with their children also reported reduced frequency of activities for five out of the seven science learning opportunities listed. Mothers, compared to fathers, reported less confidence in doing science activities with their children. Findings indicate that parents’ confidence in science may impact early science experiences and highlight parents as a key support for increasing early science engagement

    De novo growth of methanogenic granules indicates a biofilm life-cycle with complex ecology

    Methanogenic sludge granules are densely packed, small (diameter, approx. 0.5-2.0 mm) spherical biofilms found in anaerobic digesters used to treat industrial wastewaters, where they underpin efficient organic waste conversion and biogas production. A single digester contains millions of individual granules, each of which is a highly-organised biofilm comprised of a complex consortium of likely billions of cells from across thousands of species – but not all granules are identical. Whilst each granule theoretically houses representative microorganisms from all of the trophic groups implicated in the successive and interdependent reactions of the anaerobic digestion process, parallel granules function side-by-side in digesters to provide a ‘meta-organism’ of sorts. Granules from a full-scale bioreactor were size-separated into small, medium and large granules. Laboratory-scale bioreactors were operated using only small (0.6–1 mm), medium (1–1.4 mm) or large (1.4–1.8 mm) granules, or unfractionated (naturally distributed) sludge. After >50 days of operation, the granule size distribution in each of the small, medium and large bioreactor types had diversified beyond – to both bigger and smaller than – the size fraction used for inoculation. ‘New’ granules were analysed by studying community structure based on high-throughput 16S rRNA gene sequencing. Methanobacterium, Aminobacterium, Propionibacteriaceae and Desulfovibrio represented the majority of the community in new granules. Importance: Methanogenic granules are spherical biofilms found in the built environment, where despite their importance for anaerobic digestion of wastewater in bioreactors, little is understood about the fate of granules across their entire life. Information on exactly how, and at what rates, methanogenic granules develop will be important for more precise and innovative management of environmental biotechnologies. Microbial aggregates also spark interest as subjects in which to study fundamental concepts from microbial ecology, including immigration and species sorting affecting the assembly of microbial communities. This experiment is the first, of which we are aware, to compartmentalise methanogenic granules into discrete, size-resolved fractions, which were then used to separately start up bioreactors to investigate the granule life cycle. The evidence, and extent, of de novo granule growth, and the identification of key microorganisms shaping new granules at different life-cycle stages, is important for environmental engineering and microbial ecology

    Supply of Vertebrate Necropsy and Sample Recovery Services Merged Final Reports

    The Marine Institute issued a tender for the Supply of Vertebrate Necropsy and Sample Recovery Services Tender (2017-2018). The results are presented in this report. These tenders required i) the recovery and standardised necropsy of three cetacean species with associated case history reporting, ii) the provision of sampling kits to be used for the recovery of tissue samples collected from bycaught animals (birds, seals and cetaceans) by observers on commercial inshore and offshore fishing vessels in Irish waters in order to provide additional data to the MI’s existing catch sampling programme and iii) the storage and subsequent delivery of all samples and associated databases to the client. Recovery of 24 animals from two geographical lots was the target for the initial contract (2017) and was achieved. The contract extension (early 2018) required the collection and necropsy of a further 16 animals, which was exceeded as 19 animals were collected. A target of 30 animals to be recovered was required in the second contract (2018) and was also successfully reached. These 73 animals comprised 53 common dolphins (Delphinus delphis), ten striped dolphins (Stenella coeruleoalba) and ten harbour porpoises (Phocoena phocoena). Forty-one of the 73 (56%) individuals were recovered from Lot 1 from counties Clare to Donegal, while 32 (44%) individuals were recovered from Lot 2 from counties Wexford to Kerry. Thirty-five sampling kits were assembled and delivered to the Marine Institute, and additional kits were prepared and available upon request for distribution to their panel of fishery observers. Between 24 July and 14 December 2017, two adult male grey seals were incidentally captured in tangle nets and reported to the Project Coordinator. Results from these tenders will inform the development of a long-term strategy for necropsy of stranded cetaceans and recovery of tissue samples for further biological studies. Samples requiring long-term storage at -20°C, including those for virology, were transported in a portable refrigerator to the IWDG office in Kilrush, Co. Clare and stored at facilities on site. Formalin fixed tissues, such as reproductive organs and adrenal glands, were stored at the RVL in Cork. All samples have been delivered to the Marine Institute in Oranmore, Co. Galway along with a detailed database of samples collected.EMF

    A smartphone intervention for adolescent obesity: study protocol for a randomised controlled non-inferiority trial

    Background There are few evidence-based mobile health solutions for treating adolescent obesity. The primary aim of this parallel non-inferiority trial is to assess the effectiveness of an experimental smartphone application in reducing obesity at 12 months, compared to the Temple Street W82GO Healthy Lifestyles intervention. Methods/design The primary outcome measure is change in body mass index standardised deviation score at 12 months. The secondary aim is to compare the effect of treatment on secondary outcomes, including waist circumference, insulin sensitivity, quality of life, physical activity and psychosocial health. Adolescents with a body mass index at or above the 98th percentile (12 to 17 years) will be recruited from the Obesity clinic at Temple Street Children’s University Hospital in Dublin, Ireland. W82GO is a family-based lifestyle change intervention delivered in two phases over 12 months. In the current study, participants will be randomised for phase two of treatment to either usual care or care delivered via smartphone application. One hundred and thirty-four participants will be randomised between the two study arms. An intention-to-treat analysis will be used to compare treatment differences between the groups at 12 months. Discussion The results of this study will be disseminated via open access publication and will provide important information for clinicians, patients and policy makers regarding the use of mobile health interventions in the management of adolescent obesity.     Hadrons at high temperature: an update from the FASTSUM collaboration

    We present the most recent results from the FASTSUM collaboration for hadron properties at high temperature. This includes the temperature dependence of the light and charmed meson and baryon spectrum, as well as properties of heavy quarkonia. The results are obtained using anisotropic lattices with a fixed scale approach. We also present the status of our next generation gauge ensembles.Comment: 8 pages, 8 figures.
